Evaluation of a commercial intravaginal thermometer to predict calving in a Hungarian Holstein‐Friesian dairy farm
In this study, the utility of a commercial intravaginal thermometer was evaluated as an automated method for the prediction of calving in a total of 257 healthy pregnant Holstein–Friesian female cattle. The accuracy and the sensitivity of predicting calving within 48 hr before calving were also evaluated. The intravaginal temperature changes from 72 hr before and up to calving were significantly (p ≤ .001) affected by parity, season (summer vs. autumn), the time of day (8 a.m. or 8 p.m.) and the 6-hr time intervals (38.19°C: first interval 0 to 6 hr before calving vs. 38.78°C: twelfth interval 66 to 72 hr before calving), while the gender (p = .943), and the weight of the calf (p = .610), twinning (p = .300), gestation length (p = .186), foetal presentation (p = .123), dystocia (p = .197) and retention of foetal membranes (p = .253) did not affect it significantly. The sensitivity of the SMS of expecting calving within 48 hr and the positive predictive value were 62.4% and 75%, respectively, while the sensitivity and the positive predictive value for the SMS of expulsion reached 100%. It can be concluded that the investigated thermometer is not able to predict calving within 48 hr accurately; however, imminent calving can be accurately alerted
Fetal heart rate and fetal heart rate variability in Lipizzaner broodmares
Monitoring fetal heart rate (FHR) and fetal heart rate variability (FHRV) helps to understand and evaluate normal and pathological conditions in the foal. The aim of this study was to establish normal heart rate reference values for the ongoing equine pregnancy and to perform a heart rate variability (HRV) time-domain analysis in Lipizzaner mares. Seventeen middle- and late-term (days 121–333) pregnant Lipizzaner mares were examined using fetomaternal electrocardiography (ECG). The mean FHR (P = 0.004) and the standard deviation of FHR (P = 0.012) significantly decreased during the pregnancy. FHR ± SD values decreased from 115 ± 35 to 79 ± 9 bpm between months 5 and 11. Our data showed that HRV in the foal decreased as the pregnancy progressed, which is in contrast with the findings of earlier equine studies. The standard deviation of normal-normal intervals (SDNN) was higher (70 ± 25 to 166 ± 108 msec) than described previously. The root mean square of successive differences (RMSSD) decreased from 105 ± 69 to 77 ± 37 msec between the 5th and 11th month of gestation. Using telemetric ECG equipment, we could detect equine fetal heartbeat on day 121 for the first time. In addition, the large differences observed in the HR values of four mare-fetus pairs in four consecutive months support the assumption that there might be ‘high-HR’ and ‘low-HR’ fetuses in horses. It can be concluded that the analysis of FHR and FHRV is a promising tool for the assessment of fetal well-being but the applicability of these parameters in the clinical setting and in studs requires further investigation
Biochemical and pharmacological characterization of three opioid-nociceptin hybrid peptide ligands reveals substantially differing modes of their actions
In an attempt to design opioid-nociceptin hybrid peptides, three novel bivalent ligands, H-YGGFGGGRYYRIK-NH2, H-YGGFRYYRIK-NH2 and Ac-RYYRIKGGGYGGFL-OH were synthesized and studied by biochemical, pharmacological, biophysical and molecular modelling tools. These chimeric molecules consist of YGGF sequence, a crucial motif in the N-terminus of natural opioid peptides, and Ac-RYYRIK-NH2, which was isolated from a combinatorial peptide library as an antagonist or partial agonist that inhibits the biological activity of the endogenously occurring heptadecapeptide nociceptin. Solution structures for the peptides were studied by analysing their circular dichroism spectra. Receptor binding affinities were measured by equilibrium competition experiments using four highly selective radioligands. G-protein activating properties of the multitarget peptides were estimated in [35S]GTPγS binding tests. The three compounds were also measured in electrically stimulated mouse vas deferens (MVD) bioassay. H-YGGFGGGRYYRIK-NH2 (BA55), carrying N-terminal opioid and C-terminal nociceptin-like sequences interconnected with GGG tripeptide spacer displayed a tendency of having either unordered or β-sheet structures, was moderately potent in MVD and possessed a NOP/KOP receptor preference. A similar peptide without spacer H-YGGFRYYRIK-NH2 (BA62) exhibited the weakest effect in MVD, more α-helical periodicity was present in its structure and it exhibited the most efficacious agonist actions in the G-protein stimulation assays. The third hybrid peptide Ac-RYYRIKGGGYGGFL-OH (BA61) unexpectedly displayed opioid receptor affinities, because the opioid message motif is hidden within the C-terminus. The designed chimeric peptide ligands presented in this study accommodate well into a group of multitarget opioid compounds that include opioid-non-opioid peptide dimer analogues, dual non-peptide dimers and mixed peptide- non-peptide bifunctional ligands
Age-dependent parathormone levels and different CKD-MBD treatment practices of dialysis patients in Hungary - results from a nationwide clinical audit
