Growing and testing mycelium bricks as building insulation materials
In order to improve energy performance of buildings, insulation materials (such as mineral glass and rock wools, or fossil fuel-based plastic foams) are being used in increasing quantities, which may lead to potential problem with materials depletions and landfill disposal. One sustainable solution suggested is the use of bio-based, biodegradable materials. A number of attempts have been made to develop biomaterials, such as sheep wood, hemcrete or recycled papers. In this paper, a novel type of bio insulation materials ? mycelium is examined. The aim is to produce mycelium materials that could be used as insulations. The bio-based material was required to have properties that matched existing alternatives, such as expanded polystyrene, in terms of physical and mechanical characteristics but with an enhanced level of biodegradability. The testing data showed mycelium bricks exhibited good thermal performance. Strong and weak lensing united III: Measuring the mass distribution of the merging galaxy cluster 1E0657-56
The galaxy cluster 1E0657-56 (z = 0.296) is remarkably well-suited for
addressing outstanding issues in both galaxy evolution and fundamental physics.
We present a reconstruction of the mass distribution from both strong and weak
gravitational lensing data. Multi-color, high-resolution HST ACS images allow
detection of many more arc candidates than were previously known, especially
around the subcluster. Using the known redshift of one of the multiply imaged
systems, we determine the remaining source redshifts using the predictive power
of the strong lens model. Combining this information with shape measurements of
"weakly" lensed sources, we derive a high-resolution, absolutely-calibrated
mass map, using no assumptions regarding the physical properties of the
underlying cluster potential. This map provides the best available
quantification of the total mass of the central part of the cluster. We also

Progress Report 2008: A Scalable and Extensible Earth System Model for Climate Change Science
This project employs multi-disciplinary teams to accelerate development of the Community Climate System Model (CCSM), based at the National Center for Atmospheric Research (NCAR). A consortium of eight Department of Energy (DOE) National Laboratories collaborate with NCAR and the NASA Global Modeling and Assimilation Office (GMAO). The laboratories are Argonne (ANL), Brookhaven (BNL) Los Alamos (LANL), Lawrence Berkeley (LBNL), Lawrence Livermore (LLNL), Oak Ridge (ORNL), Pacific Northwest (PNNL) and Sandia (SNL). The work plan focuses on scalablity for petascale computation and extensibility to a more comprehensive earth system model. The human squamous oesophagus has widespread capacity for clonal expansion from cells at diverse stages of differentiation.
OBJECTIVE: Knowledge of the cellular mechanisms involved in homeostasis of human squamous oesophagus in the steady state and following chronic injury is limited. We aimed to better understand these mechanisms by using a functional 3D approach. DESIGN: Proliferation, mitosis and the expression of progenitor lineage markers were assessed in normal squamous oesophagus from 10 patients by immunofluorescence on 3D epithelial whole mounts. Cells expressing differential levels of epithelial and progenitor markers were isolated using flow cytometry sorting and characterised by qPCR and IF. Their self-renewing potential was investigated by colony forming cells assays and in vitro organotypic culture models. RESULTS: Proliferation and mitotic activity was highest in the interpapillary basal layer and decreased linearly towards the tip of the papilla (p<0.0001). The orientation of mitosis was random throughout the basal layer, and asymmetric divisions were not restricted to specific cell compartments. Cells sorted into distinct populations based on the expression of epithelial and progenitor cell markers (CD34 and EpCAM) showed no difference in self-renewal in 2D culture, either as whole populations or as single cells. In 3D organotypic cultures, all cell subtypes were able to recapitulate the architecture of the tissue of origin and the main factor determining the success of the 3D culture was the number of cells plated, rather than the cell type. CONCLUSIONS: Oesophageal epithelial cells demonstrate remarkable plasticity for self-renewal. This situation could be viewed as an ex vivo wounding response and is compatible with recent findings in murine models
Expression and regulation of drug transporters in vertebrate neutrophils.
