Background: Leptospirosis is currently a source of significant economic losses in the agribusiness; as such, experimental studies on this infection are required to develop a better understanding of the pathogenesis, treatment, and immunoprophylaxis of the disease. Sheep may represent a good model for ruminants in such models. Despite the extent of the studies that has been conducted thus far, researchers have yet to reach a consensus on the experimental practices to apply for leptospirosis in this animal species, and several gaps in understanding remain. To bridge these gaps, the present study aimed to assess the usage of several tools for the monitoring of experimental leptospirosis in sheep.Material, Methods & Results: Twelve Santa Ines sheep of different ages were each allocated to one of four groups (A, B, C, and D). The subjects in groups A, B, and C received different doses of Leptospira interrogans serogroup Icterohemorrhagiae by intraperitoneal route, 1x102, 1x105, and 1x108 respectively. Group D was the control. Hematological, biochemical and clinical parameters were evaluated daily. Serology by microscopic agglutination test (MAT) and PCR were performed to evaluate the infection status. The most remarkable clinical signs were fever (41ºC) and dehydration, and acute pain (cub). Two animals from Group C presented leukocytosis. Only those in Group C exhibited positive results according to serology, while positivity in PCR was observed in animals in groups A and C. The results of the experiment indicated that sheep may be experimentally infected and can, therefore, be used as a model for leptospirosis in ruminants. Clinical signs cannot be considered to represent a reliable parameter for evaluating the development of leptospirosis in experimentally infected sheep. We recommend the use of urine PCR and serology to confirm the infection in experimentally infected animals and daily complete blood count (CBC) as a follow-up tool.Discussion: It was observed that the clinical signs cannot be considered as a reliable parameter to evaluate the pathogenesis in experimentally infected ewes, being important to emphasize that the age of the animals does not seem to alter their susceptibility to the infection. This finding is in agreement with other experimental studies, which report that leptospirosis infection in ruminants occurs asymptomatic and subclinical. Hematological and biochemical tests proved to be adequate tools to monitor the experimental infection. Studies have shown that the complete blood count has been used to monitor the acute phase of leptospirosis and is effective in detecting anemia and leukocytosis with neutrophilia in ruminants. Despite the lack of clinical signs, the serological and molecular results confirmed the experimental infection. PCR has been used as an important tool in the diagnosis of leptospirosis. In addition, the current study is the first of its kind to use PCR to detect the carrier status in experimentally infected ewes. Despite this limitation, PCR was very effective in confirming the infection and should be considered for use in experimental studies. Sheep have been used as a good experimental model in several studies, sheep are relatively small compared to other ruminants and can be easily allocated in smaller pens and pens, facilitating the management of research and minimizing the costs of experimentation. In this context, we suggest that sheep represent a good model for the study of leptospirosis in ruminants and therefore a reliable protocol for experimental infection by leptospirosis is necessary

Cocos nucifera L. (Arecaceae) is a widely distributed species around the tropical areas. Popular uses have been reported in the treatment of arthritis and diarrhea. This study evaluates the antimicrobial activity of husk fiber aqueous extract from C. nucifera and performed the identification of some biological active substances. The minimal inhibitory concentration (MIC) against human pathogen microorganisms was determined. Chromatographic and spectrometric procedures were also performed to isolate and identify the components present in the extract. In the MIC assay of crude aqueous extract, only the methicillin sensible and the resistant (MRSA) Staphylococcus aureus strains were susceptible at 156 μg/mL. The ethyl acetate partition taken from crude extract was more promising (MIC of 78 μg/mL). No fungal growth inhibition was observed. Catechin, epicatechin, two procyanidin dimers and condensed tannins were found in the organic phase. In addition, gallic and ellagic acids were detected for the first time in C. nucifera husk fiber. Gallic acid showed MIC of 39 μg/mL and minimal bactericidal concentration (MBC) at 78 μg/mL. Ellagic acid was not active against the tested strains, as well as catechin and epicatechin. Additionally catechin, epicatechin, two procyanidin dimers and condensed tannins were also detected. The antimicrobial activity observed was selective to S. aureus strains. Keywords: Antimicrobial analyses, Cocos nucifera, arecaceae, ellagic acid, gallic acid, procyanidins, Staphylococcus aureus strainsAfrican Journal of Biotechnology Vol. 12(18), pp. 2478-248

The beneficial effects of a water-based aerobic exercise session on the blood lipids of women with dyslipidemia are independent of their training status

OBJECTIVES: To evaluate the acute effects of a session of water-based aerobic exercise on the blood lipid levels of women with dyslipidemia and to compare these results according to their training status. METHOD: Fourteen premenopausal women with dyslipidemia, aged 40–50 years, participated in two waterbased aerobic exercise sessions, the first when they were generally sedentary and the second after they were trained with a water-based aerobic training program for 12 weeks. Both experimental sessions were performed using the same protocol, lasted 45 min, and incorporated an interval method, alternating 3 min at a rating of perceived exertion (RPE) of 13 and 2 min at an RPE of 9. Total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), high-density lipoprotein (HDL), and lipoprotein lipase enzyme (LPL) were obtained through venous blood collection before and immediately after each session. A generalized estimating equation method and Bonferroni tests were conducted (with time and training status as factors) for statistical analyses. RESULTS: At enrollment, the mean age of the participants was 46.57 years (95% confidence interval [CI] 44.81– 48.34). The statistical analyses showed a significant time effect for all variables (TC: p=0.008; TG: p=0.012; HDL: po0.001; LPL: po0.001) except for LDL (p=0.307). However, the training status effect was not significant for any variable (TC: p=0.527; TG: p=0.899; HDL: p=0.938; LDL: p=0.522; LPL: p=0.737). These results indicate that the TC and TG levels reduced and the HDL and LPL concentrations increased from pre- to post-session in similar magnitudes in both sedentary and trained women. CONCLUSIONS: A single water-based aerobic exercise session is sufficient and effective to beneficially modify the lipid profile of women with dyslipidemia, regardless of their training status

Since the advent of systems and synthetic biology, many studies have sought to harness microbes as cell factories through genetic and metabolic engineering approaches. Yeast and filamentous fungi have been successfully harnessed to produce fine and high value-added chemical products. In this review, we present some of the most promising advances from recent years in the use of fungi for this purpose, focusing on the manipulation of fungal strains using systems and synthetic biology tools to improve metabolic flow and the flow of secondary metabolites by pathway redesign. We also review the roles of bioinformatics analysis and predictions in synthetic circuits, highlighting in silico systemic approaches to improve the efficiency of synthetic modules
