    Development of sustainable georesources for the built environment in the United Kingdom

    The character of the UK’s built heritage has been largely determined by the country’s diverse geology. Indigenous natural stone forms a major component of the nation’s pre-1919 building stock. Stone has been used traditionally for roofing, roads, pavements, bridges, engineering works, and all forms of walling. Today it is mostly employed as thin panel cladding to concrete frameworks in modern construction and is now increasingly being used in large volumes for new city streetscapes.This paper outlines the material requirements for the repair and maintenance of the stone-built heritage and illustrates a range of initiatives across the UK aimed at safeguarding and redeveloping indigenous resources. The importance, particularly for the repair and conservation sector, of selecting appropriate replacement stone is being recognized by architectural and conservation professionals and by local authority officials. There is also increasing recognition of the importance to the economy of the local character of the built environment in terms of its value to tourism and to architectural, historical, and cultural identity. The paper also examines the historical sources of information on stone in the UK and offers recommendations for databasing and disseminating stone resource information. This may assist the redevelopment of a healthy indigenous stone industry and ensure that the unique built heritage character of the UK is maintained and enhanced

    Dose-response between frequency of breaks in sedentary time and glucose control in type 2 diabetes: a proof of concept study

    Objectives This study aimed to investigate dose-response between frequency of breaks in sedentary time and glucose control.DesignRandomised three-treatment, two-period balanced incomplete block trial.MethodsTwelve adults with type 2 diabetes (age, 60 ± 11 years; body mass index, 30.2 ± 4.7 kg/m2) participated in two of the following treatment conditions: sitting for 7 h interrupted by 3 min light-intensity walking breaks every (1) 60 min (Condition 1), (2) 30 min (Condition 2), and (3) 15 min (Condition 3). Postprandial glucose incremental area under the curves (iAUCs) and 21-h glucose total area under the curve (AUC) were measured using continuous glucose monitoring. Standardised meals were provided. Results Compared with Condition 1 (6.7 ± 0.8 mmol L−1 × 3.5 h−1), post-breakfast glucose iAUC was reduced for Condition 3 (3.5 ± 0.9 mmol L−1 × 3.5 h−1, p ˂ 0.04). Post-lunch glucose iAUC was lower in Condition 3 (1.3 ± 0.9 mmol L−1 × 3.5 h−1, p ˂ 0.03) and Condition 2 (2.1 ± 0.7 mmol L−1 × 3.5 h−1, p ˂ 0.05) relative to Condition 1 (4.6 ± 0.8 mmol L−1 × 3.5 h−1). Condition 3 (1.0 ± 0.7 mmol L−1 × 3.5 h−1, p = 0.02) and Condition 2 (1.6 ± 0.6 mmol L−1 × 3.5 h−1, p ˂ 0.04) attenuated post-dinner glucose iAUC compared with Condition 1 (4.0 ± 0.7 mmol L−1 × 3.5 h−1). Cumulative 10.5-h postprandial glucose iAUC was lower in Condition 3 than Condition 1 (p = 0.02). Condition 3 reduced 21-h glucose AUC compared with Condition 1 (p < 0.001) and Condition 2 (p = 0.002). However, post-breakfast glucose iAUC, cumulative 10.5-h postprandial glucose iAUC and 21-h glucose AUC were not different between Condition 2 and Condition 1 (p ˃ 0.05).Conclusions There could be dose-response between frequency of breaks in sedentary time and glucose. Interrupting sedentary time every 15 min could produce better glucose control

    The relationship between members of the canonical NF-κB pathway, components of tumour microenvironment and survival in patients with invasive ductal breast cancer

