    Alternatives to Animal Experimentation: Its Institutional Teaching and Scientific

    Although it is desirable to replace scientific procedures with live animals by other methods that do not use them, the use of animals in scientific procedures should be restricted to those areas that benefit human, animal, and environmental health. The use of animals as experimental models of observation of biological phenomena has evolved with man, to this day. The use of animals for scientific or educational purposes should be considered only when there is no other alternative and it is governed by the principles of replacement, reduction, and refinement. The scientists should be sure that the information obtainable with the experiments is not yet available or that the protocol was designed taking into account animal protection considerations. The chosen methods must use the least number of animals; provide satisfactory results; use the species with the least ability to experience pain, suffering, anguish, and damage; and be optimal for the extrapolation of results to the target species such as humans. It will be fundamental to guarantee on a scientific and ethical basis that the use of an animal is subject to a careful evaluation regarding the scientific or educational validity

    Social sustainability in the agroindustrial sector of Trujillo state - Venezuela. A Relevant construct for development

    This paper was to analyze the social sustainability of the Agroindustry of the Trujillo-Venezuela state. Realism was used as an ontological reference, consistent with the inductive empirical epistemological approach of quantitative studies, with a field design, not experimental and cross. The population was constituted by twentytwo (22) companies, whose administrators were given a semistructured questionnaire with 17 items. The processing of the information was done through descriptive statistics. The results show that these companies have a medium level of social sustainability, influenced by the degree of competitiveness of the five dimensions that make it up.El objetivo de esta investigación fue analizar la sostenibilidad social del sector agroindustrial del estado Trujillo-Venezuela. Se utilizó como referente ontológico el realismo, coherente con el enfoque epistemológico empírico inductivo de los estudios de carácter cuantitativo, con un diseño de campo, no experimental y transeccional. La población estuvo constituida por veintidós (22) empresas, a cuyos administradores se le aplicó un cuestionario semiestructurado con 17 items. El procesamiento de la información se realizó a través de la estadística descriptiva. Los resultados demuestran que estas empresas tienen un nivel medio de sostenibilidad social, influenciado por el grado de competitividad de las cinco dimensiones que la conforman

    Macrolide resistance and molecular typing of Mycoplasma pneumoniae infections during a 4 year period in Spain

    Mycoplasma pneumoniae (MP) causes community-acquired pneumonia affecting mainly children, and tends to produce cyclic outbreaks. The widespread use of macrolides is increasing resistance rates to these antibiotics. Molecular tools can help in diagnosis, typing and resistance detection, leading to better patient management. To assess the MP genotypes and resistance pattern circulating in our area while comparing serological and molecular diagnosis of MP. Molecular and serological diagnosis of MP was performed in 821 samples collected in Badalona (Barcelona, Spain) from 2013 to 2017. Multiple locus variable number tandem repeat analysis (MLVA) and macrolide resistance detection by pyrosequencing were performed in those cases positive by PCR. Presence of respiratory viruses and relevant clinical data were also recorded. MP was detected in 16.8% of cases by PCR, with an overall agreement with serology of 76%. Eleven different MLVA types were identified, with 4-5-7-2 (50.1%) and 3-5-6-2 (29.2%) being the most abundant, with the latter showing a seasonal increase during the study. A total of 8% of the strains harboured a point substitution associated with macrolide resistance, corresponding mainly to an A2063G 23S rRNA mutation and directly related to previous macrolide therapy. Analysis of respiratory viruses showed viral coinfections in most cases. Serological and molecular tools combined could improve MP diagnosis and the analysis of its infection patterns. Macrolide resistance is associated with previous therapy. Given that MP pneumonia usually resolves spontaneously, it should be reconsidered whether antibiotic treatment is suitable for all cases

    Psychosocial profile of juvenile and adult offenders who acknowledge having committed child-to-parent violence

    The main objective of this study was to establish the psychosocial profile of adolescents and adults who have admitted to committing child-to-parent violence (CPV) and were serving a judicial sanction or prison sentence, respectively. Two groups of participants took part in this study. The first group was made up of 89 male youths who were serving judicial sanctions, and the second group was made up of 70 men serving a prison sentence. A cross-sectional retrospective design with concurrent measurements was used in this study. Group differences in the exposure-to-violence variables were conducted. Automatic regression models were used to estimate a self-reported CPV. In relation to the variables of indirect exposure to violence, statistically significant differences between those who admitted having committed CPV and those who did not, irrespective of being adults or adolescents, were found for seeing violence in class and at home but not for seeing violence on the street or on television. Regarding the variables related to experiencing violence, the results showed statistically significant differences in experiencing violence at home but not in class or on the street. The best predictive model of CPV includes some of the dimensions of self-concept, specifically academic and family self-concept, as well as the avoidant and rational problem-solving styles and the negative orientation toward problems. The results have shown the existence of a CPV offender profile that is common to minors and adults

