    Use of Foliar Chemical Treatments to Induce Disease Resistance in Rhododendrons Inoculated with Phytophthora ramorum

    A field study was conducted at the National Ornamental Research Site at Dominican University California (NORS-DUC). The study goal was to evaluate three chemical inducers applied as foliar treatments for controlling Phytophthora ramorum, on Rhododendron x ‘Cunningham’s White’ nursery plants. The inducers were chlorine dioxide (ElectroBiocide), hydrogen peroxide (OxiDate 2.0), and acibenzolar-s methyl (Actigard). Water samples from the electrostatic sprayer were measured for three physicochemical water properties. Visual assessment of plant foliage, based on the Horsfall- Barratt scale, was conducted at three and five months after chemical treatments. Foliar fluorescence (Fv/Fm) was measured over three dates. The success of P. ramorum inoculations were determined using qPCR methods. Visual assessment across both months showed no signs of P. ramorum infection or chemical injury symptoms. However, P. ramorum infection vis-à-vis qPCR analysis was confirmed. The September Fv/Fm results revealed that all the chemical inducer treatments were equivalent to the water treatment, except for Actigard. The qPCR results were in general agreement with the Fv/Fm results indicating that the rhododendrons were successfully inoculated with P. ramorum but were non-symptomatic. The electrostatic sprayer ionized the water droplets, resulting in increased Fv/Fm values for the water treatments 90 days after application. There was a three-month delay in fluorescence responses to the most effective chemical applications, indicating that woody plants may need to be monitored over the long term to determine accurate responses to foliar treatments

    Evaluation of an inter-professional workshop to develop a psychosocial assessment and child-centred communication training programme for paediatricians in training

    BACKGROUND: The quality of psychosocial assessment of children in consultations varies widely. One reason for this difference is the variability in effective mental health and communication training at undergraduate and post-qualification levels. In recognition of this problem, the Royal College of Paediatrics and Child Health in the United Kingdom have developed the Child in Mind Project that aims to meet this deficit in medical training. This paper describes the evaluation of a workshop that explored the experiences and expectations of health care professionals in the development of a training programme for doctors. METHODS: The one-day inter-professional workshop was attended by 63 participants who were invited to complete evaluation forms before and immediately after the workshop. RESULTS: The results showed that the workshop was partially successful in providing an opportunity for an inter-professional group to exchange ideas and influence the development of a significant project. Exploring the content and process of the proposed training programme and the opportunity for participants to share experiences of effective practice were valued. Participants identified that the current culture within many health care settings would be an obstacle to successful implementation of a training programme. Working within existing training structures will be essential. Areas for improvement in the workshop included clearer statement of goals at the outset and a more suitable environment for the numbers of participants. CONCLUSIONS: The participants made a valuable contribution to the development of the training programme identifying specific challenges. Inter-professional collaborations are likely to result in more deliverable and relevant training programmes. Continued consultation with potential users of the programme – both trainers and trainees will be essential

    Impact of maternal ART on mother-to-child transmission (MTCT) of HIV at six weeks postpartum in Rwanda

