    Electrochemical characterisation of Formate dehydrogenase and its catalytic properties

    In recent years the amount of CO2 release into the planet atmosphere has increase exponentially due to a global dependency in fossil fuels. This is one of the major contributors to the climate change that is seen nowadays. On the other hand, due to the CO2 abundance, there has been an interest in developing methods to harvest and use this CO2 in the production of green energy and sustainable chemistry. Formate dehydrogenase (FDH) proteins are a class of metalloenzymes with different subunit composition containing either molybdenum or tungsten at the active site. FDH catalyses the oxidation of formate into CO2. Recently it was shown that some of the FDH enzymes have the ability to perform the reverse reaction that is, these can catalyse the reversible interconversion of CO2 and formate. However, to date, the electrochemical characterisation of the protein has not yet been attained using direct, non-mediated methods. In this thesis the isolation of FDH from Desulfovibrio desulfuricans, its biochemical and non-mediated electrochemical characterisation was attained. The redox features of the Mo catalytic centre of the enzyme, including thermodynamic and kinetic parameters such as its formal reduction potential (E0’ = -245 ± 8 mV vs NHE) and heterogeneous electron transfer constant, were determined, for the first time, using direct electrochemical methods. The catalytic activity towards the CO2 reduction was also observed, for the first time, by direct electrochemical techniques

    Este proyecto de investigación se justifica teniendo en cuenta los siguientes parámetros para desarrollar un proyecto de grado: La relación entre la generación de conocimiento y el proyecto a desarrollar, la innovación social y la relación directa que hay entre nuestro trabajo, la televisión educativa como estrategia audiovisual y la licenciatura en comunicación e informática. La televisión educativa le apuesta a fortalecer los procesos de enseñanza/aprendizaje en el aula, ya que en nuestra actualidad, las opciones para poder aprovechar lo audiovisual en la educación, son mucho más asequibles. Dado que la educación actual de nuestra región, cuenta con las estrategias pedagógicas de los docentes asignados, estrategias con las cuales los estudiantes van construyendo su sendero de conocimiento, la televisión educativa surge como un apoyo a esos procesos pedagógicos, para fortalecer y brindar al estudiante una educación que aprovecha los medios de comunicación y su influencia en la sociedad

    Relatório de estágio de mestrado, Tradução, Universidade de Lisboa, Faculdade de Letras, 2012Língua de Especialidade e Tradução Técnica na Onoma consiste num relatório de estágio de dois semestres, realizado, no ano lectivo 2010-11, na Onoma, num prestigiado gabinete de Tradução, sediado em Lisboa. Este relatório visa um estudo dos originais ingleses e das respectivas traduções para Português Europeu, tomando como instrumento de aferição os parâmetros textuais presentes em diferentes tipos de texto no quadro da Tradução Técnica, mais concretamente em textos do domínio das Ciências Médicas, do Direito, da Engenharia, nos subramos da Engenharia Civil, Mecânica, Informática e Electrotécnica, bem como do domínio do discurso empresarial. Na base da descrição das especificidades dos diferentes tipos de texto de cada um dos domínios, que motivam soluções de tradução diversas, ilustradas por excertos das traduções realizadas, foi identificado um conjunto de diferenças na estruturação textual dos vários domínios da tradução técnica. Tendo em vista a compilação dos acervos terminógicos usados durante o estágio, bem como a identificação das ferramentas usadas, foram elaborados quatro glossários terminológicos bilingues Inglês-Português, relativos a estas quatro áreas de Tradução, em que cada um dos termos deles constantes é classificado por recurso a parâmetros da classificação [Resolução interna] ou [Resolução externa] que permitem evidenciar isoladamente, quer no seu conjunto, as ferramentas usadas pelo tradutor, não só na realização da tradução de cada texto, mas também, no cômputo geral, no conjunto dos textos traduzidos, no decurso do estágio curricular.Abstract: Língua da Especialidade e Tradução Técnica na Onoma consists of a two-semester translation Internship Report at Onoma, a prestigious translation company in Lisbon, in the time span of 2010-2011. The present report aims at characterizing English source texts and their translations to European Portuguese, taking to an array of textual parameters to be found in technical texts in the domains of Medical Sciences, Law , Engineering (namely Civil, Mechanical, Electrical and Computer Engineering), as well as in the domain of Enterprise discourse. The identification of text type specificities from each of the above-mentioned technical domains, necessarily illustrated with our translation solutions, led to the grounding of the diverse translation solutions in the different text domains. Hence, four bilingual glossaries of technical terminology (English-Portuguese) in the four technical translation domains were elaborated, resorting to the classification parameters [Internal Resolution] or [External Resolution], respectively, in reference to translation solutions provided by the translator herself or obtained from external sources, aiming both at highlighting the importance of the different translation tools in the different text domains, as well as at emphasizing the trainee´s progression in course of the internship period

