    A 520 year record of summer sunshine for the eastern European Alps based on stable carbon isotopes in larch tree rings

    A 520-year stable carbon isotope chronology from tree ring cellulose in high altitude larch trees (Larix decidua Mill.), from the eastern European Alps, correlates more strongly with summer temperature than with summer sunshine hours. However, when instrumental records of temperature and sunshine diverge after AD 1980, the tree ring time series does not follow warming summer temperatures but more closely tracks summer sunshine trends. It is concluded that sunshine is the dominant control on carbon isotope fractionation in these trees, via the influence of photosynthetic rate on the internal partial pressure of CO2, and that high summer (July-August) sunshine hours is a suitable target for climate reconstruction. We thus present the first reconstruction of summer sunshine for the eastern Alps and compare it with the regional temperature evolution

    RpfC (Rv1884) atomic structure shows high structural conservation within the resuscitation promoting factor catalytic domain

    We report the first structure of the catalytic domain of RpfC (Rv1884), one of theresuscitation-promoting factors (RPFs) from Mycobacterium tuberculosis. The structure was solved using molecular replacement, once the space group had been correctly identified as twinned P21 rather than the apparent C2221 by searching for anomalous scattering sites in P1. The structure displays a very high degree of structural conservation with the structures of the catalytic domains of RpfB (Rv1009) and RpfE (Rv2450) already published. This structural conservation highlights the importance of the versatile domain composition of the RPF family

    Spatio‐Temporal Variations in Carbon Isotope Discrimination Predicted by the JULES Land Surface Model

    Stable carbon isotopes in plants can help evaluate and improve the representation of carbon and water cycles in land-surface models, increasing confidence in projections of vegetation response to climate change. Here, we evaluated the predictive skills of the Joint UK Land Environmental Simulator (JULES) to capture spatio-temporal variations in carbon isotope discrimination (Δ13C) reconstructed by tree rings at twelve sites in the United Kingdom over the period 1979-2016. Modelled and measured Δ13C time series were compared at each site and their relationships with local climate investigated. Modelled Δ13C time series were significantly correlated (p < 0.05) with tree-ring Δ13C at eight sites, but JULES underestimated mean Δ13C values at all sites, by up to 2.6‰. Differences in mean Δ13C may result from post-photosynthetic isotopic fractionations that were not considered in JULES. Inter-annual variability in Δ13C was also underestimated by JULES at all sites. While modelled Δ13C typically increased over time across the UK, tree-ring Δ13C values increased only at five sites located in the northern regions but decreased at the southern-most sites. Considering all sites together, JULES captured the overall influence of environmental drivers on Δ13C but failed to capture the direction of change in Δ13C caused by air temperature, atmospheric CO2 and vapour pressure deficit at some sites. Results indicate that the representation of carbon-water coupling in JULES could be improved to reproduce both the trend and magnitude of interannual variability in isotopic records, the influence of local climate on Δ13C, and to reduce uncertainties in predicting vegetation-environment interactions

    Experimental results for nulling the effective thermal expansion coefficient of fused silica fibres under a static stress

    We have experimentally demonstrated that the effective thermal expansion coefficient of a fused silica fibre can be nulled by placing the fibre under a particular level of stress. Our technique involves heating the fibre and measuring how the fibre length changes with temperature as the stress on the fibre was systematically varied. This nulling of the effective thermal expansion coefficient should allow for the complete elimination of thermoelastic noise and is essential for allowing second generation gravitational wave detectors to reach their target sensitivity. To our knowledge this is the first time that the cancelation of the thermal expansion coefficient with stress has been experimentally observed

    Death or survival from invasive pneumococcal disease in Scotland: associations with serogroups and multilocus sequence types

    We describe associations between death from invasive pneumococcal disease (IPD) and particular serogroups and sequence types (STs) determined by multilocus sequence typing (MLST) using data from Scotland. All IPD episodes where blood or cerebrospinal fluid (CSF) culture isolates were referred to the Scottish Haemophilus, Legionella, Meningococcal and Pneumococcal Reference Laboratory (SHLMPRL) from January 1992 to February 2007 were matched to death certification records by the General Register Office for Scotland. This represented 5959 patients. The median number of IPD cases in Scotland each year was 292. Deaths, from any cause, within 30 days of pneumococcal culture from blood or CSF were considered to have IPD as a contributing factor. Eight hundred and thirty-three patients died within 30 days of culture of Streptococcus pneumoniae from blood or CSF [13.95%; 95% confidence interval (13.10, 14.80)]. The highest death rates were in patients over the age of 75. Serotyping data exist for all years but MLST data were only available from 2001 onward. The risk ratio of dying from infection due to particular serogroups or STs compared to dying from IPD due to all other serogroups or STs was calculated. Fisher's exact test with Bonferroni adjustment for multiple testing was used. Age adjustment was accomplished using the Cochran-Mantel-Haenszel test and 95% confidence intervals were reported. Serogroups 3, 11 and 16 have increased probability of causing fatal IPD in Scotland while serogroup 1 IPD has a reduced probability of causing death. None of the 20 most common STs were significantly associated with death within 30 days of pneumococcal culture, after age adjustment. We conclude that there is a stronger association between a fatal outcome and pneumococcal capsular serogroup than there is between a fatal outcome and ST