    BACKGROUND: In 2010, Rwanda adopted ART for prevention of mother to child transmission of HIV from pregnant women living with HIV during pregnancy and breasfeeding period. This study examines rates of mother-to-childtransmission of HIV at 6–10 weeks postpartum and risk factors for mother-to-child transmission of HIV (MTCT) among HIV infected women on ART during pregnancy and breastfeeding. METHODS: A cross-sectional survey study was conducted between July 2011–June 2012 among HIV-exposed infants aged 6–10 weeks and their mothers/caregivers. Stratified multi-stage, probability proportional to size and systematic sampling to select a national representative sample of clients. Consenting mothers/caregivers were interviewed on demographic and program interventions. Dry blood spots from HIV-exposed infants were collected for HIV testing using DNA PCR technique. Results are weighted for sample realization. Univariable analysis of socio-demographic and programmatic determinants of early mother-to-child transmission of HIV was conducted. Variables were retained for final multivariable models if they were either at least of marginal significance (p-value < 0.10) or played a confounding role (the variable had a noticeable impact > 10% change on the effect estimate). RESULTS: The study sample was 1639 infants with HIV test results. Twenty-six infants were diagnosed HIV-positive translating to a weighted MTCT estimate of 1.58% (95% CI 1.05–2.37%). Coverage of most elimination of MTCT (EMTCT) program interventions, was above 80, and 90.4% of mother-infant pairs received antiretroviral treatment or prophylaxis. Maternal ART and infant antiretroviral prophylaxis (OR 0.01; 95%CI 0.001–0.17) and maternal age older than 25 years were significantly protective (OR 0.33; 95%CI 0.14–0.78). No disclosure of HIV status, not testing for syphilis during pregnancy and preterm birth were significant risk factors for MTCT. Factors suggesting higher sociodemographic status (flush toilet, mother self-employed) were borderline risk factors for MTCT. CONCLUSION: ART for all women during pregnancy and breastfeeding was associated with the estimated low MTCT rate of 1.58%. Mothers who did not receive a full package of anti-retroviral therapy according to the Rwanda EMTCT protocol, and young and single mothers were at higher risk of MTCT and should be targeted for support in preventing HIV infection

    Cytotoxic and HIV-1 enzyme inhibitory activities of Red Sea marine organisms

    BACKGROUND: Cancer and HIV/AIDS are two of the greatest public health and humanitarian challenges facing the world today. Infection with HIV not only weakens the immune system leading to AIDS and increasing the risk of opportunistic infections, but also increases the risk of several types of cancer. The enormous biodiversity of marine habitats is mirrored by the molecular diversity of secondary metabolites found in marine animals, plants and microbes which is why this work was designed to assess the anti-HIV and cytotoxic activities of some marine organisms of the Red Sea. METHODS: The lipophilic fractions of methanolic extracts of thirteen marine organisms collected from the Red Sea (Egypt) were screened for cytotoxicity against two human cancer cell lines; leukaemia (U937) and cervical cancer (HeLa) cells. African green monkey kidney cells (Vero) were used as normal non-malignant control cells. The extracts were also tested for their inhibitory activity against HIV-1 enzymes, reverse transcriptase (RT) and protease (PR). RESULTS: Cytotoxicity results showed strong activity of the Cnidarian Litophyton arboreum against U-937 (IC50; 6.5 μg/ml ±2.3) with a selectivity index (SI) of 6.45, while the Cnidarian Sarcophyton trochliophorum showed strong activity against HeLa cells (IC50; 5.2 μg/ml ±1.2) with an SI of 2.09. Other species showed moderate to weak cytotoxicity against both cell lines. Two extracts showed potent inhibitory activity against HIV-1 protease; these were the Cnidarian jelly fish Cassiopia andromeda (IC50; 0.84 μg/ml ±0.05) and the red algae Galaxura filamentosa (2.6 μg/ml ±1.29). It is interesting to note that the most active extracts against HIV-1 PR, C. andromeda and G. filamentosa showed no cytotoxicity in the three cell lines at the highest concentration tested (100 μg/ml). CONCLUSION: The strong cytotoxicity of the soft corals L. arboreum and S. trochliophorum as well as the anti-PR activity of the jelly fish C. andromeda and the red algae G. filamentosa suggests the medicinal potential of crude extracts of these marine organisms.The Medical Research Council, the Technology Innovation Agency and the University of Pretoria, South Africa.http://www.biomedcentral.com/bmccomplementalternmedam201

    Genome-wide association study identifies 32 novel breast cancer susceptibility loci from overall and subtype-specific analyses.