There remains a need to identify novel pro-resolution drugs for treatment of inflammatory disease. To date, there are no neutrophil-specific anti-inflammatory treatments in clinical use, perhaps due to our lack of understanding of how drugs access this complex cell type. Here we present the first comprehensive description and expression of both major classes of drug transporters, SLC and ABC, in resting human blood neutrophils. Moreover, we have studied the expression of these carriers in the tractable model system, the zebrafish (Danio rerio), additionally examining the evolutionary relationship between drug transporters in zebrafish and humans. We anticipate that this will be a valuable resource to the field of inflammation biology and will be an important asset in future anti-inflammatory drug design
Location, location, location: modelling of noise mitigation by urban woodland shows the benefit of targeted tree planting in cities
Noise pollution from road traffic is ubiquitous in modern cities and is the second greatest environmental risk to health in Western Europe. Urban woodland can provide substantial noise mitigation if located properly, yet such considerations are often absent from the urban planning process. Current approaches for quantifying this important ecosystem service (ES) do not account adequately for important spatial factors and are unable to identify effectively the best locations to place new woodland for noise mitigation. We present new methods, in which we exploit the concept of least-cost-distance, to map and value the mitigating effect of urban woodland, and to identify optimal locations to place new woodland. Applying these methods, we show that urban woodland currently provides Birmingham City (UK) with over GBP 3.8 million in noise mitigation benefits, annually. We also show that our new ‘opportunity’ mapping methods effectively identify the best locations for new woodland, achieving close to a maximum service with less than a quarter of the additional woodland needed to achieve it. This has important implications for the design and implementation of urban tree planting for noise mitigation, and these methods can be adapted for other ES, allowing consideration of multiple service outcomes
Focusing Cosmic Telescopes: Exploring Redshift z~5-6 Galaxies with the Bullet Cluster 1E0657-56
The gravitational potential of clusters of galaxies acts as a cosmic
telescope allowing us to find and study galaxies at fainter limits than
otherwise possible and thus probe closer to the epoch of formation of the first
galaxies. We use the Bullet Cluster 1E0657-56 (z = 0.296) as a case study,
because its high mass and merging configuration makes it one of the most
efficient cosmic telescopes we know. We develop a new algorithm to reconstruct
the gravitational potential of the Bullet Cluster, based on a non-uniform
adaptive grid, combining strong and weak gravitational lensing data derived
from deep HST/ACS F606W-F775W-F850LP and ground-based imaging. We exploit this
improved mass map to study z~5-6 Lyman Break Galaxies (LBGs), which we detect
as dropouts. One of the LBGs is multiply imaged, providing a geometric
confirmation of its high redshift, and is used to further improve our mass
model. We quantify the uncertainties in the magnification map reconstruction in
the intrinsic source luminosity, and in the volume surveyed, and show that they
are negligible compared to sample variance when determining the luminosity
function of high-redshift galaxies. With shallower and comparable magnitude
limits to HUDF and GOODS, the Bullet cluster observations, after correcting for
magnification, probe deeper into the luminosity function of the high redshift
galaxies than GOODS and only slightly shallower than HUDF. We conclude that
accurately focused cosmic telescopes are the most efficient way to sample the
bright end of the luminosity function of high redshift galaxies and - in case

The social value of a QALY : raising the bar or barring the raise?
Background: Since the inception of the National Institute for Health and Clinical Excellence (NICE) in England,
there have been questions about the empirical basis for the cost-per-QALY threshold used by NICE and whether
QALYs gained by different beneficiaries of health care should be weighted equally. The Social Value of a QALY
(SVQ) project, reported in this paper, was commissioned to address these two questions. The results of SVQ were
released during a time of considerable debate about the NICE threshold, and authors with differing perspectives
have drawn on the SVQ results to support their cases. As these discussions continue, and given the selective use of
results by those involved, it is important, therefore, not only to present a summary overview of SVQ, but also for
those who conducted the research to contribute to the debate as to its implications for NICE.
Discussion: The issue of the threshold was addressed in two ways: first, by combining, via a set of models, the
current UK Value of a Prevented Fatality (used in transport policy) with data on fatality age, life expectancy and
age-related quality of life; and, second, via a survey designed to test the feasibility of combining respondents’
answers to willingness to pay and health state utility questions to arrive at values of a QALY. Modelling resulted in
values of £10,000-£70,000 per QALY. Via survey research, most methods of aggregating the data resulted in values
of a QALY of £18,000-£40,000, although others resulted in implausibly high values. An additional survey, addressing
the issue of weighting QALYs, used two methods, one indicating that QALYs should not be weighted and the
other that greater weight could be given to QALYs gained by some groups.
