    Importance of Estrus Expression Before Fixed-time AIon Conception Rates in Beef Cattle

    Expression of estrus prior to fixed-time AI has been reported to strongly impact overall pregnancy success. Behavioral estrus is a visual indicator that a cow or heifer’s internal environment is prepared for breeding. Insemination of a cow or heifer after estrus has been expressed will yield greater pregnancy success due to adequate uterine environment, increased fertilization rates, increased accessory sperm numbers, and increased overall embryo survival. It can be difficult to analyze the effects of estrus on pregnancy success across studies due to differences in number of animals and proportion of animals exhibiting estrus per study. In order to accurately analyze such data, a meta-analysis was used to place all studies on an equal level, thus, eliminating study bias. In the present study, a meta-analysis was conducted using data available on 10,116 beef cows and heifers in 26 studies that utilized the 5 most common fixed-time AI protocols to examine the effect of expression of estrus prior to insemination on conception rates. The overall model indicated a positive effect of estrus on conception rates with cows expressing estrus before fixed-time AI having greater conception rates compared with those not exhibiting estrus. There are also numerous management factors that can influence expression of estrus. Data were available on 547 cows that were synchronized with a CIDR based fixed-time AI protocol for estrus for 2 to 4 years. Analysis of these cows indicated that days postpartum did not impact estrus expression. In contrast, Body Condition Score (BCS) influenced estrus expression with cows in a BCS of ≤ 4 having decreased expression of estrus compared to those with a BCS \u3e 4. Initiation of estrous cycles before the breeding season also influenced estrus expression, with anestrous cows having greater expression of estrus compared with estrus-cycling cows. Fixed-time AI protocols offer producers the added benefit of reduced labor needed for heat detection, but the results of this study indicate the importance of detecting an animal in estrus prior to breeding. In conclusion, among all currently recommended fixed-time AI protocols, cows expressing estrus before fixed-time AI had improved conception rates, and BCS and estrus-cycling status had the greatest influence on expression of estrus

    Mucosal biomarkers of colorectal cancer risk do not increase at 6 months following sleeve gastrectomy, unlike gastric bypass

    YesObjective The hypothesis that sleeve gastrectomy (SG) is not associated with an increase in mucosal colorectal cancer (CRC) biomarkers, unlike Roux-en-Y gastric bypass (RYGB), was tested. Design and Methods Rectal mucosa, blood, and urine were obtained from morbidly obese patients (n = 23) before and after (median 28 months) SG, as well as from nonobese controls (n = 20). Rectal epithelial cell mitosis and apoptosis, crypt size/fission, and pro-inflammatory gene expression were measured, as well as systemic inflammatory biomarkers, including C-reactive protein (CRP). Results The mean pre-operative body mass index in SG patients was 65.7 kg/m2 (24.7 kg/m2 in controls). Mean excess weight loss post-SG was 38.2%. There was a significant increase in mitosis frequency, crypt size, and crypt fission (all P < 0.01) in SG patients versus controls, as well as evidence of a chronic inflammatory state (raised CRP and mononuclear cell p65 NFκB binding), but there was no significant change in these biomarkers after SG, except CRP reduction. Macrophage migration inhibitory factor mRNA levels were increased by 39% post-SG (P = 0.038). Conclusions Mucosal biomarkers of CRC risk do not increase at 6 months following SG, unlike RYGB.     Comparison of Three CIDR Based Fixed-time AI Protocols for Beef Heifers