    Estilo de vida y su asociación con variables sociodemográficas en universitarios

    Introduction: Lifestyles are defined as the set of personal characteristics and daily behavior at home, work, daily activities, diet, physical activity, among others. According to the WHO, healthy lifestyles prevent the onset of NCDs. Objective: To identify the lifestyle and its association with the sociodemographic variables of the nursing students of the FET-UAT. Methodology: Cross-sectional, correlational descriptive design, with a random sample of 134 students. Lifestyle was measured with the FANTASTIC questionnaire. Results: The reliability of the instrument was .77. Students mostly have a "good job, you're on the right track" lifestyle (46.3). In the association between the lifestyle with the age, sex, shift and academic period of the students, greater affectation was found in the dimensions of family, alcohol and tobacco, with men and those over 21 years of age consuming more alcohol and being the evening shift who smokes the most. Conclusions: It was determined that the lifestyle is mostly "good", but the students showed greater consumption of alcohol and tobacco and have a poor relationship with the family.Introducción: Los estilos de vida se definen cómo el conjunto de características personales y comportamiento diario en el hogar, trabajo, actividades cotidianas, dieta, actividad física, entre otros. Según la OMS, los estilos de vida saludable previenen la aparición de ECNT. Objetivo: Identificar el estilo de vida y su asociación con las variables sociodemográficas de los estudiantes de enfermería de la FET-UAT. Metodología: El diseño descriptivo transversal, correlacional, con una muestra al azar de 134 estudiantes. El estilo de vida se midió con el cuestionario FANTÁSTICO. Resultados: La confiabilidad del instrumento fue de .77. Los estudiantes en su mayoría tienen un estilo de vida de “buen trabajo, estás en buen camino” (46.3%). En la asociación entre el estilo de vida con la edad, sexo, turno y periodo académico de los estudiantes se encontró mayor afectación a las dimensiones de familia, alcohol y tabaco, siendo los hombres y los mayores de 21 años quien consumen más alcohol y es el turno vespertino quien más fuma. Conclusiones: Se determinó que el estilo de vida en su mayoría es “bueno”, pero los estudiantes mostraron mayor consumo de alcohol y tabaco y tener mala relación con la familia

    Obesity attenuates the effect of sleep apnea on active TGF-ss 1 levels and tumor aggressiveness in patients with melanoma

    Active transforming growth factor-β1 (TGF-β1), a cytokine partially regulated by hypoxia and obesity, has been related with poor prognosis in several tumors. We determine whether obstructive sleep apnea (OSA) increases serum levels of active TGF-β1 in patients with cutaneous melanoma (CM), assess their relationship with melanoma aggressiveness and analyze the factors related to TGF-β1 levels in obese and non-obese OSA patients. In a multicenter observational study, 290 patients with CM were underwent sleep studies. TGF-β1 was increased in moderate-severe OSA patients vs. non-OSA or mild OSA patients with CM. In OSA patients, TGF-β1 levels correlated with mitotic index, Breslow index and melanoma growth rate, and were increased in presence of ulceration or higher Clark levels. In CM patients, OSA was associated with higher TGF-β1 levels and greater melanoma aggressiveness only in non-obese subjects. An in vitro model showed that IH-induced increases of TGF-β1 expression in melanoma cells is attenuated in the presence of high leptin levels. In conclusion, TGF-β1 levels are associated with melanoma aggressiveness in CM patients and increased in moderate-severe OSA. Moreover, in non-obese patients with OSA, TGF-β1 levels correlate with OSA severity and leptin levels, whereas only associate with leptin levels in obese OSA patients

    Impacto de la COVID-19 en los servicios de cirugía cardiovascular en España: Análisis de los grupos relacionados con el diagnóstico (Estudio SECCE-COVID-19 fase 2)

