    As áreas urbanizadas do Município do Funchal têm vindo nas últimas décadas, a crescer para as cotas mais elevadas, hoje conhecidas como Zonas Altas do Funchal. Esta expansão urbana fez-se em boa parte sem planeamento territorial e urbanístico, num contexto territorial onde o fenómeno da construção ilegal é uma realidade incontornável, estando na origem de diversos problemas provocados pela ocupação descontrolada do território mais periférico, que tem como consequência a falta de espaços públicos, de equipamentos, áreas verdes e acessos adequados para a generalidade da população residente, fundamentais à vivência humana. Em resultado geram-se situações de carência urbana e de conflito, com efeitos na qualidade vida dos residentes. Tendo em conta esta realidade, a par da recente adaptação à Região da legislação nacional que permite a reconversão de áreas urbanas de génese ilegal, abre-se uma janela de oportunidade que, aproveitando todo este processo, poderá promover à reconversão destas Zonas Altas do Funchal. No âmbito desta dissertação propõe-se caraterizar os principais problemas destas áreas residenciais e aportar contributos que permitam, em primeiro lugar, a legalização, mas também abordar a sua regeneração urbana.The urbanized areas of the Funchal’s county have been growing over the last decades to the highest levels, now known as Funchal High Zones. This urban sprawl was largely done without any territorial and urban planning, in a territorial context where the phenomenon of illegal construction is an unavoidable reality, being the source of several problems caused by the uncontrolled occupation of the peripheral territory, which results in the lack of public spaces, equipment, green areas and adequate access for the general population, fundamental to the human experience. As a result, urban deprivation and conflict are generated, which creates effects on the quality of life of residents. Given this reality, along with the recent adaptation by the Region of national legislation that allows the conversion of urban area of illegal genesis, a window of opportunity opens up, taking advantage of this process, it may promote the conversion of these areas Funchal High Zone. This dissertation aims to characterize the main problems of these residential areas and contributes that allows, in the first place, a legalization, but also to address their urban regeneration

    Who are the patients with tuberculosis who are diagnosed in emergency facilities? An analysis of treatment outcomes in the state of São Paulo, Brazil.

    OBJECTIVE: Early tuberculosis diagnosis and treatment are determinants of better outcomes and effective disease control. Although tuberculosis should ideally be managed in a primary care setting, a proportion of patients are diagnosed in emergency facilities (EFs). We sought to describe patient characteristics by place of tuberculosis diagnosis and determine whether the place of diagnosis is associated with treatment outcomes. A secondary objective was to determine whether municipal indicators are associated with the probability of tuberculosis diagnosis in EFs. METHODS: We analyzed data from the São Paulo State Tuberculosis Control Program database for the period between January of 2010 and December of 2013. Newly diagnosed patients over 15 years of age with pulmonary, extrapulmonary, or disseminated tuberculosis were included in the study. Multiple logistic regression models adjusted for potential confounders were used in order to evaluate the association between place of diagnosis and treatment outcomes. RESULTS: Of a total of 50,295 patients, 12,696 (25%) were found to have been diagnosed in EFs. In comparison with the patients who had been diagnosed in an outpatient setting, those who had been diagnosed in EFs were younger and more socially vulnerable. Patients diagnosed in EFs were more likely to have unsuccessful treatment outcomes (adjusted OR: 1.54; 95% CI: 1.42-1.66), including loss to follow-up and death. At the municipal level, the probability of tuberculosis diagnosis in EFs was associated with low primary care coverage, inequality, and social vulnerability. In some municipalities, more than 50% of the tuberculosis cases were diagnosed in EFs. CONCLUSIONS: In the state of São Paulo, one in every four tuberculosis patients is diagnosed in EFs, a diagnosis of tuberculosis in EFs being associated with poor treatment outcomes. At the municipal level, an EF diagnosis of tuberculosis is associated with structural and socioeconomic indicators, indicating areas for improvement

    Characterization of PlyPl23, an endolysin from a Paenibacillus larvae bacteriophage

The estimated world value for the contribution of pollinators, especially bees, to the production of crops used directly for human food was estimated in 153 billion. Yet, beekeeping is facing many challenges, as changes in agricultural practices, extensive use of pesticides and the presence of pathogens. One of the most widespread and the most destructive of the honeybee brood diseases is the American foulbrood (AFB), caused by Paenibacillus larvae, a Gram-positive bacterium wherein the spore is the infectious form. In most situations P. larvae is highly virulent and when contracted will kill a complete colony. This represents an economic threat to commercial beekeepers and apiculture worldwide. The European regulation limits the presence of antibiotics in honey, excluding its use for therapy. The antibacterial features of (bacterio)phages and endolysins were considered for the development of a therapeutic product for this disease. A P. larvae phage, phiIBB_Pl23, belonging to the Siphoviridae family was isolated and its genome has been sequenced and analyzed. Annotation of the phiIBB_Pl23 genome identified gp21 as an endolysin protein, designated as PlyPl23, which was further characterized and is presented herein. In silico analysis revealed that PlyPl23, is a single domain globular protein, which is a rare feature of Gram-positive endolysins. The catalytic domain was identified as an N-acetylmuramoyl-L-alanine amidase (pfam Amidase_2 family) with a molecular weight of 25.8 kDa and isoelectric point of 5.74. Activity tests were performed at 37oC, which is similar to the hive temperature (about 34-35ºC) and revealed that this is a broad-spectrum endolysin for P. larvae with an optimal pH for reactivity between 3 and 5. This matches the pH levels of honey, nectar, pollen, and royal jelly (pH 3 to 4). The enzyme also displays a high antimicrobial activity between 5 and 7 when supplemented with NaCl, matching also the intestinal pH of bee larvae (pH 6.8) and adult bees (pH 5.6 to 6.3). Furthermore, tests performed with honeybee larvae juice revealed a higher bacterial decrease reduction, which may be a good indicator for the in vivo effectiveness of the endolysin. The present work describes, to our knowledge, the first characterization of an endolysin from a P. larvae phage.     Comparação de performance entre telas de poliutretano autolimpantes e telas de aço aplicadas em peneiramento a umidade natural de minério de ferro

