    Saisir la vie amoureuse par le croisement des méthodes

    In this article, I would like to explain how the crossing of different data gathering methods can improve the study of romantic relationships. I will only talk about Western societies, my fieldwork taking place in France, USA (mainly California) and Spain (mainly Catalonia). Love is an inter-personal process commonly experienced, but it is also very personal and intimate. Grounded Theory offers a methodological frame for dealing with the variety of representations of love. But it does not suggest which are the best techniques for data gathering. On the contrary, it leaves the possibility of working on diversified data in order to explore the various dimensions of love. I shall expose here my own methodological path, following the chronological order of the study that I conduct for a PhD in Sociology on the subject of the love life of SCI men. It turned out that this order is also logical, going from public through private data, until intimate ones. They all complement each other disclosing how love life stories take place.Dans cet article, je souhaiterais montrer l’intĂ©rĂȘt de croiser plusieurs mĂ©thodes de recueil de donnĂ©es pour Ă©tudier un processus interpersonnel comme l’amour. De l’amour, il n’est question ici que de l’amour eros, l’amour entre partenaires, ce que les anglophones appellent romantic relationships. La rĂ©flexion se limite aussi uniquement Ă  des sociĂ©tĂ©s occidentales, mes terrains comprenant la France, les USA (principalement la Californie), et l’Espagne (principalement la Catalogne). L’expĂ©rience amoureuse est Ă  la fois rĂ©pandue, partagĂ©e, ordinaire, elle est co-construite dans des interactions, mais elle est aussi personnelle, intime et profonde. La Grounded Theory offre un cadre mĂ©thodologique permettant de gĂ©rer la variĂ©tĂ© des reprĂ©sentations de l’amour. Mais elle ne prĂ©sume pas des techniques de recueil de donnĂ©es Ă  mettre en Ɠuvre. Au contraire, elle laisse la possibilitĂ© de travailler sur des donnĂ©es diversifiĂ©es afin d’explorer les diffĂ©rentes dimensions de l’amour. J’exposerai ici mon propre cheminement mĂ©thodologique, suivant l’ordre chronologique du dĂ©ploiement de la recherche que je mĂšne dans le cadre d’un doctorat en sociologie sur la vie amoureuse d’hommes lĂ©sĂ©s mĂ©dullaires.     Allelopathy Effect of the Blue Diatom HasleaOstrearia (Gaillon) Simonsen: Growth Inhibition in Aquaculture Relevant Microalgae

    We are well known that the marine pennate diatom Hasleaostrearia (Gaillon) Simonsen produces the blue-green pigment called marennine. Marennine was identified as a polyphenolic compound. This compound is capable to inhibit the growth of herpes simplex virus type 1 (HSV-1) and several bacteria, including pathogens as Vibrio sp. The aim of this study was to determine how strong the allelophatic effect of marennine is generated byH. ostrearia and identifying its minimal concentration that inhibits the growth of other microalgae. The experiment consisted of two phases of research activities. The first phase was biovolumecomparaison between H. ostrearia and microalgae suitable for aquaculture tested to take into account the differences in size of the microalgae tested(Tetraselmissuecica, Chaetocerosgracilis, Skeletonemacostatum, PavlovalutheriandIsochrysisgalbana (T-Iso) in term of H. ostrearia biomass and production of marennine. The second phase was the bioactivity tests, which is include growth kinetics, allelophatic effects and concentration of marennine in the culture medium, measured with spectrophotometer. The results of this study showed that H. ostrearia could inhibit several species of microalgae currently used in aquaculture (T. suecica, C. gracilis and S. costatum). The statistical analysis (One way ANOVA) showed that there are significant differences (P < 0, 05) between treatments andwhen co-cultured with H. ostrearia, these microalgae exhibited a significant growth inhibition. The highest inhibition value was 97.77% (on the treatementH. ostrearia+C. gracilis). In this treatment, the concentration of marennine in the culture was 5.35 mg L-1. According to the results, we can conclude that the supernatant of H. ostrearia with marennine acts as an allelochemical. The minimal concentration is 0.23 mg L-1 and the highest is 5.35 mg L-1. The treatments H. ostreariawithP. lutheri and T-Iso did not performed any inhibition

    Including 38 kHz in the standardization protocol for hydroacoustic fish surveys in temperate lakes

    Hydroacoustics has become a requisite method to assess fish populations and allows to describe the relationships of fish with other elements of the aquatic ecosystem. This nonintrusive method is currently an integral part of the sampling procedures recommended for fish stock assessment by the Water Framework Directive and has been standardized by the European Committee for Standardization [CEN (2014) CSN EN 15910 - Water quality - Guidance on the estimation of fish abundance with mobile hydroacoustic methods, Category: 7577 Water quality. Biological.]. In Europe, hydroacoustic surveys are performed in freshwater using different frequencies. Consequently, there is a need to evaluate if survey results can be compared. This study aimed to carry out in situ comparisons at the 38 kHz frequency (noted f) with two other commonly used frequencies, 70 and 200 kHz. The 38 kHz frequency has seldom been compared with other frequencies in freshwater although it is widely used worldwide, especially in the Great Lakes of North America and in Sweden. In 2016, hydroacoustic data were acquired in Lakes Annecy and Bourget using methods validated in previous studies that compared the frequencies 70, 120 and 200 kHz. This study showed similar density and biomass estimations as a function of frequency, density(f) and biomass(f), between the frequencies studied for low to moderate fish densities. For higher fish densities, the results were more variable and need to be verified. Fish density(f) and biomass(f) estimations sometimes exhibit differences between frequencies, which is not fully in agreement with theoretical calculations. The aim of this study was to evaluate frequency comparisons in practise. However, if the differences on acoustic metrics, density(f) or biomass(f) between frequencies were occasionally statistically significant, the differences were small enough to be considered negligible for fish population management. These analyses led to better knowledge of the responses from fish in temperate lakes for the studied frequencies. Our findings should be considered when revising the CEN standard

