    Maintenance of a Protein Structure in the Dynamic Evolution of TIMPs over 600 Million Years

    Deciphering the events leading to protein evolution represents a challenge, especially for protein families showing complex evolutionary history. Among them, TIMPs represent an ancient eukaryotic protein family widely distributed in the animal kingdom. They are known to control the turnover of the extracellular matrix and are considered to arise early during metazoan evolution, arguably tuning essential features of tissue and epithelial organization. To probe the structure and molecular evolution of TIMPs within metazoans, we report the mining and structural characterization of a large data set of TIMPs over approximately 600 Myr. The TIMPs repertoire was explored starting from the Cnidaria phylum, coeval with the origins of connective tissue, to great apes and humans. Despite dramatic sequence differences compared with highest metazoans, the ancestral proteins displayed the canonical TIMP fold. Only small structural changes, represented by an α-helix located in the N-domain, have occurred over the evolution. Both the occurrence of such secondary structure elements and the relative solvent accessibility of the corresponding residues in the three-dimensional structures raises the possibility that these sites represent unconserved element prone to accept variations

    The nucleic acid-binding protein PcCNBP is transcriptionally regulated during the immune response in red swamp crayfish Procambarus clarkii

    Gene family encoding cellular nucleic acid binding proteins (CNBP) is well conserved among vertebrates; however, there is limited knowledge in lower organisms. In this study, a CNBP homolog from the red swamp crayfish Procambarus clarkii was characterised. The full-length cDNA of PcCNBP was of 1257 bp with a 5′-untranslated region (UTR) of 63 bp and a 3′-UTR of 331 bp with a poly (A) tail, and an open-reading frame (ORF) of 864 bp encoding a polypeptide of 287 amino acids with the predicted molecular weight of about 33 kDa. The predicted protein possesses 7 tandem repeats of 14 amino acids containing the CCHC zinc finger consensus sequence, two RGG-rich single-stranded RNA-binding domain and a nuclear localization signal, strongly suggesting that PcCNBP was a homolog of vertebrate CNBP. The PcCNBP transcript was constitutively expressed in all tested tissues of unchallenged crayfish, including hepatopancreas, gill, eyestalk, haemocytes, intestine, stomach and cuticle with highest expression in haemocytes, intestine, gills and hepatopancreas. The mRNA expression of PcCNBP in haemocytes was modulated at transcriptional level by different immune challenges, suggesting its involvement in the immune response of P. clarkii during both bacteria and viruses infection

    European anchovy (Engraulis encrasicolus) age structure and growth rate in two contrasted areas of the Mediterranean Sea: the paradox of faster growth in oligotrophic seas

    Fishery production is highly dependent on fish growth and environmental factors; primary production and temperature are the two most important variables affecting almost all biological rates, especially individual growth or population production rate. The growth of European anchovy, Engraulis encrasicolus, was compared between two populations inhabiting different ecosystems: the Tyrrhenian Sea, characterized by riverine inputs and the Strait of Sicily, an oligotrophic area. The effects of habitat conditions on growth was also evaluated using literature data on other of European anchovy populations. Water temperature was confirmed to be a determinant factor for anchovy growth at the species level positively affecting the length at age-1. On the contrary, a negative relationship was found between chlorophyll-a and the length at age-1. These findings suggested that size at age-1 is strongly dependent on habitat conditions. The age structure differed significantly between the two populations, with the Tyrrhenian stock dominated by age-1 fish

    Density dependence in the spatial behaviour of anchovy and sardine across Mediterranean systems

    A spatial indicator—the spreading area index—is used to describe anchovy and sardine spatial distribution in relation to biomass variation and to look for ecosystem differences within the Mediterranean basin. Specifically, the variation in the spreading area index in relation to biomass was examined for different areas of the Mediterranean Sea (i.e. Aegean Sea, western Adriatic Sea, Strait of Sicily, Gulf of Lion, and Spanish Mediterranean waters). In order to capture the spatial variability of the population at different levels of fish density, acoustic survey data for the years of highest, lowest, and intermediate abundance were used. In a subsequent step standardized values of spreading area and biomass were estimated to allow comparisons. Results showed pronounced area differences. A significant relationship was revealed in the case of anchovy for areas with extended continental shelf (i.e. Aegean Sea, Adriatic Sea, and Gulf of Lion), indicating an increase in biomass with an increase in the spreading area. No relationship was found for areas dominated by narrow continental shelf and strong currents (i.e. Spanish Mediterranean waters and the Strait of Sicily). With regard to sardine, an increase in biomass was followed by an increase in the spreading area when estimates from the Aegean Sea, the Adriatic Sea, and the Strait of Sicily were considered together. The relationship was even more Abstracts–Theme Session B 9 pronounced when analysis was limited to the Aegean Sea and the Strait of Sicily. No relationship was found for the Spanish Mediterranean waters and the Gulf of Lion. This clearly implies that spatial indicators should be integrated into ecosystem management, taking into account that they can be area‐ or ecosystem‐dependent

    Sound production mechanism in the semiterrestrial crab Neohelice granulata (Brachyura, Varunidae)

