    Coating process efficiency for polymer coated broccoli seeds in spouted bed

    Broccoli seeds were coated in a conical-cylindrical spouted bed with an aqueous suspension of hydroxy ethyl cellulose aiming to improve the seeds coating technique using a fluid-dynamic process. An experimental design was applied to investigate the effects of the operating variables: gas temperature, atomizing air pressure and suspension flow rate on the germination of the seeds and on the process efficiency. Results indicated that the operating variables affect both the coating process efficiency and the germination ability. However, the analysis didn t identify differences between the germination potential of coated and uncoated seeds. Coated seeds absorbed up to 10 percent less moisture than the uncoated ones, when the environment temperature and humidity were controlled over a period of time.Neste trabalho, sementes de brócolos foram recobertas em leito de jorro cone-cilíndrico, com suspensão aquosa de hidroxietilcelulose, visando a aprimorar a técnica do recobrimento de sementes, utilizando o processo fluidodinâmico. Foram investigados os efeitos das variáveis operacionais temperatura do ar de jorro, pressão de ar de atomização e vazão de suspensão de recobrimento, na eficiência do processo e na germinação das sementes. Os resultados revelaram que houve influência das variáveis operacionais na eficiência do processo e na germinação. Verificou-se, também, que as sementes recobertas apresentaram de 2 a 10% de umidade a menos que as sementes não-recobertas, quando em ambiente com temperatura controlada e saturado de vapor de água em determinado período.30531

    Characterization of Soybean Cultivars for Biodiesel Production

    Due to environmental issues involving the polluting gasesemission, Brazil has adopted the policy of using oil and biodiesel. For biodiesel production, the main raw material used in Brazil is soybean oil. The development of the numerous genotypes of this culture has always considered quantitative aspects. The objective was to qualitatively characterize 12 soybean cultivars for biodiesel production. The experimental design was randomized blocks with three replicates. The cultivars were sown in December 2016, in no-tillage system, in Ponta Grossa, Paraná, Brazil (-25.093056, -50.063327 UTM). The analyzed variables were: oil and protein contents, acidity index and specific mass. It was concluded that there were no significant differences among the cultivars for oil and protein contents. For the variables acidity index and specific mass, there were significant differences among the cultivars, being below the limits established by the Brazilian legislation for vegetable oil, but with potential for biodiesel production

    RANK links senescence to stemness in the mammary epithelia, delaying tumor onset but increasing tumor aggressiveness

    Rank signaling enhances stemness in mouse and human mammary epithelial cells (MECs) and mediates mammary tumor initiation. Mammary tumors initiated by oncogenes or carcinogen exposure display high levels of Rank and Rank pathway inhibitors have emerged as a new strategy for breast cancer prevention and treatment. Here, we show that ectopic Rank expression in the mammary epithelia unexpectedly delays tumor onset and reduces tumor incidence in the oncogene-driven Neu and PyMT models. Mechanistically, we have found that ectopic expression of Rank or exposure to Rankl induces senescence, even in the absence of other oncogenic mutations. Rank leads to DNA damage and senescence through p16/p19. Moreover, RANK-induced senescence is essential for Rank-driven stemness, and although initially translates into delayed tumor growth, eventually promotes tumor progression and metastasis. We uncover a dual role for Rank in the mammary epithelia: Rank induces senescence and stemness, delaying tumor initiation but increasing tumor aggressiveness

    Lower production of IL-17A and increased susceptibility to Mycobacterium bovis in mice coinfected with Strongyloides venezuelensis

    The presence of intestinal helminths can down-regulate the immune response required to control mycobacterial infection. BALB/c mice infected with Mycobacterium bovis following an infection with the intestinal helminth Strongyloides venezuelensis showed reduced interleukin-17A production by lung cells and increased bacterial burden. Also, small granulomas and a high accumulation of cells expressing the inhibitory molecule CTLA-4 were observed in the lung. These data suggest that intestinal helminth infection could have a detrimental effect on the control of tuberculosis (TB) and render coinfected individuals more susceptible to the development of TB

    Design of Novel Relaxase Substrates Based on Rolling Circle Replicases for Bioconjugation to DNA Nanostructures

    During bacterial conjugation and rolling circle replication, HUH endonucleases, respectively known as relaxases and replicases, form a covalent bond with ssDNA when they cleave their target sequence (nic site). Both protein families show structural similarity but limited amino acid identity. Moreover, the organization of the inverted repeat (IR) and the loop that shape the nic site differs in both proteins. Arguably, replicases cleave their target site more efficiently, while relaxases exert more biochemical control over the process. Here we show that engineering a relaxase target by mimicking the replicase target, results in enhanced formation of protein-DNA covalent complexes. Three widely different relaxases, which belong to MOBF, MOBQ and MOBP families, can properly cleave DNA sequences with permuted target sequences. Collaterally, the secondary structure that the permuted targets acquired within a supercoiled plasmid DNA resulted in poor conjugation frequencies underlying the importance of relaxase accessory proteins in conjugative DNA processing. Our results reveal that relaxase and replicase targets can be interchangeable in vitro.     Flowability of inert particle beds with fruit pulp: effects on the drying in spouted bed

