Exo1 phosphorylation inhibits exonuclease activity and prevents fork collapse in rad53 mutants independently of the 14-3-3 proteins
The S phase checkpoint is crucial to maintain genome stability under conditions that threaten DNA replication. One of its critical functions is to prevent Exo1-dependent fork degradation, and Exo1 is phosphorylated in response to different genotoxic agents. Exo1 seemed to be regulated by several post-translational modifications in the presence of replicative stress, but the specific contribution of checkpoint-dependent phosphorylation to Exo1 control and fork stability is not clear. We show here that Exo1 phosphorylation is Dun1-independent and Rad53-dependent in response to DNA damage or dNTP depletion, and in both situations Exo1 is similarly phosphorylated at multiple sites. To investigate the correlation between Exo1 phosphorylation and fork stability, we have generated phospho-mimic exo1 alleles that rescue fork collapse in rad53 mutants as efficiently as exo1-nuclease dead mutants or the absence of Exo1, arguing that Rad53-dependent phosphorylation is the mayor requirement to preserve fork stability. We have also shown that this rescue is Bmh1–2 independent, arguing that the 14-3-3 proteins are dispensable for fork stabilization, at least when Exo1 is downregulated. Effect of the methacrylate-based endodontic sealer Epiphany on rat peritoneal macrophages viability
Objective:
To evaluate the effect of the endodontic sealer Epiphany on rat peritoneal macrophages viability.
Materials and methods:
Peritoneal macrophages were obtained from Wistar rats and resuspended in RPMI-
1640 medium. Undiluted (crude extract) and diluted extracts to 10%, 1%, 0.1%, 0.01%, 0.001% and 0.0001% of
Epiphany, AH 26 and AH Plus sealers on RPMI-1640 medium were tested for cytotoxicity to rat peritoneal macrophages using the trypan blue dye exclusion assay. Data were analyzed statistically by the Kruskal-Wallis and
Mann-Whitney tests at 5% significance level.
Results:
Crude extract of Epiphany killed 51% of cells, but was less cytotoxic that crude extracts of AH Plus and
AH 26, which killed 81% and 86% of cells, respectively. Ten-fold dilutions of Epiphany, AH Plus and AH 26 killed
44%, 56%, 62% of macrophages, respectively. A hundred dilution of Epiphany only killed 7% of macrophages,
but the same dilution of AH Plus and AH 26 killed 10% and 31% of macrophages, respectively. Lower dilutions of
sealer extracts caused minimal cell death as compared to the control groups (p>0.05).
Conclusions:
The methacrylate-based endodontic sealer Epiphany showed lower cytotoxicity on macrophages than
resin-based sealers AH Plus and AH 26. Dilution of elutes of the three materials by tenfold markedly reduced their
effect
High incidence of clinical fragility fractures in postmenopausal women with rheumatoid arthritis. A case-control study.
Objectives: To estimate the incidence of clinical fragility fractures in postmenopausal women with rheumatoid arthritis (RA) and analyze risk factors for fracture. Methods: Incidence of clinical fragility fractures in 330 postmenopausal women with RA was compared to that of a control population of 660 age-matched postmenopausal Spanish women. Clinical fractures during the previous five years were recorded. We analyzed associations with risk factors for fracture in both populations and with disease-related variables in RA patients. Results: Median age of RA patients was 64 years; median RA duration was eight years. Sixty-nine percent were in remission or on low activity. Eighty-five percent had received glucocorticoids (GCs); 85 %, methotrexate; and 40 %, ≥1 biologic DMARD. Fifty-four patients and 47 controls had ≥1 major osteoporotic fracture (MOF). Incidence of MOFs was 3.55 per 100 patient-year in patients and 0.72 in controls (HR: 2.6). Risk factors for MOFs in RA patients were age, previous fracture, parental hip fracture, years since menopause, BMD, erosions, disease activity and disability, and cumulative dose of GCs. Previous fracture in RA patients was a strong risk for MOFs (HR: 10.37). Conclusion: Of every 100 postmenopausal Spanish women with RA, 3-4 have a MOF per year. This is more than double that of the general population. A previous fracture poses a high risk for a new fracture. Other classic risk factors for fracture, RA disease activity and disability, and the cumulative dose of GCs are associated with fracture development