    Introducción y objetivos La pandemia por COVID-19 causada por infección del virus SARS-CoV-2 ha saturado al sistema sanitario español, afectándose la atención de las enfermedades cardiovasculares. Queremos cuantificar el impacto de la pandemia en el número de las intervenciones quirúrgicas cardíacas analizando los grupos relacionados con el diagnóstico (GRD) más prevalentes de nuestra especialidad. Métodos A instancias de la Sociedad Española de Cirugía Cardiovascular y Endovascular, se solicitó a todos los centros nacionales que quisieron participar, los datos de los códigos de GRD números 162 (cirugía sobre válvulas cardíacas con infarto o diagnóstico complejo), 163 (cirugía sobre válvulas cardíacas sin infarto o diagnóstico complejo), 165 (bypass coronario con infarto o diagnóstico complejo), 166 (bypass coronario sin infarto o diagnóstico complejo) y 167 (otros procedimientos cardiotorácicos o vasculares torácicos) entre el 1 de marzo de 2020 y el 30 de septiembre de 2020 (siete meses), y como período control las mismas fechas de 2019. Resultados Se recibieron los datos de 24 hospitales, 22 públicos y dos privados. Existió un descenso global en el número de intervenciones del 30% (rango -19 a -42%, p < 0,001) de 4.648 en 2019 a 3.262 en 2020 (-1.386 de diferencia), siendo +7% para el GRD 162 (p = 0,500), -37% para el 163 (p = 0,001), -9% para el 165 (p = 0,304), -32% para el 166 (p = 0,001), y -16% para el 167 (p = 0,062). Conclusiones Existió un descenso global de cirugías estadísticamente significativo en 2020 del 30% respecto del 2019 entre el 1 de marzo y el 30 de septiembre

    One-year breakthrough SARS-CoV-2 infection and correlates of protection in fully vaccinated hematological patients

    The long-term clinical efficacy of SARS-CoV-2 vaccines according to antibody response in immunosuppressed patients such as hematological patients has been little explored. A prospective multicenter registry-based cohort study conducted from December 2020 to July 2022 by the Spanish Transplant and Cell Therapy group, was used to analyze the relationship of antibody response over time after full vaccination (at 3-6 weeks, 3, 6 and 12 months) (2 doses) and of booster doses with breakthrough SARS-CoV-2 infection in 1551 patients with hematological disorders. At a median follow-up of 388 days after complete immunization, 266 out of 1551 (17%) developed breakthrough SARS-CoV-2 infection at median of 86 days (range 7-391) after full vaccination. The cumulative incidence was 18% [95% confidence interval (C.I.), 16-20%]. Multivariate analysis identified higher incidence in chronic lymphocytic leukemia patients (29%) and with the use of corticosteroids (24.5%), whereas female sex (15.5%) and more than 1 year after last therapy (14%) were associated with a lower incidence (p < 0.05 for all comparisons). Median antibody titers at different time points were significantly lower in breakthrough cases than in non-cases. A serological titer cut-off of 250 BAU/mL was predictive of breakthrough infection and its severity. SARS-CoV-2 infection-related mortality was encouragingly low (1.9%) in our series. Our study describes the incidence of and risk factors for COVID-19 breakthrough infections during the initial vaccination and booster doses in the 2021 to mid-2022 period. The level of antibody titers at any time after 2-dose vaccination is strongly linked with protection against both breakthrough infection and severe disease, even with the Omicron SARS-CoV-2 variant

    Expression of HMGCS2 in intestinal epithelial cells is downregulated in inflammatory bowel disease associated with endoplasmic reticulum stress.

    INTRODUCTION The Unfolded Protein Response, a mechanism triggered by the cell in response to Endoplasmic reticulum stress, is linked to inflammatory responses. Our aim was to identify novel Unfolded Protein Response-mechanisms that might be involved in triggering or perpetuating the inflammatory response carried out by the Intestinal Epithelial Cells in the context of Inflammatory Bowel Disease. METHODS We analyzed the transcriptional profile of human Intestinal Epithelial Cell lines treated with an Endoplasmic Reticulum stress inducer (thapsigargin) and/or proinflammatory stimuli. Several genes were further analyzed in colonic biopsies from Ulcerative Colitis patients and healthy controls. Lastly, we generated Caco-2 cells lacking HMGCS2 by CRISPR Cas-9 and analyzed the functional implications of its absence in Intestinal Epithelial Cells. RESULTS Exposure to a TLR ligand after thapsigargin treatment resulted in a powerful synergistic modulation of gene expression, which led us to identify new genes and pathways that could be involved in inflammatory responses linked to the Unfolded Protein Response. Key differentially expressed genes in the array also exhibited transcriptional alterations in colonic biopsies from active Ulcerative Colitis patients, including NKG2D ligands and the enzyme HMGCS2. Moreover, functional studies showed altered metabolic responses and epithelial barrier integrity in HMGCS2 deficient cell lines.
