    Inflammation in stroke: the role of cholinergic, purinergic and glutamatergic signaling

    The inflammatory response is a major factor in stroke pathophysiology and contributes to secondary neuronal damage in both acute and chronic stages of the ischemic injury. Recent work in experimental cerebral ischemia has demonstrated the involvement of neurotransmitter signaling in the modulation of neuroinflammation. The present review discusses recent findings on the therapeutic potential and diagnostic perspectives of cholinergic, purinergic and glutamatergic receptors and transporters in experimental stroke. It provides evidence of the role of neurotransmission signaling as a promising inflammatory biomarker in stroke. Finally, recent molecular imaging studies using positron emission tomography of cholinergic receptors and glutamatergic transporters are outlined along with their potential as novel anti-inflammatory therapy to reduce the outcome of cerebral ischemia.We acknowledge financial support by MINECO SAF2014-54070-JIN (A.M.) and SAF2016-75292-R (C.M.)

    Type of judge and decisions in dismissal cases: An analysis of Labour Courts in Spain

    El objetivo de este artículo es profundizar en la relación existente entre las decisiones judiciales de lo social en España en los casos de despido y los costes asociados a la extinción de los contratos laborales. Primero justificamos que los cambios en la probabilidad de que un juez falle a favor de un trabajador afectan a los costes de despido del empleador. En segundo lugar, examinamos cómo difiere dicha probabilidad cuando el juez es titular o sustituto. Se observa una mayor dispersión en la resolución de los casos de los jueces sustitutos, y un significativo repunte en los años de crisis económica de los fallos judiciales a favor del trabajador al considerar los jueces titulares, siendo este más débil en los casos de los sustitutosThe aim of this paper is to examine in detail the relationship between the decisionsof the Spanish labour courts regarding dismissal, and the costs associated with the terminationof employment contracts. We explain the changes in the probability that a judge ruling infavour of a worker affects the effective firing costs that the employer faces. We also examine how this probability differs depending on whether the judge is in charge of the court or is asubstitute. There is a greater dispersion in the resolution of cases when there are substitute judges, with a significant rise, during the economic crisis, of court decisions in favour of theworkers with permanent judges, which is less so with substitute judge

    An Expedient Method for the Umpolung Coupling of Enols with Heteronucleophiles

    [EN] In this paper, we present an unprecedented and general umpolung protocol that allows the functionalization of silyl enol ethers and of 1,3-dicarbonyl compounds with a large range of heteroatom nucleophiles, including carboxylic acids, alcohols, primary and secondary amines, azide, thiols, and also anionic carbamates derived from CO2. The scope of the reaction also extends to carbon-based nucleophiles. The reaction relies on the use of 1-bromo-3,3-dimethyl-1,3-dihydro-1 lambda(3)[d][1,2]iodaoxole, which provides a key alpha-brominated carbonyl intermediate. The reaction mechanism has been studied experimentally and by DFT, and we propose formation of an unusual enolonium intermediate with a halogen-bonded bromide.The authors are grateful for support from the Swedish Research Council through Vetenskapsradet, and from the Goran Gustafsson Foundation.     In Vitro Culture of ‘Myrobalan’ (Prunus cerasifera Ehrh.) Embryos

This tool has been used to germinate seeds of early-maturing or hybrid Prunus species. ‘Myrobalan’ (Prunus cerasifera Ehrh.) is a widely used rootstock for plum and apricot cultivars and its interspecific hybrids have a clear potential for breeding purposes. However, early seed abortion is often a problem in interspecific crosses because no protocol has been established yet for ‘Myrobalan’ seeds. In this work, we developed a procedure for in vitro germination of embryos of different sizes. Various factors affecting embryo germination such as the culture media, the presence of cotyledons, the stratification temperature, and the embryo size were tested in three different ‘Myrobalan’ clones. The developed protocol includes the use of full embryos that were stratified at 4 °C and cultured in C2d culture medium. The germination rate was strongly affected by the embryo size and reached 90% germination with intermediate- to large-sized embryos (6.5 to 10 mm). However, smaller embryos could also be germinated, and up to 30% germination was achieved with 0.5- to 2-mm long embryos.     Characterization of goat colostrum oligosaccharides by nano-liquid chromatography on chip quadrupole time-of-flight mass spectrometry and hydrophilic interaction liquid chromatography-quadrupole mass spectrometry

    A detailed qualitative and quantitative characterization of goat colostrum oligosaccharides (GCO) has been carried out for the first time. Defatted and deproteinized colostrum samples, previously treated by size exclusion chromatography (SEC) to remove lactose, were analyzed by nanoflow liquid chromatography–quadrupole-time of flight mass spectrometry (Nano-LC-Chip–Q-TOF MS). Up to 78 oligosaccharides containing hexose, hexosamine, fucose, N-acetylneuraminic acid or Nglycolylneuraminic acid monomeric units were identified in the samples, some of them detected for the first time in goat colostra. As a second step, a hydrophilic interaction liquid chromatography coupled to mass spectrometry (HILIC-MS) methodology was developed for the separation and quantitation of the main GCO, both acidic and neutral carbohydrates. Among other experimental chromatographic conditions, mobile phase additives and column temperature were evaluated in terms of retention time, resolution, peak width and symmetry of target carbohydrates. Narrow peaks (wh: 0.2–0.6 min) and good symmetry (As: 0.8–1.4) were obtained for GCO using an acetonitrile:water gradient with 0.1% ammonium hydroxide at 40 ◦C. These conditions were selected to quantify the main oligosaccharides in goat colostrum samples. Values ranging from 140 to 315 mg L−1 for neutral oligosaccharides and from 83 to 251 mg L−1 for acidic oligosaccharides were found.     Multidisciplinary consensus statement on the clinical management of patients with pancreatic cancer

    Pancreatic cancer (PC) remains one of the most aggressive tumors with an increasing incidence rate and reduced survival. Although surgical resection is the only potentially curative treatment for PC, only 15-20% of patients are resectable at diagnosis. To select the most appropriate treatment and thus improve outcomes, the diagnostic and therapeutic strategy for each patient with PC should be discussed within a multidisciplinary expert team. Clinical decision-making should be evidence-based, considering the staging of the tumor, the performance status and preferences of the patient. The aim of this guideline is to provide practical and evidence-based recommendations for the management of PC