Retos del Programa de Acción Tutorial: la adaptación curricular y nuevas vías de interacción con los estudiantes
El programa de adaptación curricular pretende adaptar el proceso de enseñanza-aprendizaje tanto a las necesidades específicas del alumnado universitario (movilidad, deporte de élite, violencia de género, vida laboral) como a las características particulares de los estudiantes con necesidades educativas especiales derivadas de una situación de discapacidad, física o psíquica, para que puedan acceder al currículo y alcanzar los objetivos de las distintas asignaturas y planes de estudio en igualdad de condiciones que el resto de estudiantes. En este sentido, la Facultad de Derecho ha resuelto varias solicitudes del alumnado de adaptación curricular durante el curso 2015-2016, en cuyo procedimiento se han implicado distintos agentes: la dirección del Centro, el voluntariado, el profesorado afectado y el CAE, junto con los tutores del PAT. La revisión por la Red del procedimiento de actuación previsto en el artículo 5 del Reglamento de Adaptación curricular en la Universidad de Alicante y su implementación en la Facultad de Derecho ha puesto de manifiesto la necesidad de acomodar las medidas de adaptación sobre las solicitudes presentadas por alumnos con discapacidad psíquica que requieren la intervención del Centro de Apoyo al Estudiante de la Universidad de Alicante
Effects of intubation timing in patients with COVID-19 throughout the four waves of the pandemic : a matched analysis
The primary aim of our study was to investigate the association between intubation timing and hospital mortality in critically ill patients with COVID-19-associated respiratory failure. We also analysed both the impact of such timing throughout the first four pandemic waves and the influence of prior non-invasive respiratory support on outcomes. This is a secondary analysis of a multicentre, observational and prospective cohort study that included all consecutive patients undergoing invasive mechanical ventilation due to COVID-19 from across 58 Spanish intensive care units (ICU) participating in the CIBERESUCICOVID project. The study period was between 29 February 2020 and 31 August 2021. Early intubation was defined as that occurring within the first 24 h of intensive care unit (ICU) admission. Propensity score (PS) matching was used to achieve balance across baseline variables between the early intubation cohort and those patients who were intubated after the first 24 h of ICU admission. Differences in outcomes between early and delayed intubation were also assessed. We performed sensitivity analyses to consider a different timepoint (48 h from ICU admission) for early and delayed intubation. Of the 2725 patients who received invasive mechanical ventilation, a total of 614 matched patients were included in the analysis (307 for each group). In the unmatched population, there were no differences in mortality between the early and delayed groups. After PS matching, patients with delayed intubation presented higher hospital mortality (27.3% versus 37.1%, p =0.01), ICU mortality (25.7% versus 36.1%, p=0.007) and 90-day mortality (30.9% versus 40.2%, p=0.02) when compared to the early intubation group. Very similar findings were observed when we used a 48-hour timepoint for early or delayed intubation. The use of early intubation decreased after the first wave of the pandemic (72%, 49%, 46% and 45% in the first, second, third and fourth wave, respectively; first versus second, third and fourth waves p<0.001). In both the main and sensitivity analyses, hospital mortality was lower in patients receiving high-flow nasal cannula (n=294) who were intubated earlier. The subgroup of patients undergoing NIV (n=214) before intubation showed higher mortality when delayed intubation was set as that occurring after 48 h from ICU admission, but not when after 24 h. In patients with COVID-19 requiring invasive mechanical ventilation, delayed intubation was associated with a higher risk of hospital mortality. The use of early intubation significantly decreased throughout the course of the pandemic. Benefits of such an approach occurred more notably in patients who had received high-flow nasal cannul
Insights from the genome of the biotrophic fungal plant pathogen Ustilago maydis
