    Q: Is ketamine effective and safe for treatment-resistant depression?

    Evidence-based answer: Maybe, but it’s too soon to tell. There is limited evidence that ketamine by itself is effective in the very short term. Single-dose intravenous (IV) ketamine is more likely than placebo (odds ratio = 11-13) to produce improvement (> 50%) in standardized depression scores in 1 to 3 days, lasting up to a week. Twice- or thriceweekly IV ketamine improves symptom scores by 20%-25% over 2 weeks (strength of recommendation [SOR]: B, meta-analysis of small, low-quality, randomized controlled trials [RCTs] and a single small RCT). Augmentation of sertraline with daily oral ketamine moderately improves symptom scores for 6 weeks in patients with moderate depression (SOR: B, small, low-quality RCTs). Augmentation of oral antidepressants (duloxetine, escitalopram, sertraline, venlafaxine) with intranasal esketamine spray improves response and remission rates at 4 weeks (16% for both outcomes) in patients with predominantly treatment-resistant major depression (SOR: A, meta-analysis of RCTs). Ketamine therapy is associated with confusion, emotional blunting, headache, dizziness, and blurred vision (SOR: A, meta-analyses).     Developing User‐Friendly Habitat Suitability Tools from Regional Stream Fish Survey Data

    We developed user‐friendly fish habitat suitability tools (plots) for fishery managers in Michigan; these tools are based on driving habitat variables and fish population estimates for several hundred stream sites throughout the state. We generated contour plots to show patterns in fish biomass for over 60 common species (and for 120 species grouped at the family level) in relation to axes of catchment area and low‐flow yield (90% exceedance flow divided by catchment area) and also in relation to axes of mean and weekly range of July temperatures. The plots showed distinct patterns in fish habitat suitability at each level of biological organization studied and were useful for quantitatively comparing river sites. We demonstrate how these plots can be used to support stream management, and we provide examples pertaining to resource assessment, trout stocking, angling regulations, chemical reclamation of marginal trout streams, indicator species, instream flow protection, and habitat restoration.     The Actin Targeting Compound Chondramide Inhibits Breast Cancer Metastasis via Reduction of Cellular Contractility

    Background: A major player in the process of metastasis is the actin cytoskeleton as it forms key structures in both invasion mechanisms, mesenchymal and amoeboid migration. We tested the actin binding compound Chondramide as potential anti-metastatic agent. Methods: In vivo, the effect of Chondramide on metastasis was tested employing a 4T1-Luc BALB/c mouse model. In vitro, Chondramide was tested using the highly invasive cancer cell line MDA-MB-231 in Boyden-chamber assays, fluorescent stainings, Western blot and Pull down assays. Finally, the contractility of MDA-MB-231 cells was monitored in 3D environment and analyzed via PIV analysis. Results: In vivo, Chondramide treatment inhibits metastasis to the lung and the migration and invasion of MDA-MB-231 cells is reduced by Chondramide in vitro. On the signaling level, RhoA activity is decreased by Chondramide accompanied by reduced MLC-2 and the stretch induced guanine nucleotide exchange factor Vav2 activation. At same conditions, EGF-receptor autophosphorylation, Akt and Erk as well as Rac1 are not affected. Finally, Chondramide treatment disrupted the actin cytoskeleton and decreased the ability of cells for contraction. Conclusions: Chondramide inhibits cellular contractility and thus represents a potential inhibitor of tumor cell invasion

    The H1 Forward Proton Spectrometer at HERA

    The forward proton spectrometer is part of the H1 detector at the HERA collider. Protons with energies above 500 GeV and polar angles below 1 mrad can be detected by this spectrometer. The main detector components are scintillating fiber detectors read out by position-sensitive photo-multipliers. These detectors are housed in so-called Roman Pots which allow them to be moved close to the circulating proton beam. Skin burn tissues were non-invasively characterized by OCT images and biopsied for further histopathology analysis, SHG imaging and FTIR spectroscopy at 3, 5, 7 and 14 days after burn. The laser protocol was found as efficient as the classical treatment for promoting the healing process. The study concludes to the validation of femtosecond ultra-short pulses laser treatment for skinburns, with the advantage of minimizing operatory trauma.     ifo Konjunkturprognose 2011/2012: Schuldenkrise bremst deutsche Wirtschaft aus

