    Unraveling cellular and molecular mechanisms of acid stress tolerance and resistance in marine species: New frontiers in the study of adaptation to ocean acidification

    Since the industrial revolution, fossil fuel combustion has led to a 30 %-increase of the atmospheric CO2 con- centration, also increasing the ocean partial CO2 pressure. The consequent lowered surface seawater pH is termed ocean acidification (OA) and severely affects marine life on a global scale. Cellular and molecular re- sponses of marine species to lowered seawater pH have been studied but information on the mechanisms driving the tolerance of adapted species to comparatively low seawater pH is limited. Such information may be obtained from species inhabiting sites with naturally low water pH that have evolved remarkable abilities to tolerate such conditions. This review gathers information on current knowledge about species naturally facing low water pH conditions and on cellular and molecular adaptive mechanisms enabling the species to survive under, and even benefit from, adverse pH conditions. Evidences derived from case studies on naturally acidified systems and on resistance mechanisms will guide predictions on the consequences of future adverse OA scenarios for marine biodiversity

    Trichoderma harzianum T-22 induces systemic esistance in tomato Infected by Cucumber mosaic virus

    Understanding the induction of plant defenses against viruses using biocontrol agents is essential for developing new strategies against these pathogens, given the ineffectiveness of chemical treatments. The ability of Trichoderma harzianum, strain T-22 (T22) to control Cucumber mosaic virus (CMV) in Solanum lycopersicum var. cerasiforme plants and the changes in the physiology of tomato treated/infected with T22/CMV were examined. Plant growth-promoting effects, photosynthetic performance, reactive oxygen species scavenging enzymes, and phytohormones were investigated. T22 improved tomato growth in terms of plant height and improved photosynthesis, total chlorophyll content and plant gas exchange. In contrast, CMV induced a negative effect on dry matter accumulation and inhibited the photosynthetic capacity. The analysis of plant hormones demonstrated that treating with T22 before or simultaneously to CMV infection, led to a systemic resistance by jasmonic acid/ethylene and salicylic acid signaling pathways. Conversely, systemic resistance was abscissic acid-dependent when T22 treatment was administered after the CMV infection. In conclusion, the data reported here indicate that the T22-based strategy may be the most effective measure against CMV

    Ambient vibrations of age-old masonry towers: results of long-term dynamic monitoring in the historic centre of Lucca

    The paper presents the results of an ambient vibration monitoring campaign conducted on so-called Clock Tower (Torre delle Ore), one the best known and most visited monuments in the historic centre of Lucca. The vibrations of the tower were continuously monitored from November 2017 to March 2018 using high-sensitivity instrumentation. In particular, four seismic stations provided by the Istituto Nazionale di Geofisica e Vulcanologia and two three-axial accelerometers developed by AGI S.r.l., spin-off of the Istituto Nazionale di Astrofisica, were installed on the tower. The measured vibration level was generally very low, since the structure lies in the middle of a limited traffic area. Nevertheless, the availability of two different types of highly sensitive and accurate instruments allowed the authors to follow the dynamic behaviour of the tower during the entire monitoring period and has moreover provided cross-validation of the results

    Esta investigación se desarrolla en un compañía del sector construcción, la cual tienen como actividad principal la ejecución y elaboración de proyectos de construcción, siendo el objetivo principal de este trabajo aplicar la Sección 23 de Ingresos por Actividades Ordinarias de Norma Internacionales de Información Financiera para Pequeñas y Medianas Entidades en la Empresa Constructora del Sur S.A en el Contrato de Construcción “Rehabilitación del Canal No. 1 de la Planta Carlos Fonseca Amador III de febrero del año 2018, desarrollando las bases para la adopción de la sección 23 como es el Método de Porcentaje de terminación en los contratos de Construcción, destacando la importancia que tiene para la empresa la utilización un método que le permita el cálculo apropiado de sus ingresos. El método utilizado en el trabajo fue el método investigativo descriptivo con un enfoque de estudio en las Norma Internacionales de Información Financiera para Pequeñas y Medianas Entidades especificamnete la Sección 23 de Ingresos por Actividades ordinarias, para esto se solicitó a la empresa se brindara la información necesaria para poder aplicar y desarrollar un caso práctico logrando los resultados esperados. Dentro de las conclusiones se puede resumir la importancia que tienen para Constructora del Sur S.A la adopción del Método de Porcentaje de Terminación en los Contratos de Construcción establecido en la Sección 23 de Ingresos por Actividades Ordinarias de Norma Internacionales de Información Financiera para Pequeñas y Medianas, ya que a traves de este, se pueden determinar los avances fisicos y financieros, facilitando el reconocimiento y medicion de los ingresos para la compañía

