    Cyclization of 1,2:3,4-di-O-isopropylidene-α-D-galacto-1,6-hexodialdo-1,5-pyranose acylhydrazone and semicarbazone

    Cyclization of 1,2:3,4-di-O-isopropylidene-α-D-galacto-1,6-hexodialdo-1,5-pyranose benzoylhydrazone using acetylating mixtures led us to the corresponding (2R)- and (2S)-5- phenyl-1,3,4-oxadiazoline derivatives. The same conditions applied to the semicarbazone produced the 5-methyl-1,3,4-oxadiazoline derivative as the main compound, which is formed with acetylating mixtures even at room temperature.     Síntesis y caracterización de un nuevo monómero para aplicaciones optoeléctricas

    Dentro del marco de nuestras investigaciones, donde se busca obtener nuevos polímeros capaces de presentar propiedades optoeléctricas, y con el objeto de analizar el efecto de un espaciador flexible entre la función polimerizable y un cluster conjugado, se exploró la reacción entre dos reactivos comerciales: la naftilamina y la caprolactona. La introducción de este espaciador flexible obedece a la hipótesis de que su presencia podría favorecer la auto estructuración del material y modificar su comportamiento. Como resultado de esta tarea y luego de analizar diferentes condiciones de reacción, se ha podido obtener un nuevo monómero, con un espaciador de longitud variable, conteniendo un promedio de entre dos y tres unidades de caprolactona. Actualmente se analiza la reproducibilidad del método y se trabaja en la obtención de mayores cantidades de masa del monómero, lo que permitirá finalmente obtener y analizar el material buscado, corroborando o refutando nuestra hipótesis inicial.Fil: Compte Alvaredo, Juan.     Interaction between HFE and haptoglobin polymorphisms and its relation with plasma glutathione levels in obese children

    Short CommunicationObesity among children has emerged as a serious public health problem. The growing prevalence of childhood obesity has led to the appearance of serious complications, including a chronic systemic inflammation associated with oxidative stress. In the present study, we analysed the interaction between two genes related with iron metabolism - HFE and haptoglobin – and the plasmatic concentration of glutathione, as a way to evaluate the antioxidant response capacity in obesity. To achieve this, 118 obese children and 89 eutrophic children were recruited for the study. Results showed that although obese children present a significantly decreased tGSH levels, once we analysed separately children based on their haptoglobin phenotype, the decreased tGSH levels is significant only for the Hp 2 allele. Additionally, Hp 2.2 obese children carrying H63D polymorphism show significantly lower tGSH/GSSG values.     Conhecimento da equipe multiprofissional acerca do trabalho da terapia ocupacional em uma unidade de internação pediátrica

    INTRODUÇÃO: Os últimos anos foram marcados por uma ampliação do mercado de trabalho para os terapeutas ocupacionais no Distrito Federal. Além disso, houve reforço no corpo clínico de profissionais da equipe multiprofissional no âmbito da SES-DF com o advento do programa de residência em saúde da criança pela Escola Superior de Ciências e Saúde de Brasília. Os profissionais foram inseridos em vários hospitais do Distrito Federal, porém, o papel desse profissional nem sempre é claro para as equipes multiprofissionais. OBJETIVO: verificar o grau de conhecimento que a equipe multiprofissional de saúde das enfermarias do Hospital Materno Infantil de Brasília tem quanto à atuação do terapeuta ocupacional no contexto hospitalar pediátrico. MÉTODO: A pesquisa foi do tipo transversal, quantitativa com aplicação de questionário. Foram 54 participantes do estudo, entre  servidores e residentes que atuam no período diurno e que aceitaram participar da pesquisa. RESULTADOS: As categorias profissionais dos participantes da pesquisa foram médicos, enfermeiros, técnicos de enfermagem e os especialistas da equipe multiprofissional, entre os quais, 90,7% responderam que tinham conhecimento quanto à profissão. CONSIDERAÇÕES FINAIS: Apesar dos profissionais participantes mostrarem ter um conhecimento acerca das atribuições e competências do terapeuta ocupacional, nota-se uma imperícia quanto ao domínio deste profissional, sendo necessário pesquisas mais minuciosa abrangendo o quadro fixo de servidores alocados nas enfermarias

