    Some pregnancies are abnormal in human genetically and end with the spontaneous abortion, which is the most common problem of pregnancy. The recurrent abortions are often referred to as multifactorial disease that one of which is thrombosis. The thrombosis in placenta capillaries seems to disturb the blood circulation between the mother and the fetus and eventually lead to abortion. Recently, studies have shown that genetic basis for thrombophilia relates with recurrent abortion. The aim of this study is the survey of G1691A and G4070A mutations in the Factor V gene, -455G>A mutation in the gen of XIII factor, G103T mutation in Beta fibrinogen and A20210G mutation in the thrombin gene. The samples were collected from 60 patients referred to Tehran Imam Khomeini hospital .DNA was extracted from patients' blood samples by multiple PCR simultaneously containing different mutations were duplicated then the existence of mutation was evaluated by the strip technique. The genes mutation of G1691A in Factor V, G4070A in Factor V, G103T in Beta fibrinogen, -455G>A in the XIII factor and G20210A were identified 6.6, 45, 36, 40 and 3.3 respectively. Studies on the other population showed that frequency of examined mutations varies with other communities. Anyway, more samples are required in order to obtain more accurate statistics related to the frequency of mutations

    Transdifferentiation of Human Dental Pulp Stem Cells Into Oligoprogenitor Cells

    Introduction: The nerve fibers in central nervous system are surrounded by myelin sheet which is formed by oligodendrocytes. Cell therapy based on oligodendrocytes and their precursors transplantation can hold a promising alternative treatment for myelin sheet repair in demyelinating diseases. Methods: Human Dental Pulp Stem Cells (hDPSCs) are noninvasive, autologous and easy available source with multipotency characteristics, so they are in focus of interest in regenerative medicine. In the present study, hDPSCs were differentiated into oligoprogenitor using glial induction media, containing Retinoic Acid (RA), basic Fibroblast Growth Factor (bFGF), Platelet- Derived Growth Factor (PDGF), N2 and B27. The differentiated Oligoprogenitor Cells (OPCs) were evaluated for nestin, Olig2, NG2 and O4 using immunocytochemistry. Also, the expression of nestin, Olig2 and PDGFR-alpha gens (neuroprogenitor and oligoprogenitor markers) were investigated via RT-PCR technique. Results: The results indicate that glial differentiation medium induces the generation of oligoprogenitor cells as revealed via exhibition of specific glial markers, including Olig2, NG2 and O4. The expersion of nestin gene (neuroprogenitor marker) and Olig2 and PDGFR-alpha genes (oligoprogentor markers) were detected in treated hDPSCs at the end of the induction stage. Conclusion: hDPSCs can be induced to transdifferentiate into oligoprogenitor cells and respond to the routinely applied regents for glial differentiation of mesanchymal stem cells. These data suggest the hDPSCs as a valuable source for cell therapy in neurodegenerative diseases

    Anti-cancer effects of cinnamon: Insights into its apoptosis effects

    Cancer is known as a leading cause of death worldwide. In the last two decades, the incidence of cancer has been dramatically increased mostly due to lifestyle changes. The importance of this issue has attracted further attention to discover novel therapies to prevent and treat cancers. According to previous studies, drugs used to treat cancer have shown significant limitations. Therefore, the role of herbal medicines alone or in combination with chemotherapy drugs has been extensively studied in cancer treatment. Cinnamon is a natural component showing a wide range of pharmacological functions including anti-oxidant, anti-microbial and anti-cancer activities. Impaired apoptosis plays critical roles in the initiation and progression of cancer. Increasing evidence indicates that cinnamon, as a therapeutic agent, has anti-cancer effects via affecting numerous apoptosis-related pathways in cancer cells. Here, we highlighted anticancer properties of cinnamon, particularly through targeting apoptosis-related mechanisms. © 2019 Elsevier Masson SA

    STAT4 deficiency reduces the development of atherosclerosis in mice

    Atherosclerosis is a chronic inflammatory process that leads to plaque formation in large and medium sized vessels. T helper 1 (Th1) cells constitute the majority of plaque infiltrating pro-atherogenic T cells and are induced via IFNγ-dependent activation of T-box (Tbet) and/or IL-12-dependent activation of signal transducer and activator of transcription 4 (STAT4). We thus aimed to define a role for STAT4 in atherosclerosis. STAT4-deficiency resulted in a ∼71% reduction (p < 0.001) in plaque burden in Stat4(-/-)Apoe(-/-) vs Apoe(-/-) mice fed chow diet and significantly attenuated atherosclerosis (∼31%, p < 0.01) in western diet fed Stat4(-/-)Apoe(-/-) mice. Surprisingly, reduced atherogenesis in Stat4(-/-)Apoe(-/-) mice was not due to attenuated IFNγ production in vivo by Th1 cells, suggesting an at least partially IFNγ-independent pro-atherogenic role of STAT4. STAT4 is expressed in T cells, but also detected in macrophages (MΦs). Stat4(-/-)Apoe(-/-)in vitro differentiated M1 or M2 MΦs had reduced cytokine production compare to Apoe(-/-) M1 and M2 MΦs that was accompanied by reduced induction of CD69, I-A(b), and CD86 in response to LPS stimulation. Stat4(-/-)Apoe(-/-) MΦs expressed attenuated levels of CCR2 and demonstrated reduced migration toward CCL2 in a transwell assay. Importantly, the percentage of aortic CD11b(+)F4/80(+)Ly6C(hi) MΦs was reduced in Stat4(-/-)Apoe(-/-) vs Apoe(-/-) mice. Thus, this study identifies for the first time a pro-atherogenic role of STAT4 that is at least partially independent of Th1 cell-derived IFNγ, and primarily involving the modulation of MΦ responses

