    Effects of coastal orientation and depth on the distribution of subtidal benthic assemblages

    Copyright © 2013 Blackwell Verlag GmbH.A better understanding of biological systems can only be gained if we understand what processes are important and how they operate to determine the distribution of organisms. Coastal orientation and depth can influence environmental conditions, including the degree of water motion and availability of light, which in turn may influence the horizontal and vertical patterns of organism distribution. Here, we used a mixed-model design to examine the effects of coastal orientation and depth on the structure of benthic assemblages by comparing the abundance and distribution of macroalgae and invertebrates in shallow and deep waters on the opposing coasts of Sa˜o Miguel. Generally, coastal orientation had little influence on the distribution of most taxa. In contrast, significant differences were generally associated with depth, although patterns were spatially variable at the scale of locations. This study suggests that depth, and processes operating at the scale of location, but not at the scale of the coast, have an important influence on these assemblages, and that failure to recognise such a scale of variability may hamper our ability to better understand the processes that structure these communities

    Chromosomal mapping of rDNAs and H3 histone sequences in the grasshopper rhammatocerus brasiliensis (acrididae, gomphocerinae): extensive chromosomal dispersion and co-localization of 5S rDNA/H3 histone clusters in the A complement and B chromosome

    <p>Abstract</p> <p>Background</p> <p>Supernumerary B chromosomes occur in addition to standard karyotype and have been described in about 15% of eukaryotes, being the repetitive DNAs the major component of these chromosomes, including in some cases the presence of multigene families. To advance in the understanding of chromosomal organization of multigene families and B chromosome structure and evolution, the distribution of rRNA and H3 histone genes were analyzed in the standard karyotype and B chromosome of three populations of the grasshopper <it>Rhammatocerus brasiliensis</it>.</p> <p>Results</p> <p>The location of major rDNA was coincident with the previous analysis for this species. On the other hand, the 5S rDNA mapped in almost all chromosomes of the standard complement (except in the pair 11) and in the B chromosome, showing a distinct result from other populations previously analyzed. Besides the spreading of 5S rDNA in the genome of <it>R. brasiliensis </it>it was also observed multiple sites for H3 histone genes, being located in the same chromosomal regions of 5S rDNAs, including the presence of the H3 gene in the B chromosome.</p> <p>Conclusions</p> <p>Due to the intense spreading of 5S rRNA and H3 histone genes in the genome of <it>R. brasiliensis</it>, their chromosomal distribution was not informative in the clarification of the origin of B elements. Our results indicate a linked organization for the 5S rRNA and H3 histone multigene families investigated in <it>R. brasiliensis</it>, reinforcing previous data concerning the association of both genes in some insect groups. The present findings contribute to understanding the organization/evolution of multigene families in the insect genomes.</p

    Acolhimento residencial em Portugal

    In Portugal there are 8000 children and young people in Residential Care (RC). The concept of RC quality emerges as the adequacy of the characteristics of the centres to the needs of children. Being a complex and multidimensional construct requires different perspectives and an evaluation by various sources, assuming the voice of children as the main reference in measuring the quality. The quality of the RC is closely related to psychological adjustment, well-being and satisfaction with life of children and adolescents. Until now it has not been performed in Portugal an assessment of the quality of the RC system. At this time there is an ongoing study that aims to assess the quality of Portuguese RC system through needs assessment and psychological adjustment of children in RC, the evaluation of the quality of services provided by centres and understanding of the relationship between these two variables. This symposium aims to present some emerging results of the pilot study of national character research, still in progress, which aims to assess the quality of the RC Portuguese system. In this sense, a brief contextualization of the RC will be in Portugal, followed by the presentation of the main methodology that is used for this assessment – A Comprehensive Evaluation System for Residential Care in Portugal (ARQUA-P). Subsequently, it will be presented the data related to the psychological adjustment of adolescents that are in RC, their self-esteem, well-being and happiness subjective, relating these variables with the quality of the RC.info:eu-repo/semantics/publishedVersio

    Peripheral axonal ensheathment is regulated by RalA GTPase and the exocyst complex

Defects in axonal insulation lead to debilitating conditions, but, despite its importance, the molecular players responsible are poorly defined. Here, we identify RalA GTPase as a key player in axon ensheathment in Drosophila larval peripheral nerves. We demonstrate through genetic analysis that RalA action through the exocyst complex is required in wrapping glial cells to regulate their growth and development. We suggest that the RalA-exocyst pathway controls the targeting of secretory vesicles for membrane growth or for the secretion of a wrapping glia-derived factor that itself regulates growth. In summary, our findings provide a new molecular understanding of the process by which axons are ensheathed in vivo, a process that is crucial for normal neuronal function.publishersversionpublishe