    The aim of the present study was to examine the relationship between tumour NF-κB activation, tumour microenvironment including local inflammatory response (LIR) and cancer-specific survival in patients with operable ductal breast cancer. Immunohistochemistry (tissue microarray of 376 patients) and western blotting (MCF7 and MDA-MB-231 breast cancer cells) was performed to assess expression of key members of the canonical NF-κB pathway (inhibitory kappa B kinase (IKKβ) and phosphorylated p65 Ser-536 (p-p65)). Following silencing of IKKβ, cell viability and apoptosis was assessed in both MCF7 and MDA-MB-231 cell lines. P-p65 was associated with cancer-specific survival (CSS) (nuclear P=0.042 and total P=0.025). High total p-p65 was associated with increase grade tumour grade (P=0.010), ER positivity (P=0.023), molecular subtype (P=0.005), lower Klintrup- Makinen grade (P=0.013) and decreased CD138 count (P=0.032). On multivariate analysis, total p-p65 expression independently associated with poorer CSS (P=0.020). In vitro work demonstrated that the canonical NF-κB pathway was inducible by exposure to TNFα in ER-positive MCF7 cells and to a lesser extent in ER-negative MDAMB- 231 cells. Reduction of IKKβ expression by siRNA transfection increased levels of apoptosis and reduced cell viability in both MCF7 (P= 0.001, P=0.002, respectively). This is consistent with the hypothesis that canonical IKKβ-NF-κB signalling drives tumour survival. These results suggest that activation of the canonical NF-κB pathway is an important determinant of poor outcome in patients with invasive ductal breast cancer

    Synthesis of Tetragonal and Orthorhombic Polymorphs of Hf3N4 by High-Pressure Annealing of a Prestructured Nanocrystalline Precursor

    Hf3N4 in nanocrystalline form is produced by solution phase reaction of Hf(NEtMe)4 with ammonia followed by low-temperature pyrolysis in ammonia. Understanding of phase behavior in these systems is important because early transition-metal nitrides with the metal in maximum oxidation state are potential visible light photocatalysts. A combination of synchrotron powder X-ray diffraction and pair distribution function studies has been used to show this phase to have a tetragonally distorted fluorite structure with 1/3 vacancies on the anion sites. Laser heating nanocrystalline Hf3N4 at 12 GPa and 1500 K in a diamond anvil cell results in its crystallization with the same structure type, an interesting example of prestructuring of the phase during preparation of the precursor compound. This metastable pathway could provide a route to other new polymorphs of metal nitrides and to nitrogen-rich phases where they do not currently exist. Importantly it leads to bulk formation of the material rather than surface conversion as often occurs in elemental combination reactions at high pressure. Laser heating at 2000 K at a higher pressure of 19 GPa results in a further new polymorph of Hf3N4 that adopts an anion deficient cottunite-type (orthorhombic) structure. The orthorhombic Hf3N4 phase is recoverable to ambient pressure and the tetragonal phase is at least partially recoverable

    The Lick Planet Search: Detectability and Mass Thresholds

    We analyse 11 years of precise radial velocities for 76 solar type stars from the Lick survey. Eight stars in this sample have previously reported planetary-mass companions, all with mass (m sin i) less than 8 Jupiter masses (MJ). For the stars without a detected companion, we place upper limits on possible companion mass. For most stars, we can exclude companions with m sin i > 0.7 MJ (a/AU)^1/2 for orbital radii a < 5 AU. We use our results to interpret the observed masses and orbital radii of planetary-mass companions. For example, we show that the finite duration of the observations makes detection of Jupiter mass companions more and more difficult for orbital radii beyond 3 AU. Thus it is possible that the majority of solar type stars harbor Jupiter-mass companions much like our own, and if so these companions should be detectable in a few years. To search for periodicities, we adopt a "floating-mean" periodogram, which improves on the traditional Lomb-Scargle periodogram by accounting for statistical fluctuations in the mean of a sampled sinusoid.     Gap to Transition Temperature Ratio in Density Wave Ordering: a Dynamical Mean Field Study

