    Controlling thermal chaos in the mantle by positive feedback from radiative thermal conductivity

    International audienceThe thermal conductivity of mantle materials has two components, the lattice component klat from phonons and the radiative component krad due to photons. These two contributions of variable thermal conductivity have a nonlinear dependence in the temperature, thus endowing the temperature equation in mantle convection with a strongly nonlinear character. The temperature derivatives of these two mechanisms have different signs, with ?klat /?T negative and dkrad /dT positive. This offers the possibility for the radiative conductivity to control the chaotic boundary layer instabilities developed in the deep mantle. We have parameterized the weight factor between krad and klat with a dimensionless parameter f , where f = 1 corresponds to the reference conductivity model. We have carried out two-dimensional, time-dependent calculations for variable thermal conductivity but constant viscosity in an aspect-ratio 6 box for surface Rayleigh numbers between 106 and 5 Ă— 106. The averaged PĂ©clet numbers of these flows lie between 200 and 2000. Along the boundary in f separating the chaotic and steady-state solutions, the number decreases and the Nusselt number increases with internal heating, illustrating the feedback between internal heating and radiative thermal conductivity. For purely basal heating situation, the time-dependent chaotic flows become stabilized for values of f of between 1.5 and 2. The bottom thermal boundary layer thickens and the surface heat flow increases with larger amounts of radiative conductivity. For magnitudes of internal heating characteristic of a chondritic mantle, much larger values of f , exceeding 10, are required to quench the bottom boundary layer instabilities. By isolating the individual conductive mechanisms, we have ascertained that the lattice conductivity is partly responsible for inducing boundary layer instabilities, while the radiative conductivity and purely depth-dependent conductivity exert a stabilizing influence and help to control thermal chaos developed in the deep mantle. These results have been verified to exist also in three-dimensional geometry and would argue for the need to consider the potentially important role played by radiative thermal conductivity in controlling chaotic flows in time-dependent mantle convection, the mantle heat transfer, the number of hotspots and the attendant mixing of geochemical anomalies

    Characterization of the monocyte-specific esterase (MSE) gene

    Carboxylic esterases are widely distributed in hematopoietic cells. Monocytes express the esterase isoenzyme (termed 'monocyte-specific esterase', MSE) that can be inhibited by NaF in the alpha-naphthyl acetate cytochemical staining. We examined the expression of MSE in normal cells and primary and cultured leukemia-lymphoma cells. The MSE protein was demonstrated by isoelectric focusing (IEF); MSE mRNA expression was investigated by Northern blotting and reverse transcriptase-polymerase chain reaction (RT-PCR). The following samples were positive for MSE protein and Northern mRNA expression: 20/24 monocytic, 4/32 myeloid, and 1/20 erythroid-megakaryocytic leukemia cell lines, but none of the 112 lymphoid leukemia or lymphoma cell lines; of the normal purified cell populations only the monocytes were positive whereas, T, B cells, and granulocytes were negative; of primary acute (myelo) monocytic leukemia cells (CD14-positive, FAB M4/M5 morphology) 14/20 were Northern mRNA and 11/14 IEF protein positive. RT-PCR revealed MSE expression in 29/49 Northern-negative lymphoid leukemia-lymphoma cell lines. The RT-PCR signals in monocytic cell lines were on average 50-fold stronger than the mostly weak trace expression in lymphoid specimens. On treatment with various biomodulators, only all-trans retinoic acid significantly upregulated MSE message and protein levels but could not induce new MSE expression in several leukemia cell lines; lipopolysaccharide and interferon-gamma increased MSE expression in normal monocytes. Analysis of DNA methylation with sensitive restriction enzymes showed no apparent regulation of gene expression by differential methylation; the MSE gene is evolutionarily conserved among mammalian species; the half-life of the human MSE transcripts was about 5-6 h. The extent of MSE expression varied greatly among different monocytic leukemia samples. However, the MSE overexpression in a significant number of specimens was not associated with gene amplification, gross structural rearrangements or point mutations within the cDNA region. Taken together, the results suggest that MSE expression is not absolutely specific for, but strongly associated with cells of the monocytic lineage; MSE is either not expressed at all or expressed at much lower levels in cells from other lineages. The biological significance, if any, of rare MSE messages in lymphoid cells detectable only by the hypersensitive RT-PCR remains unclear. Further studies on the regulation of this gene and on the physiological function of the enzyme will no doubt be informative with respect to its striking overexpression in some malignant cells and to a possible role in the pathobiology of monocytic leukemias

    TreeDomViewer: a tool for the visualization of phylogeny and protein domain structure

