    Revisão taxonÎmica e anålise filogenética em Bathytropidae Vandel, 1952 (CRUSTACEA: ISOPODA: ONISCIDEA)

    A ordem Isopoda Ă© um dos o grupo mais diverso dentre os crustĂĄceos, sendo composta por dez subordens. Os representantes da subordem Oniscidea, conhecidos como “tatuzinhos de jardim”, obtiveram grande sucesso no domĂ­nio do ambiente terrestre. Para isso, foram necessĂĄrias vĂĄrias adaptaçÔes morfolĂłgicas, fisiolĂłgicas e comportamentais, tais como comportamento gregĂĄrio para diminuição da perda de ĂĄgua, presença de marsĂșpio fechado para reprodução e a presença de pulmĂ”es pleopodais. Apesar do grupo apresentar ampla uma distribuição geogrĂĄfica, as espĂ©cies apresentam limitada capacidade de dispersĂŁo. Essa baixa dispersĂŁo resulta em isolamento de populaçÔes e alta diversidade genĂ©tica. Dessa forma, o nĂșmero de espĂ©cies dos isĂłpodos Ă© subestimado, sendo necessĂĄrios mais esforços taxonĂŽmicos a fim de ampliar o conhecimento para o grupo. Frequentemente, revisĂ”es taxonĂŽmicas resultam em ampliação do nĂșmero de espĂ©cies, reforçando a importĂąncia destes estudos para o grupo. IdentificaçÔes usualmente se baseiam em pequenas variaçÔes morfolĂłgicas, o que muitas vezes dificulta a distinção entre espĂ©cies, sendo dados moleculares representam uma importante ferramenta para a delimitação das relaçÔes de parentesco entre espĂ©cies. O gĂȘnero Neotroponiscus ocorre na America do Sul e possui uma grande diversidade de espĂ©cies distribuĂ­das ao longo da Mata AtlĂąntica Brasileira. Oito espĂ©cies sĂŁo conhecidas na literatura e foram revisadas para o levantamento de caracteres, necessĂĄrios para os estudos filogenĂ©ticos. AlĂ©m disso, neste trabalho, cinco novas espĂ©cies novas foram identificadas. Com base em dados morfolĂłgicos e moleculares a monofilia do gĂȘnero foi testada, e o grupo foi recuperado como monofilĂ©tico. As relaçÔes de parentesco entre Neotroponiscus e os integrantes da famĂ­lia Bathytropidae foi testada e a famĂ­lia foi recuperada como um grupo parafilĂ©tico, corroborando para hipĂłteses anteriores. Estudos futuros com uma abordagem biogeogrĂĄfica serĂŁo importantes para elucidar os padrĂ”es de distribuição das espĂ©cies de Neotroponiscus.The order Isopoda is one of the most diverse groups among crustaceans. Species from the suborder Oniscidea, commonly known as woodlice, were very successful in conquering the terrestrial environment. For that, many morphological, physiological and behavioral adaptations were necessary, such as, closed marsupium for reproduction, presence of pleopodal lungs, and gregarious behavior to diminish water losses. Although the group has large geographical distribution, species have limited dispersion capacity. This low dispersion results in population isolation and high genetic diversity. Therefore, the number of species of terrestrial isopods is greatly underestimated and more taxonomical studies are necessary in order to broaden the knowledge on the group. Taxonomical revisions on terrestrial isopods frequently result in higher number of species reinforcing the importance of these studies for the fore the mentioned group. Identification is usually based on subtle morphological differences, so molecular data is an important tool to elucidate the relationship within the group. The genus Neotroponiscus occurs in South America and has high species diversity throughout the Atlantic Forest. A total of eight species are known from the literature and were reviewed to identify characters that are necessary for phylogenetic studies. In this study, five new species were identified and described. Based on morphological and molecular data, the monophyly of the group was tested and the group was confirmed as monophyletic. The relationship between Neotroponiscus and the other species from the family Bathytropidae were also tested, showing that the family represents a paraphyletic group, which corroborates previous suggestions. Future studies with biogeographic approuch will be important to elucidate the distribution pattern of the genus Neotroponiscus

    A Novel Electrostimulated Drug Delivery System Based on PLLA Composites Exploiting the Multiple Functions of Graphite Nanoplatelets