    A user experience analysis for a mobile Mixed Reality application for cultural heritage

    Mixed Reality has emerged as a valuable tool for the promotion of cultural heritage. In this context, in particular, the metaphor of virtual portals allows the virtual visit of monuments that are inaccessible or no longer exist in their original form, integrating them into the real environment. This paper presents the development of a Mixed Reality mobile application that proposes a virtual reconstruction of the church of Sant’Elia in Ruggiano, in the southern province of Lecce (Italy). By placing the virtual portal in the same place where the entrance of the church was located, the user can cross this threshold to enter inside and make a virtual journey into the past. The user experience was evaluated by administering a questionnaire to 60 users who tried the application. From the data collected, four user experience factors were identified (interest, focus of attention, presence and usability), which were compared between young and old, male and female users, and between users who had already visited the church in person and all other users. In general, the scores reveal a total independence of the other three factors from usability and a very high level of interest

    Plexiform architecture in gastrointestinal stromal tumors is not restricted to succinate dehydrogenase-deficient cases

    Accumulating evidence reveals the heterogeneous features of gastrointestinal stromal tumors (GISTs), primarily distinguished by their various molecular triggers defining well characterized subgroups. The identification of the pathogenetic group a given GIST belongs to, in combination with the currently adopted GIST prognosticators, is pivotal for the correct management of GIST patients. Epidemiological, anatomical and morphological features are more or less strictly associated with the various possible GIST molecular pathogenesis; therefore, they can concur to addressing molecular analysis or even influence the identification of GIST subsets by themselves. This is particularly true in a cost/benefit perspective aimed at cutting the expenses of pathology labs. Under these circumstances, a correct classical pathological analysis still appears a fundamental step to achieve an optimal GIST characterization.We herein report a gastric epithelioid PDGFRA-mutant GIST displaying the multinodular/plexiform architecture distinctive of succinate dehydrogenase (SDH)-deficient GISTs. Immunohistochemistry and molecular analysis led to the correct tumor characterization. The reported case constitutes a valuable contribution to GIST pathology in that it demonstrates that multinodular/plexiform architecture is not restricted to SDH-deficient GISTs, but can be found also in PDGFRA-mutant ones; this is an event to be aware of, given the predilection for gastric location and epithelioid morphology shared by these two GIST subgroups, only the latter of which includes imatinib-sensitive cases. Tetrapods have been grown in a reproducible way and separated from the other possible ZnO nano-morphologies, by the optimization of growth parameters. The further deposition of these nanostructures on an alumina substrate with contacts and heater, allowed us to test the gas sensing properties of the obtained ZnO tetrapods with different gases

    Bioactive compound profiling of olive fruit: the contribution of genotype

    The health, therapeutic, and organoleptic characteristics of olive oil depend on functional bioactive compounds, such as phenols, tocopherols, squalene, and sterols. Genotype plays a key role in the diversity and concentration of secondary compounds peculiar to olive. In this study, the most important bioactive compounds of olive fruit were studied in numerous international olive cultivars during two consecutive seasons. A large variability was measured for each studied metabolite in all 61 olive cultivars. Total phenol content varied on a scale of 1–10 (3831–39,252 mg kg1) in the studied cultivars. Squalene values fluctuated over an even wider range (1–15), with values of 274 to 4351 mg kg1. Total sterols ranged from 119 to 969 mg kg1, and total tocopherols varied from 135 to 579 mg kg1 in fruit pulp. In the present study, the linkage among the most important quality traits highlighted the scarcity of cultivars with high content of at least three traits together. This work provided sound information on the fruit metabolite profile of a wide range of cultivars, which will facilitate the studies on the genomic regulation of plant metabolites and development of new olive genotypes through genomics-assisted breeding.EEA San JuanFil: Mousavi, Soraya.     TRPV1 channels are critical brain inflammation detectors and neuropathic pain biomarkers in mice

    The capsaicin receptor TRPV1 has been widely characterized in the sensory system as a key component of pain and inflammation. A large amount of evidence shows that TRPV1 is also functional in the brain although its role is still debated. Here we report that TRPV1 is highly expressed in microglial cells rather than neurons of the anterior cingulate cortex and other brain areas. We found that stimulation of microglial TRPV1 controls cortical microglia activation per se and indirectly enhances glutamatergic transmission in neurons by promoting extracellular microglial microvesicles shedding. Conversely, in the cortex of mice suffering from neuropathic pain, TRPV1 is also present in neurons affecting their intrinsic electrical properties and synaptic strength. Altogether, these findings identify brain TRPV1 as potential detector of harmful stimuli and a key player of microglia to neuron communication
