    Study of the effect of different fermenting microorganisms on the Se, Cu, Cr, and Mn contents in fermented goat and cow milks

    The aim of this study was to determine the Se, Cu, Cr, and Mn concentrations of different types of goat- and cow-milk fermented products and evaluate the influence of fermenting bacteria (classical fermenting starters and a probiotic strain) on these concentrations. Atomic absorption spectrometry with hydride generation was used to measure Se and electrothermal atomization to measure Cu, Cr and Mn. Analytical parameters determined in the fermented milks demonstrated that the procedures used were adequate for Se, Cu, Cr, and Mn analyses. Se levels were significantly lower in fermented goat milk products than in fermented cow milk products (p < 0.05). Se, Cu, Cr, and Mn levels did not differ as a function of the fermenting bacteria used in commercial fermented goat or cow milks or in the lab-produced goat yoghurt. Given the Se, and Cr intakes for healthy adults, goat and cow yogurts may be important dietary sources.Departamento de Nutrición y Bromatología, AGR-14

    Quantifying the motor efficiency via information theory

    En este trabajo se propone un método para cuantificar la eficiencia motriz, basado en métricas específicas de la teoría de la información. Así, una medida de información es obtenida a partir de registros electromiográficos (EMG) y características cinemáticas de la actividad motora. Las señales de EMG fueron obtenidas del músculo deltoides medio durante contracciones dinámicas evocadas por elevaciones laterales (movimientos de abducción y aducción del brazo) en el plano escapular. Esta actividad motora fue realizada a diferentes velocidades y bajo diferentes condiciones de carga. La eficiencia de la actividad motora fue evaluada para cada situación. El método propuesto resultó adecuado para situaciones en el cual la actividad motora es realizada bajo condiciones no estacionarias (contracciones dinámicas).A method based on Information Theory for quantify the movement efficiency was proposed. The measurement of information provides a quantitative metric of motor efficiency. This measure is obtained from electromyographic (EMG) recordings and the kinematic of motor activity. Here, the EMG signals from middle right deltoid were recorded during dynamic contraction. These contractions were evoked by abduction-adduction arm movements at scapular plane. The arm movements were realized at different speeds and under different loading conditions. The efficiency of motor activity was then evaluated for each situation.     Challenges in Diabetic Macular Edema Management: An Expert Consensus Report

    Purpose: This paper aimed to present daily-practice recommendations for the management of diabetic macular edema (DME) patients based on available scientific evidence and the clinical experience of the consensus panel. Methods: A group of Spanish retina experts agreed to discuss different aspects related with the clinical management of DME patients. Results: Panel was mainly focused on therapeutic objectives in DME management; defini-tion terms; and role of biomarkers as prognostic and predictive factors to intravitreal treatment response. The panel recommends to start DME treatment as soon as possible in those eyes with a visual acuity less than 20/25 (always according to the retina unit capacity). Naive patient was defined, in a strict manner, as a patient who, up to that moment, had never received any treatment. A refractory DME patient may be defined as the one who did not achieve a complete resolution of the disease, regardless of the treatment administered. Different optical coherence tomography biomarkers, such as disorganization of the retinal inner layers, hyperreflective dots, and cysts, have been identified as prognostic factors. Conclusion: This document has sought to lay down a set of recommendations and to identify key issues that may be useful for the daily management of DME patients

    Elevated α-Ketoglutaric Acid Concentrations and a Lipid-Balanced Signature Are the Key Factors in Long-Term HIV Control