    Several effective fixed-time AI (FTAI) protocols have been developed to facilitate AI while eliminating the need for estrus detection. Among these are the 5-d CO-Synch+CIDR (5d), PG 6-d CIDR (PG-CIDR), and 14-d CIDR-PG (CIDR-PG) protocols. While each of these protocols varies in duration and approach to synchronizing estrus and ovulation, each has been reported as an effective method to facilitate FTAI in beef heifers. Therefore, the objective of this study was to compare FTAI pregnancy rates in beef heifers synchronized with these three CIDR based protocols. Virgin beef heifers (n = 801) at four locations were synchronized with one of three protocols: 1) (5-day CO-Synch + CIDR) an injection of GnRH (100 μg; i.m.) and insertion of a CIDR on d -5, PG (25 mg; i.m.) and CIDR removal on d 0 with a second injection of PG (\u3e4 h after CIDR removal) on d 0 and FTAI at 72 h after CIDR removal, 2) (PG 6-day CIDR) PG (25 mg; i.m.) on d -9, GnRH (100 μg; i.m.) and insertion of a CIDR on d -6, PG and CIDR removal on d 0, and FTAI at 66 h after CIDR removal, or 3) (14-day CIDR-PG) a 14-day CIDR insert from d -30 to -16, PG (25 mg; i.m.) on d 0, and FTAI at 66 h after PG. All heifers received an injection of GnRH (100 μg; i.m.) concurrent with FTAI. Timing of treatment initiation was offset to allow all heifers to receive FTAI concomitantly and at random. Pregnancy success was determined between 35 and 40 d after FTAI by transrectal ultrasonography. Blood samples were collected approximately 12 d before the beginning of each protocol and at the initiation of each protocol to determine estrous cycling status (77%). Data were analyzed using the GLIMMIX procedures of SAS. Fixed-time AI pregnancy success did not differ between treatments (P = 0.13; 62.5%, 56.9%, and 53.3%, for 5-day CO-Synch + CIDR, PG 6-day CIDR, and 14-day CIDR-PG; respectively) or location (P = 0.16; 51.5%, 62.7%, 56.1%, and 58.6% for location 1, 2, 3, and 4; respectively). However, heifers that had reached puberty by initiation of synchronization had greater (P \u3c 0.01) pregnancy success compared to heifers that were prepubertal (60.7% and 47.3%; respectively). In summary, all three protocols had similar FTAI pregnancy success, and puberty status had the greatest impact on pregnancy success

    Cilengitide in newly diagnosed glioblastoma: biomarker expression and outcome.

    Integrins αvβ3 and αvβ5 regulate angiogenesis and invasiveness in cancer, potentially by modulating activation of the transforming growth factor (TGF)-β pathway. The randomized phase III CENTRIC and phase II CORE trials explored the integrin inhibitor cilengitide in patients with newly diagnosed glioblastoma with versus without O6-methylguanine DNA methyltransferase (MGMT) promoter methylation. These trials failed to meet their primary endpoints.Immunohistochemistry was used to assess the levels of the target integrins of cilengitide, αvβ3 and αvβ5 integrins, of αvβ8 and of their putative target, phosphorylation of SMAD2, in tumor tissues from CENTRIC (n=274) and CORE (n=224).αvβ3 and αvβ5 expression correlated well in tumor and endothelial cells, but showed little association with αvβ8 or pSMAD2 levels. In CENTRIC, there was no interaction between the biomarkers and treatment for prediction of outcome. In CORE, higher αvβ3 levels in tumor cells were associated with improved progression-free survival by central review and with improved overall survival in patients treated with cilengitide.Integrins αvβ3, αvβ5 and αvβ8 are differentially expressed in glioblastoma. Integrin levels do not correlate with the activation level of the canonical TGF-β pathway. αvβ3 integrin expression may predict benefit from integrin inhibition in patients with glioblastoma lacking MGMT promoter methylation

    Crystal structure, electronic, and magnetic properties of the bilayered rhodium oxide Sr3Rh2O7

    The bilayered rhodium oxide Sr3Rh2O7 was synthesized by high-pressure and high-temperature heating techniques. The single-phase polycrystalline sample of Sr3Rh2O7 was characterized by measurements of magnetic susceptibility, electrical resistivity, specific heat, and thermopower. The structural characteristics were investigated by powder neutron diffraction study. The rhodium oxide Sr3Rh2O7 [Bbcb, a = 5.4744(8) A, b = 5.4716(9) A, c = 20.875(2) A] is isostructural to the metamagnetic metal Sr3Ru2O7, with five 4d electrons per Rh, which is electronically equivalent to the hypothetic bilayered ruthenium oxide, where one electron per Ru is doped into the Ru-327 unit. The present data show the rhodium oxide Sr3Rh2O7 to be metallic with enhanced paramagnetism, similar to Sr3Ru2O7. However, neither manifest contributions from spin fluctuations nor any traces of a metamagnetic transition were found within the studied range from 2 K to 390 K below 70 kOe.Comment: To be published in PR