    In this work six types of inert particles were characterized and analyzed for drying tropical fruit pulps. The repose angle was determined with and without the addition of water and pulp of various tropical fruits. The bed flowability was related to the particle properties and chemical composition of pulps. The influence of the repose angle on the drying performance was analyzed. It was also verified that the composition of pulps influenced the bed flowability. The global analysis showed that the pulps with high lipids and insoluble solids content and low reducing sugar content improved the bed flowability. The results obtained with the drying of the fruit pulps using low-density polystyrene granules as inert particles showed that high flowabilities lead to lower minimum spout flow rates, but do not necessarily lead to the highest powder production.Neste trabalho, foram caracterizados seis tipos de material inerte, utilizados na secagem de polpa de frutas em leito de jorro. Determinou-se o ângulo de repouso das partículas, com e sem adição de água e de polpa de diversas frutas tropicais. Correlacionou-se a escoabilidade com as propriedades das partículas e com a composição química das polpas. Analisou-se a influência do ângulo de repouso sobre o desempenho do secador, no que se refere à produção. Os resultados mostraram que, em geral, as polpas com elevadas concentrações de gordura e sólidos insolúveis e baixos teores de açúcares redutores, facilitam a escoabilidade. Uma análise dos resultados obtidos na secagem de polpa de frutas tropicais, utilizando-se partículas de poliestireno de baixa densidade, como material inerte, mostrou que, embora a escoabilidade permita a obtenção de menores vazões de jorro mínimo, em relação ao desempenho do secador, pode não favorecer uma produção maior de pó.47548

    Low prevalence of renal, cardiac, pulmonary, and neurological extra-articular clinical manifestations in spondyloarthritis: analysis of the Brazilian Registry of Spondyloarthritis

    OBJECTIVE: To describe the extra-articular manifestations (cardiac, renal, pulmonary, and neurological), usually not related to spondyloarthritis (SpA), in a large cohort of Brazilian patients. MATERIALS AND METHODS: This retrospective study analyzed 1,472 patients diagnosed with SpA and cared for at 29 health care centers distributed in the five major geographic regions in the country, participating in the Brazilian Registry of Spondyloarthritis (BRS). All patients were assessed for the prevalence of major extra-articular manifestations (cardiac, renal, pulmonary, and neurological), classified according to the diagnosis [ankylosing spondylitis (AS), psoriatic arthritis (PsA), reactive arthritis (ReA), arthritis associated with inflammatory bowel disease (IBD), undifferentiated spondyloarthritis (uSpA), and juvenile SpA], and according to the clinical presentation (axial, peripheral, mixed, and enthesitis). RESULTS: Of the patients with SpA assessed, 963 had AS, 271 PsA, 49 ReA, 48 arthritis associated with IBD, 98 uSpA, and 43 juvenile SpA. Cardiac involvement was reported in 44 patients (3.0%), pulmonary involvement in 19 (1.3%), renal involvement in 17 (1.2%), and neurological involvement in 13 patients (0.9%). Most patients with visceral involvement had AS or PsA, and the mixed (axial + peripheral) and/or predominantly axial clinical form. CONCLUSION: Cardiac, renal, pulmonary, and neurological extra-articular manifestations are quite infrequent in SpA, ranging from 0.9% to 3% in this large Brazilian cohort, and affected predominantly patients with AS and PsA.OBJETIVO: Descrever as manifestações extra-articulares (cardíacas, renais, pulmonares e neurológicas) geralmente não relacionadas às espondiloartrites (EpA) em uma grande coorte de pacientes brasileiros. MÉTODOS: Este estudo retrospectivo analisou 1.472 pacientes com o diagnóstico de EpA atendidos em 29 centros distribuídos pelas cinco principais regiões geográficas do Brasil, integrantes do Registro Brasileiro de Espondiloartrites. Todos os pacientes foram avaliados para a prevalência das principais manifestações extra-articulares (cardíacas, renais, pulmonares e neurológicas), divididas por diagnóstico [espondilite anquilosante (EA), artrite psoriásica (AP), artrite reativa (ARe), artrite associada a doença inflamatória intestinal (DII), EpA indiferenciada (EI) e EpA juvenil] e por forma clínica (axial, periférica, mista e entesítica). RESULTADOS: Dentre os pacientes avaliados com EpA, 963 apresentavam EA, 271 AP, 49 ARe, 48 artrite associada a DII, 98 EI e 43 EpA juvenil. Acometimento cardíaco foi observado em 44 pacientes (3,0%), seguido por acometimento pulmonar em 19 (1,3%), renal em 17 (1,2%) e neurológico em 13 pacientes (0,9%). A maioria dos casos de acometimento visceral ocorreu nos pacientes com EA ou AP e naqueles com forma clínica mista (axial e periférica) e/ou predominantemente axial. CONCLUSÃO: As manifestações extra-articulares cardíacas, renais, pulmonares e neurológicas são muito pouco frequentes nas EpA, variando de 0,9%-3% nesta grande coorte brasileira, estando mais associadas a EA e AP.37938
