Anwendung von Synchrotronstrahlung zur Charakterisierung von Biomineralien
In der vorliegenden Arbeit wurden mehrere Projekte beschrieben, bei denen Röntgenpulverdiffraktometrie und/oder Mikrocomputertomographie mit Synchrotron-strahlung eingesetzt wurden.
Die Untersuchung des ZebrabĂ€rblings hat gezeigt, dass auch die hochauflösende SR”CT fĂŒr âgroĂeâ Objekte, wie einen drei Zentimeter langen Fisch, möglich ist. Der ZebrabĂ€rbling selber eignet sich gut als Modelorganismus fĂŒr die Untersuchung des Mineralisationsprozesses in ZĂ€hnen, da diese permanent ersetzt werden. Hier könnte der Einfluss verschiedener biologischer Faktoren und UmwelteinflĂŒsse untersucht werden. Die Zahnbildung im ZebrabĂ€rbling ist ein schneller Prozess, der in ca. neun bis elf Tagen ablĂ€uft. Somit könnte auch der Einfluss der Umweltbedingungen auf die Zahnbildung in diesem Organismus untersucht werden.
Ein anderer Modelorganismus in der Biologie ist der Medaka. Da das Genom und der Aufbau der Zellen unter den Wirbeltieren hoch konserviert sind, eignen sich Fische wie der Medaka und der ZebrabÀrbling gut als Modelorganismen um Knochenkrankheiten zu untersuchen. In dieser Arbeit wurde das Skelett eines gesunden und eines Medaka mit idiopatischen Deformationen untersucht. Es ist von höheren Wirbeltieren bekannt, dass die Verformung der WirbelsÀule durch missgebildete oder fehlende Teile hervorgerufen werden kann. Die Ursache der Deformierung des Medakas dieser Arbeit waren deformierte Knochen. Die Deformation betraf nicht nur die WirbelsÀule, sondern auch andere Knochen wie bespielsweise die Rippen. SR”CT ist hier eine hervorragend geeignete Methode, da eine Untersuchung mit Hilfe der Alizarin-Rot-FÀrbung beim ZebrabÀrbling gezeigt hat, dass damit die Knochenstruktur verÀndert wird. Um die UmstÀnde der idiopatischen Deformierung genauer zu untersuchen, könnten SR”CT Experimente an Embryonen und jungen Fischen bei verschiedenen Stadien weitere Informationen liefern.
Im Projekt das im Folgenden beschrieben wird, wurden Teile von Lachswirbeln untersucht, die wĂ€hrend der Wirbelbildung nacheinander gebildet wurden. Die Teile wurden mit Röntgenpulverdiffraktometrie und Thermogravimetrie untersucht. Es konnte nur ein signifikanter Unterschied gefunden werden. Der Carbonatgehalt des zu Beginn gebildeten Minerals war doppelt so hoch wie bei den anderen Proben. Nach Aussage der Projektpartner wird dieses Material nicht von Osteoblasten mineralisiert, somit könnte dies der Grund fĂŒr den Unterschied sein. Ansonsten stimmten KristallgröĂe und Zusammensetzung weitestgehend untereinander und verglichen mit humanen Knochen ĂŒberein.
Analog zu den Wirbelteilen wurden Mausknochen untersucht. Diese Knochen stammten von normalen und sogenannten Fetuin A-knock-out MĂ€usen. Bei den knock-out MĂ€usen allgemein handelt es sich um Tiere, bei denen ein Gen ausgeschaltet wurde. In diesem Fall wurde das Gen fĂŒr Fetuin-A ausgeschaltet. Fetuin-A verhindert im Körper das Ausfallen von Hydroxylapatit. Projektpartner hatten Fetuin-A im Knochen nachgewiesen und deshalb eine Beteiligung bei der Knochenbildung vermutet. Die Knochen der verschiedenen Tiere zeigten keine signifikanten Unterschiede in Zusammensetzung und KristallitgröĂe. Auf Grund der Ergebnisse konnte eine Funktion des Fetuin-A bei der Knochenbildung nicht bestĂ€tigt, aber auch nicht ausgeschlossen werden. Hier sind weitere Untersuchungen nötig, um einen Aufschluss zu liefern.
