A Genomewide Functional Network for the Laboratory Mouse
Establishing a functional network is invaluable to our understanding of gene function, pathways, and systems-level properties of an organism and can be a powerful resource in directing targeted experiments. In this study, we present a functional network for the laboratory mouse based on a Bayesian integration of diverse genetic and functional genomic data. The resulting network includes probabilistic functional linkages among 20,581 protein-coding genes. We show that this network can accurately predict novel functional assignments and network components and present experimental evidence for predictions related to Nanog homeobox (Nanog), a critical gene in mouse embryonic stem cell pluripotency. An analysis of the global topology of the mouse functional network reveals multiple biologically relevant systems-level features of the mouse proteome. Specifically, we identify the clustering coefficient as a critical characteristic of central modulators that affect diverse pathways as well as genes associated with different phenotype traits and diseases. In addition, a cross-species comparison of functional interactomes on a genomic scale revealed distinct functional characteristics of conserved neighborhoods as compared to subnetworks specific to higher organisms. Thus, our global functional network for the laboratory mouse provides the community with a key resource for discovering protein functions and novel pathway components as well as a tool for exploring systems-level topological and evolutionary features of cellular interactomes. A neuroscientist's guide to lipidomics
Nerve cells mould the lipid fabric of their membranes to ease vesicle fusion, regulate ion fluxes and create specialized microenvironments that contribute to cellular communication. The chemical diversity of membrane lipids controls protein traffic, facilitates recognition between cells and leads to the production of hundreds of molecules that carry information both within and across cells. With so many roles, it is no wonder that lipids make up half of the human brain in dry weight. The objective of neural lipidomics is to understand how these molecules work together; this difficult task will greatly benefit from technical advances that might enable the testing of emerging hypotheses
Rates, Associations, and Predictors of Psychopathology in a Convenience Sample of School-Aged Latino Youth: Identifying Areas for Mental Health Outreach
The Latino youth population is rapidly growing and expected to comprise nearly 40 % of the total youth population by 2060. Unfortunate disparities exist in the United States (U.S.), such that young Latinos are less likely than non-Hispanic Whites to receive and benefit from mental health services. In order to identify and prioritize specific areas of mental health outreach, the current study examined preliminary rates, associations, and predictors of child psychopathology in a convenience sample of Latino youth. 123 Spanish and English speaking Latino parents of school-aged children completed a series of questionnaires regarding child and family functioning. Latino youth in the current sample demonstrated comparable rates of psychopathology to non-referred, normative samples. Parental acculturation (particularly Separated parental acculturation status: high orientation to Latino culture and low orientation to U.S. mainstream culture) was associated with an increased prevalence of clinically significant psychopathology across several domains, and socioeconomic status was associated with an increased prevalence of thought problems. Additionally, Separated parental acculturation status significantly predicted the prevalence of clinically significant anxious/depressed problems, such that youth of parents displaying Separated acculturation status were significantly more represented in the clinically-elevated groups than the functional groups. These preliminary results suggest that prioritizing outreach to Latino youth of parents maintaining orientation to Latino culture but not U.S. mainstream culture may be necessary in order to begin addressing existing mental health disparities in the U.S
