    Get PDF

This Lesson Study forms part of the research work carried out by the Spanish team, in order to analyse the potential of this professional teacher development methodology for the reconstruction of teachers' practical thinking. It is a qualitative research process which involves the monitoring of seven infant school teachers during two school cycles. The teachers developed a Lesson Study over this period, with a practical thinking case study being carried out both before and after. This paper focuses on the Lesson Study case study. The Lesson Study was developed independently by the group of teachers, who were advised throughout by the undersigned researchers. At the same time, the researchers carried out a case study focusing on the Lesson Study being developed by the teachers. Findings/Conclusions: - From the start of the Lesson Study, we can find evidence of changes in the practical thinking of the teachers involved, which led to immediate changes in their classroom practice. - The development of the Lesson Study brought about significant reflection with regards to the Infant Education curriculum, in some cases resulting in substantial modification to the way it is implemented in their different classrooms. - The teachers pointed out the value of cooperative work on several different occasions. - They also emphasised the high level of learning they achieved by observing how their colleagues developed the lesson designed by the group using different strategies and data collection instruments, such as documentation inspired by the Reggio Emilia approach, when reflecting on their own practice. On the whole, we can state that the cooperative design work of this lesson showed the contrast between the espoused theories of the participants and their theories-in-use, conditioned fundamentally by their work contexts and previous experience. Lesson Study is a way to bring these two poles closer together, enriching teachers with strategies to help detect contradictions in their practice and to resolve them and feel more comfortable with what they do.Universidad de Málaga. Amyloid β Oligomers Increase ER-Mitochondria Ca2+ Cross Talk in Young Hippocampal Neurons and Exacerbate Aging-Induced Intracellular Ca2+ Remodeling

Alzheimer’s disease (AD) is the most common neurodegenerative disorder and strongly associated to aging. AD has been related to excess of neurotoxic oligomers of amyloid β peptide (Aβo), loss of intracellular Ca2+ homeostasis and mitochondrial damage. However, the intimate mechanisms underlying the pathology remain obscure. We have reported recently that long-term cultures of rat hippocampal neurons resembling aging neurons are prone to damage induced by Aβ oligomers (Aβo) while short-term cultured cells resembling young neurons are not. In addition, we have also shown that aging neurons display critical changes in intracellular Ca2+ homeostasis including increased Ca2+ store content and Ca2+ transfer from the endoplasmic reticulum (ER) to mitochondria. Aging also promotes the partial loss of store-operated Ca2+ entry (SOCE), a Ca2+ entry pathway involved in memory storage. Here, we have addressed whether Aβo treatment influences differentially intracellular Ca2+ homeostasis in young and aged neurons. We found that Aβo exacerbate the remodeling of intracellular Ca2+ induced by aging. Specifically, Aβo exacerbate the loss of SOCE observed in aged neurons. Aβo also exacerbate the increased resting cytosolic Ca2+ concentration, Ca2+ store content and Ca2+ release as well as increased expression of the mitochondrial Ca2+ uniporter (MCU) observed in aging neurons. In contrast, Aβo elicit none of these effects in young neurons. Surprisingly, we found that Aβo increased the Ca2+ transfer from ER to mitochondria in young neurons without having detrimental effects. Consistently, Aβo increased also colocalization of ER and mitochondria in both young and aged neurons. However, in aged neurons, Aβo suppressed Ca2+ transfer from ER to mitochondria, decreased mitochondrial potential, enhanced reactive oxygen species (ROS) generation and promoted apoptosis. These results suggest that modulation of ER—mitochondria coupling in hippocampal neurons may be a novel physiological role of Aβo. However, excess of Aβo in the face of the remodeling of intracellular Ca2+ homeostasis associated to aging may lead to loss of ER—mitochondrial coupling and AD

The Impact of Additive Manufacturing on Supply Chain Management from a System Dynamics Model-Scenario: Traditional, Centralized, and Distributed Supply Chain

