    Use of hyperspectral transmittance imaging to evaluate the internal quality of nectarines

    [EN] The internal quality of nectarines (Prunus persica L. Batsch var. nucipersica) cv. 'Big Top' (yellow flesh) and 'Magique' (white flesh) has been inspected using hyperspectral transmittance imaging. Hyperspectral images of intact fruits were acquired in the spectral range of 630-900 nm using transmittance mode during their ripening under controlled conditions. The detection of split pit disorder and classification according to an established firmness threshold were performed using PLS-DA. The prediction of the Internal Quality Index (IQI) related to ripeness was performed using PLS-R. The most important variables were selected using interval-PLS. As a result, an accuracy of 94.7% was obtained in the detection of fruits with split pit of the 'Big Top' cultivar. Accuracies of 95.7% and 94.6% were achieved in the classification of the 'Big Top' and 'Magique' cultivars, respectively, according to the firmness threshold. The internal quality was predicted through the IQI with R-2 values of 0.88 and 0.86 for the two cultivars. The results obtained indicate the great potential of hyperspectral transmittance imaging for the assessment of the internal quality of intact nectarines.This work was partially funded by INIA and FEDER funds through project RTA2015-00078-00-00.     Evaluation of ATM Kinase Inhibitor KU-55933 as Potential Anti-Toxoplasma gondii Agent

    Toxoplasma gondii is an apicomplexan protozoan parasite with a complex life cycle composed of multiple stages that infect mammals and birds. Tachyzoites rapidly replicate within host cells to produce acute infection during which the parasite disseminates to tissues and organs. Highly replicative cells are subject to Double Strand Breaks (DSBs) by replication fork collapse and ATM, a member of the phosphatidylinositol 3-kinase (PI3K) family, is a key factor that initiates DNA repair and activates cell cycle checkpoints. Here we demonstrate that the treatment of intracellular tachyzoites with the PI3K inhibitor caffeine or ATM kinase-inhibitor KU-55933 affects parasite replication rate in a dose-dependent manner. KU-55933 affects intracellular tachyzoite growth and induces G1-phase arrest. Addition of KU-55933 to extracellular tachyzoites also leads to a significant reduction of tachyzoite replication upon infection of host cells. ATM kinase phosphorylates H2A.X (γH2AX) to promote DSB damage repair. The level of γH2AX increases in tachyzoites treated with camptothecin (CPT), a drug that generates fork collapse, but this increase was not observed when co-administered with KU-55933. These findings support that KU-55933 is affecting the Toxoplasma ATM-like kinase (TgATM). The combination of KU-55933 and other DNA damaging agents such as methyl methane sulfonate (MMS) and CPT produce a synergic effect, suggesting that TgATM kinase inhibition sensitizes the parasite to damaged DNA. By contrast, hydroxyurea (HU) did not further inhibit tachyzoite replication when combined with KU-55933

    Beyond calendars and maps: Rethinking time and space for effective knowledge governance in protected areas

    Protected area managers rely on relevant, credible, and legitimate knowledge. However, an increase in the rate, extent, severity, and magnitude of the impacts of drivers of change (e.g., climate change, altered land use, and demand for natural resources) is affecting the response capacity of managers and their agencies. We address temporal aspects of knowledge governance by exploring time-related characteristics of information and decision-making processes in protected areas. These areas represent artefacts where the past (e.g., geological periods and evolutionary processes), the present (e.g., biodiversity richness), and the future (e.g., protection of ecosystem services for future generations) are intimately connected and integrated. However, temporal horizons linked with spatial scales are often neglected or misinterpreted in environmental management plans and monitoring programs. In this paper, we present a framework to address multi-dimensional understandings of knowledge-based processes for managing protected areas to guide researchers, managers, and practitioners to consider temporal horizons, spatial scales, different knowledge systems, and future decisions. We propose that dealing with uncertain futures starts with understanding the knowledge governance context that shapes decision-making processes, explicitly embracing temporal dimensions of information in decision-making at different scales. We present examples from South Africa and Colombia to illustrate the concepts. This framework can help to enable a reflexive practice, identify pathways or transitions to enable actions and connect knowledge for effective conservation of protected areas

    Randomized Controlled Trial of Difelikefalin for Chronic Pruritus in Hemodialysis Patients

    INTRODUCTION: There is an unmet medical need for pruritus associated with chronic kidney disease, a distressing complication characterized by generalized and persistent itch affecting 20% to 40% of patients undergoing hemodialysis. Here we report the results of a phase 2 trial evaluating the efficacy and safety of a novel peripherally restricted kappa opioid receptor agonist, difelikefalin, in adult patients undergoing hemodialysis with pruritus. METHODS: In this study, 174 hemodialysis patients with moderate-to-severe pruritus were randomly assigned to receive difelikefalin (0.5, 1.0, or 1.5 μg/kg) or placebo intravenously thrice weekly after each hemodialysis session for 8 weeks in a double-blind, controlled trial. The primary endpoint was the change from baseline at week 8 in the weekly mean of the 24-hour Worst Itching Intensity Numerical Rating Scale score. The secondary efficacy endpoint was the change in itch-related quality of life measured by the Skindex-10 questionnaire. Other endpoints included safety, sleep quality, and additional measures including the 5-D itch scale. RESULTS: A significant reduction from baseline in itch intensity scores at week 8 favored all difelikefalin doses combined versus placebo ( CONCLUSION: In this trial, difelikefalin effectively reduced itching intensity and improved sleep and itch-related quality of life

