    Cattle identification based in biometric features of the muzzle

    Regional and temporal changes in bivalve diversity off the south coast of Portugal

Seasonal tendencies in diversity varied according the year, thus the interpretation of long-term and regional scales is essential before drawing any conclusions in other studies. Richness and diversity consistently decreased with depth and increased with sediment grain size (from low in very coarse sand to high in coarse silt). Diversity decreased progressively from 3 to 16 m depth, thus the harsher shallower environments (due to waves and tidal air exposure) showed greater diversity than the most stable areas. Communities in finer sediments were more diverse than those in coarser sand. Evenness showed patterns opposite to diversity, overall.Diversity and evenness maps (produced with multivariate universal kriging), showed that most geographic areas with greater diversity were farer from river outflows and wastewater treatment plants. Two types of geographic pattern were observed: areas with persistently greater bivalve diversity through time and areas that changed locally from year to year. This spatial analysis can be used to establish priority conservation areas for management purposes, and to analyse the persistency of regional diversity patterns. The area with most habitat heterogeneity (Sotavento) corresponded to greatest diversity.There was a positive relationship between Spisula solida and Chamelea gallina landings and bivalve diversity 2 years and 1 year later, respectively. Possibly, local fisheries, by selectively withdrawing the commercial numerically dominant species from the ecosystem, increased diversity 1 to 2 years later, as the ecological niches of the dominants are quickly filled by several other species thereby creating a more even community.     Análise da percepção ergonómica de postos de trabalho dotados de microscópio

    A adequabilidade do dimensionamento dos postos e equipamentos de trabalho aos trabalhadores e às tarefas desenvolvidas é fundamental para a promoção de condições de conforto e prevenção do desenvolvimento de LMERT. A percepção dos trabalhadores é importante para a análise e intervenção ergonómica. Este estudo teve como objectivo analisar a percepção dos trabalhadores sobre a adequabilidade dos postos de trabalho que envolvam tarefas ao microscópio. Foi analisada a adequabilidade das dimensões dos elementos do mobiliário e do microscópio, e desenvolvido e aplicado o Questionário de Análise Ergonómica de Tarefas ao Microscópio (QAETM). O questionário contemplou questões associadas a características pessoais, actividades desenvolvidas, percepções de risco e sensações de dor e/ou desconforto. A análise das dimensões dos postos de trabalho mostrou que estas se apresentam em geral inadequadas às tarefas realizadas. Verificou-se ainda que alguns postos de trabalho tinham sofrido intervenções por parte dos profissionais. A maioria dos inquiridos considera as dimensões das cadeiras adequadas, contrariamente à das bancadas. No que respeita ao microscópio, foram sugeridas alterações ao nível dos parafusos de focagem, movimentação de platina e altura e inclinação das oculares. Este estudo permitiu verificar que alguns dos profissionais apresentam uma baixa percepção ergonómica, não sendo na sua maioria capazes de identificar a falta de adequabilidade das dimensões dos seus postos de trabalho. Apesar do referido, alguns revelaram preocupações ergonómicas, sugerindo e identificando, nomeadamente ao nível do microscópio, situações a alterar, tendo mesmo a iniciativa de intervir no seu posto de trabalho de modo a promover uma melhor postura.info:eu-repo/semantics/publishedVersio

Abstract Whipple disease (WD) is a rare systemic infection caused by Tropheryma whippelii. Neurological involvement has been recognised in 40% of patients, either as initial manifestations or during the course of the disease. We report on a 45 years-old man with WD with initial, persistent and irresistible episodes of daytime somnolence. The patient was HLA-DQB1*0602 positive (genetic marker for narcolepsy). WD diagnosis was suspected on clinical and MRI basis and confirmed by histological and immunohistochemical study of duodenal biopsy. Forty months later all clinical features improved, narcoleptic-like episodes disappeared and cerebral MRI and CSF normalised. Longitudinal neurophysiological studies revealed persistent sleep pattern abnormalities with sleep fragmentation, paucity of slow wave and of REM sleep. The disruption of the hypocretin circuitry in the hypothalamic - diencephalic region triggered by the infection was the probable cause of the hypersomnia and narcopleptic symptoms. WD should be added to the list of causes of secondary hypersomnia

    Study of prevalence, severity and risk factors of periodontal disease in a Portuguese population

    Periodontal disease is a common worldwide oral inflammation/infection affecting tissues that surround and support teeth. This study aims to evaluate the prevalence, extent and severity of periodontal diseases and its risk factors, according to the most recent periodontal classification, in an adult population of Northern Portugal. This observational study collected data from clinical records of patients who attended the University Clinic of Gandra between April 2021 and April 2022. Of a total of 941 patients included, 457 (48.6%) had periodontitis, 253 (26.9%) had gingivitis and the remaining 231 (24.5%) were healthy patients. The prevalence of stage III severe periodontitis was 51.2%, more prevalent in males, and in the age group of 61–70 years. Gingivitis was more prevalent in females, and in the age group of 31–40 years; in both diseases, the most prevalent extension was the generalized one. Using a binary logistic regression, we observe a significant relation of the risk of periodontitis with age (p = 0.019; OR 1.033; 95% CI 1.005–1.062), tooth brushing (p = 0.002; OR 0.25; 95% CI 0.105–0.599) and dental flossing (p = 0.015; OR 0.63; 95% CI 0.09–0.768). This study revealed a high prevalence of periodontitis. Increased age, lack of tooth brushing and flossing were identified as potential risk factors for periodontitis in the investigated Portuguese population.This research was funded by Instituto Universitário de Ciencias da Saúde (CESPU) and by CESPU Diagnóstico.     Homogeneous batch micro-crystallization of proteins from ammonium sulfate