[EN] In order to describe the impact that the appropriation of additive manufacturing (AM) has on the supply chain (SC), a validated system dynamics model representing vectorially multiple products and multiple demands in different periods was used as a basis to apply to a case study of medical implant manufacturing, configuring three chain scenarios: 1. traditional supply chain with subtractive manufacturing, 2. centralized supply chain with additive manufacturing, and 3. decentralized supply chain with additive manufacturing. It was possible to notice that the production time is longer in additive manufacturing compared to traditional manufacturing and the cycle time and total demand closure were lower in traditional manufacturing. In addition, it was observed that the AM performance is significantly better in conditions of lower demand, which can be attributed to the characteristics of customization and small batches that this type of production approach implies. The aim of this research project is to analyze the school coexistence in the Salesian Educational Institutions “Miguel Unía” and “María Inmaculada Sede Tarqui”, is intended to generate strategies to minimize the negative impact on the processes of school coexistence, since it can be seen that at the time of the school’s rest, there are considerable difficulties in the environment due to intolerance and a lack of motivation towards the use of values. On the other hand, teachers rarely commit themselves to attending this space and in other circumstances the presence of the educator is more to monitor than to accompany. After approaching the reality, we find it opportune to offer a didactic material as a result that enriches the playful and recreational experience of rest so that it is not simply a moment between classes, It should be a real experience of sharing the Educational Community and also provide spaces for reflection, for recognizing the other, helping to enhance their cognitive and affective dimensions among learner

Abstract

Background

DNA-damage assays, quantifying the initial number of DNA double-strand breaks induced by radiation, have been proposed as a predictive test for radiation-induced toxicity. Determination of radiation-induced apoptosis in peripheral blood lymphocytes by flow cytometry analysis has also been proposed as an approach for predicting normal tissue responses following radiotherapy. The aim of the present study was to explore the association between initial DNA damage, estimated by the number of double-strand breaks induced by a given radiation dose, and the radio-induced apoptosis rates observed.

Methods

Peripheral blood lymphocytes were taken from 26 consecutive patients with locally advanced breast carcinoma. Radiosensitivity of lymphocytes was quantified as the initial number of DNA double-strand breaks induced per Gy and per DNA unit (200 Mbp). Radio-induced apoptosis at 1, 2 and 8 Gy was measured by flow cytometry using annexin V/propidium iodide.

Results

Radiation-induced apoptosis increased in order to radiation dose and data fitted to a semi logarithmic mathematical model. A positive correlation was found among radio-induced apoptosis values at different radiation doses: 1, 2 and 8 Gy (p < 0.0001 in all cases). Mean DSB/Gy/DNA unit obtained was 1.70 ± 0.83 (range 0.63-4.08; median, 1.46). A statistically significant inverse correlation was found between initial damage to DNA and radio-induced apoptosis at 1 Gy (p = 0.034). A trend toward 2 Gy (p = 0.057) and 8 Gy (p = 0.067) was observed after 24 hours of incubation.

Conclusions

An inverse association was observed for the first time between these variables, both considered as predictive factors to radiation toxicity.</p

IL-17RA-signaling modulates CD8+ T Cell survival and exhaustion during trypanosoma cruzi infection

The IL-17 family contributes to host defense against many intracellular pathogens by mechanisms that are not fully understood. CD8+ T lymphocytes are key elements against intracellular microbes, and their survival and ability to mount cytotoxic responses are orchestrated by several cytokines. Here, we demonstrated that IL-17RA-signaling cytokines sustain pathogen-specific CD8+ T cell immunity. The absence of IL-17RA and IL-17A/F during Trypanosoma cruzi infection resulted in increased tissue parasitism and reduced frequency of parasite-specific CD8+ T cells. Impaired IL-17RA-signaling in vivo increased apoptosis of parasite-specific CD8+ T cells, while in vitro recombinant IL-17 down-regulated the pro-Apoptotic protein BAD and promoted the survival of activated CD8+ T cells. Phenotypic, functional, and transcriptomic profiling showed that T. cruzi-specific CD8+ T cells derived from IL-17RA-deficient mice presented features of cell dysfunction. PD-L1 blockade partially restored the magnitude of CD8+ T cell responses and parasite control in these mice. Adoptive transfer experiments established that IL-17RA-signaling is intrinsically required for the proper maintenance of functional effector CD8+ T cells. Altogether, our results identify IL-17RA and IL-17A as critical factors for sustaining CD8+ T cell immunity to T. cruzi.Fil: Tosello Boari, Jimena.