    Long-term elite controllers (LTECs) are a fascinating small subset of HIV individuals with viral and immunological HIV control in the long term that have been designated as models of an HIV functional cure. However, data on the LTEC phenotype are still scarce, and hence, the metabolomics and lipidomics signatures in the LTEC-extreme phenotype, LTECs with more than 10 years of viral and immunological HIV control, could be pivotal to finding the keys for functional HIV remission. Metabolomics and lipidomics analyses were performed using high-resolution mass spectrometry (ultra-high-performance liquid chromatography-electrospray ionization-quadrupole time of flight [UHPLC-(ESI) qTOF] in plasma samples of 13 patients defined as LTEC-extreme, a group of 20 LTECs that lost viral and/or immunological control during the follow-up study (LTEC-losing) and 9 EC patients with short-term viral and immunological control (less than 5 years; no-LTEC patients). Long-term viral and immunological HIV-1 control was found to be strongly associated with elevated tricarboxylic acid (TCA) cycle function. Interestingly, of the nine metabolites identified in the TCA cycle, α-ketoglutaric acid (p = 0.004), a metabolite implicated in the activation of the mTOR complex, a modulator of HIV latency and regulator of several biological processes, was found to be a key metabolite in the persistent control.     SPARC mediates metastatic cooperation between CSC and non-CSC prostate cancer cell subpopulations

    Background Tumor cell subpopulations can either compete with each other for nutrients and physical space within the tumor niche, or co-operate for enhanced survival, or replicative or metastatic capacities. Recently, we have described co-operative interactions between two clonal subpopulations derived from the PC-3 prostate cancer cell line, in which the invasiveness of a cancer stem cell (CSC)-enriched subpopulation (PC-3M, or M) is enhanced by a non-CSC subpopulation (PC-3S, or S), resulting in their accelerated metastatic dissemination. Methods M and S secretomes were compared by SILAC (Stable Isotope Labeling by Aminoacids in Cell Culture). Invasive potential in vitro of M cells was analyzed by Transwell-Matrigel assays. M cells were co-injected with S cells in the dorsal prostate of immunodeficient mice and monitored by bioluminescence for tumor growth and metastatic dissemination. SPARC levels were determined by immunohistochemistry and real-time RT-PCR in tumors and by ELISA in plasma from patients with metastatic or non-metastatic prostate cancer. Results Comparative secretome analysis yielded 213 proteins differentially secreted between M and S cells. Of these, the protein most abundantly secreted in S relative to M cells was SPARC. Immunodepletion of SPARC inhibited the enhanced invasiveness of M induced by S conditioned medium. Knock down of SPARC in S cells abrogated the capacity of its conditioned medium to enhance the in vitro invasiveness of M cells and compromised their potential to boost the metastatic behavior of M cells in vivo. In most primary human prostate cancer samples, SPARC was expressed in the epithelial tumoral compartment of metastatic cases. Conclusions The matricellular protein SPARC, secreted by a prostate cancer clonal tumor cell subpopulation displaying non-CSC properties, is a critical mediator of paracrine effects exerted on a distinct tumor cell subpopulation enriched in CSC. This paracrine interaction results in an enhanced metastatic behavior of the CSC-enriched tumor subpopulation. SPARC is expressed in the neoplastic cells of primary prostate cancer samples from metastatic cases, and could thus constitute a tumor progression biomarker and a therapeutic target in advanced prostate cancer

    Phage-Derived and Aberrant HaloTag Peptides Immobilized on Magnetic Microbeads for Amperometric Biosensing of Serum Autoantibodies and Alzheimer's Disease Diagnosis

    An electrochemical biosensing platform for serum autoantibodies (AAbs) detection is reported in this work, exploiting for the first time six Alzheimer's disease (AD)-specific phage-derived and frameshift aberrant HaloTag peptides as receptors, immobilized on magnetic microbeads (MBs) surface and captured on disposable electrodes to perform amperometric detection. Operational analytical characteristics and clinical diagnostic ability of the bioplatform were probed in optimized key experimental conditions by analysing serum AAbs of AD patients and healthy subjects. The value of 100% obtained for AUC, sensitivity, and selectivity from the all peptides combined ROC curve, indicate full AD-diagnostic capability of the methodology, which was further implemented, as proof of concept, in a POC multiplexing platform to detect the signature in a single test over clinically actionable times (1 h 15 min), opening great promise for the type of diagnosis and AD patients’ monitoring follow-up currently pursued

