    Phospholipid metabolism by phagocytic cells. Phospholipases A2 associated with rabbit polymorphonuclear leukocyte granules

    Polymorphonuclear leukocytes obtained from sterile peritoneal exudates in rabbits contain two phospholipid-splitting activities (phosphatidylacylhydrolases EC 3.1.1.4), one most active at pH 5.5 and the other between pH 7.2 and 9.0. Hydrolysis of phospholipid was demonstrated using Escherichia coli labeled during growth with [1-(14)C]oleate and then autoclaved to inactivate E. coli phospholipases and to increase the accessibility of the microbial phospholipid substrates. The acid and alkaline phospholipase activities are both membrane bound, calcium dependent, and heat stable, and they appear to be specific for the 2-acyl position of phospholipids. Evidence was also obtained suggesting that the E. coli envelope phospholipids with oleate in position 2 are more readily degraded than those with palmitate. The two activities are associated with azurophilic as well as specific granules (obtained by zonal centrifugation) and with phagosomes (isolated after ingestion of paraffin particles by the granulocytes). Phospholipase A activities at pH 5.5 and pH 7.5 degrade the two major phospholipids of E. coli, phosphatidylethanolamine and phosphatidylglycerol, to the same extent, but the phospholipase activity at acid pH does not hydrolyze micellar dispersions of phosphatidylethanolamine. By contrast, phospholipase A(2) activity at pH 7.5 degrades both types of phosphatidylethanolamine substrates. Heparin and chondroitin sulfate inhibit phospholipase activity at pH 5.5 but have little effect on activity at pH 7.5. All detergents tested inhibited phospholipase activity, and both activities are inhibited by reaction products, free fatty acid and lysophosphatidylethanolamine. This product inhibition is only partially prevented by addition of albumin. Supernatant fractions of granulocyte homogenates contain a heat-labile inhibitor of granule phospholipase activity at pH 7.5. Boiling the fraction not only removes the inhibition but actually results in stimulation of hydrolysis at pH 7.5 as well as pH 5.5. These granule-associated phospholipase A activities of polymorphonuclear leukocytes differ in several of their properties from granule or lysosomal phospholipases of other phagocytic cells

    Histidine-Rich Glycoprotein Protects from Systemic Candida Infection

    Fungi, such as Candida spp., are commonly found on the skin and at mucosal surfaces. Yet, they rarely cause invasive infections in immunocompetent individuals, an observation reflecting the ability of our innate immune system to control potentially invasive microbes found at biological boundaries. Antimicrobial proteins and peptides are becoming increasingly recognized as important effectors of innate immunity. This is illustrated further by the present investigation, demonstrating a novel antifungal role of histidine-rich glycoprotein (HRG), an abundant and multimodular plasma protein. HRG bound to Candida cells, and induced breaks in the cell walls of the organisms. Correspondingly, HRG preferentially lysed ergosterol-containing liposomes but not cholesterol-containing ones, indicating a specificity for fungal versus other types of eukaryotic membranes. Both antifungal and membrane-rupturing activities of HRG were enhanced at low pH, and mapped to the histidine-rich region of the protein. Ex vivo, HRG-containing plasma as well as fibrin clots exerted antifungal effects. In vivo, Hrg−/− mice were susceptible to infection by C. albicans, in contrast to wild-type mice, which were highly resistant to infection. The results demonstrate a key and previously unknown antifungal role of HRG in innate immunity

    The Impact of Symbolic and Substantive Actions on Environmental Legitimacy

    Drawing on institutional theory and insights from stakeholder theory and impression management, we empirically analyze the impact of both environmental symbolic polices (participation in voluntary environmental programs, green trademarks, environmental-dedicated board committees, environmental pay policies and community communication) and substantive actions (environmental patents and pollution prevention practices) on environmental legitimacy. We show that (1) symbolic actions have a weaker positive effect on legitimacy than substantive actions, (2) that the impact of symbolic actions is greater when they are combined with substantive actions, (3) that this impact is only short-term while substantive actions have both short- and long-term effects