    Identification and ranking of causes of delay in container handling operation by TOPSIS method

    This applied research has been conducted to identify and rank the causes of delay in container handling operation in two stages. In the first, having considered the daily census of container handling operation in the pertinent terminal of the port, As well, brain storming cessions attended by experts from the studied container terminals were held during which twenty seven numbers of causes of delay were detected. In the second stage, having considered the detected factors from the first stage, the probability of the causes occurrence, the extent of impact of causes on process after their occurrence and probability of causes detection before having effect on process have been determined by experts in form of a scale ranging from one to ten, Finally based on the obtained scores by each one of the causes, the decision matrix was formed and the mentioned causes were ranked by TOPSIS method. The abstained results showed that the causes of delays in loading and unloading operations in studied container terminals have significant different in the terms of probability of the causes occurrence, the extent of impact of causes on process after their occurrence and probability of causes detection before having effect on process. Finally to mitigation of these causes several strategies have been presented

    Neotethyan Subduction Ignited the Iran Arc and Backarc Differently

    Most arcs show systematic temporal and spatial variations in magmatism with clear shifts in igneous rock compositions between those of the magmatic front (MF) and those in the backarc (BA). It is unclear if similar magmatic polarity is seen for extensional continental arcs. Herein, we use geochemical and isotopic characteristics coupled with zircon U‐Pb geochronology to identify the different magmatic style of the Iran convergent margin, an extensional system that evolved over 100 Myr. Our new and compiled U‐Pb ages indicate that major magmatic episodes for the NE Iran BA occurred at 110–80, 75–50, 50–35, 35–20, and 15–10 Ma. In contrast to NE Iran BA magmatic episodes, compiled data from MF display two main magmatic episodes at 95–75 and 55–5 Ma, indicating more continuous magmatism for the MF than for the BA. We show that Paleogene Iran serves as a useful example of a continental arc under extension. Our data also suggest that there is not a clear relationship between the subduction velocity of Neotethyan Ocean beneath Iran and magmatic activity in Iran. Our results imply that the isotopic compositions of Iran BA igneous rocks do not directly correspond to the changes in tectonic processes or geodynamics, but other parameters such as the composition of lithosphere and melt source(s) should be considered. In addition, changes in subduction zone dynamics and contractional versus extensional tectonic regimes influenced the composition of MF and BA magmatic rocks.     Induced vertical disparity effects on local and global stereopsis

    Purpose: Although significant amounts of vertical misalignment could have a noticeable effect on visual performance, there is no conclusive evidence about the effect of very small amount of vertical disparity on stereopsis and binocular vision. Hence, the aim of this study was to investigate the effects of induced vertical disparity on local and global stereopsis at near. Materials and Methods: Ninety participants wearing best-corrected refraction had local and global stereopsis tested with 0.5 and 1.0 prism diopter (Δ) vertical prism in front of their dominant and non-dominant eye in turn. This was compared to local and global stereopsis in the same subjects without vertical prism. Data were analyzed in SPSS.17 software using the independent samples T and the repeated measures ANOVA tests. Results: Induced vertical disparity decreases local and global stereopsis. This reduction is greater when vertical disparity is induced in front of the non-dominant eye and affects global more than local stereopsis. Repeated measures ANOVA showed differences in the mean stereopsis between the different measured states for local and global values. Local stereopsis thresholds were reduced by 10s of arc or less on average with 1.0Δ of induced vertical prism in front of either eye. However, global stereopsis thresholds were reduced by over 100s of arc by the same 1.0Δ of induced vertical prism. Conclusion: Induced vertical disparity affects global stereopsis thresholds by an order of magnitude (or a factor of 10) more than local stereopsis. Hence, using a test that measures global stereopsis such as the TNO is more sensitive to vertical misalignment than a test such as the Stereofly that measures local stereopsis. © 2014 Informa Healthcare USA, Inc.     Designer Exosomes: A New Platform for Biotechnology Therapeutics

    Abstract: Desirable features of exosomes have made them a suitable manipulative platform for biomedical applications, including targeted drug delivery, gene therapy, cancer diagnosis and therapy, development of vaccines, and tissue regeneration. Although natural exosomes have various potentials, their clinical application is associated with some inherent limitations. Recently, these limitations inspired various attempts to engineer exosomes and develop designer exosomes. Mostly, designer exosomes are being developed to overcome the natural limitations of exosomes for targeted delivery of drugs and functional molecules to wounds, neurons, and the cardiovascular system for healing of damage. In this review, we summarize the possible improvements of natural exosomes by means of two main approaches: parental cell-based or pre-isolation exosome engineering and direct or post-isolation exosome engineering. Parental cell-based engineering methods use genetic engineering for loading of therapeutic molecules into the lumen or displaying them on the surface of exosomes. On the other hand, the post-isolation exosome engineering approach uses several chemical and mechanical methods including click chemistry, cloaking, bio-conjugation, sonication, extrusion, and electroporation. This review focuses on the latest research, mostly aimed at the development of designer exosomes using parental cell-based engineering and their application in cancer treatment and regenerative medicine.
