    El estudio se centra en el conocimiento y comprensión de los procesos de modelización científica desarrollados en contextos multilingües dentro de la formación inicial del profesorado de primaria. Los datos corresponden a la asignatura Didáctica de las Ciencias desarrollada bajo el enfoque CLIL (Content and Language Integrated Learning) que promueve la enseñanza de contenidos disciplinares a través de una lengua vehicular (inglés). Se analizan las interacciones comunicativas de dos equipos de estudiantes durante actividades experimentales al estudiar el modelo de ser vivo. Las categorías de análisis estuvieron pautadas por tres ámbitos de reflexión, que explican la modelización científica: pensamiento-modelo, experiencia y lenguaje. Caracterizamos el papel que juega cada ámbito en momentos clave para construir un modelo explicativo más completo y complejo

    Antifungal activity against Fusarium culmorum of stevioside, Silybum marianum seed extracts, and their conjugate complexes

    Fusarium head blight (FHB) is a disease that poses a major challenge in cereal production that has important food and feed safety implications due to trichothecene contamination. In this study, the effect of stevioside—a glycoside found in the leaves of candyleaf (Stevia rebaudiana Bertoni)—was evaluated in vitro against Fusarium culmorum (W.G. Smith) Sacc., alone and in combination (in a 1:1 molar ratio) with polyphenols obtained from milk thistle seeds (Silybum marianum (L.) Gaertn). Different concentrations, ranging from 32 to 512 µg·mL−1, were assayed, finding EC50 and EC90 inhibitory concentrations of 156 and 221 µg·mL−1, respectively, for the treatment based only on stevioside, and EC50 and EC90 values of 123 and 160 µg·mL−1, respectively, for the treatment based on the stevioside–polyphenol conjugate complexes. Colony formation inhibition results were consistent, reaching full inhibition at 256 µg·mL−1. Given that synergistic behavior was observed for this latter formulation (SF = 1.43, according to Wadley’s method), it was further assessed for grain protection at storage, mostly directed against mycotoxin contamination caused by the aforementioned phytopathogen, confirming that it could inhibit fungal growth and avoid trichothecene contamination. Moreover, seed tests showed that the treatment did not affect the percentage of germination, and it resulted in a lower incidence of root rot caused by the pathogen in Kamut and winter wheat seedlings. Hence, the application of these stevioside–S. marianum seed extract conjugate complexes may be put forward as a promising and environmentally friendly treatment for the protection of cereal crops and stored grain against FHB

    The bacterial DNA binding protein matp involved in linking the nucleoid terminal domain to the divisome at midcell interacts with lipid membranes

Division ring formation at midcell is controlled by various mechanisms in Escherichia coli, one of them being the linkage between the chromosomal Ter macrodomain and the Z-ring mediated by MatP, a DNA binding protein that organizes this macrodomain and contributes to the prevention of premature chromosome segregation. Here we show that, during cell division, just before splitting the daughter cells, MatP seems to localize close to the cytoplasmic membrane, suggesting that this protein might interact with lipids. To test this hypothesis, we investigated MatP interaction with lipids in vitro. We found that, when encapsulated inside vesicles and microdroplets generated by microfluidics, MatP accumulates at phospholipid bilayers and monolayers matching the lipid composition in the E. coli inner membrane. MatP binding to lipids was independently confirmed using lipid-coated microbeads and biolayer interferometry assays, which suggested that the recognition is mainly hydrophobic. Interaction of MatP with the lipid membranes also occurs in the presence of the DNA sequences specifically targeted by the protein, but there is no evidence of ternary membrane/protein/DNA complexes. We propose that the association of MatP with lipids may modulate its spatiotemporal localization and its recognition of other ligands. IMPORTANCE The division of an E. coli cell into two daughter cells with equal genomic information and similar size requires duplication and segregation of the chromosome and subsequent scission of the envelope by a protein ring, the Z-ring. MatP is a DNA binding protein that contributes both to the positioning of the Z-ring at midcell and the temporal control of nucleoid segregation. Our integrated in vivo and in vitro analysis provides evidence that MatP can interact with lipid membranes reproducing the phospholipid mixture in the E. coli inner membrane, without concomitant recruitment of the short DNA sequences specifically targeted by MatP. This observation strongly suggests that the membrane may play a role in the regulation of the function and localization of MatP, which could be relevant for the coordination of the two fundamental processes in which this protein participates, nucleoid segregation and cell division

    Cap Formation Induced by Concanavalin A in Pathogenic Free-living Amoebae

    Morphological differences in cap formation were found when trophozoites of different free-living amoebae were treated with the lectin Concanavalin A, which resulted in a rapid redistribution of certain surface components to form small clusters and membrane-folded structures of diverse sizes. Acanthamoeba castellanii, Acanthamoba polyphaga and Naegleria lovaniensis exhibited characteristic caps, however, in A. castellanii this structure was larger and included several folds of the plasma membrane; furthermore, some of these caps had vacuoles containing a fi bro granular content. In contrast, the caps formed by A. polyphaga and N. lovaniensis lacked vacuoles. Regarding Naegleria fowleri, the trophozoites did not produce a defi ned cap, and only small patches of lectin-bound surface receptor complexes were observed at one pole of the cell body. In the free-living amoebae studied, it was not possible to correlate the shape and size of cap with pathogenicity

    Sugar beet agronomic performance evolution in NW Spain in future scenarios of climate change

