    Hypothalamic AMPK-ER Stress-JNK1 Axis Mediates the Central Actions of Thyroid Hormones on Energy Balance

    Thyroid hormones (THs) act in the brain to modulate energy balance. We show that central triiodothyronine (T3) regulates de novo lipogenesis in liver and lipid oxidation in brown adipose tissue (BAT) through the parasympathetic (PSNS) and sympathetic nervous system (SNS), respectively. Central T3 promotes hepatic lipogenesis with parallel stimulation of the thermogenic program in BAT. The action of T3 depends on AMP-activated protein kinase (AMPK)-induced regulation of two signaling pathways in the ventromedial nucleus of the hypothalamus (VMH): decreased ceramide-induced endoplasmic reticulum (ER) stress, which promotes BAT thermogenesis, and increased c-Jun N-terminal kinase (JNK) activation, which controls hepatic lipid metabolism. Of note, ablation of AMPKalpha1 in steroidogenic factor 1 (SF1) neurons of the VMH fully recapitulated the effect of central T3, pointing to this population in mediating the effect of central THs on metabolism. Overall, these findings uncover the underlying pathways through which central T3 modulates peripheral metabolism

    Recognition of emotional facial expressions in Alexithymia

    Alexithymia is a personality trait which is associated with difficulties in identifying and verbalizing emotions. Previous studies have shown a significant association between alexithymia and a lack of ability to decode emotional facial expressions. Three groups of university students (N = 1645) were formed by splitting the sample based on Toronto Alexithymia Scale (TAS-20) scores. All participants performed an emotional expression recognition task, using the "Reading the Mind in the Eyes'" method. The experimenter presented images of the eye-region of the faces of actors and actresses. As expected, results showed that alexithymics performed worse on ability to infer others' emotional states expressed by the eyes. These results suggest that alexithymia is associated with impaired emotion recognition, that is most apparent when processing capacity is restricted, high-alexithymic individuals could develop less detailed perceptual representations of facial expression which might impair the process of drawing conclusions about its emotional significance

    Weight Loss and Sleep, Current Evidence in Animal Models and Humans

    Sleep is a vital process essential for survival. The trend of reduction in the time dedicated to sleep has increased in industrialized countries, together with the dramatic increase in the prevalence of obesity and diabetes. Short sleep may increase the risk of obesity, diabetes and cardiovascular disease, and on the other hand, obesity is associated with sleep disorders, such as obstructive apnea disease, insomnia and excessive daytime sleepiness. Sleep and metabolic disorders are linked; therefore, identifying the physiological and molecular pathways involved in sleep regulation and metabolic homeostasis can play a major role in ameliorating the metabolic health of the individual. Approaches aimed at reducing body weight could provide benefits for both cardiometabolic risk and sleep quality, which indirectly, in turn, may determine an amelioration of the cardiometabolic phenotype of individuals.     En 1854 con el descubrimiento del espejito laríngeo por Manuel García, profesor de canto español, nació la laringología como especialidad. Desde entonces han sido numerosos los avances en el terreno de la valoración visual de la dinámica laríngea durante la fonación. En los últimos años la tecnología digital ha permitido no sólo la obtención y registro de imágenes de manera precisa y fiable sino que permite realizar mediciones de fenómenos extremadamente rápidos y delicados, como son el desplazamiento de la mucosa de las cuerdas vocales, los cambios del área glótica durante el ciclo vocal, etc. Se revisa la historia de la estroboscopia, los protocolos de valoración, las técnicas de la estroboscopia digital y sus posibilidades diagnósticas así como sus limitaciones. Se propone así mismo dos nuevas técnicas que han irrumpido recientemente en el panorama otorrinolaringológico que son la videokimografía digital y la grabación en vídeo de alta velocidad que superan las limitaciones de la estroboscopia y probablemente se convertirán en un medio de exploración rutinario en un futuro próximo

    Current professional standing of young medical oncologists in Spain : a nationwide survey by the Spanish Society of Medical Oncology + MIR section

