    Lack of effect of pravastatin on cerebral blood flow or parenchymal volume loss in elderly at risk for vascular disease

    <p><b>Background and Purpose:</b> Ageing is associated with a decline in cerebral blood flow. Animal studies have shown that cholesterol-lowering therapy with statins might preserve cerebral blood flow (CBF). We examined the effect of 40 mg pravastatin on the decline in CBF and brain volume in a subset of elderly subjects participating in the PROspective Study of Pravastatin in the Elderly at Risk (PROSPER) trial.</p> <p><b>Methods:</b> Randomization was not stratified according to whether or not subjects participated in the MRI substudy. In 391 men (n=226) and women (n=165) aged 70 to 82 years (mean±SD, 75±3.2), we measured total CBF (in mL/min) at baseline and after a mean±SD follow-up of 33±1.4 months with a gradient-echo phase-contrast MRI technique. Total CBF was defined as the summed flows in both internal carotid and vertebral arteries. Parenchymal volume (whole brain) was segmented with the use of in-house–developed semiautomatic software.</p> <p><b>Results:</b> Total CBF significantly declined in the placebo-allocated group, from 521±83 to 504±92 mL/min (P=0.0036) and in the pravastatin-allocated group from 520±94 to 506±92 mL/min (P=0.018). This decline was not significantly different between treatment groups (P=0.56). There was also a significant reduction in brain volume over time (P<0.001), which was not different between the treatment groups (P=0.47). When expressed per unit of parenchymal volume, the decline in CBF over time was no longer statistically significant.</p> <p><b>Conclusions:</b> Elderly people at risk for cerebral vascular disease had a significant decline in CBF with increasing age that was explained by a concomitant reduction in brain volume. Treatment with 40 mg pravastatin daily had no beneficial effect on total CBF.</p&gt

    Measurement of throughput variation across a large format volume-phase holographic grating

    In this paper, we report measurements of diffraction efficiency and angular dispersion for a large format (~ 25 cm diameter) Volume-Phase Holographic (VPH) grating optimized for near-infrared wavelengths (0.9 ~ 1.8 μm). The aim of this experiment is to see whether optical characteristics vary significantly across the grating. We sampled three positions in the grating aperture with a separation of 5 cm between each. A 2 cm diameter beam is used to illuminate the grating. At each position, throughput and diffraction angle were measured at several wavelengths. It is found that whilst the relationship between diffraction angle and wavelength is nearly he same at the three positions, the throughputs vary by up to ~ 10% from position to position. We explore the origin of the throughput variation by comparing the data with predictions from coupled-wave analysis. We find that it can be explained by a combination of small variations over the grating aperture in gelatin depth and/or refractive index modulation amplitude, and amount of energy loss by internal absorption and/or surface reflection

In this paper, we investigate a simplified one-dimensional version of the model. This model yields travelling wave solutions and we analyse the behaviour of the travelling waves. Approximations for the rate of healing and maximum values for both the active healing and the healed cell densities are obtained

    Hierarchical Gaussian process mixtures for regression

    As a result of their good performance in practice and their desirable analytical properties, Gaussian process regression models are becoming increasingly of interest in statistics, engineering and other fields. However, two major problems arise when the model is applied to a large data-set with repeated measurements. One stems from the systematic heterogeneity among the different replications, and the other is the requirement to invert a covariance matrix which is involved in the implementation of the model. The dimension of this matrix equals the sample size of the training data-set. In this paper, a Gaussian process mixture model for regression is proposed for dealing with the above two problems, and a hybrid Markov chain Monte Carlo (MCMC) algorithm is used for its implementation. Application to a real data-set is reported

    Suppression of Mitochondrial Electron Transport Chain Function in the Hypoxic Human Placenta: A Role for miRNA-210 and Protein Synthesis Inhibition

    Fetal growth is critically dependent on energy metabolism in the placenta, which drives active exchange of nutrients. Placental oxygen levels are therefore vital, and chronic hypoxia during pregnancy impairs fetal growth. Here we tested the hypothesis that placental hypoxia alters mitochondrial electron transport chain (ETS) function, and sought to identify underlying mechanisms. We cultured human placental cells under different oxygen concentrations. Mitochondrial respiration was measured, alongside levels of ETS complexes. Additionally, we studied placentas from sea-level and high-altitude pregnancies. After 4 d at 1% O2 (1.01 KPa), complex I-supported respiration was 57% and 37% lower, in trophoblast-like JEG3 cells and fibroblasts, respectively, compared with controls cultured at 21% O2 (21.24 KPa); complex IV-supported respiration was 22% and 30% lower. Correspondingly, complex I levels were 45% lower in placentas from high-altitude pregnancies than those from sea-level pregnancies. Expression of HIF-responsive microRNA-210 was increased in hypoxic fibroblasts and high-altitude placentas, whilst expression of its targets, iron-sulfur cluster scaffold (ISCU) and cytochrome c oxidase assembly protein (COX10), decreased. Moreover, protein synthesis inhibition, a feature of the high-altitude placenta, also suppressed ETS complex protein levels. Our results demonstrate that mitochondrial function is altered in hypoxic human placentas, with specific suppression of complexes I and IV compromising energy metabolism and potentially contributing to impaired fetal growth. \ua9 2013 Colleoni et al

    Theory of Double-Sided Flux Decorations

    A novel two-sided Bitter decoration technique was recently employed by Yao et al. to study the structure of the magnetic vortex array in high-temperature superconductors. Here we discuss the analysis of such experiments. We show that two-sided decorations can be used to infer {\it quantitative} information about the bulk properties of flux arrays, and discuss how a least squares analysis of the local density differences can be used to bring the two sides into registry.     Co-precipitation synthesis of nickel-rich cathodes for Li-ion batteries

    The preparation of Ni-rich cathode materials is challenging due to the Ni2+ ion sensitivity to oxidation during synthesis. The synthesis conditions during the manufacture of Ni-rich materials such as LiNi0.8Mn0.1Co0.1O2 (NMC811) therefore require stringent control. The co-precipitation step, applied in the synthesis of the metal hydroxide precursor, determines the secondary particle assembly formation, where it is typically desirable to produce uniform, spherical, ∼10μm-diameter structures. A stirred tank reactor is often employed to maintain a constant temperature of 60 °C and a controlled pH of between 10.5 and 11.5 in an inert atmosphere to maintain a high Ni2+/Ni3+ ion ratio. This promotes the formation of an NMC hydroxide precursor (NixMnyCoz(OH)2) which is typically milled with a lithium salt and calcined to form LiNixMnyCozO2 with a layered α-NaFeO2 crystalline structure.
