Amyloid precursor-like protein 2 (APLP2) affects the actin cytoskeleton and increases pancreatic cancer growth and metastasis.
Amyloid precursor-like protein 2 (APLP2) is aberrantly expressed in pancreatic cancer. Here we showed that APLP2 is increased in pancreatic cancer metastases, particularly in metastatic lesions found in the diaphragm and intestine. Examination of matched human primary tumor-liver metastasis pairs showed that 38.1% of the patients had positive APLP2 expression in both the primary tumor and the corresponding liver metastasis. Stable knock-down of APLP2 expression (with inducible shRNA) in pancreatic cancer cells reduced the ability of these cells to migrate and invade. Loss of APLP2 decreased cortical actin and increased intracellular actin filaments in pancreatic cancer cells. Down-regulation of APLP2 decreased the weight and metastasis of orthotopically transplanted pancreatic tumors in nude mice
Use of different RT-QuIC substrates for detecting CWD prions in the brain of Norwegian cervids
Chronic wasting disease (CWD) is a highly contagious prion disease affecting captive and free-ranging cervid populations. CWD has been detected in United States, Canada, South Korea and, most recently, in Europe (Norway, Finland and Sweden). Animals with CWD release infectious prions in the environment through saliva, urine and feces sustaining disease spreading between cervids but also potentially to other non-cervids ruminants (e.g. sheep, goats and cattle). In the light of these considerations and due to CWD unknown zoonotic potential, it is of utmost importance to follow specific surveillance programs useful to minimize disease spreading and transmission. The European community has already in place specific surveillance measures, but the traditional diagnostic tests performed on nervous or lymphoid tissues lack sensitivity. We have optimized a Real-Time Quaking-Induced Conversion (RT-QuIC) assay for detecting CWD prions with high sensitivity and specificity to try to overcome this problem. In this work, we show that bank vole prion protein (PrP) is an excellent substrate for RT-QuIC reactions, enabling the detection of trace-amounts of CWD prions, regardless of prion strain and cervid species. Beside supporting the traditional diagnostic tests, this technology could be exploited for detecting prions in peripheral tissues from live animals, possibly even at preclinical stages of the disease
Taking the strain? Impact of glaucoma on patients' informal caregivers
Purpose: To estimate informal caregiver (ICG) strain in people from a glaucoma clinic.
Methods: Patients with glaucoma were consecutively identified from a single clinic in England for a cross-sectional postal survey. The sample was deliberately enriched with a number of patients designated as having advanced glaucoma (visual field [VF] mean deviation worse than -12 dB in both eyes). Patients were asked to identify an ICG who recorded a Modified Caregiver Strain Index (MCSI), a validated 13 item instrument scored on a scale of 0-26. Previous research has indicated mean MCSI to be >10 in Multiple Sclerosis and Parkinson’s disease. All participants gave a self-reported measure of general health (EQ5D).
Results: Responses from 105 patients (43% of those invited) were analysed; only 38 of the 105 named an ICG. Mean (95% confidence interval [CI]) MCSI was 2.4 (1.3, 3.6) and only three ICGs recorded a MCSI > 7. The percentage of patients with an ICG was much higher in patients with advanced VF loss (82%; 9/11) when compared to those with non-advanced VF loss (31%; 29/94; p=0.001). Mean (standard deviation) MCSI was considerably inflated in the advanced patients (5.6 [4.9] vs 1.5 [2.2] for non-advanced; p=0.040). Worsening VF and poorer self-reported general health (EQ5D) of the patient were associated with worsening MCSI.