    Environmental determinants of health: NOVA National School of Public Health research to tackle ongoing threats and challenges

    The environment in which we live represents a major determinant of our health and well-being. The complex human-animal-ecosystem interface imposes an increasing risk to public health with population health influenced by the interaction of multiple environmental determinants. Recognizing how the environment influences human health and disease allows us to identify the most relevant risk factors and the most suitable measures to mitigate environmental factors that adversely affect human health and ecological systems. Climate change represents a major threat to human health, and it is nowadays undermining every dimension of global health monitored, increasing the fragility of the global systems, and increasing the vulnerability of populations, with a special emphasis on the already most vulnerable. The world is now facing frequent extreme weather events such as heatwaves, floods, droughts, and hurricanes. Rising temperatures and changes in precipitation patterns also affect the seasonality and geographic range of many infectious diseases, particularly those transmitted by vectors. Predictably, air quality will be negatively impacted, leading to an increase in the burden of noncommunicable diseases. Biological and chemical contaminants in food are also impacted by climate change, modifying their virulence, occurrence, and distribution patterns, which may increase the risk of human exposure to foodborne hazards. Agricultural productivity and clean water availability may decrease, resulting in malnutrition, foodborne illnesses, and waterborne diseases, endangering food security and safety. The transition to more sustainable and healthier diets constitutes a significant challenge for human populations already under the pressure of climate change-associated events but is considered one of the solutions to tackle this global threat.info:eu-repo/semantics/publishedVersio

    Validação da Versão Portuguesa do Questionário EHP-30 (The Endometriosis Health Profile-30)

    INTRODUCTION: Endometriosis Health Profile Questionnaire-30 is currently the most used questionnaire for quality of life measurement in women with endometriosis. The aim of this study is to evaluate the psychometric properties and to validate the Portuguese Endometriosis Health Profile Questionnaire-30 version. MATERIAL AND METHODS: A sequential sample of 152 patients with endometriosis, followed in a Portugal reference center, were asked to complete a questionnaire on social and demographic features, the Portuguese version of the Endometriosis Health Profile Questionnaire-30 and of the Short Form Health Survey 36 Item â version 2. Appropriate statistical analysis was performed using descriptive statistics, factor analysis, internal consistency, item-total correlation and convergent validity. RESULTS: Factorial analysis confirmed the validity of the five-dimension structure of the Endometriosis Health Profile Questionnaire-30 core questionnaire, which explained 83.2% of the total variance. All item-total correlations presented acceptable results and high internal consistency, with Cronbach's alpha ranging between 0.876 and 0.981 for the core questionnaire and between 0.863 and 0.951 for the modular questionnaire. Significant negative associations between similar scales of Endometriosis Health Profile Questionnaire-30 and Short Form Health Survey 36 Item â version 2 were demonstrated. Data completeness achieved was high for all dimensions. The emotional well-being scale in the core questionnaire and the infertility scale in the modular section had the highest median scores, and therefore the most negative impact on the quality of life of participating women. DISCUSSION: The test-retest reliability and responsiveness of the questionnaire should be evaluated in future studies. CONCLUSION: The present study demonstrates that the Portuguese version of the Endometriosis Health Profile Questionnaire-30 is a valid, reliable and acceptable tool for evaluating the health-related quality of life of Portuguese women with endometriosis

    Relationship between Ketones, Ghrelin, and, Appetite on Isocaloric Diets with Varying Carbohydrate Quality and Amount: Results from a Randomized Controlled Trial in People with Obesity (CARBFUNC)