    Characterisation of Bombyx mori odorant-binding proteins reveals that a general odorant-binding protein discriminates between sex pheromone components

    In many insect species, odorant-binding proteins (OBPs) are thought to be responsible for the transport of pheromones and other semiochemicals across the sensillum lymph to the olfactory receptors (ORs) within the antennal sensilla. In the silkworm Bombyx mori, the OBPs are subdivided into three main subfamilies; pheromone-binding proteins (PBPs), general odorant-binding proteins (GOBPs) and antennal-binding proteins (ABPs). We used the MotifSearch algorithm to search for genes encoding putative OBPs in B. mori and found 13, many fewer than are found in the genomes of fruit flies and mosquitoes. The 13 genes include seven new ABP-like OBPs as well as the previously identified PBPs (three), GOBPs (two) and ABPx. Quantitative examination of transcript levels showed that BmorPBP1, BmorGOBP1, BmorGOBP2 and BmorABPx are expressed at very high levels in the antennae and so could be involved in olfaction. A new two-phase binding assay, along with other established assays, showed that BmorPBP1, BmorPBP2, BmorGOBP2 and BmorABPx all bind to the B. mori sex pheromone component (10E,12Z)-hexadecadien-1-ol (bombykol). BmorPBP1, BmorPBP2 and BmorABPx also bind the pheromone component (10E,12Z)-hexadecadienal (bombykal) equally well, whereas BmorGOBP2 can discriminate between bombykol and bombykal. X-ray structures show that when bombykol is bound to BmorGOBP2 it adopts a different conformation from that found when it binds to BmorPBP1. Binding to BmorGOBP2 involves hydrogen bonding to Arg110 rather than to Ser56 as found for BmorPBP1

    Rotating Resonator-Oscillator Experiments to Test Lorentz Invariance in Electrodynamics

    In this work we outline the two most commonly used test theories (RMS and SME) for testing Local Lorentz Invariance (LLI) of the photon. Then we develop the general framework of applying these test theories to resonator experiments with an emphasis on rotating experiments in the laboratory. We compare the inherent sensitivity factors of common experiments and propose some new configurations.     Trends in serotypes and sequence types among cases of invasive pneumococcal disease in Scotland, 1999-2010.

    INTRODUCTION: The 7-valent pneumococcal conjugate vaccine (Prevenar(Âź), Wyeth; PCV7) was introduced to the UK paediatric immunisation schedule in 2006. This study investigates trends in serotypes and multi locus sequence types (STs) among cases of invasive pneumococcal disease (IPD) in Scotland prior to, and following, the introduction of PCV7. METHODS: Scottish Invasive Pneumococcal Disease Enhanced Surveillance has records of all cases of IPD in Scotland since 1999. Cases diagnosed from blood or cerebrospinal fluid isolates until 2010 were analysed. Logistic and poisson regression modelling was used to assess trends prior to and following the introduction of PCV7. RESULTS: Prior to PCV7 use, on average 650 cases of IPD were reported each year; 12% occurred in those aged <5 years and 35% affected those aged over 65 years. Serotypes in PCV7 represented 47% of cases (68% in <5 year olds). The serotype and ST distribution was relatively stable with only serotype 1 and associated ST 306 showing an increasing trend. PCV7 introduction was associated with a 69% (95% CI: 50%, 80%) reduction in the incidence of IPD among those aged <5 years, a 57% (95% CI: 47%, 66%) reduction among those aged 5-64 years but no significant change among those aged 65 years and over where increases in non-PCV7 serotypes were observed. Serotypes which became more prevalent post-PCV7 are those which were associated with STs related to the PCV7 serotypes. CONCLUSIONS: Routine serotyping and sequence typing in Scotland allowed the assessment of the relationship between the capsule and the clones in the post vaccination era. Changes in the distribution of serotypes post PCV7 introduction appear to be driven by associations between serotypes and STs prior to PCV7 introduction. This has implications for the possible effects of the introduction of higher valency vaccines and could aid in predicting replacement serotypes in IPD

    On the elimination of the sweeping interactions from theories of hydrodynamic turbulence

    In this paper, we revisit the claim that the Eulerian and quasi-Lagrangian same time correlation tensors are equal. This statement allows us to transform the results of an MSR quasi-Lagrangian statistical theory of hydrodynamic turbulence back to the Eulerian representation. We define a hierarchy of homogeneity symmetries between incremental homogeneity and global homogeneity. It is shown that both the elimination of the sweeping interactions and the derivation of the 4/5-law require a homogeneity assumption stronger than incremental homogeneity but weaker than global homogeneity. The quasi-Lagrangian transformation, on the other hand, requires an even stronger homogeneity assumption which is many-time rather than one-time but still weaker than many-time global homogeneity.
