On the shopfloor: exploring the impact of teacher trade unions on school-based industrial relations
Teachers are highly unionised workers and their trade unions exert an important influence on the shaping and implementation of educational policy. Despite this importance there is relatively little analysis of the impact of teacher trade unions in educational management literature. Very little empirical research has sought to establish the impact of teacher unions at school level. In an era of devolved management and quasi-markets this omission is significant. New personnel issues continue to emerge at school level and this may well generate increased trade union activity at the workplace. This article explores the extent to which devolved management is drawing school-based union representation into a more prominent role. It argues that whilst there can be significant differences between individual schools, increased school autonomy is raising the profile of trade union activity in the workplace, and this needs to be better reflected in educational management research
Metadata schema registries in the partially Semantic Web: The CORES experience
The CORES metadata schemas registry is designed to enable users to discover and navigate metadata element sets. The paper reflects on some of the experiences of implementing the registry, and examines some of the issues of promoting such services in the context of a "partially Semantic Web" where metadata applications are evolving and many have not yet adopted the RDF model
What terms does your metadata use? Application profiles as machine-understandable narratives
The SCHEMAS Registry aims at providing a selected and annotated overview of metadata vocabularies and their use in application environments. Based on harvested metadata in RDF (Resource Description Framework), the registry allows users to explore links between "namespace schemas", which declare standard definitions of metadata terms, and "application profiles" ? RDF statements about the use or adaptation of namespace terms for particular domains, services, or projects. Where instance metadata does not follow standard namespaces or explicit data models, this style allows implementors to assert an explicit mapping to standard terms. Registering profiles can help harmonize metadata usage in particular domains and, in the longer term, could provide a machine-processable basis for automating crosswalks and conversions
PRAME Is a Golgi-Targeted Protein That Associates with the Elongin BC Complex and Is Upregulated by Interferon-Gamma and Bacterial PAMPs
Preferentially expressed antigen in melanoma (PRAME) has been described as a cancer-testis antigen and is associated with leukaemias and solid tumours. Here we show that PRAME gene transcription in leukaemic cell lines is rapidly induced by exposure of cells to bacterial PAMPs (pathogen associated molecular patterns) in combination with type 2 interferon (IFNγ). Treatment of HL60 cells with lipopolysaccharide or peptidoglycan in combination with IFNγ resulted in a rapid and transient induction of PRAME transcription, and increased association of PRAME transcripts with polysomes. Moreover, treatment with PAMPs/IFNγ also modulated the subcellular localisation of PRAME proteins in HL60 and U937 cells, resulting in targeting of cytoplasmic PRAME to the Golgi. Affinity purification studies revealed that PRAME associates with Elongin B and Elongin C, components of Cullin E3 ubiquitin ligase complexes. This occurs via direct interaction of PRAME with Elongin C, and PRAME colocalises with Elongins in the Golgi after PAMP/IFNγ treatment. PRAME was also found to co-immunoprecipitate core histones, consistent with its partial localisation to the nucleus, and was found to bind directly to histone H3 in vitro. Thus, PRAME is upregulated by signalling pathways that are activated in response to infection/inflammation, and its product may have dual functions as a histone-binding protein, and in directing ubiquitylation of target proteins for processing in the Golgi
Delineation of a unique protein-protein interaction site on the surface of the estrogen receptor
Recent studies have identified a series of estrogen receptor (ER)interacting peptides that recognize sites that are distinct from the classic coregulator recruitment (AF2) region. Here, we report the structural and functional characterization of an ER alpha-specific peptide that binds to the liganded receptor in an AF2-independent manner. The 2-angstrom crystal structure of the ER/peptide complex reveals a binding site that is centered on a shallow depression on the beta-hairpin face of the ligand-binding domain. The peptide binds in an unusual extended conformation and makes multiple contacts with the ligand-binding domain. The location and architecture of the binding site provides an insight into the peptide's ER subtype specificity and ligand interaction preferences. In vivo, an engineered coactivator containing the peptide motif is able to strongly enhance the transcriptional activity of liganded ER alpha, particularly in the presence of 4-hydroxytamoxifen. Furthermore, disruption of this binding surface alters ER's response to the coregulator TIF2. Together, these results indicate that this previously unknown interaction site represents a bona fide control surface involved in regulating receptor activity
Analyses of 123 Peripheral Human Immune Cell Subsets: Defining Differences with Age and between Healthy Donors and Cancer Patients not Detected in Analysis of Standard Immune Cell Types