Summary: Although we conducted only a feasibility study and a modelling exercise, neither present compelling
evidence for moving the NICE threshold up or down. Some preliminary evidence would indicate it could be
moved up for some types of QALY and down for others. While many members of the public appear to be open to
the possibility of using somewhat different QALY weights for different groups of beneficiaries, we do not yet have
any secure evidence base for introducing such a system
Backyard poultry cases in UK small animal practices: Demographics, health conditions and pharmaceutical prescriptions
Background: Backyard poultry ownership is of keen interest in the United Kingdom. However, despite this, little is known about veterinary care engagement and outcomes of visits in this group of species. Methods: This study described and characterised veterinary practice‐visiting backyard poultry, utilising electronic health record data supplied by veterinary practices voluntarily participating in the Small Animal Veterinary Surveillance Network between 1st April 2014 and 31st March 2019. Results: In total, 4424 recorded poultry consultations originating from 197 veterinary practices (352 sites) were summarised. Chicken consultation (n = 3740) peak incidence was in early summer (April‐June), relative to all recorded species. More chickens resided in rural (incident rate ratio = 2.5, confidence interval [CI] 2.3–2.6, p <0.001) or less deprived areas. Non‐specific clinical signs were commonly recorded (17.6% of chicken consultations, CI 15.9–19.2), as were those indicative of advanced disease. This latter finding was reflected in prescribed management strategies, with euthanasia comprising 29.8% (CI 27.0–32.6) of consultations. Antimicrobials were commonly prescribed (33.0% of consultations, CI 29.8–36.2), 43.8% of which included antimicrobials considered ‘highest priority critically important’ by the World Health Organisation. Conclusion: Our findings indicate a need to tailor antimicrobial prescription guidance to the backyard poultry setting. In addition, late presentation of disease, vague clinical descriptions in clinical narratives and high euthanasia rates show that disease identification, management and knowledge of poultry health and welfare among owners and veterinary surgeons can be improved
Genetic validation of bipolar disorder identified by automated phenotyping using electronic health records
Bipolar disorder (BD) is a heritable mood disorder characterized by episodes of mania and depression. Although genomewide association studies (GWAS) have successfully identified genetic loci contributing to BD risk, sample size has become a rate-limiting obstacle to genetic discovery. Electronic health records (EHRs) represent a vast but relatively untapped resource for high-throughput phenotyping. As part of the International Cohort Collection for Bipolar Disorder (ICCBD), we previously validated automated EHR-based phenotyping algorithms for BD against in-person diagnostic interviews (Castro et al. Am J Psychiatry 172:363–372, 2015). Here, we establish the genetic validity of these phenotypes by determining their genetic correlation with traditionally ascertained samples. Case and control algorithms were derived from structured and narrative text in the Partners Healthcare system comprising more than 4.6 million patients over 20 years. Genomewide genotype data for 3330 BD cases and 3952 controls of European ancestry were used to estimate SNP-based heritability (h2g) and genetic correlation (rg) between EHR-based phenotype definitions and traditionally ascertained BD cases in GWAS by the ICCBD and Psychiatric Genomics Consortium (PGC) using LD score regression. We evaluated BD cases identified using 4 EHR-based algorithms: an NLP-based algorithm (95-NLP) and three rule-based algorithms using codified EHR with decreasing levels of stringency—“coded-strict”, “coded-broad”, and “coded-broad based on a single clinical encounter” (coded-broad-SV). The analytic sample comprised 862 95-NLP, 1968 coded-strict, 2581 coded-broad, 408 coded-broad-SV BD cases, and 3 952 controls. The estimated h2g were 0.24 (p = 0.015), 0.09 (p = 0.064), 0.13 (p = 0.003), 0.00 (p = 0.591) for 95-NLP, coded-strict, coded-broad and coded-broad-SV BD, respectively. The h2g for all EHR-based cases combined except coded-broad-SV (excluded due to 0 h2g) was 0.12 (p = 0.004). These h2g were lower or similar to the h2g observed by the ICCBD + PGCBD (0.23, p = 3.17E−80, total N = 33,181). However, the rg between ICCBD + PGCBD and the EHR-based cases were high for 95-NLP (0.66, p = 3.69 × 10–5), coded-strict (1.00, p = 2.40 × 10−4), and coded-broad (0.74, p = 8.11 × 10–7). The rg between EHR-based BD definitions ranged from 0.90 to 0.98. These results provide the first genetic validation of automated EHR-based phenotyping for BD and suggest that this approach identifies cases that are highly genetically correlated with those ascertained through conventional methods. High throughput phenotyping using the large data resources available in EHRs represents a viable method for accelerating psychiatric genetic research