    The Adsorption of H2O on TiO2 and SnO2(110) Studied by First-Principles Calculations

    First-principles calculations based on density functional theory and the pseudopotential method have been used to investigate the energetics of H2_2O adsorption on the (110) surface of TiO2_2 and SnO2_2. Full relaxation of all atomic positions is performed on slab systems with periodic boundary conditions, and the cases of full and half coverage are studied. Both molecular and dissociative (H2_2O \rightarrow OH^- + H+^+) adsorption are treated, and allowance is made for relaxation of the adsorbed species to unsymmetrical configurations. It is found that for both TiO2_2 and SnO2_2 an unsymmetrical dissociated configuration is the most stable. The symmetrical molecularly adsorbed configuration is unstable with respect to lowering of symmetry, and is separated from the fully dissociated configuration by at most a very small energy barrier. The calculated dissociative adsorption energies for TiO2_2 and SnO2_2 are in reasonable agreement with the results of thermal desorption experiments. Calculated total and local electronic densities of states for dissociatively and molecularly adsorbed configurations are presented and their relation with experimental UPS spectra is discussed

    Mujeres víctimas del conflicto armado. Análisis de su reparación en el marco de la Ley 1448 de 2011

    Con el fin reflexionar sobre el impacto que ha tenido el enfoque de género consagrado en la Ley 1448 de 2001, cuyo propósito es reparar a las víctimas del conflicto armado interno, el presente artículo desarrollará algunos apuntes respecto a las experiencias de un grupo de mujeres víctimas, y de un grupo de funcionarios/as públicos, líderes sociales y ONG. Para la recolección de la información se utilizó la metodología cualitativa, con entrevistas semiestructuradas en profundidad y talleres de trabajo con diferentes actores. El análisis conlleva a identificar algunos elementos clave que empañan los éxitos legislativos y jurisprudenciales que ha logrado el Estado colombiano en materia de reparación con enfoque de género. Los resultados indican la necesidad de ir más allá del género, como categoría diferenciadora, para lograr una mayor efectividad en la reparación propuesta a las víctimas.Agencia Andaluza de Cooperación Internacional para el Desarroll

    Este artículo interpreta los discursos de mujeres víctimas del conflicto armado colombiano, residentes en el Valle de Aburrá (Antioquia-Colombia), en el momento de acceder a la reparación que le reconoce el Estado en la Ley 1448 de 2011. A partir de una metodología cualitativa y fenomenológica, fueron entrevistadas setenta mujeres y desarrollado tres talleres con funcionarios/as públicos y organizaciones sociales de víctimas. Los resultados exponen la falta de rigor en la comprensión de la heterogeneidad de la situación de las mujeres víctimas y la inefectividad de la ley para una atención integral de toda la población víctima, limitándose, en la mayoría de los casos, a una reparación monetaria.Agencia Andaluza de Cooperación Internacional para el Desarroll

    Engineering the Controlled Assembly of Filamentous Injectisomes in E. coli K-12 for Protein Translocation into Mammalian Cells.

    Bacterial pathogens containing type III protein secretion systems (T3SS) assemble large needle-like protein complexes in the bacterial envelope, called injectisomes, for translocation of protein effectors into host cells. The application of these molecular syringes for the injection of proteins into mammalian cells is hindered by their structural and genomic complexity, requiring multiple polypeptides encoded along with effectors in various transcriptional units (TUs) with intricate regulation. In this work, we have rationally designed the controlled expression of the filamentous injectisomes found in enteropathogenic Escherichia coli (EPEC) in the nonpathogenic strain E. coli K-12. All structural components of EPEC injectisomes, encoded in a genomic island called the locus of enterocyte effacement (LEE), were engineered in five TUs (eLEEs) excluding effectors, promoters and transcriptional regulators. These eLEEs were placed under the control of the IPTG-inducible promoter Ptac and integrated into specific chromosomal sites of E. coli K-12 using a marker-less strategy. The resulting strain, named synthetic injector E. coli (SIEC), assembles filamentous injectisomes similar to those in EPEC. SIEC injectisomes form pores in the host plasma membrane and are able to translocate T3-substrate proteins (e.g., translocated intimin receptor, Tir) into the cytoplasm of HeLa cells reproducing the phenotypes of intimate attachment and polymerization of actin-pedestals elicited by EPEC bacteria. Hence, SIEC strain allows the controlled expression of functional filamentous injectisomes for efficient translocation of proteins with T3S-signals into mammalian cells

    lncRNA requirements for mouse acute myeloid leukemia and normal differentiation

    A substantial fraction of the genome is transcribed in a cell type-specific manner, producing long non-coding RNAs (lncRNAs), rather than protein-coding transcripts. Here we systematically characterize transcriptional dynamics during hematopoiesis and in hematological malignancies. Our analysis of annotated and de novo assembled lncRNAs showed many are regulated during differentiation and mis-regulated in disease. We assessed lncRNA function via an in vivo RNAi screen in a model of acute myeloid leukemia. This identified several lncRNAs essential for leukemia maintenance, and found that a number act by promoting leukemia stem cell signatures. Leukemia blasts show a myeloid differentiation phenotype when these lncRNAs were depleted, and our data indicates that this effect is mediated via effects on the c-MYC oncogene. Bone marrow reconstitutions showed that a lncRNA expressed across all progenitors was required for the myeloid lineage, whereas the other leukemia-induced lncRNAs were dispensable in the normal setting
