    Diacylglycerol triggers Rim101 pathway dependent necrosis in yeast: a model for lipotoxicity

    The loss of lipid homeostasis can lead to lipid overload and is associated with a variety of disease states. However, little is known as to how the disruption of lipid regulation or lipid overload affects cell survival. In this study we investigated how excess diacylglycerol (DG), a cardinal metabolite suspected to mediate lipotoxicity, compromises the survival of yeast cells. We reveal that increased DG achieved by either genetic manipulation or pharmacological administration of 1,2-dioctanoyl-sn-glycerol (DOG) triggers necrotic cell death. The toxic effects of DG are linked to glucose metabolism and require a functional Rim101 signaling cascade involving the Rim21 dependent sensing complex and activation of a calpain-like protease. The Rim101 cascade is an established pathway that triggers a transcriptional response to alkaline or lipid stress. We propose that the Rim101 pathway senses DG-induced lipid perturbation and conducts a signaling response that either facilitates cellular adaptation or triggers lipotoxic cell death. Using established models of lipotoxicity i.e. high fat diet in Drosophila and palmitic acid administration in cultured human endothelial cells, we present evidence that the core mechanism underlying this calpain-dependent lipotoxic cell death pathway is phylogenetically conserved

    Segmentation of the ECG Signal by Means of a Linear Regression Algorithm

    The monitoring and processing of electrocardiogram (ECG) beats have been actively studied in recent years: new lines of research have even been developed to analyze ECG signals using mobile devices. Considering these trends, we proposed a simple and low computing cost algorithm to process and analyze an ECG signal. Our approach is based on the use of linear regression to segment the signal, with the goal of detecting the R point of the ECG wave and later, to separate the signal in periods for detecting P, Q, S, and T peaks. After pre-processing of ECG signal to reduce the noise, the algorithm was able to efficiently detect fiducial points, information that is transcendental for diagnosis of heart conditions using machine learning classifiers. When tested on 260 ECG records, the detection approach performed with a Sensitivity of 97.5% for Q-point and 100% for the rest of ECG peaks. Finally, we validated the robustness of our algorithm by developing an ECG sensor to register and transmit the acquired signals to a mobile device in real time

    Inter-regional variation on leaf surface defenses in native and non-native Centaurea solstitialis plants

    Artículo de publicación ISIPlant chemical defenses can be qualitative (toxins) to face generalist herbivores and quantitative (digestibility reducers) to specialists. Trichomes can produce chemicals, but also acts as a quantitative defense and in water loss. The shifting defense hypothesis (SDH) poses that invasive plants reduce the production of qualitative defenses against specialist herbivores because those are frequently absent in invaded regions, while increasing defenses against generalist herbivores. Chemical and physical leaf surface defensive traits could give information about direct plant-herbivore interaction in native and non-native regions. We studied leaf surface morphology and epicuticular chemistry of the invasive C. solstitialis in plants from native and non-native regions. Across regions, the main chemical components were sesquiterpene lactones, similar densities of sessile glandular trichomes and a variable number of large multicellular trichomes. Exotic plants face different sets of challenges in the new regions. In accordance with SDH, plants from non-native regions presented higher total sesquiterpene lactones concentrations (qualitative defense). Trichome density was possibly associated to differences in local climatic conditions. Sesquiterpene lactones are likely to play an important role on the invasion success of Centaurea. Leaf surface components provide meaningful information that should be consider in future studies to unravel the mechanisms involved in plant invasions. (C) 2015 Elsevier Ltd.     Inhibition of the NorA efflux pump of Staphylococcus aureus by synthetic riparins

    Aim: The goal of this study was to increase knowledge about the antimicrobial activity of some synthetic Riparin-derived compounds, alone or in combination with fluoroquinolone antibiotics, against a strain of Staphylococcus aureus resistant to fluoroquinolone by way of overexpression of the NorA efflux pump. Methods and Results: Microdilution tests showed that Riparins A and B did not show any significant antibacterial activity against Staph. aureus strains. On the other hand, the intrinsic antibacterial activity increased with increasing lipophilicity of the compounds, in the following order: Riparin-D (MIC 256 μg ml−1; Log P 2·95) < Riparin-C (MIC 102 μg ml−1; Log P 3·22) < Riparin-E (MIC 16 μg ml−1; Log P 3·57). The addition of all riparins to growth media at subinhibitory concentrations caused an increase in the antibacterial activity of antibiotics against the NorA-overexpressing test strain. Riparin-B, which has two methoxyl groups at the phenethyl moiety, showed the best modulatory effect. Conclusions: Riparin-E is a good anti-staphylococci agent, while Riparin-B functions as a NorA efflux pump inhibitor. Significance and Impact of the Study: Our data suggest the possibility of using Riparin-B in combination with norfloxacin or ciprofloxacin for therapy of infections caused by multi-drug resistant Staph. aureus.Fil: Costa, L.