    Geographical and climatic limits of needle types of one- and two-needled pinyon pines

    Aim The geographical extent and climatic tolerances of one- and two-needled pinyon pines (Pinus subsect. Cembroides) are the focus of questions in taxonomy, palaeoclimatology and modelling of future distributions. The identification of these pines, traditionally classified by one- versus two-needled fascicles, is complicated by populations with both one- and two-needled fascicles on the same tree, and the description of two more recently described one-needled varieties: the fallax-type and californiarum-type. Because previous studies have suggested correlations between needle anatomy and climate, including anatomical plasticity reflecting annual precipitation, we approached this study at the level of the anatomy of individual pine needles rather than species. Location Western North America. Methods We synthesized available and new data from field and herbarium collections of needles to compile maps of their current distributions across western North America. Annual frequencies of needle types were compared with local precipitation histories for some stands. Historical North American climates were modeled on a c. 1-km grid using monthly temperature and precipitation values. A geospatial model (ClimLim), which analyses the effect of climate modulated physiological and ecosystem processes, was used to rank the importance of seasonal climate variables in limiting the distributions of anatomical needle types. Results The pinyon needles were classified into four distinct types based upon the number of needles per fascicle, needle thickness and the number of stomatal rows and resin canals. The individual needles fit well into four categories of needle types, whereas some trees exhibit a mixture of two needle types. Trees from central Arizona containing a mixture of Pinus edulis and fallax-type needles increased their percentage of fallax-type needles following dry years. All four needle types occupy broader geographical regions with distinctive precipitation regimes. Pinus monophylla and californiarum-type needles occur in regions with high winter precipitation. Pinus edulis and fallax-type needles are found in regions with high monsoon precipitation. Areas supporting californiarum-type and fallax-type needle distributions are additionally characterized by a more extreme May–June drought. Main conclusions These pinyon needle types seem to reflect the amount and seasonality of precipitation. The single needle fascicle characterizing the fallax type may be an adaptation to early summer or periodic drought, while the single needle of Pinus monophylla may be an adaptation to summer–autumn drought. Although the needles fit into four distinct categories, the parent trees are sometimes less easily classified, especially near their ancestral Pleistocene ranges in the Mojave and northern Sonoran deserts. The abundance of trees with both one- and two-needled fascicles in the zones between P. monophylla, P. edulis and fallax-type populations suggest that needle fascicle number is an unreliable characteristic for species classification. Disregarding needle fascicle number, the fallax-type needles are nearly identical to P. edulis, supporting Little’s (1968) initial classification of these trees as P. edulis var. fallax, while the californiarum-type needles have a distinctive morphology supporting Bailey’s (1987) classification of this tree as Pinus californiarum

    A deep learning system accurately classifies primary and metastatic cancers using passenger mutation patterns.

    In cancer, the primary tumour's organ of origin and histopathology are the strongest determinants of its clinical behaviour, but in 3% of cases a patient presents with a metastatic tumour and no obvious primary. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, we train a deep learning classifier to predict cancer type based on patterns of somatic passenger mutations detected in whole genome sequencing (WGS) of 2606 tumours representing 24 common cancer types produced by the PCAWG Consortium. Our classifier achieves an accuracy of 91% on held-out tumor samples and 88% and 83% respectively on independent primary and metastatic samples, roughly double the accuracy of trained pathologists when presented with a metastatic tumour without knowledge of the primary. Surprisingly, adding information on driver mutations reduced accuracy. Our results have clinical applicability, underscore how patterns of somatic passenger mutations encode the state of the cell of origin, and can inform future strategies to detect the source of circulating tumour DNA