    Neuropsychological predictors of quality of life in focal epilepsy

    Spontaneous complaints of outpatients with focal epilepsy often stress the relationship between cognitive deficits and Quality of Life (QOL). Consequently, the aim of the present study was to find the best neuropsychological predictors of QOL in individuals with focal epilepsy, in order to guide their ambulatory health care. A sample of 71 Portuguese patients was studied: 40 female, 47 married, with a mean age of 37.48 years (S.D. = 11.79, 16–62), mean education of 7.93 (S.D. = 4.05, 3–17), and focal epilepsy of moderate severity. A Socio-demographic and Clinical Questionnaire, the SF-36 v1, the Cognitive Functioning Scale from the ESI-55, a Seizure Control scale (items from the Liverpool Seizure Severity Scale), and several neuropsychological tests were used. Semantic Fluency was the only predictor of Physical Functioning, Role Functioning – Physical, and Mental Health; I.A. Test predicted Bodily Pain; and AttentiveMatrices predicted General Health, Vitality, and Role Functioning – Emotional. The Mental Component of the SF-36 v1 was predicted by Attentive Matrices, and the Physical Component was predicted by Semantic Fluency. Cognitive Functioning was predicted by the Token Test. Social Functioning and Seizure Control presented no statistically significant correlation with the neuropsychological indicators used. These results underscore the importance of cognitive performance to the QOL of individuals with focal epilepsy, supporting the systematic screening of cognitive performance in this population. Additionally, they suggest cognitive rehabilitation has the potential to improve these individuals’ QOL

    Novel Perspectives in Management of Angiogenesis and Cancer Therapy

FL-C PhD fellowship was funded by FCT (PD/BD/128337/2017).The activation of endothelial cells (ECs) is a crucial step on the road map of tumor angiogenesis and expanding evidence indicates that a pro-oxidant tumor microenvironment, conditioned by cancer metabolic rewiring, is a relevant controller of this process. Herein, we investigated the contribution of oxidative stress-induced ferroptosis to ECs activation. Moreover, we also addressed the anti-angiogenic effect of Propranolol. We observed that a ferroptosis-like mechanism, induced by xCT inhibition with Erastin, at a non-lethal level, promoted features of ECs activation, such as proliferation, migration and vessel-like structures formation, concomitantly with the depletion of reduced glutathione (GSH) and increased levels of oxidative stress and lipid peroxides. Additionally, this ferroptosis-like mechanism promoted vascular endothelial cadherin (VE-cadherin) junctional gaps and potentiated cancer cell adhesion to ECs and transendothelial migration. Propranolol was able to revert Erastin-dependent activation of ECs and increased levels of hydrogen sulfide (H2S) underlie the mechanism of action of Propranolol. Furthermore, we tested a dual-effect therapy by promoting ECs stability with Propranolol and boosting oxidative stress to induce cancer cell death with a nanoformulation comprising selenium-containing chrysin (SeChry) encapsulated in a fourth generation polyurea dendrimer (SeChry@PUREG4). Our data showed that novel developments in cancer treatment may rely on multi-targeting strategies focusing on nanoformulations for a safer induction of cancer cell death, taking advantage of tumor vasculature stabilization.publishersversionpublishe

Saccharomyces pastorianus, used to produce Lager beer, is widely reused, not changing its fermentation performance. However, S. cerevisiae, used to produce Ale beer, is partial or not even reused, due to its poor performance. It is hypothesized that cells modulate their wall polysaccharides to increase the cell-wall strength. In this work industrial S. cerevisiae and S. pastorianus brewer's spent yeasts with different repitching numbers were studied. Glucans were the main component of S. cerevisiae whereas mannoproteins were abundant in S. pastorianus. The major changes were noticed on glucans of both species, β1,3-glucans decrease more pronounced in S. cerevisiae. The increase of α1,4-Glc, related with osmotolerance, was higher in S. cerevisiae while β1,4-Glc, related with cell-wall strength, had a small increase. In addition, these structural details showed different binding profiles to immune receptors, important to develop tailored bioactive applications.publishersversionpublishe

    Loss of postprandial insulin clearance control by Insulin-degrading enzyme drives dysmetabolism traits

