Difficulty Scaling in Proof of Work for Decentralized Problem Solving
We propose DIPS Difficulty-based Incentives for Problem Solving), a simple
modification of the Bitcoin proof-of-work algorithm that rewards blockchain
miners for solving optimization problems of scientific interest. The result is
a blockchain which redirects some of the computational resources invested in
hash-based mining towards scientific computation, effectively reducing the
amount of energy `wasted' on mining. DIPS builds the solving incentive directly
in the proof-of-work by providing a reduction in block hashing difficulty when
optimization improvements are found. A key advantage of this scheme is that
decentralization is preserved and no additional protocol layers are required on
top of the standard blockchain. We study two incentivization schemes and
provide simulation results showing that DIPS is able to reduce the amount of
hash-power used in the network while generating solutions to optimization
problems
Membrane with supported internal passages
The invention provides an improved proton exchange membrane for use in electrochemical cells having internal passages parallel to the membrane surface comprising permanent tubes preferably placed at the ends of the fluid passages. The invention also provides an apparatus and process for making the membrane, membrane and electrode assemblies fabricated using the membrane, and the application of the membrane and electrode assemblies to a variety of devices, both electrochemical and otherwise. The passages in the membrane extend from one edge of the membrane to another and allow fluid flow through the membrane and give access directly to the membrane
A human phospholipid phosphatase activated by a transmembrane control module
In voltage-sensitive phosphatases (VSPs), a transmembrane voltage sensor domain (VSD) controls an intracellular phosphoinositide phosphatase domain, thereby enabling immediate initiation of intracellular signals by membrane depolarization. The existence of such a mechanism in mammals has remained elusive, despite the presence of VSP-homologous proteins in mammalian cells, in particular in sperm precursor cells. Here we demonstrate activation of a human VSP (hVSP1/TPIP) by an intramolecular switch. By engineering a chimeric hVSP1 with enhanced plasma membrane targeting containing the VSD of a prototypic invertebrate VSP, we show that hVSP1 is a phosphoinositide-5-phosphatase whose predominant substrate is PI(4,5)P(2). In the chimera, enzymatic activity is controlled by membrane potential via hVSP1\u27s endogenous phosphoinositide binding motif. These findings suggest that the endogenous VSD of hVSP1 is a control module that initiates signaling through the phosphatase domain and indicate a role for VSP-mediated phosphoinositide signaling in mammals

Pancreatic Duct Glands Are Distinct Ductal Compartments That React to Chronic Injury and Mediate Shh-Induced Metaplasia
BACKGROUND & AIMS: Pancreatic intraepithelial neoplasia (PanIN) are pancreatic cancer precursor lesions of unclear origin and significance. PanIN aberrantly express sonic hedgehog (Shh), an initiator of pancreatic cancer, and gastrointestinal mucins. A majority of PanIN are thought to arise from ducts. We identified a novel ductal compartment that is gathered in gland-like outpouches (pancreatic duct glands [PDG]) of major ducts and characterized its role in injury and metaplasia. METHODS: The ductal system was analyzed in normal pancreata and chronic pancreatitis in humans and mice. Anatomy was assessed by serial hematoxylin and eosin sections and scanning electron microscopy of corrosion casts. Expression of mucins and developmental genes and proliferation were assessed by immunohistochemistry or real-time quantitative polymerase chain reaction. Effects of Shh on ductal cells were investigated by exposure to Shh in vitro and transgenic misexpression in vivo. RESULTS: Three-dimensional analysis revealed blind-ending outpouches of ducts in murine and human pancreata. These PDG are morphologically and molecularly distinct from normal ducts; even in normal pancreata they display PanIN and metaplastic features, such as expression of Shh and gastric mucins. They express other developmental genes, such as Pdx-1 and Hes-1. In injury, Shh is up-regulated along with gastric mucins. Expansion of the PDG compartment results in a mucinous metaplasia. Shh promotes this transformation in vitro and in vivo. CONCLUSIONS: PDG are distinct gland-like mucinous compartments with a distinct molecular signature. In response to injury, PDG undergo an Shh-mediated mucinous gastrointestinal metaplasia with PanIN-like features. PDG may provide a link between Shh, mucinous metaplasia, and neoplasia
Distinct facial expressions represent pain and pleasure across cultures
open access articleReal-world studies show that the facial expressions produced during pain and orgasm—two different and intense affective experiences—are virtually indistinguishable. However, this finding is counterintuitive, because facial expressions are widely considered to be a powerful tool for social interaction. Consequently, debate continues as to whether the facial expressions of these extreme positive and negative affective states serve a communicative function. Here, we address this debate from a novel angle by modeling the mental representations of dynamic facial expressions of pain and orgasm in 40 observers in each of two cultures (Western, East Asian) using a data-driven method. Using a complementary approach of machine learning, an information-theoretic analysis, and a human perceptual discrimination task, we show that mental representations of pain and orgasm are physically and perceptually distinct in each culture. Cross-cultural comparisons also revealed that pain is represented by similar face movements across cultures, whereas orgasm showed distinct cultural accents. Together, our data show that mental representations of the facial expressions of pain and orgasm are distinct, which questions their nondiagnosticity and instead suggests they could be used for communicative purposes. Our results also highlight the potential role of cultural and perceptual factors in shaping the mental representation of these facial expressions. We discuss new research directions to further explore their relationship to the production of facial expressions
Marine Citizen Science: Current State in Europe and New Technological Developments
Marine citizen science is emerging with promising opportunities for science, policy and
public but there is still no comprehensive overview of the current state in Europe.
Based on 127 projects identified for the North Sea area we estimate there might be
as much as 500 marine and coastal citizen science projects running in Europe, i.e.,
one marine citizen science project per 85 km of coastline, with an exponential growth
since 1990. Beach-based projects are more accessible and hence most popular (60%
of the projects), and the mean duration of the projects is 18–20 years. Current trends,
topics, organizers, aims, and types of programme in terms of participation are presented
in this overview. Progress in marine citizen science is specially enabled and promoted
through technological developments. Recent technological advances and best practise
examples are provided here, untapping the potential of smart mobile apps, do-it-yourself
(DIY) technologies, drones, and artificial intelligence (AI) web servicesVersión del edito
Explicit BCJ Numerators from Pure Spinors
We derive local kinematic numerators for gauge theory tree amplitudes which
manifestly satisfy Jacobi identities analogous to color factors. They naturally
emerge from the low energy limit of superstring amplitudes computed with the
pure spinor formalism. The manifestation of the color--kinematics duality is a
consequence of the superstring computation involving no more than (n-2)!
kinematic factors for the full color dressed n-point amplitude. The bosonic
part of these results describe gluon scattering independent on the number of
supersymmetries and captures any N^kMHV helicity configuration after
dimensional reduction to D=4 dimensions.Comment: 32 pages, harvma