    The Anti-Inflammatory and Antibacterial Basis of Human Omental Defense: Selective Expression of Cytokines and Antimicrobial Peptides

    BACKGROUND: The wound healing properties of the human omentum are well known and have extensively been exploited clinically. However, the underlying mechanisms of these effects are not well understood. We hypothesize that the omentum tissue promotes wound healing via modulation of anti-inflammatory pathways, and because the omentum is rich in adipocytes, the adipocytes may modulate the anti-inflammatory response. Factors released by human omentum may affect healing, inflammation and immune defense. METHODOLOGY: Six human omentum tissues (non obese, free from malignancy, and any other systemic disorder) were obtained during diagnostic laparoscopies having a negative outcome. Healthy oral mucosa (obtained from routine oral biopsies) was used as control. Cultured adipocytes derived from human omentum were exposed to lipopolysaccharide (LPS) (1-50 ng/mL) for 12-72 hours to identify the non-cytotoxic doses. Levels of expression (mRNA and protein) were carried out for genes associated with pro- and anti-inflammatory cytokine responses and antibacterial/antimicrobial activity using qRT-PCR, western blotting, and cell-based ELISA assays. RESULTS: The study shows significant higher levels of expression (mRNA and protein) of several specific cytokines, and antibacterial peptides in the omentum tissues when compared to oral sub-mucosal tissues. In the validation studies, primary cultures of adipocytes, derived from human omentum were exposed to LPS (5 and 10 ng/mL) for 24 and 48 h. The altered expressions were more pronounced in cultured adipocytes cells when exposed to LPS as compared to the omentum tissue. CONCLUSIONS/SIGNIFICANCE: Perhaps, this is the first report that provides evidence of expressional changes in pro- and anti-inflammatory cytokines and antibacterial peptides in the normal human omentum tissue as well as adipocytes cultured from this tissue. The study provides new insights on the molecular and cellular mechanisms of healing and defense by the omentum, and suggests the potential applicability of cultured adipocytes derived from the omentum for future therapeutic applications

    Antimicrobial proteins and polypeptides in pulmonary innate defence

    Inspired air contains a myriad of potential pathogens, pollutants and inflammatory stimuli. In the normal lung, these pathogens are rarely problematic. This is because the epithelial lining fluid in the lung is rich in many innate immunity proteins and peptides that provide a powerful anti-microbial screen. These defensive proteins have anti-bacterial, anti- viral and in some cases, even anti-fungal properties. Their antimicrobial effects are as diverse as inhibition of biofilm formation and prevention of viral replication. The innate immunity proteins and peptides also play key immunomodulatory roles. They are involved in many key processes such as opsonisation facilitating phagocytosis of bacteria and viruses by macrophages and monocytes. They act as important mediators in inflammatory pathways and are capable of binding bacterial endotoxins and CPG motifs. They can also influence expression of adhesion molecules as well as acting as powerful anti-oxidants and anti-proteases. Exciting new antimicrobial and immunomodulatory functions are being elucidated for existing proteins that were previously thought to be of lesser importance. The potential therapeutic applications of these proteins and peptides in combating infection and preventing inflammation are the subject of ongoing research that holds much promise for the future

    ‘I was there from the start’: The identity maintenance strategies used by fans to combat the threat of losing

    On-field performances are a key, yet uncontrollable, determinant of team identification. In this research, we explore how fans of a new team, with an overwhelming loss to win ratio, maintain a positive social identity. Qualitative data gathered from 20 semi-structured interviews were used to address this research objective. Our findings indicated fans use social creativity and social mobility strategies to help preserve a positive and distinctive group identity. In the absence of success, fans evaluated the group on dimensions that reflected positively on, and emphasised the distinctiveness of, group membership. Fans also sought to increase their status in the group to increase the positivity of this association. We use these findings to extend understanding of social identity theory and provide recommendations for sport organisations with unfavourable performance records. Recommendations are themed around highlighting the unique nature of the group and and favourable status comparisons between members of the in-group