Ustilago maydis is a ubiquitous pathogen of maize and a well-established model organism for the study of plant-microbe interactions. This basidiomycete fungus does not use aggressive virulence strategies to kill its host. U. maydis belongs to the group of biotrophic parasites (the smuts) that depend on living tissue for proliferation and development. Here we report the genome sequence for a member of this economically important group of biotrophic fungi. The 20.5-million-base U. maydis genome assembly contains 6,902 predicted protein-encoding genes and lacks pathogenicity signatures found in the genomes of aggressive pathogenic fungi, for example a battery of cell-wall-degrading enzymes. However, we detected unexpected genomic features responsible for the pathogenicity of this organism. Specifically, we found 12 clusters of genes encoding small secreted proteins with unknown function. A significant fraction of these genes exists in small gene families. Expression analysis showed that most of the genes contained in these clusters are regulated together and induced in infected tissue. Deletion of individual clusters altered the virulence of U. maydis in five cases, ranging from a complete lack of symptoms to hypervirulence. Despite years of research into the mechanism of pathogenicity in U. maydis, no 'true' virulence factors had been previously identified. Thus, the discovery of the secreted protein gene clusters and the functional demonstration of their decisive role in the infection process illuminate previously unknown mechanisms of pathogenicity operating in biotrophic fungi. COVID-19 vaccine failure
COVID-19 affects the population unequally with a higher impact on aged and
immunosuppressed people. Hence, we assessed the effect of SARS-CoV-2 vaccination
in immune compromised patients (older adults and oncohematologic patients),
compared with healthy counterparts. While the acquired humoral and cellular memory
did not predict subsequent infection 18 months after full immunization, spectral and
computational cytometry revealed several subsets within the CD8+ T-cells, B-cells, NK
cells, monocytes and CD45RA+
CCR7- Tγδ cells differentially expressed in further
infected and non-infected individuals not just following immunization, but also prior to
that. Of note, up to 7 subsets were found within the CD45RA+
CCR7-
Tγδ population with
some of them being expanded and other decreased in subsequently infected individuals.
Moreover, some of these subsets also predicted COVID-induced hospitalization in
oncohematologic patients. Therefore, we hereby have identified several cellular subsets
that, even before vaccination, strongly related to COVID-19 vulnerability as opposed to

Key Factors Associated With Pulmonary Sequelae in the Follow-Up of Critically Ill COVID-19 Patients
Introduction: Critical COVID-19 survivors have a high risk of respiratory sequelae. Therefore, we aimed to identify key factors associated with altered lung function and CT scan abnormalities at a follow-up visit in a cohort of critical COVID-19 survivors. Methods: Multicenter ambispective observational study in 52 Spanish intensive care units. Up to 1327 PCR-confirmed critical COVID-19 patients had sociodemographic, anthropometric, comorbidity and lifestyle characteristics collected at hospital admission; clinical and biological parameters throughout hospital stay; and, lung function and CT scan at a follow-up visit. Results: The median [p25–p75] time from discharge to follow-up was 3.57 [2.77–4.92] months. Median age was 60 [53–67] years, 27.8% women. The mean (SD) percentage of predicted diffusing lung capacity for carbon monoxide (DLCO) at follow-up was 72.02 (18.33)% predicted, with 66% of patients having DLCO < 80% and 24% having DLCO < 60%. CT scan showed persistent pulmonary infiltrates, fibrotic lesions, and emphysema in 33%, 25% and 6% of patients, respectively. Key variables associated with DLCO < 60% were chronic lung disease (CLD) (OR: 1.86 (1.18–2.92)), duration of invasive mechanical ventilation (IMV) (OR: 1.56 (1.37–1.77)), age (OR [per-1-SD] (95%CI): 1.39 (1.18–1.63)), urea (OR: 1.16 (0.97–1.39)) and estimated glomerular filtration rate at ICU admission (OR: 0.88 (0.73–1.06)). Bacterial pneumonia (1.62 (1.11–2.35)) and duration of ventilation (NIMV (1.23 (1.06–1.42), IMV (1.21 (1.01–1.45)) and prone positioning (1.17 (0.98–1.39)) were associated with fibrotic lesions. Conclusion: Age and CLD, reflecting patients’ baseline vulnerability, and markers of COVID-19 severity, such as duration of IMV and renal failure, were key factors associated with impaired DLCO and CT abnormalities
