    Analysis of ecosystem services provision in the Colombian Amazon using participatory research and mapping techniques

    Over the last two decades indigenous peoples in the lower Caquetá River basin in Colombia have experienced detrimental changes in the provision of important ecosystem services in ways that have significant implications for the maintenance of their traditional livelihoods. To assess these changes we conducted eight participatory mapping activities and convened 22 focus group discussions. We focused the analysis on two types of change: (1) changes in the location of ecosystem services provisioning areas and (2) changes in the stock of ecosystem services. The focal ecosystem services include services such as provision of food, raw materials and medicinal resources. Results from the study show that in the past two decades the demand for food and raw materials has intensified and, as a result, locations of provisioning areas and the stocks of ecosystem services have changed. We found anecdotal evidence that these changes correlate well with socio-economic factors such as greater need for income generation, change in livelihood practices and consumption patterns. We discuss the use of participatory mapping techniques in the context of marginalized and data-poor regions. We also show how this kind of information can strengthen existing ecosystem-based management strategies used by indigenous peoples in the Colombian Amazon

    Protocolos diseñados para el biobanco de dientes de la Universidad Nacional de Colombia.

    En un banco de dientes los protocolos son indispensables para garantizar: la correcta organización y el buen funcionamiento, la reproducción y estandarización que disminuyan la contaminación cruzada, conserven las características y propiedades sicoquímicas originales de los dientes, el cumplimiento de la normatividad ético–legal y un correcto manejo de los desechos. Objetivo: proponer los protocolos de desinfección, almacenamiento y transporte para el Biobanco de dientes en la Universidad Nacional de Colombia. Materiales y métodos: se revisaron las bases de datos Pubmed, Science Direct y Scielo utilizando las palabras “teeth, human, tooth bank, disinfection, sterilization, storage, organization, biosecurity, biobank, protocol, prevention”. Fueron incluidos 37 artículos, publicados entre 1988 y 2014, sobre banco de dientes, lineamientos para su limpieza, desinfección y almacenamiento. Se consultaron las normas internacionales y las ético–legales vigentes en Colombia para donación, manejo e investigación con órganos. Se estudiaron, analizaron y modicaron críticamente los procesos desde la recepción hasta su transformación de órganos en muestras aptas para investigación teniendo en cuenta la literatura, las experiencias previas en el laboratorio a través de trabajos de grado y tesis de posgrado, y los conocimientos en química inorgánica. Resultados: se planearon y elaboraron los protocolos de recolección, transporte, limpieza/desinfección y almacenamiento. Conclusión: basados en la literatura cientíca, en normas internacionales y nacionales vigentes y la experiencia en el laboratorio se propusieron los protocolos de desinfección, transporte particulares para el Biobanco de dientes de la Universidad Nacional de Colombia.Protocols in a Tooth Bank are essential in order to assure smooth operation, reproducibility and standardization that minimize cross contamination, maintain original characteristics and physicochemical properties of teeth, fulll ethical and legal regulations and a proper disposal of residues. Objective: to propose the disinfection, storing and transportation protocols for the UNTB. Methods: A literature search was conducted using the words “teeth, human, tooth bank, disinfection, sterilization, storage, organization, biosecurity, biobank, protocol, prevention” in the Pubmed, Science Direct and Scielo databases. 37 papers ranging from 1988 up to 2014 were selected. International and Colombian ethical and legal regulations for organ donation, handling and investigation were taken into account as well as laboratory observations and chemical basic principles gained through several undergraduate and graduate thesis. All this input was carefully studied, analysed and critically modied for setting the recommended processes for the conversion of donated teeth into organs suitable for research. Results: Collection, transportation, cleaning/disinfection and storing protocols were planned and elaborated. Conclusions: Based on scientic literature, national and international regulations and experimental experience, several protocols for the UNTB were presented

    A novel, stain-free, natural auto-fluorescent signal, Sig M, identified from cytometric and transcriptomic analysis of infectivity of Cryptosporidium hominis and Cryptosporidium parvum

    Cryptosporidiosis is a worldwide diarrheal disease caused by the protozoan Cryptosporidium. The primary symptom is diarrhea, but patients may exhibit different symptoms based on the species of the Cryptosporidium parasite they are infected with. Furthermore, some genotypes within species are more transmissible and apparently virulent than others. The mechanisms underpinning these differences are not understood, and an effective in vitro system for Cryptosporidium culture would help advance our understanding of these differences. Using COLO-680N cells, we employed flow cytometry and microscopy along with the C. parvum-specific antibody Sporo-Glo™ to characterize infected cells 48 h following an infection with C. parvum or C. hominis. The Cryptosporidium parvum-infected cells showed higher levels of signal using Sporo-Glo™ than C. hominis-infected cells, which was likely because Sporo-Glo™ was generated against C. parvum. We found a subset of cells from infected cultures that expressed a novel, dose-dependent auto-fluorescent signal that was detectable across a range of wavelengths. The population of cells that expressed this signal increased proportionately to the multiplicity of infection. The spectral cytometry results confirmed that the signature of this subset of host cells closely matched that of oocysts present in the infectious ecosystem, pointing to a parasitic origin. Present in both C. parvum and C. hominis cultures, we named this Sig M, and due to its distinct profile in cells from both infections, it could be a better marker for assessing Cryptosporidium infection in COLO-680N cells than Sporo-Glo™. We also noted Sig M’s impact on Sporo-Glo™ detection as Sporo-Glo™ uses fluoroscein–isothiocynate, which is detected where Sig M also fluoresces. Lastly, we used NanoString nCounter® analysis to investigate the transcriptomic landscape for the two Cryptosporidium species, assessing the gene expression of 144 host and parasite genes. Despite the host gene expression being at high levels, the levels of putative intracellular Cryptosporidium gene expression were low, with no significant difference from controls, which could be, in part, explained by the abundance of uninfected cells present as determined by both Sporo-Glo™ and Sig M analyses. This study shows for the first time that a natural auto-fluorescent signal, Sig M, linked to Cryptosporidium infection can be detected in infected host cells without any fluorescent labeling strategies and that the COLO-680N cell line and spectral cytometry could be useful tools to advance the understanding of Cryptosporidium infectivity

    Urticaria : recommendations from the Italian Society of Allergology, Asthma and Clinical Immunology and the Italian Society of Allergological, Occupational and Environmental Dermatology

    Background: Urticaria is a disorder affecting skin and mucosal tissues characterized by the occurrence of wheals, angioedema or both, the latter defining the urticaria-angioedema syndrome. It is estimated that 12-22% of the general population has suffered at least one subtype of urticaria during life, but only a small percentage (estimated at 7.6-16%) has acute urticaria, because it is usually self-limited and resolves spontaneously without requiring medical attention. This makes likely that its incidence is underestimated. The epidemiological data currently available on chronic urticaria in many cases are deeply discordant and not univocal, but a recent Italian study, based on the consultation of a national registry, reports a prevalence of chronic spontaneous urticaria of 0.02% to 0.4% and an incidence of 0.1-1.5 cases/1000 inhabitants/year. Methods: We reviewed the recent international guidelines about urticaria and we described a methodologic approach based on classification, pathophysiology, impact on quality of life, diagnosis and prognosis, differential diagnosis and management of all the types of urticaria. Conclusions: The aim of the present document from the Italian Society of Allergology, Asthma and Clinical Immunology (SIAAIC) and the Italian Society of Allergological, Occupational and Environmental Dermatology (SIDAPA) is to provide updated information to all physicians involved in diagnosis and management of urticaria and angioedema