    The emergence of X-ray free-electron lasers has led to the development of serial macromolecular crystallography techniques, making it possible to study smaller and more challenging crystal systems and to perform time-resolved studies on fast time scales. For most of these studies the desired crystal size is limited to a few micrometres, and the generation of large amounts of nanocrystals or microcrystals of defined size has become a bottleneck for the wider implementation of these techniques. Despite this, methods to reliably generate microcrystals and fine-tune their size have been poorly explored. Working with three different enzymes, L-aspartate alpha-decarboxylase, copper nitrite reductase and copper amine oxidase, the precipitating properties of ammonium sulfate were exploited to quickly transition from known vapour-diffusion conditions to reproducible, large-scale batch crystallization, circumventing the tedious determination of phase diagrams. Furthermore, the specific ammonium sulfate concentration was used to fine-tune the crystal size and size distribution. Ammonium sulfate is a common precipitant in protein crystallography, making these findings applicable to many crystallization systems to facilitate the production of large amounts of microcrystals for serial macromolecular crystallography experiments.Peer reviewe

    Safety of oral ivermectin during pregnancy: a systematic review and meta-analysis

    Background: About 3·7 billion doses of ivermectin have been distributed in mass drug administration (MDA) campaigns globally over the past 30 years. At 10–100 times higher than current human doses, ivermectin is a known teratogen in mammals. During these campaigns with recommended doses, pregnant women might be inadvertently exposed. We therefore aimed to evaluate the existing evidence for serious and non-serious adverse events after ivermectin exposure in pregnant women. Methods: For this systematic review and meta-analysis, we searched relevant databases and trial registry platforms on July 15, 2018, for randomised controlled trials (RCTs) and observational studies that reported adverse events in pregnant women. We did not use language or date restrictions. Outcomes of interest were spontaneous abortions, stillbirths, congenital anomalies, and neonatal death (serious adverse events), as well as maternal morbidity, preterm births, and low birthweight (adverse events). The risk of bias was assessed using the Newcastle-Ottawa Scale for observational studies and the Cochrane Risk of Bias Tool for RCTs. We did the meta-analysis of observational studies and RCTs separately. The quality of evidence was assessed using the GRADE approach. The study protocol is registered with PROSPERO, protocol CRD42016046914. Findings: We identified 147 records, of which only five observational studies and one RCT were included for quantitative analysis; these studies were published between 1990 and 2008, and were done in six African countries. 893 women with 899 pregancy outcomes were included, of whom 496 pregnant women (500 pregnancy outcomes) received ivermectin inadvertently during MDA campaigns in the observational studies and 397 pregnant women (399 pregnancy outcomes) purposely received ivermectin as part of the open-label RCT. No study reported neonatal deaths, maternal morbidity, preterm births, or low birthweight. It is unclear whether exposure to ivermectin during pregnancy increases the risk of spontaneous abortions and stillbirths (odds ratio [OR] 1·15 [95% CI 0·75–1·78] with very low certainty of evidence for the four observational studies and 0·62 [0·18–2·14] with very low certainty of evidence for the RCT) or congenital anomalies (OR 1·69 [95% CI 0·83–3·41] with very low certainty of evidence for the five observational studies and 1·10 [0·07–17·65] with very low certainty of evidence for the RCT). Interpretation: There is insufficient evidence to conclude on the safety profile of ivermectin during pregnancy. Treatment campaigns should focus additional efforts on preventing inadvertent treatment of pregnant women

    Tlx3 exerts direct control in specifying excitatory over inhibitory neurons in the dorsal spinal cord

No use, distribution or reproduction is permitted which does not comply with these terms.The spinal cord dorsal horn is a major station for integration and relay of somatosensory information and comprises both excitatory and inhibitory neuronal populations. The homeobox gene Tlx3 acts as a selector gene to control the development of late-born excitatory (dILB) neurons by specifying glutamatergic transmitter fate in dorsal spinal cord. However, since Tlx3 direct transcriptional targets remain largely unknown, it remains to be uncovered how Tlx3 functions to promote excitatory cell fate. Here we combined a genomics approach based on chromatin immunoprecipitation followed by next generation sequencing (ChIP-seq) and expression profiling, with validation experiments in Tlx3 null embryos, to characterize the transcriptional program of Tlx3 in mouse embryonic dorsal spinal cord. We found most dILB neuron specific genes previously identified to be directly activated by Tlx3. Surprisingly, we found Tlx3 also directly represses many genes associated with the alternative inhibitory dILA neuronal fate. In both cases, direct targets include transcription factors and terminal differentiation genes, showing that Tlx3 directly controls cell identity at distinct levels. Our findings provide a molecular frame for the master regulatory role of Tlx3 in developing glutamatergic dILB neurons.