    Phylogenetic analysis and examination of protein domains allow accurate genome annotation and are invaluable to study proteins and protein complex evolution. However, two sequences can be homologous without sharing statistically significant amino acid or nucleotide identity, presenting a challenging bioinformatics problem. We present TreeDomViewer, a visualization tool available as a web-based interface that combines phylogenetic tree description, multiple sequence alignment and InterProScan data of sequences and generates a phylogenetic tree projecting the corresponding protein domain information onto the multiple sequence alignment. Thereby it makes use of existing domain prediction tools such as InterProScan. TreeDomViewer adopts an evolutionary perspective on how domain structure of two or more sequences can be aligned and compared, to subsequently infer the function of an unknown homolog. This provides insight into the function assignment of, in terms of amino acid substitution, very divergent but yet closely related family members. Our tool produces an interactive scalar vector graphics image that provides orthological relationship and domain content of proteins of interest at one glance. In addition, PDF, JPEG or PNG formatted output is also provided. These features make TreeDomViewer a valuable addition to the annotation pipeline of unknown genes or gene products.     A fully integrated microfluidic device for point of care monitoring of antithrombotics

    © 2016 The Royal Society of Chemistry. The simplicity and efficiency of point of care diagnostics have revolutionised patient care. Current methods for measuring hypercoagulability often require trained technicians, large blood volumes, and result in long turnaround times. Standard testing for hypercoagulable disorders is performed in the central laboratory using automated coagulation analysers. However the trend is moving towards the development and implementation of point of care testing, as a result of the ever increasing number of patients on antithrombotic therapy. We present a novel microfluidic device and assay for monitoring the effect of two anticoagulants, unfractionated heparin (UFH) and low molecular weight heparin (LMWH). The assay is based on the anti-Xa assay principle but uses fluorescence detection. Our device is a disposable laminate microfluidic strip, fabricated from the cyclic polyolefin (COP), Zeonor®, which is extremely suitable for application to fluorescent device platforms. We present data on the execution of the anti-Xa assay in this microfluidic format, demonstrating that the assay can be used to measure both UFH and LMWH in human plasma samples from 0 to 1 U mL-1, with a rapid result obtained within 30-60 seconds

    A Study of Cross-Cultural Groups at Malcolm Price Laboratory School; University of Northern Iowa

    A brief history of the integration efforts of the school and the resulting interracial conflict is presented. From this background evolved the cross-cultural groups which are the focus of this paper. Essentially; discussion was used for exploring the issues which were identified as impediments to good relationships within the school; (1) too few black faculty and students; (2) the impact of Tomism; (3) geographic location of students\u27 homes; (4) differing interests of black and white students; (5) the need for extra-school interracial contact; and (6) inhibitions; both social and parental; against interracial contact; especially dating. The results of a questionnaire; administered to 51 black and white students in the school; suggests the impact of the cross-cultural groups. While a number of negative aspects are revealed in the data; the authors focus on two positives: (1) the overwhelming enthusiasm of students to continue participating in the groups; and (2) principal and staff perceptions of an improved school environment.     Cross-Cultural Counseling: Bridge Between Desegregation and Integration. A Study Relative to Group Counseling and its Impact on Students and Faculty

    For the past several years; much emphasis has been placed on open enrollment plans involving bussing that enable black and white students to attend schools together. The Laboratory School of the University of Northern Iowa in Cedar Falls has been involved in such a program since 1968. As the project developed; the potential for very serious problems relating to racial tension and student frustration became evident. To approach these problems and to promote more openness and understanding between students from different communities and differing cultural backgrounds; cross-cultural group counseling sessions were developed. The groups during the past two years have consisted of combined 7th and 8th graders and combined 9th through 12th graders. Due to administrative support; student requests; and observed behavior change; a similar grouping of students will be employed during the 1972-73 academic year. Counselor observations of the program are examined; and student and faculty questionnaires and analyses of them are also included in this paper. (Author

    The Large-scale J=3-2 and J=2-1 CO Emission from M17 and its Implications for Extragalactic CO Observations

    We observed a 10x20 pc region of the molecular cloud M17 in the 12CO and 13CO J=3-2 and J=2-1 transitions to determine their global behavior and to assess the reliability of using ratios of CO line intensities integrated over an entire cloud to determine the physical conditions within the cloud. Both the 12CO/13CO J=2-1 and J=3-2 line ratios correlate with the 13CO integrated intensity, with smaller line ratios observed at locations with large integrated intensities. This correlation is likely due to variations in the column density from one position to another within M17. The 12CO and 13CO (J=3-2/J=2-1) line ratios show no significant variation from place to place within M17, even on the peak of the photon-dominated region. A Large Velocity Gradient analysis of globally averaged line ratios gives results in reasonable agreement with the results obtained for individual lines-of-sight through the cloud, which suggests that the typical physical conditions in a molecular cloud can be determined using CO line ratios integrated over the entire cloud. There appears to be a clear trend of increasing 12CO/13CO J=2-1 and J=3-2 line ratios as one moves from Galactic molecular cloud cores to entire Galactic molecular clouds to normal galaxies. The most likely explanation of the high line ratios for normal galaxies is a significant contribution to the CO emission by low column density material, such as diffuse molecular clouds or the outer envelopes of giant molecular clouds.Comment: 26 pages, 6 figures, 2 tables.