    Breast cancer susceptibility variants frequently show heterogeneity in associations by tumor subtype1-3. To identify novel loci, we performed a genome-wide association study including 133,384 breast cancer cases and 113,789 controls, plus 18,908 BRCA1 mutation carriers (9,414 with breast cancer) of European ancestry, using both standard and novel methodologies that account for underlying tumor heterogeneity by estrogen receptor, progesterone receptor and human epidermal growth factor receptor 2 status and tumor grade. We identified 32 novel susceptibility loci (P < 5.0 × 10-8), 15 of which showed evidence for associations with at least one tumor feature (false discovery rate < 0.05). Five loci showed associations (P < 0.05) in opposite directions between luminal and non-luminal subtypes. In silico analyses showed that these five loci contained cell-specific enhancers that differed between normal luminal and basal mammary cells. The genetic correlations between five intrinsic-like subtypes ranged from 0.35 to 0.80. The proportion of genome-wide chip heritability explained by all known susceptibility loci was 54.2% for luminal A-like disease and 37.6% for triple-negative disease. The odds ratios of polygenic risk scores, which included 330 variants, for the highest 1% of quantiles compared with middle quantiles were 5.63 and 3.02 for luminal A-like and triple-negative disease, respectively. These findings provide an improved understanding of genetic predisposition to breast cancer subtypes and will inform the development of subtype-specific polygenic risk scores

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Factors Associated with Revision Surgery after Internal Fixation of Hip Fractures

    Background: Femoral neck fractures are associated with high rates of revision surgery after management with internal fixation. Using data from the Fixation using Alternative Implants for the Treatment of Hip fractures (FAITH) trial evaluating methods of internal fixation in patients with femoral neck fractures, we investigated associations between baseline and surgical factors and the need for revision surgery to promote healing, relieve pain, treat infection or improve function over 24 months postsurgery. Additionally, we investigated factors associated with (1) hardware removal and (2) implant exchange from cancellous screws (CS) or sliding hip screw (SHS) to total hip arthroplasty, hemiarthroplasty, or another internal fixation device. Methods: We identified 15 potential factors a priori that may be associated with revision surgery, 7 with hardware removal, and 14 with implant exchange. We used multivariable Cox proportional hazards analyses in our investigation.      Agricultural machinery and farm equipment are potential sources of infectious material that can lead to the contamination and spread of diseases if proper action isn’t taken. Two stage decontamination methods, involving power washing followed by disinfectant applications, are needed to clean farm equipment, agricultural transport vehicles, and storage units. The field experiments confirm that pressure washing surfaces is an extremely important step in enhancing spore efficacy. Log10 reduction values were 5.45 and 2.90 for disinfectant applications with and without power washing, respectively. Both experiments show that the commercial chlorine dioxide disinfectant Electro-Biocide was an effective disinfectant alone and when mixed with adjuvants. Increasing the concentration of some tested adjuvants resulted in more spores being removed or killed, however this was not true for all adjuvants tested in these two experiments

    Two Stage Decontamination of Agricultural Equipment Using Power Washing Followed by Disinfectant Treatments

     Foreign agricultural pests can become problematic to the environment, economy, animal, plant, and human health if widely transported on contaminated equipment or vehicles. Two equipment decontamination studies were conducted using a mobile power washer and disinfectant treatments. The first study factors were: steel and fabric surfaces, power washing conditions, disinfectants, and disinfectant adjuvants. The second study factors were: relative humidity conditions, disinfectant type, disinfectant additive, and number of repeat disinfectant applications. Efficacy for the power washing and disinfectant treatments was based on log10 reduction of Bacillus subtilis spores attached to the two surface types. Power washing increased log10 reduction of spores by 3 to 4 log, when washing was followed by disinfectant treatments. The optimal decontamination treatment in the first study was power washing for 30 seconds at a distance of 10 cm, using a commercial chlorine dioxide formulation (Electro-Biocide) with Reign (1%) that resulted in a 4.7 log10 reduction of B. subtilis spores on steel washers. In the second study the optimal treatment was power washing for 10 seconds at a nozzle distance of 20 cm with a commercial disinfectant (Easy Decon DF-200) mixed with 20% glycerol resulting in a 5.1 log10 reduction of B. subtilis spores on wool fabric samples