    As telas para peneiramento são de basicamente 3 tipos: aço, poliuretano e borracha, sendo que só os dois primeiros são utilizados para operações à úmido ou umidade natural. As telas de aço, são comumente adotadas principalmente por possuírem maior área aberta e menor custo, mas, possuem vida útil curta quando comparadas as de poliuretano. Assim, para comparação entre o desempenho dessas telas no tratamento de minério de ferro, foram utilizadas duas peneiras iguais, uma com telas de aço e outra com telas de poliuretano. Apesar de menor área aberta, tanto nos quesitos taxa de alimentação, custo, produção de sucata metálica, peso, vida útil e tempo de limpeza, as telas de poliuretano tiveram performance superior, principalmente a vida útil 6x maior que a de aço. Assim, considerando produtividade, as telas de poliuretano performam maior utilização física e disponibilidade física além dos quesitos sustentabilidade, pela menor geração de sucata, do mesmo modo que maior segurança e ergonomia na movimentação das telas para os funcionários.&nbsp

    Effect of 0.9% saline solution and phosphate buffer saline at different temperatures and incubation times on the morphology of goat preantral follicles

    The present work investigated the efficiency of 0.9% saline solution and Phosphate Buffered Saline (PBS) in the preservation of goat preantral follicles in situ at different temperatures and incubation times. The ovarian pair of each animal was divided into 19 fragments. One ovarian fragment was taken randomly and fixed (control). The other 18 fragments were randomly distributed in tubes containing 0.9% saline solution or PBS at 4, 20 or 39 ºC for 4, 12 or 24 h. A total of 5,921 preantral follicles were examined. The quality of preantral follicles was evaluated by classical histology. The storage of ovarian fragments in 0.9% saline solution or PBS at 4 ºC did not reduce significantly the percentage of morphologically normal follicles when compared with the control, except after preservation in 0.9% saline solution for 24 h. The storage of ovarian fragments at 20 or 39°C reduced the percentage of normal preantral follicles when compared to the control, except after preservation in PBS at 20°C for 4 h. In conclusion, this study showed for the first time that goat preantral follicles can be stored in situ successfully at 4 ºC in 0.9% saline solution for 12 h and in PBS for 24 h, and at 20 ºC in PBS for 4 h.O presente trabalho investigou a eficiência da solução salina 0,9% e tampão fosfato salina (PBS) na conservação de folículos pré-antrais caprinos in situ a diferentes temperaturas e tempos de incubação. O par ovariano de cada animal foi dividido em 19 fragmentos. Um fragmento foi escolhido aleatoriamente e fixado (controle). Os outros 18 fragmentos foram distribuídos aleatoriamente em tubos contendo solução salina 0,9% ou PBS a 4, 20 ou 39 °C por 4, 12 ou 24 h. Um total de 5.921 folículos pré-antrais foram analisados. A qualidade dos folículos pré-antrais foi avaliada através de histologia clássica. A incubação de fragmentos ovarianos em solução salina 0,9% ou PBS a 4 ºC não reduziu significativamente a percentagem de folículos morfologicamente normais quando comparados com o controle, exceto após a conservação em solução salina 0,9% por 24 h. A incubação de fragmentos ovarianos a 20 ou 39°C reduziu a percentagem de folículos pré-antrais normais quando comparados com o controle, exceto após conservação em PBS a 20°C por 4 h. Em conclusão, este estudo mostrou pela primeira vez que folículos pré-antrais caprinos podem ser conservados in situ com sucesso a 4 ºC em solução salina 0,9% por 12 h e em PBS por 24 h, e a 20 ºC em PBS por 4 h

    Bilastine: stability-indicating a method using environmentally friendly by reversed-phase high-performance liquid chromatography (RP-HPLC)

    This study describes the development and validation of a new environmentally friendly analytical method for the determination of bilastine in coated tablets and the evaluation of its capacity to be stability-indicating as well. The ecofriendly analytical method was validated by specificity, linearity, accuracy, precision and robustness by reversed-phase high-performance liquid chromatography (RP-HPLC) according to International Conference on Harmonization guidelines (ICH) and Association of Official Analytical Chemists (AOAC). Isocratic LC separation was achieved on a RP18 column using a mobile phase of sodium dihydrogen phosphate aqueous buffer solution adjusted to pH (6.0 ± 0.1) with o-phosphoric acid (85% v/v) and triethylamine (0,3% v/v) and ethanol (EtOH) in the following proportions (60:40 v/v), at a flow rate of 1.0 mL·min-1 at temperature-controlled at 30 °C. The analytical method showed selectivity, good recovery and precision (intra- and inter-day), robustness, and linear over a range from 5.0 to 50 μg·mL-1