Recent advances in human immunology have led to the identification of novel immune cell subsets and the biological function of many of these subsets has now been identified. The recent US Food and Drug Administration approval of several immunotherapeutics for the treatment of a variety of cancer types and the results of ongoing immunotherapy clinical studies requires a more thorough interrogation of the immune system. We report here the use of flow cytometry-based analyses to identify 123 immune cell subsets of peripheral blood mononuclear cells. The use of these panels defines multiple differences in younger (< 40 years) vs. older (≥ 40 years) individuals and between aged-matched apparently healthy individuals and metastatic cancer patients, aspects not seen in the analysis of the following standard immune cell types: CD8, CD4, natural killer, natural killer-T, regulatory T, myeloid derived suppressor cells, conventional dendritic cells (DCs), plasmacytoid DCs and B cells. The use of these panels identifying 123 immune cell subsets may aid in the identification of patients who may benefit from immunotherapy, either prior to therapy or early in the immunotherapeutic regimen, for the treatment of cancer or other chronic or infectious diseases
A signature motif mediating selective interactions of BCL11A with the NR2E/F subfamily of orphan nuclear receptors
Despite their physiological importance, selective interactions between nuclear receptors (NRs) and their cofactors are poorly understood. Here, we describe a novel signature motif (F/YSXXLXXL/Y) in the developmental regulator BCL11A that facilitates its selective interaction with members of the NR2E/F subfamily. Two copies of this motif (named here as RID1 and RID2) permit BCL11A to bind COUP-TFs (NR2F1;NR2F2;NR2F6) and Tailless/TLX (NR2E1), whereas RID1, but not RID2, binds PNR (NR2E3). We confirmed the existence of endogenous BCL11A/TLX complexes in mouse cortex tissue. No interactions of RID1 and RID2 with 20 other ligand-binding domains from different NR subtypes were observed. We show that RID1 and RID2 are required for BCL11A-mediated repression of endogenous γ-globin gene and the regulatory non-coding transcript Bgl3, and we identify COUP-TFII binding sites within the Bgl3 locus. In addition to their importance for BCL11A function, we show that F/YSXXLXXL/Y motifs are conserved in other NR cofactors. A single FSXXLXXL motif in the NR-binding SET domain protein NSD1 facilitates its interactions with the NR2E/F subfamily. However, the NSD1 motif incorporates features of both LXXLL and FSXXLXXL motifs, giving it a distinct NR-binding pattern in contrast to other cofactors. In summary, our results provide new insights into the selectivity of NR/cofactor complex formation
The crisis of public sector trade unionism: evidence from the Mid Staffordshire hospital crisis
This article contends that there is a growing, if uneven, crisis in public sector trade unionism masked by relatively high membership figures that obscure a weakening of trade unions in the workplace, leaving hollowed out organisation vulnerable to further legislative and employer-led onslaughts. The weakening is not inevitable but to overcome it requires a refocusing of organising efforts on the everyday concerns of members such as understaffing and the provision of better public services. Only with an engaged membership will national issues and wider campaigns have material force. Having outlined a general argument, the article takes as illustrative the nature and performance of trade unions, and particularly UNISON, during the Mid Staffordshire hospital crisis
Actitudes y percepciones de los docentes universitarios en el proceso de adopción de una innovación. La iniciativa OpenCourseWare en la Universidad de Valencia (España)
El proyecto OpenCourseWare, repositorio de recursos educativos en abierto, se inició en la Universidad de Valencia en 2008 y en 2009, sólo incluía 10 asignaturas, ninguna de ellas del área de ciencias de la salud. El objetivo de este trabajo es investigar las actitudes de los docentes de esta área así como sus percepciones con relación a este repositorio para determinar cuáles son las estrategias necesarias para extender su uso. Se parte de la teoría de la difusión de las innovaciones de Rogers (2003) y de un enfoque cualitativo a través del muestreo teórico, entrevistas semiestructuradas y codificación abierta. Los resultados indican la existencia de dos grandes grupos de profesores. Los adoptantes tempranos con una predisposición alta a participar en la iniciativa y con una percepción positiva de sus atributos y los mainstream o corriente principal más reacios a participar y con una percepción negativa respecto a alguno de sus atributos. La identificación de las características de estos dos grandes grupos marca las líneas de acción y de investigación futura en el ámbito del marketing y difusión, formación, sensibilización y motivación para extender el uso del OCW. The OpenCourseWare project, an open learning resource repository, was set up at the University of Valencia between 2008 and 2009. It initially consisted of only 10 subjects, none of which was in the field of health sciences. The objective of this research is to investigate the attitudes and perceptions of teaching staff in the field of health sciences with regard to the project in order to identify strategies to increase its use and expand its coverage. The diffusion of innovations theory as propounded by Rogers is used, along with a qualitative approach using theoretical sampling, semi-structured interviews and open coding. The results indicate the existence of two categories of teaching staff. Firstly, those who adopt an innovation early, have a marked inclination to participate in the initiative and a positive perception of the characteristics of the innovation. Secondly, a larger category of those more reluctant to participate in it and with a negative perception of one or more of its characteristics. Identifying the nature of these two groups provides guidelines for future action and research in the marketing and distribution of such projects, and the training, awareness and motivation required in order to extend the use of OCW
