    Diet Supplementation with Polyphenol-Rich Salicornia ramosissima Extracts Protects against Tissue Damage in Experimental Models of Cerebral Ischemia

Regular consumption of polyphenols has been shown to reduce the risk of suffering a cardiovascular event. For this reason, we have investigated the protective effect of Salicornia ramosissima, a seasonal halophyte that synthetizes high amounts of bioactive compounds, including polyphenols, in response to environmental stress. Aqueous, hydroalcoholic, and ethanolic extracts were prepared to investigate if dietary supplementation prior to ischemic challenge can prevent subsequent damage using two animal models. First, we screened the protective effect against hypoxia–reoxygenation in Drosophila melanogaster and observed that both ethanolic and hydroalcoholic extracts protected flies from the deleterious effects of hypoxia. Second, we confirmed the protective effect of S. ramosissima ethanolic extract against brain ischemia using the transient middle cerebral artery occlusion mice model. Four weeks of oral supplementation with the ethanolic extract before artery occlusion reduced infarct volume and lowered the plasma levels of the DNA peroxidant product 8-hydroxydeoxyguanosine. Phytochemical profiling of S. ramosissima ethanolic extract revealed 50 compounds. Thus, it represents a valuable source of bioactive compounds that show promising disease-modifying activities and could be further developed as an effective food supplement for the prevention or treatment of neurovascular disorders.The authors received financial support from “CSF-Proyectos estratégicos de I+D+i.     Fine-tuning a brain spheroid model as a screening platform for neuroprotective therapies against ischemic stroke.

    Motivation: Stroke is the second cause of death and a leading cause of disability worldwide. Ischemic stroke represents more than 80% of all events and it is due to the presence of a clot blocking the blood flow. European legislation has already assumed the goal of no longer needing to use animals for scientific research in the future. To improve current in vitro models, a two-stage 3D brain spheroid model was previously described to recreate the blood-brain barrier and serve as a tool to assess ischemic damage and neuroinflammation. Our objective is to fine tune this organoid model, so we can use it as a screening platform of different neuroprotective therapies against ischemic stroke. Methods: Six different human brain cells were cultured following manufacturer´s recommendations. In order to be able to visualize each cell type in the 3D model, immunocytochemistry protocols were stablished. Cells were fixed and immunolabeled using a suitable primary antibody for each type of cell followed by the appropriate Alexa 568 secondary antibody. Negative controls were also prepared following the same protocol without the addition of the primary antibody. As a fist approach for spheroid formation, astrocytes, microglia, oligodendrocytes and cortical neurons were co-cultured in round bottom 96 well plates building the spheroid core. After 48h microvascular cells and pericytes were added to the culture creating the surface area. Finally, spheroids images were taken employing bright field microscopy and diameter was measured. Results: Fluorescent images confirmed that the immunolabeling protocols for each cell type were set up appropriately. Red fluorescence staining indicated cell´s detection by their specific markers. Fluorescent signal was absent in negative controls, confirming the specific binding. Finally, two-stage spheroids (4.000 cells) were formed. After 48, one round-shaped spheroid per well was observed. Spheroid diameter was 313.9± 22 μm after 48 h and 690.9±64 μm after 4 days. Conclusions: We have fine-tuned the immunocytochemical technique to identify each cell line that will be used to analyse brain spheroids. The first trial for spheroid formation showed homogenous shape and size. Next step would be testing smaller spheroids (2.000 cells) and verifying that each cell type is correctly located in the 3D structure. If results are satisfactory these spheroids could be used as a tool to test potential neuroprotective therapies

