    Zusammenfassung: Wurde in der Vergangenheit die Verrentung als zentrale Schwelle für den Übergang in die Altersphase gesehen, so erfordert die mittlerweile zu konstatierende Destandardisierung des Lebenslaufs eine Neubestimmung des Altersübergangs. Es ist davon auszugehen, dass die Verrentung nicht länger als alleiniger Indikator für den Übergang in die Altersphase betrachtet werden kann. Die im Folgenden vorgestellten Forschungsergebnisse thematisieren Altersübergänge mit Blick auf den Lebensbereich Wohnen. Mit einem qualitativ-empirischen Zugang wurden Vorstellungen alternder Menschen hinsichtlich des Wohnens im späten Erwachsenenalter sowie am Lebensende ermittelt. Im Fokus des Beitrags stehen Beschreibungen der Interviewten hinsichtlich der von ihnen antizipierten und präferierten Wohnszenarien.     Determination of islet cell antibodies using an ELISA system with a preparation of rat insulinoma (RIN A2) cells

    An enzyme-linked immunosorbent assay (ELISA) was established for the detection of islet cell antibodies in human sera. The antigen was prepared from rat insulinoma (RIN A2) cells. Cells were dissociated in lysis buffer and the lysate was centrifuged at 100,000 x g. The supernatant was used to coat microtiter ELISA plates (10 micrograms protein/ml in PBS pH 7.2). Non-specific binding sites on the plates were blocked with 2% PBS-BSA. Human test sera were preabsorbed on separate plates using 2% PBS-BSA and incubated on precoated plates at an optimal dilution of 1/10 in 60 mM PBS for 60 min at 37 degrees C. Phosphatase-labeled anti-human IgG serum and phosphatase substrate were applied and the reaction was stopped by adding 3 M NaOH. Out of 90 sera from type I diabetic patients, 47 (52.2%) reacted in the new ELISA whereas none of 15 type II diabetics, 50 sera containing non-islet specific antibodies or 100 normal controls were positive. In the same group of patients, ICA were positive in 63.3%. When both, the ELISA and conventional ICA testing were applied, the number of positives was increased to 83%. The ICA-ELISA with the above described antigen preparation provides a well standardized and reproducible test method which is highly specific for type I diabetes. It may therefore be useful for large screening procedures

    Growth Hormone (GH)-Releasing Peptide Stimulation of GH Release from Human Somatotroph Adenoma Cells: Interaction with GH-Releasing Hormone, Thyrotropin- Releasing Hormone, and Octreotide.

    The synthetic hexapeptide GH-releasing peptide (GHRP; His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) specifically stimulates GH secretion in humans in vivo and in animals in vitro and in vivo via a still unknown receptor and mechanism. To determine the effect of GHRP on human somatotroph cells in vitro, we stimulated cell cultures derived from 12 different human somatotroph adenomas with GHRP alone and in combination with GH-releasing hormone (GHRH), TRH, and the somatostatin analog octreotide. GH secretion of all 12 adenoma cultures could be stimulated with GHRP, whereas GHRH was active only in 6 adenoma cultures. In GHRH-responsive cell cultures, simultaneous application of GHRH and GHRP had an additive effect on GH secretion. TRH stimulated GH release in 4 of 7 adenoma cultures; in TRH-responsive cell cultures there was also an additive effect of GHRP and TRH on GH secretion. In 5 of 9 adenoma cultures investigated, octreotide inhibited basal GH secretion. In these cell cultures, GHRP-induced GH release was suppressed by octreotide. In 5 of 5 cases, the protein kinase-C inhibitor phloretin partly inhibited GHRP-stimulated GH release, but not basal GH secretion. In summary, GH secretion was stimulated by GHRP in all somatotroph adenomas investigated, indicating that its unknown receptor and signaling pathway are expressed more consistently in somatotroph adenoma cells than those for GHRH, TRH, and somatostatin. Our data give further evidence that GHRP-stimulated GH secretion is mediated by a receptor different from that for GHRH or TRH, respectively, and that protein kinase-C is involved in the signal transduction pathway. Because human somatotroph adenoma cell cultures respond differently to various neuropeptides (GHRH, TRH, somatostatin, and others), they provide a model for further investigation of the mechanism of action of GHRP-induced GH secretion

    The Fate of Visible Features of Invisible Elements

    To investigate the integration of features, we have developed a paradigm in which an element is rendered invisible by visual masking. Still, the features of the element are visible as part of other display elements presented at different locations and times (sequential metacontrast). In this sense, we can “transport” features non-retinotopically across space and time. The features of the invisible element integrate with features of other elements if and only if the elements belong to the same spatio-temporal group. The mechanisms of this kind of feature integration seem to be quite different from classical mechanisms proposed for feature binding. We propose that feature processing, binding, and integration occur concurrently during processes that group elements into wholes

    RSE-box : an analysis and modelling package to study response times to multiple signals

    This work was supported by the Biotechnology and Biological Sciences Research Council (BBSRC, grant number: BB/N010108/1).Responses to two redundant sensory signals are typically faster than responses to the individual component signals. This redundant signals effect (RSE) is extensively studied not only with an impressive variety of signals but also across different subject populations focusing on development, aging, and many clinical samples. Yet, a standardized methodology to analyse and interpret the RSE is not consistently employed. Moreover, an intriguing explanation of the effect, based on the so-called race model championed by Raab (1962), is typically not fully appreciated in its explanatory power. To facilitate RSE research, here, an analysis and modelling toolbox, called RSE-box, is introduced. It provides MATLAB functions that (1) perform basic analysis steps on response times (RTs), (2) investigate the RSE at the level of RT distributions, (3) fit the most recent race model by Otto and Mamassian (2012), and (4) simulate the RSE with synthetic data. The model functions are accompanied by parameter recovery simulations to test and validate the fitting procedures. An example of the simulation results is the demonstration that the analysis of group RT distributions can be biased.     Effect of Build Orientation on the Microstructure, Mechanical and Corrosion Properties of a Biodegradable High Manganese Steel Processed by Laser Powder Bed Fusion

    In the last decade, additive manufacturing technologies like laser powder bed fusion (LPBF) have emerged strongly. However, the process characteristics involving layer-wise build-up of the part and the occurring high, directional thermal gradient result in significant changes of the microstructure and the related properties compared to traditionally fabricated materials. This study presents the influence of the build direction (BD) on the microstructure and resulting properties of a novel austenitic Fe-30Mn-1C-0.02S alloy processed via LPBF. The fabricated samples display a {011} texture in BD which was detected by electron backscatter diffraction. Furthermore, isolated binding defects could be observed between the layers. Quasi-static tensile and compression tests displayed that the yield, ultimate tensile as well as the compressive yield strength are significantly higher for samples which were built with their longitudinal axis perpendicular to BD compared to their parallel counterparts. This was predominantly ascribed to the less severe effects of the sharp-edged binding defects loaded perpendicular to BD. Additionally, a change of the Young’s modulus in dependence of BD could be demonstrated, which is explained by the respective texture. Potentiodynamic polarization tests conducted in a simulated body fluid revealed only slight differences of the corrosion properties in dependence of the build design