Fluorotische ZÀhne entstehen, wenn bei der Odontogenese zu viel Fluorid aufgenommen wird. Sie zeigen eine VerfÀrbung des Enamels und Dentins. Der Vergleich eines fluorotischen mit einem gesunden Zahn mittels SR”CT zeigte nicht die erwarteten Unterschiede in der Mineralisation.
Einen Schwerpunkt dieser Arbeit stellt die Untersuchung der Kutikula verschiedener Asselarten dar. Das Exoskelett dieser Tiere muss verschiedenen AnsprĂŒchen dienen. AuĂerdem muss es, da es nicht mitwĂ€chst, von Zeit zu Zeit abgeworfen und erneuert werden. Um den Mineralisationsprozess zu verstehen, wurde die Mineralisation der Kutikula von Porcellio scaber bei verschiedenen Zeitpunkten der HĂ€utung untersucht. Es wurden die anteriore und die posteriore HĂ€lfte der Tiere miteinander verglichen. Mittels Röntgenpulverdiffraktometrie und AAS wurde das VerhĂ€ltnis von ACC/Calcit bestimmt und ĂŒber die HĂ€utung verfolgt. Die Gesamtmineralisation wurde mittels quantitativer Auswertung der SR”CT-Daten ermittelt. Hier werden die prĂ€sentierten Ergebnisse noch durch Ramanspektroskopie und Elektronenmikroskopie durch die Projektpartner erweitert. Danach erfolgt die biologische Bewertung. Wie schon erwĂ€hnt, sind die AnsprĂŒche an die Kutikula von Tier zu Tier je nach Verhalten und Lebensraum unterschiedlich. In der Untersuchung der Kutikula von Porcellio scaber und Armadillidium vulgare wurde ein Zusammenhang zwischen Verhalten und Zusammensetzung der Kutikula beobachtet. In dieser Arbeit wurde die Zusammensetzung zusĂ€tzlich mit dem Lebensraum verglichen. Dazu wurden vier weitere terrestrische und vier marine Spezies untersucht. Die Unterschiede zwischen âRollernâ und ânon-Rollernâ sind sowohl terrestrisch als auch marin gleich. Der hohe Gehalt von ACC in terestrischen Tieren kann durch die schlechte VerfĂŒgbarkeit von Calciumcarbonat wĂ€hrend der HĂ€utung erklĂ€rt werden. ACC hat eine höhere Löslichkeit wie Calcit, was fĂŒr die Demineralisation von Vorteil ist. FĂŒr die marinen Spezies kann dies nicht erklĂ€rt werden. Da der Zusammenhang zwischen Verhalten und Zusammensetzung der Kutikula weiter untersucht werden sollte, wurden weitere Tiere analog analysiert. FĂŒr diese Ergebnisse steht die biologische Bewertung noch aus.
Die Untersuchung der Embryonen von Biomphalaria glabrata durch SR”CT und Rasterelektronenmikroskopie zeigt die Entwicklung der Schale bei verschiedenen Zeitpunkten der Entwicklung. Es konnte das Gewicht der Schale verglichen mit dem Alter der Tiere dargestellt werden.
Es wurden noch SR”CT-Aufnahmen von zwei Australseepocken erstellt, von denen eine gezielt beschÀdigt wurde. In diesem Projekt kann die Mineralisation und Selbstheilung des GehÀuses durch weitere Studien untersucht werden. ZusÀtzlich wÀre es sinnvoll, das GehÀuse auch mit Röntgenpulver-diffraktometrie und durch thermogravimetrische Analysen zu untersuchen, um weitere Informationen zu erhalten.
Die hochauflösende Röntgenpulverdiffraktometrie mit Synchrotronstrahlung zeigte sich fĂŒr diese Arbeit als besonders nĂŒtzlich, da die quantitative Phasenanalyse durch das gute VerhĂ€ltnis des Signals zum Untergrundrauschen sowie eine geringe GerĂ€teverbreiterung der Reflexe genauere Ergebnisse liefert. Auch die Bestimmung der KristallitgröĂe mit der Scherrer-Gleichung wird dadurch genauer.