    A novel drug delivery system based on poly(l-lactide) (PLLA), graphite, and porphyrin was developed. In particular, 5,10,15,20-tetrakis(4-hydroxyphenyl)porphyrin (THPP) was chosen because, besides its potential as codispersing agent of graphite, it is a pharmacologically active molecule. Graphite nanoplatelets, homogeneously dispersed in both the neat PLLA and the PLLA/porphyrin films, which were prepared by solution casting, turned out to improve the crystallinity of the polymer. Moreover, IR measurements demonstrated that unlike PLLA/porphyrin film, where the porphyrin was prone to aggregate causing variable concentration throughout the sample, the system containing also GNP was characterized by a homogeneous dispersion of the above molecule. The effect of graphite nanoplatelets on the thermal stabilization, electrical conductivity, and improvement of mechanical properties of the polymer resulted to be increased by the addition of the porphyrin to the system, thus demonstrating the role of the molecule in ameliorating the filler dispersion in PLLA. The porphyrin release from the composite film, occurring both naturally and with the application of an electrical field, was measured using an UV-vis spectrophotometer. Indeed, voltage application turned out to improve significantly the kinetic of drug release. The biocompatibility of the polymer matrix as well as the mechanical and thermal properties of the composite together with its electrical response makes the developed material extremely promising in biological applications, particularly in the drug delivery field

    The genus Novamundoniscus includes eight species with distribution in Brazil and Venezuela. The new species Novamundoniscus adhara Campos-Filho & Cardoso sp. nov. from the state of Tocantins, northern Brazil, is described, which represents the first record to the family for the state of Tocantins

    New species and records of Exalloniscus Stebbing, 1911 from southern Asia (Malacostraca, Isopoda, Oniscidea)

    The genus Exalloniscus is widely distributed in southern and eastern Asia. In this contribution, eight species are described as new: Exalloniscus schmalfussi from Nepal; E. siamensis from Thailand; E. kampucheaensis from Cambodia; E. tortilis and E. deharvengi from Vietnam; E. stilifer from southern China; E. burmaensis from Myanmar; and E. convexus from Laos. Ten more species are recorded: E. nepalensis from Nepal; E. coecus, E. brincki, E. bicoloratus, E. caudatus and E. maschwitzi from West Malaysia; E. sumatranus from Sumatra, Indonesia, and Singapore; E. papillosus from West Malaysia and Vietnam; and E. thailandensis from Laos. Exalloniscus bessoni Dalens, 1992 and E. yinae Nunomura and Xie, 2000 are considered to be junior synonyms of E. thailandensis Dalens, 1987. A map with the recorded distributions of all the 28 species of Exalloniscus is provided and their habitat is discussed

    Connecting gut microbiome changes with fish health conditions in juvenile Atlantic cod (Gadus morhua) exposed to dispersed crude oil

    Polycyclic aromatic hydrocarbons found in crude oil can impair fish health following sublethal exposure. However, the dysbiosis of microbial communities within the fish host and influence it has on the toxic response of fish following exposure has been less characterized, particularly in marine species. To better understand the effect of dispersed crude oil (DCO) on juvenile Atlantic cod (Gadus morhua) microbiota composition and potential targets of exposure within the gut, fish were exposed to 0.05 ppm DCO for 1, 3, 7, or 28 days and 16 S metagenomic and metatranscriptomic sequencing on the gut and RNA sequencing on intestinal content were conducted. In addition to assessing species composition, richness, and diversity from microbial gut community analysis and transcriptomic profiling, the functional capacity of the microbiome was determined. Mycoplasma and Aliivibrio were the two most abundant genera after DCO exposure and Photobacterium the most abundant genus in controls, after 28 days. Metagenomic profiles were only significantly different between treatments after a 28-day exposure. The top identified pathways were involved in energy and the biosynthesis of carbohydrates, fatty acids, amino acids, and cellular structure. Biological processes following fish transcriptomic profiling shared common pathways with microbial functional annotations such as energy, translation, amide biosynthetic process, and proteolysis. There were 58 differently expressed genes determined from metatranscriptomic profiling after 7 days of exposure. Predicted pathways that were altered included those involved in translation, signal transduction, and Wnt signaling. EIF2 signaling was consistently dysregulated following exposure to DCO, regardless of exposure duration, with impairments in IL-22 signaling and spermine and spermidine biosynthesis in fish after 28 days. Data were consistent with predictions of a potentially reduced immune response related to gastrointestinal disease.     Study of the long-finned pilot whale (Globicephala melas) bile content - An indicator of ocean health

