Caloric restriction blocks neuropathology and motor deficits in Machado–Joseph disease mouse models through SIRT1 pathway

    Get PDF
Machado–Joseph disease (MJD) is a neurodegenerative disorder characterized by an abnormal expansion of the CAG triplet in the ATXN3 gene, translating into a polyglutamine tract within the ataxin-3 protein. The available treatments only ameliorate symptomatology and do not block disease progression. In this study we find that caloric restriction dramatically rescues the motor incoordination, imbalance and the associated neuropathology in transgenic MJD mice. We further show that caloric restriction rescues SIRT1 levels in transgenic MJD mice, whereas silencing SIRT1 is sufficient to prevent the beneficial effects on MJD pathology. In addition, the re-establishment of SIRT1 levels in MJD mouse model, through the gene delivery approach, significantly ameliorates neuropathology, reducing neuroinflammation and activating autophagy. Furthermore, the pharmacological activation of SIRT1 with resveratrol significantly reduces motor incoordination of MJD mice. The pharmacological SIRT1 activation could provide important benefits to treat MJD patients.Fonds Europeen de Developpement Economique et Regional. The presense of C. diphteriae toxin was demonstrated in throat lesions of 47.7 per cent of case with probable diagnosis of diphtheria, by passive hemagglutination test using erithrocytes previously sensitized by diphtheria anti-toxin. From 53.0 per cent of the same patients toxigenous bacilli were isolated. Hemagglutination test was the only positive one in 13.6 per cent of cases and cultivation in 18.9 per cent. The passive hemagglutination tests performed on 42 children, normal or with pharyngitis or tonsillitis but without clinical characteristics of diphtheria were all negative. The procedure described in this paper, of very simple execution can show results in less than two hours and offers great possibility of advantageous application in the diagnosis of diphtheria.

Static and Fatigue Behaviour of the Main Section of a Fast Patrol Boat

This paper analyses the static resistance, as well as the fatigue resistance, of the main section of a fast patrol boat designed and manufactured in Portugal. The ship under study is a high-speed lightweight craft that was mainly fabricated with two types of aluminium series alloys, namely the 5083-H111 and the 6082-T6 alloys, which are commonly used in shipbuilding. The structural response of the critical section of the ship was obtained using the Finite Element Method (FEM), when the structure was submitted to different loading conditions, such as the hydrodynamic and hydrostatic sea loads, the longitudinal sagging or hogging, or the loads on fuel oil tanks. Results obtained for the load cases considered showed that the Von Mises equivalent stresses do not exceed the Yield Strength of the aluminium alloys used in the manufacturing of the ship. In addition, strain gages were placed on the main ship's bulkhead, near a structural detail in its bottom, and real-time acquisition strain data was collected using a computer code routine written in LabView. The Rainflow cycle counting method was applied to load spectrum gathered in order to obtain the Rainflow matrix and to predict the fatigue life of the critical main section of the ship.publishersversionpublishe

A survey of RNA viruses in mosquitoes from Mozambique reveals novel genetic lineages of flaviviruses and phenuiviruses, as well as frequent flavivirus-like viral DNA forms in Mansonia

Background: Mosquito-borne diseases involving arboviruses represent expanding threats to sub-Saharan Africa imposing as considerable burden to human and veterinary public health. In Mozambique over one hundred species of potential arbovirus mosquito vectors have been identified, although their precise role in maintaining such viruses in circulation in the country remains to be elucidated. The aim of this study was to screen for the presence of flaviviruses, alphaviruses and bunyaviruses in mosquitoes from different regions of Mozambique. Results: Our survey analyzed 14,519 mosquitoes, and the results obtained revealed genetically distinct insect-specific flaviviruses, detected in multiple species of mosquitoes from different genera. In addition, smaller flavivirus-like NS5 sequences, frequently detected in Mansonia seemed to correspond to defective viral sequences, present as viral DNA forms. Furthermore, three lineages of putative members of the Phenuiviridae family were also detected, two of which apparently corresponding to novel viral genetic lineages. Conclusion: This study reports for the first-time novel insect-specific flaviviruses and novel phenuiviruses, as well as frequent flavivirus-like viral DNA forms in several widely known vector species. This unique work represents recent investigation of virus screening conducted in mosquitoes from Mozambique and an important contribution to inform the establishment of a vector control program for arbovirus in the country and in the region.publishersversionpublishe

Radio‐cephalic fistula recovered with drainage to median basilic vein by straightening of the forearm cephalic vein: A case report

The distal arteriovenous fistula (AVF) has less complications and better patency than the proximal fistula, when it works properly. The complications of outflow of the fistula are complex, and it is necessary to analyze all solutions to solve the problem. We introduce a novel approach to solve outflow problems of a radio-cephalic AVF by straightening of the forearm cephalic vein with drainage into the median basilic vein.info:eu-repo/semantics/publishedVersio

Mechanics of epithelial closure over non-adherent environments

    Get PDF
International audienceThe closure of gaps within epithelia is crucial to maintain its integrity during biological processes such as wound healing and gastrulation. Depending on the distribution of extracellular matrix, gap closure occurs through assembly of multicellular actin-based contractile cables or protrusive activity of border cells into the gap. Here we show that the supracellular actomyosin contractility of cells near the gap edge exerts sufficient tension on the surrounding tissue to promote closure of non-adherent gaps. Using traction force microscopy, we observe that cell-generated forces on the substrate at the gap edge first point away from the centre of the gap and then increase in the radial direction pointing into the gap as closure proceeds. Combining with numerical simulations, we show that the increase in force relies less on localized purse-string contractility and more on large-scale remodelling of the suspended tissue around the gap. Our results provide a framework for understanding the assembly and the mechanics of cellular contractility at the tissue level