El propósito de este trabajo de profundización es, ejecutar un análisis en la empresa Textiles Industriales S.A.S., la cual fue fundada en 1978 y, es especializada en la fabricación de medios filtrantes sintéticos de uso industrial pues en ella se observó una gran oportunidad de mejora, por lo cual fue seleccionada con el fin de generar propuestas de hacer avances en sus sistematizaciones, asegurándose de que sean coherentes a sus insuficiencias. Dentro de este proyecto se observaron los procesos que llevan a cabo, consiguiendo establecer los representantes principales a nivel global y de forma detallada. También, se efectuó la revisión del contexto en el que se encuentra Colombia en términos logísticos. De igual forma, se efectuó el diseño de los procesos que se realizan en la empresa, según el modelo de Referencia de las Operaciones en la Cadena de Suministro, para lograr construir la configuración de la red de suministro y estructurar los flujos. Así mismo, por medio de un análisis logístico del Índice de Desempeño Logístico del Banco Mundial, se estableció la posición de Colombia a nivel global en procesos logísticos, los cuales fueron contextualizados según el documento CONPES 3982 Política Nacional de Logística. Para terminar, se hizo un análisis de la empresa en su gestión de inventarios, diseño de almacenes y, procesos logísticos como lo fueron el de aprovisionamiento y distribución; identificando los mejores métodos aplicables para la empresa en particular. Se concluye que, mediante el desarrollo de la investigación y análisis, se pueden revelar oportunidades de perfeccionamiento en logística y la Supply Chain Management en la empresa Textiles Industriales S.A.S., a través del fortalecimiento de los métodos y la aplicación de materiales y técnicas que permiten pulir los procesos ejecutados, manifestando no solo disminución de costos sino una lugar competitivo para la empresa, adquiriendo de esta forma, destacar la empresa Textiles Industriales S.A.S. en la industria en la cual se desenvuelve en la actualidad.Supply Chain Management into a company is gaining more and more strength, always seeking to compete in the global market and to remain at the highest levels, which generate more profits for the company. The objective of this work is to carry out an analysis in Textiles Industriales S.A.S. Company, which was founded in 1978 and it is specialized in the manufacture of synthetic filters for industrial use, since a great opportunity for improvement was observed in it. That is why; it was selected in order to generate proposals to make progress in its systematizations, ensuring that they are consistent with its insufficiencies. Within this project, the processes they carry out were observed, managing to establish the main representatives at a global level and in detail. In addition, a review of the context in which Colombia finds itself in logistical terms was done. In the same way, the design of the processes that are done in the company was carried out, according to the Supply Chain Operations Reference model, in order to build the configuration of the supply network and structure the flows. Likewise, through a logistics analysis of the Logistics Performance Index of the World Bank, the position of Colombia at a global level in logistics processes was established, which were contextualized according to the document CONPES 3982 National Logistics Policy. Finally, an analysis of the company was made in its inventory management, warehouse design and logistics processes such as supply and distribution; identifying the best applicable methods for the particular company. To sum up, through the development of research and analysis, opportunities for improvement in logistics and Supply Chain Management can be revealed in Textiles Industriales S.A.S. Company, over the strengthening of methods and the application of materials and techniques that allow polishing. The executed processes, showing not only a reduction in costs but also a competitive place for the company, thus acquiring, highlighting Textiles Industriales S.A.S. Company in the industry in which he currently operates

    En el presente documento, se abordan diferentes temáticas teniendo en cuenta la importancia del enfoque narrativo y el camino reflexivo que este ha logrado ampliar desde los diferentes contextos significantes en los procesos terapéuticos o con carácter psicosocial en los escenariosde violencia que se presentan en los sistemas. Tomando como referencia las diferentes técnicas de acción, se puede hablar de lafoto voz o (Photovoice), la cual permite ahondar en una situación problémica desde la exposiciónde diferentes imágenes que pueden generar significados a través del poder metafórico y generarcambios notorios en comunidades completas. Es imprescindible para los profesionales en psicología, el aprender a intervenir enlas diversas situaciones de violencia y los relatos de las victimas con los diferentes métodos de creación de preguntas y estrategias que aporten a la vida de las personas de manera significativa;Es por esto que se trabajara en base a casos de importancia encuadrados en problemáticas de violencia y desplazamientos forzados a causa del conflicto armado y la fuerza pública y grupos al margen de la ley como el Caso Peñas Coloradas, con los cuales se pretende adquirir diferentes fortalezas en la apropiación de conocimientos nuevos.     Effectiveness of an intervention for improving drug prescription in primary care patients with multimorbidity and polypharmacy:Study protocol of a cluster randomized clinical trial (Multi-PAP project)

    This study was funded by the Fondo de Investigaciones Sanitarias ISCIII (Grant Numbers PI15/00276, PI15/00572, PI15/00996), REDISSEC (Project Numbers RD12/0001/0012, RD16/0001/0005), and the European Regional Development Fund ("A way to build Europe").Background: Multimorbidity is associated with negative effects both on people's health and on healthcare systems. A key problem linked to multimorbidity is polypharmacy, which in turn is associated with increased risk of partly preventable adverse effects, including mortality. The Ariadne principles describe a model of care based on a thorough assessment of diseases, treatments (and potential interactions), clinical status, context and preferences of patients with multimorbidity, with the aim of prioritizing and sharing realistic treatment goals that guide an individualized management. The aim of this study is to evaluate the effectiveness of a complex intervention that implements the Ariadne principles in a population of young-old patients with multimorbidity and polypharmacy. The intervention seeks to improve the appropriateness of prescribing in primary care (PC), as measured by the medication appropriateness index (MAI) score at 6 and 12months, as compared with usual care. Methods/Design: Design:pragmatic cluster randomized clinical trial. Unit of randomization: family physician (FP). Unit of analysis: patient. Scope: PC health centres in three autonomous communities: Aragon, Madrid, and Andalusia (Spain). Population: patients aged 65-74years with multimorbidity (≥3 chronic diseases) and polypharmacy (≥5 drugs prescribed in ≥3months). Sample size: n=400 (200 per study arm). Intervention: complex intervention based on the implementation of the Ariadne principles with two components: (1) FP training and (2) FP-patient interview. Outcomes: MAI score, health services use, quality of life (Euroqol 5D-5L), pharmacotherapy and adherence to treatment (Morisky-Green, Haynes-Sackett), and clinical and socio-demographic variables. Statistical analysis: primary outcome is the difference in MAI score between T0 and T1 and corresponding 95% confidence interval. Adjustment for confounding factors will be performed by multilevel analysis. All analyses will be carried out in accordance with the intention-to-treat principle. Discussion: It is essential to provide evidence concerning interventions on PC patients with polypharmacy and multimorbidity, conducted in the context of routine clinical practice, and involving young-old patients with significant potential for preventing negative health outcomes.     CIBERER : Spanish national network for research on rare diseases: A highly productive collaborative initiative