    There is a lack of knowledge about the career paths and employment situation of young medical oncologists. The aim of our study was to evaluate the current professional standing of these professionals in Spain. The Spanish Society of Medical Oncology + MIR section conducted a national online survey in May 2021 of young medical oncology consultants (< 6 years of expertise) and final year medical oncology residents. A total of 162 responses were eligible for analysis and included participants from 16 autonomous communities; 64% were women, 80% were consultants, and 20% were residents. More than half of the participants performed routine healthcare activity and only 7% research activity. Almost three quarters (73%) were subspecialized in a main area of interest and almost half of these chose this area because it was the only option available after residency. Half of the respondents (51%) considered working abroad and 81% believed the professional standing in Spain was worse than in other countries. After finishing their residency, only 22 were offered a job at their training hospital. Just 16% of participants had a permanent employment contract and 87% were concerned (score of ≥ 5 on a scale of 1-10) about their job stability. In addition, one quarter of the participants in our study showed an interest in increasing their research activity. The choice of subspecialty in medical oncology may depend on job opportunities after residency rather than personal interest. The abundance of temporary contracts may have influenced the job stability concerns observed. Future mentoring strategies should engage in building a long-term career path for young medical oncologists.     Aurora A drives early signalling and vesicle dynamics during T-cell activation

    Aurora A is a serine/threonine kinase that contributes to the progression of mitosis by inducing microtubule nucleation. Here we have identified an unexpected role for Aurora A kinase in antigen-driven T-cell activation. We find that Aurora A is phosphorylated at the immunological synapse (IS) during TCR-driven cell contact. Inhibition of Aurora A with pharmacological agents or genetic deletion in human or mouse T cells severely disrupts the dynamics of microtubules and CD3z-bearing vesicles at the IS. The absence of Aurora A activity also impairs the activation of early signalling molecules downstream of the TCR and the expression of IL-2, CD25 and CD69. Aurora A inhibition causes delocalized clustering of Lck at the IS and decreases phosphorylation levels of tyrosine kinase Lck, thus indicating Aurora A is required for maintaining Lck active.     Growing up in the deep-sea protected development in deep-sea invertebrates: A case study in the southwestern Atlantic Ocean

    Marine invertebrates display a range of reproductive strategies, from external fertilization to several methods to protect the young. Both brooding or protected development of benthic species and direct development species (when juveniles born in a smaller version of its adult form) involves reducing the time that the larvae and/or juveniles spend in the plankton. The different forms of juvenile protection are often associated with the taxonomic group, although certain groups are much diversified in terms of their reproductive modes. However, the area where the species live also seems to play a key role. Several authors have indicated the unusual proportion of brooding species in the Southern Ocean, in particular in the southwestern Atlantic, including its adjacent deep-sea. The knowledge about the reproductive characteristics of most of the fauna in this area remains unknown. Proof of this are the great number of studies carried out in recent years, describing the reproductive features of several species (many also new), with some of them having many peculiarities about their biology.Fil: Teso, Silvia Valeria.     Safety and immediate humoral response of COVID-19 vaccines in chronic kidney disease patients:the SENCOVAC study

    BACKGROUND: Chronic kidney disease (CKD) patients are at high-risk for severe Covid-19. The multicentric, observational and prospective SENCOVAC study aims to describe the humoral response and safety of SARS-CoV-2 vaccines in CKD patients. Safety and immediate humoral response results are reported here. METHODS: Four cohorts of patients were included: kidney transplant (KT) recipients, haemodialysis (HD), peritoneal dialysis (PD) and non-dialysis CKD patients from 50 Spanish centres. Adverse events after vaccine doses were recorded. At baseline and on day 28 after the last vaccine dose, anti-Spike antibodies were measured and compared between cohorts. Factors associated with development of anti-Spike antibodies were analyzed. RESULTS: 1746 participants were recruited: 1116 HD, 171 PD, 176 non-dialysis CKD patients and 283 KT recipients. Most patients (98%) received mRNA vaccines. At least one vaccine reaction developed after the first dose in 763 (53.5%) and after the second dose in 741 (54.5%) of patients. Anti-Spike antibodies were measured in the first 301 patients. At 28 days, 95% of patients had developed antibodies: 79% of KT, 98% of HD, 99% of PD and 100% of non-dialysis CKD patients (p<0.001). In a multivariate adjusted analysis, absence of an antibody response was independently associated to KT (OR 20.56, p = 0.001) and to BNT162b2 vaccine (OR 6.03, p = 0.023). CONCLUSION: The rate of anti-Spike antibody development after vaccination in KT patients was low but in other CKD patients it approached 100%; suggesting that KT patients require persistent isolation measures and booster doses of a Covid-19 vaccine. Potential differences between Covid-19 vaccines should be explored in prospective controlled studies