Als ergĂ€nzende Methode fĂŒr Studien auf dem Gebiet der Biomineralisation eröffnet die auf Synchrotronstrahlung basierende Mikrocomputertomographie neue Möglichkeiten. Die Auflösung der Computertomographie allgemein reicht von 1-2 mm fĂŒr Proben in der GröĂenordnung von mehreren Dezimetern fĂŒr medizinische Computertomographie. Bei Laborquellen liegt die Auflösung bei wenigen 10 ”m (GröĂenordnung: wenige Zentimeter). FĂŒr SR”CT liegt sie bei 1-3 ”m (GröĂenordnung: wenige Millimeter). Es wurde auch schon von einer Auflösung von 700 nm berichtet. Ein Vergleich von LaborgerĂ€ten und Mikrocomputertomographie mit Synchrotronstrahlung zeigte eine höhere Auflösung und weniger Artefakte fĂŒr SR”CT. Das Einsatzgebiet der Computertomographie allgemein ist unter anderem die Untersuchung von Knochen, ZĂ€hnen, KontaktflĂ€chen von Implan-taten und anderen Biomineralien. Ein Nachteil fĂŒr die SR”CT ist allerdings die lange Messzeit, besonders fĂŒr groĂe Proben (von drei bis zu 36 Stunden), da der Zugriff auf Strahlzeit am HASYLAB limitiert ist. Die beeindruckenden Ergebnisse rechtfertigen jedoch diesen Aufwand
Calcite distribution and orientation in the tergite exocuticle of the isopods porcellio scaber and armadillidium vulgare (Oniscidea, Crustacea) - A combined FE-SEM, polarized SCm-RSI and EBSD study
The crustacean cuticle is a bio-composite consisting of hierarchically organized chitin-protein fibres, reinforced with calcite, amorphous calcium carbonate and phosphates. Comparative studies revealed that the structure and composition of tergite cuticle of terrestrial isopods is adapted to the habitat of the animals, and to their behavioural patterns to avoid predation. In this contribution we use FE-SEM, polarized SCm-RSI and EBSD to investigate micro- and nano-patterns of mineral phase distribution and crystal orientation within the tergite cuticle of the two terrestrial isopod species Armadillidium vulgare and Porcellio scaber. The results show that the proximal regions of the exocuticle contain both calcite and ACC, with ACC located within the pore canals. Calcite forms hierarchically organised mesocrystalline aggregates of similar crystallographic orientation. Surprisingly, c-axis orientation preference is horizontal in regard to the local cuticle surface for both species, in contrast to mollusc and brachiopod shell structures in which the c-axis is always perpendicular to the shell surface. The overall sharpness of calcite crystal orientation is weak compared to that of mollusc shells. However, there are considerable differences in texture sharpness between the two isopod species. In the thick cuticle of the slow-walking A. vulgare calcite is more randomly oriented resulting in more isotropic mechanical properties of the cuticle. In contrast, the rather thin and more flexible cuticle of the fast- running P. scaber texture sharpness is stronger with a preference of c-axis orientation being parallel to the bilateral symmetry-plane of the animal, leading to more anisotropic mechanical properties of the cuticle. These differences may represent adaptations to different external and/or internal mechanical loads the cuticle has to resist during predatory attempts

Controls on gut phosphatisation : the trilobites from the Weeks Formation LagerstÀtte (Cambrian; Utah)
Despite being internal organs, digestive structures are frequently preserved in Cambrian LagerstÀtten. However, the reasons for their fossilisation and their biological implications remain to be thoroughly explored. This is particularly true with arthropods--typically the most diverse fossilised organisms in Cambrian ecosystems--where digestive structures represent an as-yet underexploited alternative to appendage morphology for inferences on their biology. Here we describe the phosphatised digestive structures of three trilobite species from the Cambrian Weeks Formation LagerstÀtte (Utah). Their exquisite, three-dimensional preservation reveals unique details on trilobite internal anatomy, such as the position of the mouth and the absence of a differentiated crop. In addition, the presence of paired pygidial organs of an unknown function is reported for the first time. This exceptional material enables exploration of the relationships between gut phosphatisation and the biology of organisms. Indeed, soft-tissue preservation is unusual in these fossils as it is restricted to the digestive structures, which indicates that the gut played a central role in its own phosphatisation. We hypothesize that the gut provided a microenvironment where special conditions could develop and harboured a source of phosphorus. The fact that gut phosphatization has almost exclusively been observed in arthropods could be explained by their uncommon ability to store ions (including phosphorous) in their digestive tissues. However, in some specimens from the Weeks Formation, the phosphatisation extends to the entire digestive system, suggesting that trilobites might have had some biological particularities not observed in modern arthropods. We speculate that one of them might have been an increased capacity for ion storage in the gut tissues, related to the moulting of their heavily-mineralised carapace
