Exogenous coenzyme Q10 modulates MMP-2 activity in MCF-7 cell line as a breast cancer cellular model
<p>Abstract</p> <p>Background/Aims</p> <p>Matrix Metalloproteinases 2 is a key molecule in cellular invasion and metastasis. Mitochondrial ROS has been established as a mediator of MMP activity. Coenzyme Q<sub>10 </sub>contributes to intracellular ROS regulation. Coenzyme Q<sub>10 </sub>beneficial effects on cancer are still in controversy but there are indications of Coenzyme Q<sub>10 </sub>complementing effect on tamoxifen receiving breast cancer patients.</p> <p>Methods</p> <p>In this study we aimed to investigate the correlation of the effects of co-incubation of coenzyme Q10 and N-acetyl-L-cysteine (NAC) on intracellular H2O2 content and Matrix Metalloproteinase 2 (MMP-2) activity in MCF-7 cell line.</p> <p>Results and Discussion</p> <p>Our experiment was designed to assess the effect in a time and dose related manner. Gelatin zymography and Flowcytometric measurement of H2O2 by 2'7',-dichlorofluorescin-diacetate probe were employed. The results showed that both coenzyme Q10 and N-acetyl-L-cysteine reduce MMP-2 activity along with the pro-oxidant capacity of the MCF-7 cell in a dose proportionate manner.</p> <p>Conclusions</p> <p>Collectively, the present study highlights the significance of Coenzyme Q<sub>10 </sub>effect on the cell invasion/metastasis effecter molecules.</p
Extracorporeal membrane oxygenator as a bridge to successful surgical repair of bronchopleural fistula following bilateral sequential lung transplantation: a case report and review of literature
<p>Abstract</p> <p>Background</p> <p>Lung transplantation (LTx) is widely accepted as a therapeutic option for end-stage respiratory failure in cystic fibrosis. However, airway complications remain a major cause of morbidity and mortality in these patients, serious airway complications like bronchopleural fistula (BPF) are rare, and their management is very difficult.</p> <p>Case presentation</p> <p>A 47-year-old man with end-stage respiratory failure due to cystic fibrosis underwent bilateral sequential lung transplantation. Severe post-operative bleeding occurred due to dense intrapleural adhesions of the native lungs. He was re-explored and packed leading to satisfactory haemostasis. He developed a bronchopleural fistula on the 14<sup>th </sup>post-operative day. The fistula was successfully repaired using pericardial and intercostal vascular flaps with veno-venous extracorporeal membrane oxygenator (VV-ECMO) support. Subsequently his recovery was uneventful.</p> <p>Conclusion</p> <p>The combination of pedicled intercostal and pericardial flaps provide adequate vascular tissue for sealing a large BPF following LTx. Veno-venous ECMO allows a feasible bridge to recovery.</p
JWST NIRSpec+MIRI Observations of the nearby Type IIP supernova 2022acko
We present JWST spectral and photometric observations of the Type IIP
supernova (SN) 2022acko at ~50 days past explosion. These data are the first
JWST spectral observations of a core-collapse SN. We identify ~30 different H I
features, other features associated with products produced from the CNO cycle,
and s-process elements such as Sc II and Ba II. By combining the JWST spectra
with ground-based optical and NIR spectra, we construct a full Spectral Energy
Distribution from 0.4 to 25 microns and find that the JWST spectra are fully
consistent with the simultaneous JWST photometry. The data lack signatures of
CO formation and we estimate a limit on the CO mass of < 10^{-8} solar mass. We
demonstrate how the CO fundamental band limits can be used to probe underlying
physics during stellar evolution, explosion, and the environment. The
observations indicate little mixing between the H envelope and C/O core in the
ejecta and show no evidence of dust. The data presented here set a critical
baseline for future JWST observations, where possible molecular and dust
formation may be seen
Investigation of Pulse electric field effect on HeLa cells alignment properties on extracellular matrix protein patterned surface
YesCell behavior in terms of adhesion, orientation and guidance, on extracellular matrix (ECM)
molecules including collagen, fibronectin and laminin can be examined using micro contact
printing (MCP). These cell adhesion proteins can direct cellular adhesion, migration,
differentiation and network formation in-vitro. This study investigates the effect of microcontact
printed ECM protein, namely fibronectin, on alignment and morphology of HeLa cells
cultured in-vitro. Fibronectin was stamped on plain glass cover slips to create patterns of
25μm, 50μm and 100μm width. However, HeLa cells seeded on 50μm induced the best
alignment on fibronectin pattern (7.66° ±1.55SD). As a consequence of this, 50μm wide
fibronectin pattern was used to see how fibronectin induced cell guidance of HeLa cells was
influenced by 100μs and single pulse electric fields (PEF) of 1kV/cm. The results indicates that
cells aligned more under pulse electric field exposure (2.33° ±1.52SD) on fibronectin pattern
substrate. Thus, PEF usage on biological cells would appear to enhance cell surface attachment
and cell guidance. Understanding this further may have applications in enhancing tissue graft

Measurement of neutron-proton capture in the SNO+ water phase
The SNO+ experiment collected data as a low-threshold water Cherenkov
detector from September 2017 to July 2019. Measurements of the 2.2-MeV
produced by neutron capture on hydrogen have been made using an Am-Be
calibration source, for which a large fraction of emitted neutrons are produced
simultaneously with a 4.4-MeV . Analysis of the delayed coincidence
between the 4.4-MeV and the 2.2-MeV capture revealed a
neutron detection efficiency that is centered around 50% and varies at the
level of 1% across the inner region of the detector, which to our knowledge is
the highest efficiency achieved among pure water Cherenkov detectors. In
addition, the neutron capture time constant was measured and converted to a
thermal neutron-proton capture cross section of mb
Measurement of the 8B solar neutrino flux in SNO+ with very low backgrounds
A measurement of the 8B solar neutrino flux has been made using a 69.2 kt-day dataset acquired with the SNO+ detector during its water commissioning phase. At energies above 6 MeV the dataset is an extremely pure sample of solar neutrino elastic scattering events, owing primarily to the detector’s deep location, allowing an accurate measurement with relatively little exposure. In that energy region the best fit background rate is 0.25+0.09−0.07 events/kt−day, significantly lower than the measured solar neutrino event rate in that energy range, which is 1.03+0.13−0.12 events/kt−day. Also using data below this threshold, down to 5 MeV, fits of the solar neutrino event direction yielded an observed flux of 2.53+0.31−0.28(stat)+0.13−0.10(syst)×106 cm−2 s−1, assuming no neutrino oscillations. This rate is consistent with matter enhanced neutrino oscillations and measurements from other experiments
Search for invisible modes of nucleon decay in water with the SNO+ detector
This paper reports results from a search for nucleon decay through invisible modes, where no visible energy is directly deposited during the decay itself, during the initial water phase of SNO+. However, such decays within the oxygen nucleus would produce an excited daughter that would subsequently deexcite, often emitting detectable gamma rays. A search for such gamma rays yields limits of 2.5×1029 y at 90% Bayesian credibility level (with a prior uniform in rate) for the partial lifetime of the neutron, and 3.6×1029 y for the partial lifetime of the proton, the latter a 70% improvement on the previous limit from SNO. We also present partial lifetime limits for invisible dinucleon modes of 1.3×1028 y for nn, 2.6×1028 y for pn and 4.7×1028 y for pp, an improvement over existing limits by close to 3 orders of magnitude for the latter two