Conclusion: ICG strain, as measured by MCSI, for patients with non-advanced glaucoma is negligible, compared to other chronic disease. ICG strain increases moderately with worsening VFs but this could be partly explained by worse general health in our sample of patients
Precise measurement of the top quark mass in the dilepton channel at D0
We measure the top quark mass (mt) in ppbar collisions at a center of mass
energy of 1.96 TeV using dilepton ttbar->W+bW-bbar->l+nubl-nubarbbar events,
where l denotes an electron, a muon, or a tau that decays leptonically. The
data correspond to an integrated luminosity of 5.4 fb-1 collected with the D0
detector at the Fermilab Tevatron Collider. We obtain mt = 174.0 +- 1.8(stat)
+- 2.4(syst) GeV, which is in agreement with the current world average mt =
173.3 +- 1.1 GeV. This is currently the most precise measurement of mt in the
dilepton channel.Comment: 7 pages, 4 figure

We present the first measurements of the differential cross section d sigma/dp(T)(gamma) for the production of an isolated photon in association with at least two b-quark jets. The measurements consider photons with rapidities vertical bar y(gamma)vertical bar < 1.0 and transverse momenta 30 < p(T)(gamma) < 200 GeV. The b-quark jets are required to have p(T)(jet) > 15 GeVand vertical bar y(jet)vertical bar < 1.5. The ratio of differential production cross sections for gamma + 2 b-jets to gamma + b-jet as a function of p(T)(gamma) is also presented. The results are based on the proton-antiproton collision data at root s = 1.96 TeV collected with the D0 detector at the Fermilab Tevatron Collider. The measured cross sections and their ratios are compared to the next- to- leading order perturbative QCD calculations as well as predictions based on the k(T)- factorization approach and those from the sherpa and pythia Monte Carlo event generators
Direct measurement of the mass difference between top and antitop quarks
We present a direct measurement of the mass difference between top and
antitop quarks (dm) in lepton+jets top-antitop final states using the "matrix
element" method. The purity of the lepton+jets sample is enhanced for
top-antitop events by identifying at least one of the jet as originating from a
b quark. The analyzed data correspond to 3.6 fb-1 of proton-antiproton
collisions at 1.96 TeV acquired by D0 in Run II of the Fermilab Tevatron
Collider. The combination of the e+jets and mu+jets channels yields dm = 0.8
+/- 1.8 (stat) +/- 0.5 (syst) GeV, which is in agreement with the standard
model expectation of no mass difference.Comment: submitted to Phys. Rev.
Measurements of differential cross sections of Z/gamma*+jets+X events in proton anti-proton collisions at sqrt{s}=1.96 TeV
We present cross section measurements for Z/gamma*+jets+X production,
differential in the transverse momenta of the three leading jets. The data
sample was collected with the D0 detector at the Fermilab Tevatron proton
anti-proton collider at a center-of-mass energy of 1.96 TeV and corresponds to
an integrated luminosity of 1 fb-1. Leading and next-to-leading order
perturbative QCD predictions are compared with the measurements, and agreement
is found within the theoretical and experimental uncertainties. We also make
comparisons with the predictions of four event generators. Two
parton-shower-based generators show significant shape and normalization
differences with respect to the data. In contrast, two generators combining
tree-level matrix elements with a parton shower give a reasonable description
of the the shapes observed in data, but the predicted normalizations show
significant differences with respect to the data, reflecting large scale
uncertainties. For specific choices of scales, the normalizations for either
generator can be made to agree with the measurements.Comment: Published in PLB. 11 pages, 3 figure
Search for Decay
We have searched for the charmless hadronic decay of B0 mesons into two
neutral pions. Using 9.13fb^-1 taken at the Upsilon(4S) with the CLEO detector,
we obtain an improved upper limit for the branching fraction BR(B0-->pi0pi0) <
5.7*10^-6 at the 90% confidence level.Comment: pages postscript, also available through

Search for single vector-like quarks in ppbar collisions at sqrt(s) = 1.96 TeV
We present a search for hypothetical vector-like quarks in ppbar collisions
at sqrt(s) = 1.96 TeV. The data were collected by the D0 detector at the
Fermilab Tevatron Collider and correspond to an integrated luminosity of 5.4
fb^(-1). We select events with a final state composed of a W or Z boson and a
jet consistent with a heavy object decay. We observe no significant excess in
comparison to the background prediction and set limits on production cross
sections for vector-like quarks decaying to W+jet and Z+jet. These are the most
stringent mass limits for electroweak single vector-like quark production at
hadron colliders.Comment: submitted to Phys. Rev. Let
Measurements of single top quark production cross sections and |Vtb| in ppbar collisions at sqrt{s}=1.96 TeV
We present measurements of production cross sections of single top quarks in
\ppbar collisions at in a data sample corresponding
to an integrated luminosity of collected by the D0 detector
at the Fermilab Tevatron Collider. We select events with an isolated electron
or muon, an imbalance in transverse energy, and two, three, or four jets, with
one or two of them containing a bottom hadron. We obtain an inclusive cross
section of \sigma({\ppbar}{\rargap}tb+X, tqb+X) = 3.43\pm^{0.73}_{0.74}\;\rm
pb and use it to extract the CKM matrix element at
the 95% C.L. We also measure \sigma({\ppbar}{\rargap}tb+X) =
0.68\pm^{0.38}_{0.35}\;\rm pb and \sigma({\ppbar}{\rargap}tqb+X) =
2.86\pm^{0.69}_{0.63}\;\rm pb when assuming, respectively, and
production rates as predicted by the standard model.Comment: 11 pages, 8 figures, submitted to Phys. Rev.