    We use the dynamical mean-field method to determine the origin of the large ratio of the zero temperature gap to the transition temperature observed in most charge density wave materials. The method is useful because it allows an exact treatment of thermal fluctuations. We establish the relation of the dynamical mean-field results to conventional diagrammatics and thereby determine that in the physically relevant regime the origin of the large ratio is a strong inelastic scattering.Comment: 4 figure

    The effect of the systemic inflammatory response on plasma vitamin 25 (OH) D concentrations adjusted for albumin

    &lt;b&gt;Aim&lt;/b&gt;&lt;p&gt;&lt;/p&gt; To examine the relationship between plasma 25(OH)D, CRP and albumin concentrations in two patient cohorts.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Methods&lt;/b&gt;&lt;p&gt;&lt;/p&gt; 5327 patients referred for nutritional assessment and 117 patients with critical illness were examined. Plasma 25 (OH) D concentrations were measured using standard methods. Intra and between assay imprecision was &#60;10%.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Result&lt;/b&gt;&lt;p&gt;&lt;/p&gt; In the large cohort, plasma 25 (OH) D was significantly associated with CRP (rs = −0.113, p&#60;0.001) and albumin (rs = 0.192, p&#60;0.001). 3711 patients had CRP concentrations ≤10 mg/L; with decreasing albumin concentrations ≥35, 25–34 and &#60;25 g/l, median concentrations of 25 (OH) D were significantly lower from 35 to 28 to 14 nmol/l (p&#60;0.001). This decrease was significant when albumin concentrations were reduced between 25–34 g/L (p&#60;0.001) and when albumin &#60;25 g/L (p&#60;0.001). 1271 patients had CRP concentrations between 11–80 mg/L; with decreasing albumin concentrations ≥35, 25–34 and &#60;25 g/l, median concentrations of 25 (OH) D were significantly lower from 31 to 24 to 19 nmol/l (p&#60;0.001). This decrease was significant when albumin concentration were 25–34 g/L (p&#60;0.001) and when albumin &#60;25 g/L (p&#60;0.001). 345 patients had CRP concentrations &#62;80 mg/L; with decreasing albumin concentrations ≥35, 25–34 and &#60;25 g/l, median concentrations of 25 (OH) D were not significantly altered varying from 19 to 23 to 23 nmol/l. Similar relationships were also obtained in the cohort of patients with critical illness.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Conclusion&lt;/b&gt;&lt;p&gt;&lt;/p&gt; Plasma concentrations of 25(OH) D were independently associated with both CRP and albumin and consistent with the systemic inflammatory response as a major confounding factor in determining vitamin D status.&lt;p&gt;&lt;/p&gt

    Automatically extracting functionally equivalent proteins from SwissProt

    In summary, FOSTA provides an automated analysis of annotations in UniProtKB/Swiss-Prot to enable groups of proteins already annotated as functionally equivalent, to be extracted. Our results demonstrate that the vast majority of UniProtKB/Swiss-Prot functional annotations are of high quality, and that FOSTA can interpret annotations successfully. Where FOSTA is not successful, we are able to highlight inconsistencies in UniProtKB/Swiss-Prot annotation. Most of these would have presented equal difficulties for manual interpretation of annotations. We discuss limitations and possible future extensions to FOSTA, and recommend changes to the UniProtKB/Swiss-Prot format, which would facilitate text-mining of UniProtKB/Swiss-Prot

    Rapid dynamic activation of a marine-based Arctic ice cap

    We use satellite observations to document rapid acceleration and ice loss from a formerly slow-flowing, marine-based sector of Austfonna, the largest ice cap in the Eurasian Arctic. During the past two decades, the sector ice discharge has increased 45-fold, the velocity regime has switched from predominantly slow (~ 101 m/yr) to fast (~ 103 m/yr) flow, and rates of ice thinning have exceeded 25 m/yr. At the time of widespread dynamic activation, parts of the terminus may have been near floatation. Subsequently, the imbalance has propagated 50 km inland to within 8 km of the ice cap summit.