BACKGROUND: Achieving target levels of laboratory parameters of
bone and mineral metabolism in chronic kidney disease (CKD)
patients is important but also difficult in those living with
end-stage kidney disease. This study aimed to determine if there
are age-related differences in chronic kidney disease-mineral
and bone disorder (CKD-MBD) characteristics, including treatment
practice in Hungarian dialysis patients. METHODS: Data were
collected retrospectively from a large cohort of dialysis
patients in Hungary. Patients on hemodialysis and peritoneal
dialysis were also included. The enrolled patients were
allocated into two groups based on their age (=65
years). Characteristics of the age groups and differences in
disease-related (epidemiology, laboratory, and treatment
practice) parameters between the groups were analyzed. RESULTS:
A total of 5008 patients were included in the analysis and the
mean age was 63.4+/-14.2 years. A total of 47.2% of patients
were women, 32.8% had diabetes, and 11.4% were on peritoneal
dialysis. Diabetes (37.9% vs 27.3%), bone disease (42.9% vs
34.1%), and soft tissue calcification (56.3% vs 44.7%) were more
prevalent in the older group than the younger group (p<0.001 for
all). We found an inverse relationship between age and
parathyroid hormone (PTH) levels (p<0.001). Serum PTH levels
were lower in patients with diabetes compared with those without
diabetes below 80 years (p<0.001). Diabetes and age were
independently associated with serum PTH levels (interaction:
diabetes x age groups, p=0.138). Older patients were more likely
than younger patients to achieve laboratory target ranges for
each parameter (Ca: 66.9% vs 62.1%, p<0.001; PO4: 52.6% vs
49.2%, p<0.05; and PTH: 50.6% vs 46.6%, p<0.01), and for
combined parameters (19.8% vs 15.8%, p<0.001). Older patients
were less likely to receive related medication than younger
patients (66.9% vs 79.7%, p<0.001). CONCLUSIONS: The achievement
of laboratory target ranges for bone and mineral metabolism and
clinical practice in CKD depends on the age of the patients. A
greater proportion of older patients met target criteria and
received less medication compared with younger patients
The First Post-Kepler Brightness Dips of KIC 8462852
We present a photometric detection of the first brightness dips of the unique variable star KIC 8462852 since the end of the Kepler space mission in 2013 May. Our regular photometric surveillance started in October 2015, and a sequence of dipping began in 2017 May continuing on through the end of 2017, when the star was no longer visible from Earth. We distinguish four main 1-2.5% dips, named "Elsie," "Celeste," "Skara Brae," and "Angkor", which persist on timescales from several days to weeks. Our main results so far are: (i) there are no apparent changes of the stellar spectrum or polarization during the dips; (ii) the multiband photometry of the dips shows differential reddening favoring non-grey extinction. Therefore, our data are inconsistent with dip models that invoke optically thick material, but rather they are in-line with predictions for an occulter consisting primarily of ordinary dust, where much of the material must be optically thin with a size scale <<1um, and may also be consistent with models invoking variations intrinsic to the stellar photosphere. Notably, our data do not place constraints on the color of the longer-term "secular" dimming, which may be caused by independent processes, or probe different regimes of a single process
Effect of lead contamination of maize seed on its biological properties
Effect of treatment of seed with various lead concentrations (0, 10-5, 10-4, 10-3, and 10-2 mol/dm3) on accumulation and distribution of lead (Pb) in seedling, seed germination, seedling growth, and mobilization of mineral matter during seed germination was investigated. Content of Pb in the root and the shoot indicates that seeds imbibed in solutions of various Pb concentrations took up Pb intensively. Content of Pb in the root and the shoot increased with increase of Pb concentration and it was much larger in the root than in the shoot. Contrary to this, the accumulation coefficient was greater in the shoot than in the root. Treatment of seed with Pb did not significantly affect its biological properties. Increase of Pb concentration decreased germination ability, germination energy, and percentage of typical seedlings, while increasing the number of atypical seedlings and non-germinated seeds. Contamination of seed by Pb did not affect the dry matter mass and the growth of young plants shoots, while the length of the primary root, the mesocotyl root as well as the root mass at the highest Pb concentration, significantly decreased. Translocation of mobilized mineral matter from the seed during germination and growth of young plants into the root and shoot was specific, depending on elements. Only the implementation of the highest implemented Pb concentration affected mobilization and translocation of some elements. Based on the obtained results, it can be concluded that maize is characterized by significant tolerance to Pb contamination during seed germination and growth of seedlings