    Am 14. Dezember 2011 stellte das ifo Institut im Rahmen seines vorweihnachtlichen PressegesprĂ€chs seine Prognose fĂŒr die Jahre 2011 und 2012 vor. In Deutschland ist das Bruttoinlandsprodukt bis zuletzt deutlich gestiegen. Der ifo GeschĂ€ftsklimaindex und andere FrĂŒhindikatoren zeigen jedoch an, dass der deutschen Konjunktur ein Abschwung bevorsteht. Maßgeblich hierfĂŒr sind die sich abschwĂ€chende Weltwirtschaft und die europĂ€ische Schuldenkrise. Die AbhĂ€ngigkeit der Konjunkturentwicklung von den Entscheidungen der europĂ€ischen Politik erschwert die Prognose erheblich, denn es sind ganz unterschiedliche plausible Szenarien möglich. Unter der Annahme, dass sich die Eurokrise nicht weiter verschĂ€rft und sich insbesondere Italien weiter am Markt finanzieren kann, dĂŒrfte eine Rezession in Deutschland vermieden werden. Dennoch wird das deutsche Bruttoinlandsprodukt 2012 voraussichtlich nur um 0,4% zunehmen. Es ist daher zu erwarten, dass sich der BeschĂ€ftigungsaufbau deutlich verlangsamt. GestĂŒtzt von demographischen Faktoren dĂŒrfte die Arbeitslosenzahl aber noch um 140 000 auf rund 2,8 Mill. Personen zurĂŒckgehen. Dies entspricht einer Arbeitslosenquote von 6,7%. Im Gefolge der schwĂ€cheren Konjunktur wird sich das Tempo des Preisauftriebs abschwĂ€chen; im Jahresdurchschnitt 2012 ist mit einer Inflationsrate von 1,8% zu rechnen. Das staatliche Budgetdefizit in Relation zum nominalen Bruttoinlandsprodukt wird im nĂ€chsten Jahr voraussichtlich bei 0,9% liegen. Es muss betont werden, dass die makroökonomische Unsicherheit nicht zuletzt aus politischen GrĂŒnden derzeit extrem hoch ist. So könnten schon kleine Abweichungen Italiens vom geplanten Konsolidierungskurs zu neuen Verwerfungen an den ohnehin extrem angespannten FinanzmĂ€rkten und kaum abschĂ€tzbaren politischen Reaktionen fĂŒhren. Diese könnten die der Prognose zugrunde liegenden Annahmen schnell obsolet werden lassen.     Soft-core hyperon-nucleon potentials

    A new Nijmegen soft-core OBE potential model is presented for the low-energy YN interactions. Besides the results for the fit to the scattering data, which largely defines the model, we also present some applications to hypernuclear systems using the G-matrix method. An important innovation with respect to the original soft-core potential is the assignment of the cut-off masses for the baryon-baryon-meson (BBM) vertices in accordance with broken SU(3)F_F, which serves to connect the NN and the YN channels. As a novel feature, we allow for medium strong breaking of the coupling constants, using the 3P0^3P_0 model with a Gell-Mann--Okubo hypercharge breaking for the BBM coupling. We present six hyperon-nucleon potentials which describe the available YN cross section data equally well, but which exhibit some differences on a more detailed level. The differences are constructed such that the models encompass a range of scattering lengths in the ΣN\Sigma N and ΛN\Lambda N channels. For the scalar-meson mixing angle we obtained values ξS=37\theta_S=37 to 40 degrees, which points to almost ideal mixing angles for the scalar qqˉq\bar{q} states. The G-matrix results indicate that the remarkably different spin-spin terms of the six potentials appear specifically in the energy spectra of Λ\Lambda hypernuclei.Comment: 37 pages, 4 figure

    Detection of variable VHE gamma-ray emission from the extra-galactic gamma-ray binary LMC P3

    Context. Recently, the high-energy (HE, 0.1-100 GeV) Îł\gamma-ray emission from the object LMC P3 in the Large Magellanic Cloud (LMC) has been discovered to be modulated with a 10.3-day period, making it the first extra-galactic Îł\gamma-ray binary. Aims. This work aims at the detection of very-high-energy (VHE, >100 GeV) Îł\gamma-ray emission and the search for modulation of the VHE signal with the orbital period of the binary system. Methods. LMC P3 has been observed with the High Energy Stereoscopic System (H.E.S.S.); the acceptance-corrected exposure time is 100 h. The data set has been folded with the known orbital period of the system in order to test for variability of the emission. Energy spectra are obtained for the orbit-averaged data set, and for the orbital phase bin around the VHE maximum. Results. VHE Îł\gamma-ray emission is detected with a statistical significance of 6.4 σ\sigma. The data clearly show variability which is phase-locked to the orbital period of the system. Periodicity cannot be deduced from the H.E.S.S. data set alone. The orbit-averaged luminosity in the 1−101-10 TeV energy range is (1.4±0.2)×1035(1.4 \pm 0.2) \times 10^{35} erg/s. A luminosity of (5±1)×1035(5 \pm 1) \times 10^{35} erg/s is reached during 20% of the orbit. HE and VHE Îł\gamma-ray emissions are anti-correlated.