    Very few studies of sound production in the Brachyura have simultaneously identified the type of individuals (e.g. sex) producing acoustic signals, the structures involved in making sound and the social context. The emission and type of sound signals in Neohelice granulata were previously characterized, but the sex and the body structures involved in the sound production mechanism were not determined. In the present study, experiments conducted in the laboratory demonstrated that acoustic signals were produced by males through an up-down movement of the cheliped by rubbing the merus against the pterygostomial area of the carapace. The micromorphology of the merus showed that it has a ridge of tubercles which may act as a plectrum, while the pterygostomial area bears tubercles and might function as the pars stridens. Acoustic signals were displayed more frequently in the presence of receptive females. Agonistic encounters among males also occurred more often in the presence of receptive females. We propose that Neohelice granulata males use their chelipeds to produce sound signals in a mating context, probably to attract the receptive female and/or to repel other males when a receptive female is present.     Molecular Identification of Atlantic Bluefin Tuna (Thunnus thynnus, Scombridae) Larvae and Development of a DNA Character-Based Identification Key for Mediterranean Scombrids

    The Atlantic bluefin tuna, Thunnus thynnus, is a commercially important species that has been severely over-exploited in the recent past. Although the eastern Atlantic and Mediterranean stock is now showing signs of recovery, its current status remains very uncertain and as a consequence their recovery is dependent upon severe management informed by rigorous scientific research. Monitoring of early life history stages can inform decision makers about the health of the species based upon recruitment and survival rates. Misidentification of fish larvae and eggs can lead to inaccurate estimates of stock biomass and productivity which can trigger demands for increased quotas and unsound management conclusions. Herein we used a molecular approach employing mitochondrial and nuclear genes (CO1 and ITS1, respectively) to identify larvae (n = 188) collected from three spawning areas in the Mediterranean Sea by different institutions working with a regional fisheries management organization. Several techniques were used to analyze the genetic sequences (sequence alignments using search algorithms, neighbour joining trees, and a genetic character-based identification key) and an extensive comparison of the results is presented. During this process various inaccuracies in related publications and online databases were uncovered. Our results reveal important differences in the accuracy of the taxonomic identifications carried out by different ichthyoplanktologists following morphology- based methods. While less than half of larvae provided were bluefin tuna, other dominant taxa were bullet tuna (Auxis rochei), albacore (Thunnus alalunga) and little tunny (Euthynnus alletteratus). We advocate an expansion of expertise for a new generation of morphology-based taxonomists, increased dialogue between morphology-based and molecular taxonomists and increased scrutiny of public sequence databases.Versión del editor4,411

    Clinical features and outcomes of elderly hospitalised patients with chronic obstructive pulmonary disease, heart failure or both

    Background and objective: Chronic obstructive pulmonary disease (COPD) and heart failure (HF) mutually increase the risk of being present in the same patient, especially if older. Whether or not this coexistence may be associated with a worse prognosis is debated. Therefore, employing data derived from the REPOSI register, we evaluated the clinical features and outcomes in a population of elderly patients admitted to internal medicine wards and having COPD, HF or COPD + HF. Methods: We measured socio-demographic and anthropometric characteristics, severity and prevalence of comorbidities, clinical and laboratory features during hospitalization, mood disorders, functional independence, drug prescriptions and discharge destination. The primary study outcome was the risk of death. Results: We considered 2,343 elderly hospitalized patients (median age 81 years), of whom 1,154 (49%) had COPD, 813 (35%) HF, and 376 (16%) COPD + HF. Patients with COPD + HF had different characteristics than those with COPD or HF, such as a higher prevalence of previous hospitalizations, comorbidities (especially chronic kidney disease), higher respiratory rate at admission and number of prescribed drugs. Patients with COPD + HF (hazard ratio HR 1.74, 95% confidence intervals CI 1.16-2.61) and patients with dementia (HR 1.75, 95% CI 1.06-2.90) had a higher risk of death at one year. The Kaplan-Meier curves showed a higher mortality risk in the group of patients with COPD + HF for all causes (p = 0.010), respiratory causes (p = 0.006), cardiovascular causes (p = 0.046) and respiratory plus cardiovascular causes (p = 0.009). Conclusion: In this real-life cohort of hospitalized elderly patients, the coexistence of COPD and HF significantly worsened prognosis at one year. This finding may help to better define the care needs of this population

    A Gaussian model for evaluating the release of CH4 by the landfill of Palermo

    Many landfills for urban solid waste have been developed in sites very close to urbanised areas thus exposing the population to various dangerous pollutants. The landfill of "Bellolampo", located a few kilometres from the city ofPalermo is one of these. It was officially created in the eighties and only some years later it was decided convert it into a controlled landfill, soon becoming a catch basin of refuse for a large hinterland constituted by approximately 50 small towns. In recent years the city of Palermo has been equipped with a network of weather and air-quality stations for monitoring the main components that afe responsible for atmospheric pollution; it consists of 10 fixed stations located both in peripheral and centraI parts ofthe city. Four ofthem afe able to measure the CH4 concentration. In this work an analysis of the meteorological data and of the CH4 concentrations, acquired downwind the emission source, has been carried out. A dispersion Gaussian model of pollutants was applied in order to estimate the amount of the CH4 released by the landfill. The estimated values were validated by utilising the methane concentration values recorded by the biogas capture system installed in the landfill area