    Systemic insulin availability is determined by a balance between beta-cell secretion capacity and insulin clearance (IC). Insulin-degrading enzyme (IDE) is involved in the intracellular mechanisms underlying IC. The liver is a major player in IC control yet the role of hepatic IDE in glucose and lipid homeostasis remains unexplored. We hypothesized that IDE governs postprandial IC and hepatic IDE dysfunction amplifies dysmetabolic responses and prediabetes traits such as hepatic steatosis. In a European/Portuguese population-based cohort, IDE SNPs were strongly associated with postprandial IC in normoglycemic men but to a considerably lesser extent in women or in subjects with prediabetes. Liver-specific knockout-mice (LS-IDE KO) under normal chow diet (NCD), showed reduced postprandial IC with glucose intolerance and under high fat diet (HFD) were more susceptible to hepatic steatosis than control mice. This suggests that regulation of IC by IDE contributes to liver metabolic resilience. In agreement, LS-IDE KO hepatocytes revealed reduction of Glut2 expression levels with consequent impairment of glucose uptake and upregulation of CD36, a major hepatic free fatty acid transporter. Together these findings provide strong evidence that dysfunctional IC due to abnormal IDE regulation directly impairs postprandial hepatic glucose disposal and increases susceptibility to dysmetabolic conditions in the setting of Western diet/lifestyle.publishe

    Estratégias educacionais do projeto Nutrition up 65: curso de qualificação de profissionais de saúde

    INTRODUCTION: Nutrition knowledge among health professionals has a major impact on older adults’ healthcare. However, there is a lack of specialized education offer in this area. Within the Nutrition UP 65 Project, a health professional’s qualification nutrition course was created and implemented from April 2015 to April 2017. We aim to describe this process here. METHODOLOGY: At Faculdade de Ciências da Nutrição e Alimentação da Universidade do Porto, a course of twenty-seven hours was created by six professors. The course comprises eight hours of direct contact and nineteen hours of individual study and was accredited by the University of Porto and credited with one European Credit Transfer System (ECTS). Educational materials and presentations for each session were produced including posters and flyers. A final test was developed to evaluate attendee’s knowledge. Approximately 10% of national health units were identified, randomly selected in each country regional area (NUTS II – “Nomenclatura Comum das Unidades Territoriais para Fins Estatísticos”) and invited to participate. RESULTS: Thirty-five course editions were carried out by twenty-three Porto University accredited instructors for 784 professionals from 241 different health units. Concerning the knowledge evaluation, a 99% approval rate was achieved and the average mark score was of 16.8 out of 20. Course satisfaction survey revealed that 45.7% of the participants classified the course as “good” and 41.1% as “very good”. CONCLUSIONS: The overall balance of this branch of the Nutrition UP 65 project is very positive. The results on health professional’s adherence demonstrate that the interest in this area of nutrition care is high and that this nutrition course is a good opportunity to expand access to specialized education. This highlights the need for further initiatives promoting educational sessions targeting nutrition topics. These course sessions must be acknowledged as a starting point to a new approach in older adult’s nutrition knowledge in Portugal.INTRODUÇÃO: O conhecimento sobre nutrição dos profissionais de saúde tem um grande impacto na saúde dos indivíduos idosos. No entanto, há escassez de oferta de educação especializada nessa área. O Projeto Nutrition UP 65 foi criado e implementado a nível nacional um curso de qualificação em nutrição para profissionais de saúde, que decorreu entre abril de 2015 e abril de 2017. Pretende-se descrever aqui este processo. METODOLOGIA: Na Faculdade de Ciências da Nutrição e Alimentação da Universidade do Porto, seis professores elaboraram um curso de vinte e sete horas. Este curso é composto por oito horas de contacto direto e dezanove horas de estudo individual e foi acreditado e creditado pela Universidade do Porto com um European Credit Transfer System (ECTS). Foram produzidos materiais didáticos e apresentações, bem como cartazes e desdobráveis informativos. Desenvolveu-se uma prova final para avaliar os conhecimentos dos participantes. Cerca de 10% das unidades de saúde nacionais foram identificadas, selecionadas aleatoriamente em cada área regional (NUTS II) e convidadas a participar. RESULTADOS: Realizaram-se 35 edições do curso para 784 profissionais de 241 unidades de saúde diferentes por 23 instrutores credenciados pela Universidade do Porto. No teste de avaliação obteve-se uma taxa de aprovação de 99% e uma pontuação média de 16,8 em 20 valores. 45,7% dos participantes classificaram o curso como “bom” e 41,1% como “muito bom”. CONCLUSÕES: O balanço global desta vertente do Projeto Nutrition UP 65 é muito positivo. Os resultados da adesão dos profissionais de saúde demonstram que o interesse por esta área da intervenção nutricional é elevado e que este curso é uma boa oportunidade para expandir o acesso à formação especializada. Assim, destaca-se a necessidade de outras iniciativas que promovam sessões de formação voltadas para tópicos nutricionais.