    Background - Low-carbohydrate high-fat (LCHF) diets may suppress the increase in appetite otherwise seen after diet-induced fat loss. However, studies of diets without severe energy restriction are lacking, and the effects of carbohydrate quality relative to quantity have not been directly compared. Objectives - To evaluated short- (3 mo) and long-term (12 mo) changes in fasting plasma concentrations of total ghrelin, β-hydroxybutyrate (βHB), and subjective feelings of appetite on 3 isocaloric eating patterns within a moderate caloric range (2000–2500 kcal/d) and with varying carbohydrate quality or quantity. Methods - We performed a randomized controlled trial of 193 adults with obesity, comparing eating patterns based on “acellular” carbohydrate sources (e.g., flour-based whole-grain products; comparator arm), “cellular” carbohydrate sources (minimally processed foods with intact cellular structures), or LCHF principles. Outcomes were compared by an intention-to-treat analysis using constrained linear mixed modeling. This trial was registered at clinicaltrials.gov as NCT03401970. Results - Of the 193 adults, 118 (61%) and 57 (30%) completed 3 and 12 mo of follow-up. Throughout the intervention, intakes of protein and energy were similar with all 3 eating patterns, with comparable reductions in body weight (5%−7%) and visceral fat volume (12%−17%) after 12 mo. After 3 mo, ghrelin increased significantly with the acellular (mean: 46 pg/mL; 95% CI: 11, 81) and cellular (mean: 54 pg/mL; 95% CI: 21, 88) diets but not with the LCHF diet (mean: 11 pg/mL; 95% CI: −16, 38). Although βHB increased significantly more with the LCHF diet than with the acellular diet after 3 m (mean: 0.16 mmol/L; 95% CI: 0.09, 0.24), this did not correspond to a significant group difference in ghrelin (unless the 2 high-carbohydrate groups were combined [mean: −39.6 pg/mL; 95% CI: −76, −3.3]). No significant between-group differences were seen in feelings of hunger. Conclusions - Modestly energy-restricted isocaloric diets differing in carbohydrate cellularity and amount showed no significant differences in fasting total ghrelin or subjective hunger feelings. An increase in ketones with the LCHF diet to 0.3–0.4 mmol/L was insufficient to substantially curb increases in fasting ghrelin during fat loss

    Naive and Stem Cell Memory T Cell Subset Recovery Reveals Opposing Reconstitution Patterns in CD4 and CD8 T Cells in Chronic Graft vs. Host Disease

    The success of allogeneic hematopoietic stem cell transplantation (allo-HSCT) in the treatment of hematological malignancies remains hampered by life-threatening chronic graft vs. host disease (cGVHD). Although multifactorial in nature, cGVHD has been associated with imbalances between effector and regulatory T cells (Treg). To further elucidate this issue, we performed a prospective analysis of patients undergoing unrelated donor allo-HSCT after a reduced intensity conditioning (RIC) regimen containing anti-thymocyte globulin (ATG) and the same GVHD prophylaxis, at a single institution. We studied T cell subset homeostasis over a 24-month follow-up after HSCT in a comparative analysis of patients with and without cGVHD. We also quantified naive and memory T cell subsets, proliferation and expression of the apoptosis-related proteins Bcl-2 and CD95. Finally, we assessed thymic function by T cell receptor excision circle (TREC) quantification and T cell receptor (TCR) diversity by TCRVβ spectratyping. While the total number of conventional CD4 (Tcon) and CD8 T cells was similar between patient groups, Treg were decreased in cGVHD patients. Interestingly, we also observed divergent patterns of Naive and Stem Cell Memory (SCM) subset recovery in Treg and Tcon compared to CD8. Patients with cGVHD showed impaired recovery of Naive and SCM Tcon and Treg, but significantly increased frequencies and absolute numbers of Naive and SCM were observed in the CD8 pool. Markedly increased EMRA CD8 T cells were also noted in cGVHD. Taken together, these results suggest that Naive, SCM and EMRA CD8 play a role in the emergence of cGHVD. Reduced Naive and recent thymic emigrant Tcon and Treg in cGVHD was likely due to impaired thymic output, as it was accompanied by decreased CD4 TREC and TCR diversity. On the other hand, CD8 TCR diversity was similar between patient groups. Furthermore, no correlation was observed between CD8 TREC content and Naive CD8 numbers, suggesting limited thymic production of Naive CD8 T cells in patients after transplant, especially in those developing cGVHD. The mechanisms behind the opposing patterns of CD4 and CD8 subset cell recovery in cGVHD remain elusive, but may be linked to thymic damage associated with the conditioning regimen and/or acute GVHD