âA very orderly retreatâ: Democratic transition in East Germany, 1989-90
East Germany's 1989-90 democratisation is among the best known of East European transitions, but does not lend itself to comparative analysis, due to the singular way in which political reform and democratic consolidation were subsumed by Germany's unification process. Yet aspects of East Germany's democratisation have proved amenable to comparative approaches. This article reviews the comparative literature that refers to East Germany, and finds a schism between those who designate East Germany's transition âregime collapseâ and others who contend that it exemplifies âtransition through extricationâ. It inquires into the merits of each position and finds in favour of the latter. Drawing on primary and secondary literature, as well as archival and interview sources, it portrays a communist elite that was, to a large extent, prepared to adapt to changing circumstances and capable of learning from âreference statesâ such as Poland. Although East Germany was the Soviet state in which the positions of existing elites were most threatened by democratic transition, here too a surprising number succeeded in maintaining their position while filing across the bridge to market society. A concluding section outlines the alchemy through which their bureaucratic power was transmuted into property and influence in the ânew Germanyâ

HSPG-Deficient Zebrafish Uncovers Dental Aspect of Multiple Osteochondromas
Multiple Osteochondromas (MO; previously known as multiple hereditary exostosis) is an autosomal dominant genetic condition that is characterized by the formation of cartilaginous bone tumours (osteochondromas) at multiple sites in the skeleton, secondary bursa formation and impingement of nerves, tendons and vessels, bone curving, and short stature. MO is also known to be associated with arthritis, general pain, scarring and occasional malignant transformation of osteochondroma into secondary peripheral chondrosarcoma. MO patients present additional complains but the relevance of those in relation to the syndromal background needs validation. Mutations in two enzymes that are required during heparan sulphate synthesis (EXT1 or EXT2) are known to cause MO. Previously, we have used zebrafish which harbour mutations in ext2 as a model for MO and shown that ext2â/â fish have skeletal defects that resemble those seen in osteochondromas. Here we analyse dental defects present in ext2â/â fish. Histological analysis reveals that ext2â/â fish have very severe defects associated with the formation and the morphology of teeth. At 5 days post fertilization 100% of ext2â/â fish have a single tooth at the end of the 5th pharyngeal arch, whereas wild-type fish develop three teeth, located in the middle of the pharyngeal arch. ext2â/â teeth have abnormal morphology (they were shorter and thicker than in the WT) and patchy ossification at the tooth base. Deformities such as split crowns and enamel lesions were found in 20% of ext2+/â adults. The tooth morphology in ext2â/â was partially rescued by FGF8 administered locally (bead implants). Our findings from zebrafish model were validated in a dental survey that was conducted with assistance of the MHE Research Foundation. The presence of the malformed and/or displaced teeth with abnormal enamel was declared by half of the respondents indicating that MO might indeed be also associated with dental problems
Metabolic responses to high pCO2 conditions at a CO2 vent site in juveniles of a marine isopod species assemblage
We are starting to understand the relationship between metabolic rate responses and species' ability to respond to exposure to high pCO2. However, most of our knowledge has come from investigations of single species. The examination of metabolic responses of closely related species with differing distributions around natural elevated CO2 areas may be useful to inform our understanding of their adaptive significance. Furthermore, little is known about the physiological responses of marine invertebrate juveniles to high pCO2, despite the fact they are known to be sensitive to other stressors, often acting as bottlenecks for future species success. We conducted an in situ transplant experiment using juveniles of isopods found living inside and around a high pCO2 vent (Ischia, Italy): the CO2 'tolerant' Dynamene bifida and 'sensitive' Cymodoce truncata and Dynamene torelliae. This allowed us to test for any generality of the hypothesis that pCO2 sensitive marine invertebrates may be those that experience trade-offs between energy metabolism and cellular homoeostasis under high pCO2 conditions. Both sensitive species were able to maintain their energy metabolism under high pCO2 conditions, but in C. truncata this may occur at the expense of [carbonic anhydrase], confirming our hypothesis. By comparison, the tolerant D. bifida appeared metabolically well adapted to high pCO2, being able to upregulate ATP production without recourse to anaerobiosis. These isopods are important keystone species; however, given they differ in their metabolic responses to future pCO2, shifts in the structure of the marine ecosystems they inhabit may be expected under future ocean acidification conditions