This innovative research structure comprises 11 different specific areas dedicated to the main public health priorities in the National Health System. CIBERER, the thematic area of CIBER focused on rare diseases (RDs) currently consists of 75 research groups belonging to universities, research centers, and hospitals of the entire country. CIBERER's mission is to be a center prioritizing and favoring collaboration and cooperation between biomedical and clinical research groups, with special emphasis on the aspects of genetic, molecular, biochemical, and cellular research of RDs. This research is the basis for providing new tools for the diagnosis and therapy of low-prevalence diseases, in line with the International Rare Diseases Research Consortium (IRDiRC) objectives, thus favoring translational research between the scientific environment of the laboratory and the clinical setting of health centers. In this article, we intend to review CIBERER's 15-year journey and summarize the main results obtained in terms of internationalization, scientific production, contributions toward the discovery of new therapies and novel genes associated to diseases, cooperation with patients' associations and many other topics related to RD research

    Higher COVID-19 pneumonia risk associated with anti-IFN-α than with anti-IFN-ω auto-Abs in children

    We found that 19 (10.4%) of 183 unvaccinated children hospitalized for COVID-19 pneumonia had autoantibodies (auto-Abs) neutralizing type I IFNs (IFN-alpha 2 in 10 patients: IFN-alpha 2 only in three, IFN-alpha 2 plus IFN-omega in five, and IFN-alpha 2, IFN-omega plus IFN-beta in two; IFN-omega only in nine patients). Seven children (3.8%) had Abs neutralizing at least 10 ng/ml of one IFN, whereas the other 12 (6.6%) had Abs neutralizing only 100 pg/ml. The auto-Abs neutralized both unglycosylated and glycosylated IFNs. We also detected auto-Abs neutralizing 100 pg/ml IFN-alpha 2 in 4 of 2,267 uninfected children (0.2%) and auto-Abs neutralizing IFN-omega in 45 children (2%). The odds ratios (ORs) for life-threatening COVID-19 pneumonia were, therefore, higher for auto-Abs neutralizing IFN-alpha 2 only (OR [95% CI] = 67.6 [5.7-9,196.6]) than for auto-Abs neutralizing IFN-. only (OR [95% CI] = 2.6 [1.2-5.3]). ORs were also higher for auto-Abs neutralizing high concentrations (OR [95% CI] = 12.9 [4.6-35.9]) than for those neutralizing low concentrations (OR [95% CI] = 5.5 [3.1-9.6]) of IFN-omega and/or IFN-alpha 2

    Clonal chromosomal mosaicism and loss of chromosome Y in elderly men increase vulnerability for SARS-CoV-2

    The pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2, COVID-19) had an estimated overall case fatality ratio of 1.38% (pre-vaccination), being 53% higher in males and increasing exponentially with age. Among 9578 individuals diagnosed with COVID-19 in the SCOURGE study, we found 133 cases (1.42%) with detectable clonal mosaicism for chromosome alterations (mCA) and 226 males (5.08%) with acquired loss of chromosome Y (LOY). Individuals with clonal mosaic events (mCA and/or LOY) showed a 54% increase in the risk of COVID-19 lethality. LOY is associated with transcriptomic biomarkers of immune dysfunction, pro-coagulation activity and cardiovascular risk. Interferon-induced genes involved in the initial immune response to SARS-CoV-2 are also down-regulated in LOY. Thus, mCA and LOY underlie at least part of the sex-biased severity and mortality of COVID-19 in aging patients. Given its potential therapeutic and prognostic relevance, evaluation of clonal mosaicism should be implemented as biomarker of COVID-19 severity in elderly people.