    Metabolomics is a study through can be obtain a better understanding of the complexity of the biological systems, through the chemical composition and relations with the physiology of the plant. The literature describes a lot of information on this support in areas of medicinal plants, chemosystematics, adulteration of plants, etc., but it is scarce in agriculture. At present, agriculture plays a crucial role in human beings. The demand of foods has increased due to the continuous increase in the population, and this requires an increase in the production of crops, besides the crops being affected by the climatic change, attack of pest and diseases and resistance to a conventional agrochemicals. At present, scientists are doing some practices or studies of genetic improvement of crops to increase their production and avoid the problems pointed out. It is an important part of the genomic studies; the results could be the basis for a genetic improvement based on the chemical composition of the crops, and in the metabolomics studies represent a crucial role in their quality for human consumption.     Interploidy Hybridization In Sympatric Zones: The Formation Of Epidendrum Fulgens × E. Puniceoluteum Hybrids (epidendroideae, Orchidaceae).

    Interspecific hybridization is a primary cause of extensive morphological and chromosomal variation and plays an important role in plant species diversification. However, the role of interploidal hybridization in the formation of hybrid swarms is less clear. Epidendrum encompasses wide variation in chromosome number and lacks strong premating barriers, making the genus a good model for clarifying the role of chromosomes in postzygotic barriers in interploidal hybrids. In this sense, hybrids from the interploidal sympatric zone between E. fulgens (2n = 2x = 24) and E. puniceoluteum (2n = 4x = 56) were analyzed using cytogenetic techniques to elucidate the formation and establishment of interploidal hybrids. Hybrids were not a uniform group: two chromosome numbers were observed, with the variation being a consequence of severe hybrid meiotic abnormalities and backcrossing with E. puniceoluteum. The hybrids were triploids (2n = 3x = 38 and 40) and despite the occurrence of enormous meiotic problems associated with triploidy, the hybrids were able to backcross, producing successful hybrid individuals with broad ecological distributions. In spite of the nonpolyploidization of the hybrid, its formation is a long-term evolutionary process rather than a product of a recent disturbance, and considering other sympatric zones in Epidendrum, these events could be recurrent.33824-3

    Regeneração artificial de sobreiro (Quercus suber L.): efeito do ensombramento na sobrevivência e crescimento de plantas jovens

Universidade de LisboaEste trabalho centra-se no estudo do efeito do ensombramento na sobrevivência e crescimento de plantas jovens de sobreiro (Quercus suber L.), visando alcançar melhores resultados na regeneração, artificial ou assistida. O estudo teve lugar na Companhia das Lezírias, na zona do Catapereiro, Santarém, Portugal. Foram plantadas 300 árvores com 6 meses de idade, em março de 2020, distribuídas por 5 blocos, ou seja, 30 plantas por bloco. Destas 30, 15 receberam tratamento de sombra (rede de ensombramento) e outras 15 foram mantidas como grupo-controlo. Foram colocados pero das plantas sensores de medição de temperatura e humidade relativa em parte das árvores jovens durante períodos definidos. Em março foram recolhidos os dados iniciais do estudo, e em julho, outubro e novembro de 2020 foram recolhidos dados referentes à sobrevivência e crescimento, nomeadamente altura apical (cm) e diâmetro basal (mm). Em maio de 2021 foi realizada uma medição da radiação fotossinteticamente ativa (PAR). Embora a taxa de mortalidade geral tenha sido superior ao expectável (78%), o grupo de plantas que sofreu tratamento de sombra teve melhor sobrevivência (+6%) e maior crescimento em altura apical (0,09 mm), não sendo as diferenças significativasN/
