    Microbiological and Molecular Characterization of Staphylococcus hominis Isolates from Blood

    Background: Among Coagulase-Negative Staphylococci (CoNS), Staphylococcus hominis represents the third most common organism recoverable from the blood of immunocompromised patients. The aim of this study was to characterize biofilm formation, antibiotic resistance, define the SCCmec (Staphylococcal Chromosomal Cassette mec) type, and genetic relatedness of clinical S. hominis isolates. Methodology: S. hominis blood isolates (n = 21) were screened for biofilm formation using crystal violet staining. Methicillin resistance was evaluated using the cefoxitin disk test and the mecA gene was detected by PCR. Antibiotic resistance was determined by the broth microdilution method. Genetic relatedness was determined by pulsed-field gel electrophoresis (PFGE) and SCCmec typed by multiplex PCR using two different methodologies described for Staphylococcus aureus. Results: Of the S. hominis isolates screened, 47.6% (10/21) were categorized as strong biofilm producers and 23.8% (5/21) as weak producers. Furthermore, 81% (17/21) of the isolates were methicillin resistant and mecA gene carriers. Resistance to ampicillin, erythromycin, and trimethoprim was observed in .70% of isolates screened. Each isolate showed a different PFGE macrorestriction pattern with similarity ranging between 0–95%. Among mecA-positive isolates, 14 (82%) harbored a non-typeable SCCmec type: eight isolates were not positive for any ccr complex; four contained the mec complex A ccrAB1 and ccrC, one isolate contained mec complex A, ccrAB4 and ccrC, and one isolate contained the mec complex A, ccrAB1, ccrAB4, and ccrC. Two isolates harbored the association: mec complex A and ccrAB1. Only one strain was typeable as SCCmec III. Conclusions: The S. hominis isolates analyzed were variable biofilm producers had a high prevalence of methicillin resistance and resistance to other antibiotics, and high genetic diversity. The results of this study strongly suggested that S. hominis isolates harbor new SCCmec structural elements and might be reservoirs of ccrC1 in addition to ccrAB1 and mec complex A

    Conducting Polymers Films Deposited on Carbon Steel and Their Interaction with Crude Oil

    The formation of scale/solids deposits inside the pipelines is a frequent problem in the petrochemical industry. These scales can be organic as the asphaltenes and inorganic as the accumulations of salts, which apart from blocking the inside of the pipes can also cause a change in the integrity of the steel. Therefore, it is necessary to avoid the conditions where deposition occurs, together with chemical and mechanical methods of remediation to mitigate the deposition. In this work we intend to use conductive polymers in order to inhibit the deposition of asphaltenes on carbon steel surfaces, by using polypyrrole (PPy) as material capable of conducting electrical current. The electrodeposition of PPy on carbon steel were performed by cyclic voltammetry (CV) and chronoamperometry (CA). The results showed that under certain experimental conditions it is possible to make a PPy film with adequate characteristics. Important factors were the grip and electrochemical stability of the formed film on steel, which depends on the electrosynthesis technique and in some cases favoured by a pre-treatment with a 10% HNO3 solution applied to the steel prior to electropolymerization. The PPy films deposited with pre-treatment completely covered the steel surface and showed better stability, adherence and generated a hydrophobic material

    Ameghiniana was created 50 years ago. During this lapse, the late Cenozoic paleomastozoology developed exponentially in Argentina. Many of the papers dealed with systematics. However, fossil mammals were also used for establish the chronological scale based on a biostratigraphic sequence of faunas mainly from the Pampean area. This scale proved valid for other South American areas. Besides, mammals were used as proxies for understanding the continental climatic evolution and, at the same time, biogeographic studies flourished. In recent years, many paleoecological and morphofunctional studies were carried out. Certainly, the last 50 years were the most productive time in Argentina since the pioneering work of Florentino Ameghino

    Much is said about nutrition, and for a relatively short time we have been talking about the importance of nutrition on the brain and, therefore, on cognitive performance. A correct diet in sports performance is a priority, but will it guarantee success in competitions? Obviously not, but a wrong diet, almost certainly, will lead us to a sporting and health disaster. The same premise applies to cognitive performance. Will eating properly guarantee academic results? No, but it will facilitate better neuronal connections, more fluid information, and better oxygenation of the brain, which in the end will allow us to obtain better cognitive results. And these behaviours should be avoided in conditions where students are confined for long periods of study during exams, due to illness or any other academic activity. This is why the importance of sociodemographic factors in the dietary preferences of children at an early age is vital. We could even go further by echoing the findings of some studies that suggest that poor food choices, rich in saturated fat, salt and sugar, in preschool age are associated with reduced verbal and cognitive ability scores. Sometimes opinions emerge as to whether breakfast is appropriate or not, in which case the balance strongly shifts more toward “ adequate” breakfast. We wanted to go a bit further by analysing the different ingredients that influence the various brain functions and/or connections, for their integration into students’ daily diets. But to integrate this model it is not enough to tell the students what is right and what is not, but in this case, we have intended that they are the ones who discover them; and use them to develop different recipes so that the message reaches the rest of their classmates better.     Molecular epidemiology of coagulase-negative bloodstream isolates: detection of Staphylococcus epidermidis ST2, ST7 and linezolid-resistant ST23