    Uso de compuestos nutracéuticos en nuevas formulaciones de piensos acuícolas

    Trabajo presentado en las X Jornadas de Acuicultura en el Litoral Suratlántico: Nuevos retos y perspectivas para la sostenibilidad de la acuicultura, celebrado entre el 22 y 23 de noviembre de 2023 en Huelva.[EN] Different strategies have been followed to improve the aquaculture of main European farmed fish, such as genetic selection, refinements in culture conditions or enhanced feed formulation and management. However, more knowledge is required to exploit fish phenotypic plasticity to obtain those that better match aquaculture or market conditions. The overall objective of this mini-review is to identify the latest productive traits recently performed, mainly in commercial carnivorous species, and underlying biological processes that would be susceptible to improve the competitivity of aquaculture sector through nutritional issues. Thus, this work is framed within the concepts of Circular Economy and Blue Growth, where it is intended to point out the use of natural compounds extracted from seaweeds, microalgae and by-products of the industry for their inclusion in aquafeeds, allowing a more efficient use of more sustainable vegetable protein sources, and also to evaluate if these nutraceutical compounds counteract detrimental effects observed by nutritional interventions or environmentally challenged.[ES] La producción acuícola ha seguido diferentes estrategias para su optimización a lo largo de las últimas décadas, como el desarrollo de la selección genética, la mejora de las condiciones de cultivo, o el avance en la formulación y gestión de los piensos. Sin embargo, se requiere más conocimiento para explotar la plasticidad fenotípica de los peces para obtener aquellos que se ajusten mejor a las condiciones de la acuicultura o del mercado. El objetivo general de esta mini-revisión es identificar los últimos avances realizados recientemente, principalmente en especies carnívoras comerciales, y los procesos biológicos subyacentes que serían susceptibles de mejorar la competitividad del sector acuícola a través de intervenciones nutricionales.     Consumption of caffeinated beverages and kidney function decline in an elderly Mediterranean population with metabolic syndrome

    It remains unclear whether caffeinated beverages could have deleterious renal effects in elderly population with underlying comorbid conditions. We investigated the associations between coffee, tea, or caffeine intake and 1-year changes in glomerular filtration rate (eGFR) in a large Spanish cohort of overweight/obese elderly with metabolic syndrome (MetS). This prospective analysis includes 5851 overweight/obese adults (55-75 years) with MetS from the PREDIMED-Plus study. We assessed coffee, tea, and caffeine consumption from a validated food-frequency questionnaire and creatinine-based eGFR using the Chronic Kidney Disease Epidemiology Collaboration equation. Multivariate-adjusted regression models were applied to test associations between baseline coffee, tea, or caffeine intake and 1-year eGFR changes. Caffeinated coffee (> 2 cups/day) and tea (at least 1 cup/day) drinkers had 0.88 and 0.93 mL/min/1.73 m2 greater eGFR decrease respectively, compared to those with less than 1 cup/day of coffee consumption or non-tea drinkers. Furthermore, caffeinated coffee consumption of > 2 cups/day was associated with 1.19-fold increased risk of rapid eGFR decline > 3 mL/min/1.73 m2 (95% CI 1.01-1.41). Similarly, individuals in the highest (median, 51.2 mg/day) tertile of caffeine intake had a 0.87 mL/min/1.73 m2 greater eGFR decrease. Decaffeinated coffee was not associated with eGFR changes. In conclusion, higher consumption of caffeinated coffee, tea, and caffeine was associated with a greater 1-year eGFR decline in overweight/obese adults with MetS

Spain was one of the European countries with the highest incidence during the first wave. As a part of a consortium to monitor and study the evolution of the epidemic, we sequenced 2,170 samples, diagnosed mostly before lockdown measures. Here, we identified at least 500 introductions from multiple international sources and documented the early rise of two dominant Spanish epidemic clades (SECs), probably amplified by superspreading events. Both SECs were related closely to the initial Asian variants of SARS-CoV-2 and spread widely across Spain. We inferred a substantial reduction in the effective reproductive number of both SECs due to public-health interventions (Re < 1), also reflected in the replacement of SECs by a new variant over the summer of 2020.