    Changes in environmental conditions resulting from Climate Change are expected to have a major impact on crops. In order to foresee adaptation measures and to minimize yield decline, it is necessary to estimate the effect of those changes on the evapotranspiration and on the associated irrigation needs of crops. In the study presented herein, future conditions extracted from RCP4.5 scenario of IPCC, particularized for Castilla-y-León (Spain), were used as inputs for FAO crop simulation model (AquaCrop) to estimate sugar beet agronomic performance in the medium-term (2050 and 2070). A regional analysis of future trends in terms of yield, biomass and CO2 sequestration was carried out. An annual ET0 increase of up to 200 mm was estimated in 2050 and 2070 scenarios, with ETc increases of up to 40 mm/month. At current irrigation levels, temperature rise would be accompanied by a 9% decrease in yield and a ca. 6% decrease in assimilated CO2 in the 2050 and 2070 scenarios. However, it is also shown that the implementation of adequate adaptation measures, in combination with a more efficient irrigation management, may result in up to 17% higher yields and in the storage of between 9% and 13% higher amounts of CO2

    Relationship between Serum Concentration of Uric Acid and Insulin Secretion among Adults with Type 2 Diabetes Mellitus

    To determine the relationship between serum concentrations of uric acid and insulin secretion with hyperglycaemic clamp technique among adults with type 2 diabetes mellitus (DM2) without hyperuricemia, we carried out a cross-sectional study on 45 patients of both gender. We observed correlation between uric acid with male gender r = 0.710 (P = 0.001). Also correlation between uric acid and total insulin secretion was positive r = 0.295 (P = 0.049). As well as a positive correlation adjusted for body mass index was demonstrated for the first, second, and total phases of insulin secretion, respectively, r = 0.438 (P = 0.022), r = 0.433 (P = 0.022), and r = 0.439 (P = 0.024). Serum concentration of uric acid showed a positive relationship with the total phase of insulin secretion; even in states prior to hyperuricemia, uric acid can play an important role in the function of the beta cell in patients with DM2

    Pagurta: An interspecific hybrid of common seabream Pagrus pagrus (L., 1758) female and banded seabream males Pagrus auriga Valenciennes, 1843 male: phenotypic and molecular characterization

    Hybrids are of great interest in aquaculture. In the present study, we report for the first time an interespecific hybrid between common seabream Pagrus pagrus (L., 1758) females and red banded seabream Pagrus auriga (Valenciennes, 1843) males, which has been named the pagurta. This hybrid was detected in natural spawns during the period from March to April 2004. Phenotipically, the hybrid possessed morphological and meristic characteristics of both parental species. Mitochondrial and nuclear (microsatellite and satellite) molecular markers were used to confirm they were hybrids. The reciprocal cross was not detected.Los híbridos suscitan gran interés en la acuicultura. Sin embargo, su aplicación industrial en espáridos ha estado limitada por las dificultades técnicas que ocasiona su desarrollo. En este trabajo se describe el nacimiento no inducido, o espontáneo, de híbridos entre pargo Pagrus pagrus (L., 1758) y hurta Pagrus auriga Valenciennes, 1843 cuando dichas especies coexisten en un mismo tanque. Estos híbridos surgieron durante los meses de marzo y abril de 2004, cuando se registraron las puestas más abundantes de ambas especies. Fenotípicamente, presentaron características propias de ambas especies parentales. La aplicación de marcadores mitocondriales y nucleares (microsatélite y satélite) permitieron confirmar que eran híbridos intergenéricos de P. pagrus (♀ ) ˟ P. auriga (♂ ) a los que se denominó pagurta.     Effect of Diacerein on Insulin Secretion and Metabolic Control in Drug-Naïve Patients With Type 2 Diabetes: A randomized clinical trial

    OBJECTIVE - To assess the effect of diacerein on insulin secretion and metabolic control in drug-naïve patients with type 2 diabetes. RESEARCH DESIGN AND METHODS - A randomized, double-blind, placebo-controlled clinical trial was carried out in 40 drug-naïve adult patients with type 2 diabetes. A metabolic profile including interleukin (IL)-1β, tumor necrosis factor-a, IL-6, and fasting insulin levelswas carried out before the intervention and 2months afterward. A hyperglycemic-hyperinsulinemic clamp technique was performed to assess the phases of insulin secretion and insulin sensitivity. After randomization, 20 patients received diacerein (50mg once daily) for the first 15 days and twice daily for 45 additional days. The remaining patients received placebo. Intra- and intergroup differences were calculated by Wilcoxon signed rank and Mann-Whitney U tests. RESULTS-Therewere significant increases in first (102 ± 63 vs. 130 ± 75 pmol/L; P<0.01), late (219 ± 111 vs. 280 ± 135 pmol/L; P<0.01), and total insulin (178691 vs. 216699pmol/L; P<0.01) secretionswithout changes in insulin sensitivity after diacerein administration. There were significant decreases in fasting glucose (7.9 ± 1.4 vs. 6.8 ± 1.0mmol/L; P<0.01) and in A1C levels (8.3 ± 1.0 vs. 7.0 ± 0.8%; P < 0.001) after diacerein administration. There were no significant changes after placebo administration in the above-mentioned evaluations. CONCLUSIONS - Insulin secretion increased and metabolic control improved after diacerein administration in drug-naïve patients with type 2 diabetes. © 2011 by the American Diabetes Association