    The mechanisms contributing to persistence of coagulase-negative staphylococci are diverse; to better understanding of their dynamics, the characterization of nosocomial isolates is needed. Our aim was to characterize phenotypic and molecular characteristics of Staphylococcus epidermidis and Staphylococcus haemolyticus human blood isolates from two tertiary care hospitals in Mexico, the Hospital Universitario in Monterrey and the Hospital Civil in Guadalajara. Antimicrobial susceptibility was determined. Biofilm formation was assessed by crystal violet staining. Detection of the ica operon and Staphylococcal Cassette Chromosome mec typing were performed by PCR. Clonal relatedness was determined by Pulsed-fiel gel electrophoresis and Multi locus sequence typing. Methicillin-resistance was 85.5% and 93.2% for S. epidermidis and S. haemolyticus, respectively. Both species showed resistance >70% to norfloxacin, clindamycin, levofloxacin, trimethoprim/sulfamethoxazole, and erythromycin. Three S. epidermidis and two S. haemolyticus isolates were linezolid-resistant (one isolate of each species was cfr+). Most isolates of both species were strong biofilm producers (92.8% of S. epidermidis and 72.9% of S. haemolyticus). The ica operon was amplified in 36 (43.4%) S. epidermidis isolates. SCCmec type IV was found in 47.2% of the S. epidermidis isolates and SCCmec type V in 14.5% of S. haemolyticus isolates. No clonal relatedness was found in either species. Resistance to clindamycin, levofloxacin, erythromycin, oxacillin, and cefoxitin was associated with biofilm production for both species (p < 0.05). A G2576T mutation in 23S rRNA gene was detected in an S. haemolyticus linezolid-resistant isolate. All linezolid-resistant S. epidermidis isolates belonged to ST23; isolate with SCCmec type IV belonged to ST7, and isolate with SCCmec type III belonged to ST2. This is the first report of ST7 in Mexico. There was a high genetic diversity in both species, though both species shared characteristics that may contibute to virulence

    Curso modelado de nicho ecológico, version 1.0

    The suite of ideas, protocols, and software tools that has come to be known as “Ecological Niche Modeling” (ENM) — as well as those for the related “Species Distribution Modeling” (SDM)—has seen intensive exploration and research attention in recent decades. In spite of at least four syntheses, the field has grown so much in complexity that it is rather difficult to access for newcomers. Until now, accessibility to this field was achieved by in-person courses organized by universities or research centers, in some of which we have participated as instructors. However, the access to these specialized courses is limited, on one hand because they are not offered in all universities, and on the other because normally they are taught in English. To expand the access to a wider community of Spanish-speaking researchers, here we offer an entirely digital and free-of-charge course in Spanish, which was presented over 23 weeks via Internet in 2018. Although intrinsic Internet-related barriers may limit access to course materials, we have made them available in diverse formats (video, audio, pdf) in order to eliminate most of these problems.El conjunto de ideas, métodos y programas informáticos que se conoce como “Modelado de Nicho Ecológico” (MNE)—y el relacionado “Modelado de Distribución de Especies” (MDS)—han sido objeto de intensa exploración e investigación en las últimas décadas. A pesar de existir al menos cuatro síntesis publicadas, este campo ha crecido tanto en complejidad, que la formación de nuevos investigadores es difícil. Hasta ahora, dicha formación se ha hecho de manera presencial en cursos organizados por universidades o centros de investigación, de los que hemos formado parte como instructores. Sin embargo, el acceso a este tipo de cursos especializados es restringido, por un lado, porque los cursos no se ofrecen en todas las universidades, y por otro, porque normalmente se imparten en inglés. Para facilitar el acceso a una mayor comunidad de científicos de habla hispana, presentamos un curso en español, completamente digital y de acceso gratuito, que se realizó vía Internet durante 23 semanas consecutivas en 2018. Aunque las barreras intrínsecas al uso de Internet pueden dificultar la accesibilidad a los materiales del curso, hemos usado diversos formatos para la divulgación de los contenidos académicos (video, audio, pdf) con el objetivo de eliminar la mayor parte de estos problemas

    Characteristics and outcomes of adult patients in the PETHEMA registry with relapsed or refractory FLT3-ITD mutation-positive acute myeloid leukemia

    This retrospective study investigated outcomes of 404 patients with relapsed/refractory (R/R) FMS-like tyrosine kinase 3 (FLT3)-internal tandem duplication (ITD) acute myeloid leukemia (AML) enrolled in the PETHEMA registry, pre-approval of tyrosine kinase inhibitors. Most patients (63%) had received first-line intensive therapy with 3 + 7. Subsequently, patients received salvage with intensive therapy (n = 261), non-intensive therapy (n = 63) or supportive care only (n = 80). Active salvage therapy (i.e., intensive or non-intensive therapy) resulted in a complete remission (CR) or CR without hematological recovery (CRi) rate of 42%. More patients achieved a CR/CRi with intensive (48%) compared with non-intensive (19%) salvage therapy (p < 0.001). In the overall population, median overall survival (OS) was 5.5 months; 1- and 5-year OS rates were 25% and 7%. OS was significantly (p < 0.001) prolonged with intensive or non-intensive salvage therapy compared with supportive therapy, and in those achieving CR/CRi versus no responders. Of 280 evaluable patients, 61 (22%) had an allogeneic stem-cell transplant after they had achieved CR/CRi. In conclusion, in this large cohort study, salvage treatment approaches for patients with FLT3-ITD mutated R/R AML were heterogeneous. Median OS was poor with both non-intensive and intensive salvage therapy, with best long-term outcomes obtained in patients who achieved CR/CRi and subsequently underwent allogeneic stem-cell transplant.This study was supported by Centro de Investigación Biomédica en Red Cáncer (CIBERONC), Valencia, Spain [CB16/12/00284]