    In vivo estimation of pigment composition and optical absorption cross-sectionby spectroradiometry in four aquatic photosynthetic micro-organisms

    International audienceThe objective of the present study was to estimate in vivo pigment composition and to retrieve absorption cross-section values, a∗, of photosynthetic micro-organisms using a non-invasive technique of reflectance spectrometry. To test the methodology, organisms from different taxonomical groups and different pigment composition were used (Spirulina platensis a Cyanophyta, Porphyridium cruentum a Rhodophyta, Dunaliella tertiolecta a Chlorophyta and Entomoneis paludosa a Bacillariophyta) and photoacclimated to two different irradiance levels: 25 ÎŒmol photon m−2 s−1 (Low Light, LL) and 500 ÎŒmol photon m−2 s−1 (High Light, HL). Second derivative spectra from reflectance were used to identify pigment in vivo absorption bands that were linked to specific pigments detected by high performance liquid chromatography. Whereas some absorption bands such as those induced by Chlorophyll (Chl) a (416, 440, 625 and around 675 nm) were ubiquous, others were taxonomically specific (e.g. 636 nm for Chl c in E. paludosa) and/or photo-physiological dependent (e.g. 489 nm for zeaxanthin in the HL-acclimated S. platensis). The optical absorption cross-section, a∗, was retrieved from reflectance data using a radiative transfer model previously developed for microphytobenthos. Despite the cellular Chl a decrease observed from LL to HL (up to 88% for S. platensis), the a∗ increased, except for P. cruentum. This was attributed to a ‘package effect’ and to a greater absorption by photoprotective carotenoids that did not contribute to the energy transfer to the core Chl a

    Fine-scale vertical structure of sound-scattering layers over an east border upwelling system and its relationship to pelagic habitat characteristics

    Understanding the relationship between sound-scattering layers (SSLs) and pelagic habitat characteristics is a substantial step to apprehend ecosystem dynamics. SSLs are detected on echo sounders representing aggregated marine pelagic organisms. In this study, SSL characteristics of zooplankton and micronekton were identified during an upwelling event in two contrasting areas of the Senegalese continental shelf. Here a cold upwelling-influenced inshore area was sharply separated by a strong thermal boundary from a deeper, warmer, stratified offshore area. Mean SSL thickness and SSL vertical depth increased with the shelf depth. The thickest and deepest SSLs were observed in the offshore part of the shelf. Hence, zooplankton and micronekton seem to occur more frequently in stratified water conditions rather than in fresh upwelled water. Diel vertical and horizontal migrations of SSLs were observed in the study area. Diel period and physicochemical water characteristics influenced SSL depth and SSL thickness. Although chlorophyll-a concentration insignificantly affected SSL characteristics, the peak of chlorophyll a was always located above or in the middle of the SSLs, regularly matching with the peak of SSL biomass. Such observations indicate trophic relationships, suggesting SSLs to be mainly composed of phytoplanktivorous zooplankton and micronekton. Despite local hypoxia, below 30 m depth, distribution patterns of SSLs indicate no vertical migration boundary. The results increase the understanding of the spatial organization of mid-trophic species and migration patterns of zooplankton and micronekton, and they will also improve dispersal models for organisms in upwelling regions.publishedVersio

    Time-dependent upregulation of electron transport with concomitant induction of regulated excitation dissipation in Haslea diatoms

    International audiencePhotoacclimation by strains of Haslea "blue" diatom species H. ostrearia and H. silbo sp. nov. ined. was investigated with rapid light curves and induction-recovery curves using fast repetition rate fluorescence. Cultures were grown to exponential phase under 50 ”mol m −2 s −1 photosynthetic available radiation (PAR) and then exposed to non-sequential rapid light curves where, once electron transport rate (ETR) had reached saturation, light intensity was decreased and then further increased prior to returning to near growth light intensity. The non-sequential rapid light curve revealed that ETR was not proportional to the instantaneously applied light intensity, due to rapid photoacclimation. Changes in the effective absorption cross sections for open PSII reaction centres (σ PSII â€Č) or reaction centre connectivity (ρ) did not account for the observed increases in ETR under extended high light. σ PSII â€Č in fact decreased as a function of a time-dependent induction of regulated excitation dissipation Y(NPQ), once cells were at or above a PAR coinciding with saturation of ETR. Instead, the observed increases in ETR under extended high light were explained by an increase in the rate of PSII reopening, i.e. Q A − oxidation. This acceleration of electron transport was strictly light dependent and relaxed within seconds after a return to low light or darkness. The time-dependent nature of ETR upregulation and regulated NPQ induction was verified using induction-recovery curves. Our findings show a time-dependent induction of excitation dissipation, in parallel with very rapid photoacclimation of electron transport, which combine to make ETR independent of short-term changes in PAR. This supports a selective advantage for these diatoms when exposed to fluctuating light in their environment
