    Promoter Methylation of RASSF1A Associates to Adult Secondary Glioblastomas and Pediatric Glioblastomas

    While allelic losses and mutations of tumor suppressor genes implicated in the etiology of astrocytoma have been widely assessed, the role of epigenetics is still a matter of study. We analyzed the frequency of promoter hypermethylation by methylation-specific PCR (MSP) in five tumor suppressor genes (PTEN, MGMT, RASSF1A, p14ARF, and p16INK4A), in astrocytoma samples and cell lines. RASSF1A was the most frequently hypermethylated gene in all grades of astrocytoma samples, in cell lines, and in adult secondary GBM. It was followed by MGMT. PTEN showed a slight methylation signal in only one GBM and one pilocytic astrocytoma, and in two cell lines; while p14ARF and p16INK4A did not show any evidence of methylation in primary tumors or cell lines. In pediatric GBM, RASSF1A was again the most frequently altered gene, followed by MGMT; PTEN, p14 and p16 showed no alterations. Lack or reduced expression of RASSF1A in cell lines was correlated with the presence of methylation. RASSF1A promoter hypermethylation might be used as a diagnostic marker for secondary GBM and pediatric GBM. Promoter hypermethylation might not be an important inactivation mechanism in other genes like PTEN, p14ARF and p16INK4A, in which other alterations (mutations, homozygous deletions) are prevalent

    Velocity-Based Heuristic Evaluation for Path Planning and Vehicle Routing for Victim Assistance in Disaster Scenarios

In these applications, digital elevation models (DEM) can provide accurate knowledge for safe vehicle motion planning but grid representation results in very large search graphs. Furthermore, travel time, which becomes a crucial cost optimization criterion, may be affected by inclination and other challenging terrain characteristics. In this paper, our goal is to evaluate a search heuristic function based on anisotropic vehicle velocity restrictions for building the cost matrix required for multi-vehicle routing on natural terrain and disaster sites. The heuristic is applied to compute the fastest travel times between every pair of matrix elements by means of a path planning algorithm.     Passive interferometric interrogation of a magnetic field sensor using an erbium doped fiber optic laser with magnetostrictive transducer

    An erbium doped (Er3+) fiber optic laser is proposed for magnetic field measurement. A pair of FBGsglued onto a magnetostrictive material (Terfenol-D rod) modulates the laser wavelength operation whensubject to a static or a time dependent magnetic field. A passive interferometer is employed to measure thelaser wavelength changes due to the applied magnetic field. A data acquisition hardware and a LabVIEWsoftware measure three phase-shifted signals at the output coupler of the interferometer and processthem using two distinct demodulation algorithms. Results show that sensitivity to varying magneticfields can be tuned by introducing a biasing magnetic field. A maximum error of 0.79% was found, formagnetic fields higher than 2.26 mT-RMS

    Evaluation of physical and chemical soil properties under different management types in the south-western Colombian Andes

    Aim of study: Evaluating the variability of physical and chemical soil properties under different soil uses in an experimental farmland of the southwestern Colombian Andes.Area of study: This research was conducted at the Botana Experimental Farm in the mountainous area of Nariño, at the south-western Colombia.Material and methods: nine soil variables were measured under six soil uses which included traditional agriculture, agroforestry systems, and a 67-year-old secondary forest that was used as a reference for comparisons with other soil uses. Data was analyzed through Principal Component Analysis and Duncan’s tests.Main results: organic carbon, cation exchange capacity, clay contents and base saturation were the variables with higher variability among soil uses. The secondary forest and an agroforestry system with alley-cropped wax laurel showed the best soil conditions, whilst pastures and monoculture potato crop plots showed the least desirable conditions for all variables.Research highlights: We found that soils under alley-crop with wax laurel presented the characteristics most similar to the secondary forest.     QoSatAr: a cross-layer architecture for E2E QoS provisioning over DVB-S2 broadband satellite systems

    This article presents QoSatAr, a cross-layer architecture developed to provide end-to-end quality of service (QoS) guarantees for Internet protocol (IP) traffic over the Digital Video Broadcasting-Second generation (DVB-S2) satellite systems. The architecture design is based on a cross-layer optimization between the physical layer and the network layer to provide QoS provisioning based on the bandwidth availability present in the DVB-S2 satellite channel. Our design is developed at the satellite-independent layers, being in compliance with the ETSI-BSM-QoS standards. The architecture is set up inside the gateway, it includes a Re-Queuing Mechanism (RQM) to enhance the goodput of the EF and AF traffic classes and an adaptive IP scheduler to guarantee the high-priority traffic classes taking into account the channel conditions affected by rain events. One of the most important aspect of the architecture design is that QoSatAr is able to guarantee the QoS requirements for specific traffic flows considering a single parameter: the bandwidth availability which is set at the physical layer (considering adaptive code and modulation adaptation) and sent to the network layer by means of a cross-layer optimization. The architecture has been evaluated using the NS-2 simulator. In this article, we present evaluation metrics, extensive simulations results and conclusions about the performance of the proposed QoSatAr when it is evaluated over a DVB-S2 satellite scenario. The key results show that the implementation of this architecture enables to keep control of the satellite system load while guaranteeing the QoS levels for the high-priority traffic classes even when bandwidth variations due to rain events are experienced. Moreover, using the RQM mechanism the user’s quality of experience is improved while keeping lower delay and jitter values for the high-priority traffic classes. In particular, the AF goodput is enhanced around 33% over the drop tail scheme (on average)

    Surveillance of imported malaria in Spain: The useful tool of the Semi-Nested Multiplex PCR

    The use of a new PCR-based method for the diagnosis of malaria in the Spanish Malaria Reference Laboratory has promoted an increase in confirmed cases of malaria. From August 1997 to July 1998, a total of 192 whole-blood samples and 71 serum samples from 168 patients were received from the hospitals of the Spanish National Health System. Most of the patients came from west-central African countries (85%). This molecular method showed more sensitivity and specificity than microscopy, detecting 12.4% more positive samples than microscopy and 13% of mixed infections undetectable by Giemsa stain. Plasmodium falciparum was the main species detected, with 68% of the total positive malaria cases, followed by Plasmodium malariae (29%), Plasmodium vivax (14%), and Plasmodium ovale (7%), including mixed infections in all cases. This report consists of the first wide, centralized survey of malaria surveillance in Spain. The reference laboratory conducted the analysis of all imported cases in order to detect trends in acquisition. The use of a seminested multiplex PCR permitted confirmation of the origins of the infections and the Plasmodium species involved and confirmation of the effectiveness of drug treatments. This PCR also allowed the detection of the presence in Spain of primaquine-tolerant P. vivax strains from west-central Africa, as well as the detection of a P. falciparum infection induced by transfusion.This work was supported by the Fondo de Investigaciones Sanitarias (FIS) (contract number 96/0216) and the Spanish Agency of International Cooperation (AECI).