    Globicephala melas has been harvested in the Faroe Islands for centuries. Given the distances travelled by this species, tissue/body fluid samples represent unique matrices to be considered as an integration of environmental condition and pollution status of their prey. For the first time, bile samples were analysed for presence of polycyclic aromatic hydrocarbon (PAH) metabolites and protein content. Concentrations of 2- and 3-ring PAH metabolites ranged from 11 to 25 ÎŒg mL−1 pyrene fluorescence equivalents. In total, 658 proteins were identified and 61,5 % were common amongst all individuals. Identified proteins were integrated into in silico software and determined that the top predicted disease and functions were neurological diseases, inflammation, and immunological disorders. The metabolism of reactive oxygen species (ROS) was predicted to be dysregulated, which can have consequences to both the protection against ROS produced during dives and contaminant exposures.     ÎČ2-adrenergic receptor and UCP3 variants modulate the relationship between age and type 2 diabetes mellitus

    BACKGROUND: It is widely accepted that Type 2 Diabetes Mellitus (T2DM) and other complex diseases are the product of complex interplay between genetic susceptibility and environmental causes. To cope with such a complexity, all the statistical and conceptual strategies available should be used. The working hypothesis of this study was that two well-known T2DM risk factors could have diverse effect in individuals carrying different genotypes. In particular, our effort was to investigate if a well-defined group of genes, involved in peripheral energy expenditure, could modify the impact of two environmental factors like age and obesity on the risk to develop diabetes. To achieve this aim we exploited a multianalytical approach also using dimensionality reduction strategy and conservative significance correction strategies. METHODS: We collected clinical data and characterised five genetic variants and 2 environmental factors of 342 ambulatory T2DM patients and 305 unrelated non-diabetic controls. To take in account the role of one of the major co-morbidity conditions we stratified the whole sample according to the presence of obesity, over and above the 30 Kg/m(2 )BMI threshold. RESULTS: By monofactorial analyses the ADRB2-27 Glu27 homozygotes had a lower frequency of diabetes when compared with Gln27 carriers (Odds Ratio (OR) 0.56, 95% Confidence Interval (CI) 0.36 – 0.91). This difference was even more marked in the obese subsample. Multifactor Dimensionality Reduction method in the non-obese subsample showed an interaction among age, ADRB2-16 and UCP3 polymorphisms. In individuals that were UCP3 T-carriers and ADRB2-16 Arg-carriers the OR increased from 1 in the youngest to 10.84 (95% CI 4.54–25.85) in the oldest. On the contrary, in the ADRB2-16 GlyGly and UCP3 CC double homozygote subjects, the OR for the disease was 1.10 (95% CI 0.53–2.27) in the youngest and 1.61 (95% CI 0.55–4.71) in the oldest. CONCLUSION: Although our results should be confirmed by further studies, our data suggests that, when properly evaluated, it is possible to identify genetic factors that could influence the effect of common risk factors

    Chemical Characterization of Hydrogels Crosslinked with Polyethylene Glycol for Soft Tissue Augmentation

    BACKGROUND: Hyaluronic acid (HA) based hydrogels for esthetic applications found widespread use. HA should be crosslinked for this application to achieve the correct viscoelastic properties and avoid fast degradation by the hyaluronidase enzyme naturally present in the skin: these properties are controlled by the amount of crosslinker and the fraction that is effectively crosslinked (i.e. that binds two HA chains). AIM: Crosslinking by polyethylene glycol diglycidyl ether (PEGDE) has been more recently introduced and showed attractive features in terms of viscoelastic properties and reduced biodegradation. Aim of this paper is to define a method for the determination of the crosslinking properties of these recently introduced fillers, method that is lacking at the moment. MATERIAL AND METHOD: The percentage of crosslinker and the fraction that is effectively crosslinked were determined by proton Nuclear Magnetic Resonance (1H NMR) and by 13C NMR, respectively. The filler were preliminarily washed with acetonitrile to remove residual PEG and then digested by hyaluronidase to obtain a sample that can be analysed by NMR. RESULTS: The crosslinking parameters were determined in four samples of NEAUVIA PEG-crosslinked dermal fillers (produced by MatexLab S.p.A., Italy). The percentage of crosslinker was between 2.8% and 6.2% of HA, whereas the effective crosslinker ratios were between 0.07 and 0.16 (ratio between the moles of effectively crosslinked PEG and total moles of PEG). Moreover, a digestion procedure alternative to enzymatic digestion, based on acidic hydrolysis, was successfully tested for the determination of crosslinker percentage. CONCLUSIONS: The proposed method successfully determined the two crosslinking parameters in PEG-crosslinked dermal fillers. The estimated percentage of crosslinker is similar to previously reported data for other crosslinkers, whereas the effective crosslinker ratio is lower for PEG crosslinked hydrogels
