Combined aerobic and resistance training: are there additional benefits for older hypertensive adults?
OBJECTIVES: The objective of this study was to compare the effects of a combination of aerobic and resistance training to those of isolated aerobic training on blood pressure, body composition, and insulin sensitivity in hypertensive older adults. METHOD: Forty-four patients were randomly assigned to the aerobic group, resistance and aerobic group, and control group. Before and after 10 weeks, the following data were obtained: 24-hour ambulatory blood pressure data, abdominal circumference, waist circumference, body mass index, lean mass, fat mass, and insulin sensitivity. The study was conducted with 3 training sessions per week. RESULTS: Comparison revealed significant reductions in the body mass index, abdominal and waist circumferences, and ambulatory blood pressure (24-hour, wakefulness and sleep systolic/diastolic blood pressures) in both the aerobic group and the resistance and aerobic (combined) group. The fat mass only changed in the combined group. There was no difference in the insulin sensitivity in any group. CONCLUSIONS: The combined treatment and aerobic treatment alone were equally effective in reducing the blood pressure, body mass index, and abdominal and waist circumferences, although the addition of the resistance component also helped reduce the fat mass
Design and validation of the INCUE questionnaire: assessment of primary healthcare nurses’ basic training needs in palliative care
Many instruments have been created to measure knowledge and attitudes in palliative care. However, not only is it important to acquire knowledge, but also that this knowledge should reach patients and their relatives through application in clinical practice. This study aimed to develop and psychometrically test the INCUE questionnaire (Investigación Cuidados Enfermeros/Investigation into Nurses’ Care Understanding of End-of-Life) to assess the basic training needs of primary or home healthcare nurses in palliative care. A questionnaire was developed based on the classical theory of tests and factor analysis models. Initially, 18 experts developed 67 items in two blocks and determined content validity by two rounds of expert panels. Exploratory factor analysis and reliability testing were conducted with a non-probabilistic sample of 370 nurses. Some items were observed to have very low homogeneity indices or presented convergence problems and were eliminated. Questionnaire reliability was 0.700 in the theoretical block (KR20 Index) and 0.941 in the practical block (Cronbach’s alpha). The model converges and shows an adequate fit, specifically CFI = 0.977, TLI = 0.977 and RMSEA = 0.05. The correlation between the two factors in the model is ρ = 0.63. The questionnaire objectively evaluates primary or home healthcare nurses’ knowledge of palliative care and its practical application, thereby facilitating more efficient training plans
Hybrid materials based on polyethylene and MCM-41 microparticles functionalized with silanes: catalytic aspects of in situ polymerization, crystalline features and mechanical properties
New nanocomposites based on polyethylene have been prepared by in situ polymerization of ethylene in
presence of mesoporous MCM-41. The polymerization reactions were performed using a zirconocene
catalyst either under homogenous conditions or supported onto mesoporous MCM-41 particles, which
are synthesized and decorated post-synthesis with two silanes before polymerization in order to promote
an enhanced interfacial adhesion. The existence of polyethylene chains able to crystallize within
the mesoporous channels in the resulting nanocomposites is figured out from the small endothermic
process, located at around 80 C, on heating calorimetric experiments, in addition to the main melting
endotherm. These results indicate that polyethylene macrochains can grow up during polymerization
either outside or inside the MCM-41 channels, these keeping their regular hexagonal arrangements.
Mechanical response is observed to be dependent on the content in mesoporous MCM-41 and on the
crystalline features of polyethylene. Accordingly, stiffness increases and deformability decreases in the
nanocomposites as much as MCM-41 content is enlarged and polyethylene amount within channels is
raised. Ultimate mechanical performance improves with MCM-41 incorporation without varying the
final processing temperature
Repurposing Disulfiram as an Antimicrobial Agent in Topical Infections
Antimicrobial drugs applied topically offer several advantages. However, the widespread use of antibiotics has led to increasing antimicrobial resistance. One interesting approach in the drug discovery process is drug repurposing. Disulfiram, which was originally approved as an anti-alcoholism drug, offers an attractive alternative to treat topical multidrug resistance bacteria in skin human infections. This study aimed to evaluate the biopharmaceutical characteristics of the drug and the effects arising from its topical application in detail. Microdilution susceptibility testing showed antibacterial activity against Gram-positive bacteria Staphylococcus aureus and Streptococcus pyogenes. Dermal absorption revealed no permeation in pig skin. The quantification of the drug retained in pig skin demonstrated concentrations in the stratum corneum and epidermis, enough to treat skin infections. Moreover, in vitro cytotoxicity and micro-array analyses were performed to better understand the mechanism of action and revealed the importance of the drug as a metal ion chelator. Together, our findings suggest that disulfiram has the potential to be repurposed as an effective antibiotic to treat superficial human skin infections
Biopharmacy practices virtualization: dissolution test of several dosage forms
En años anteriores, la Unidad de Biofarmacia y Farmacocinética de la Universidad de Barcelona, ha
estado inmersa en la virtualización de las prácticas de la asignatura de cara a que en un futuro próximo
puedan realizarse con mayor autonomía. En un trabajo previo se llevó a cabo la virtualización de la
parte referente a las prácticas de farmacocinética. Prosiguiendo con esta labor, ahora se presenta la
virtualización de las prácticas de biofarmacia. Debido a limitaciones temporales y económicas, el
estudiante de grado, realiza la práctica del ensayo de disolución de comprimidos de forma
simplificada. Para que el alumno pueda ver como se realizan los ensayos de disolución de forma
farmacéuticas sólidas según las directrices internacionales que realizará en su vida laboral, hemos
confeccionado una serie de videos explicativos de dicha tarea, y se ha ampliado con material
audiovisual sobre ensayos de disolución de formas sólidas y de parches transdérmicos, los cuales no se
realizan de forma rutinaria durante las prácticas de la licenciatura. Dicho material audiovisual estará a
disposición de los alumnos en la plataforma virtual moodle para que pueda ser consultado cuando sea
necesario.Previously, the Biopharmaceutical and Pharmacokinetic Unit of the University of Barcelona had been
immersed in the practices’ virtualization of the grade subject, looking for an improvement of student’s
autonomy in a foreseeable future. The virtualization of the pharmacokinetic practices was made
previously, and now we are presenting the virtualization of the biopharmaceutical section. Due to the
economic and time limitations, students only carried out the tablet dissolution assay in a simplified
way. The aim of this guide is to show how to perform a different dissolution test by visualizing a video, which teaches the students, how to proceed according to the international guidelines. Students will be able to study it in the virtual platform moodle, whenever it may be necessary
The impact of replacing sugar- by artificially-sweetened beverages on brain and behavioral responses to food viewing - An exploratory study.
Several studies indicate that the outcome of nutritional and lifestyle interventions can be linked to brain 'signatures' in terms of neural reactivity to food cues. However, 'dieting' is often considered in a rather broad sense, and no study so far investigated modulations in brain responses to food cues occurring over an intervention specifically aiming to reduce sugar intake. We studied neural activity and liking in response to visual food cues in 14 intensive consumers of sugar-sweetened beverages before and after a 3-month replacement period by artificially-sweetened equivalents. Each time, participants were presented with images of solid foods differing in fat content and taste quality while high-density electroencephalography was recorded. Contrary to our hypotheses, there was no significant weight loss over the intervention period and no changes were observed in food liking or in neural activity in regions subserving salience and reward attribution. However, neural activity in response to high-fat, sweet foods was significantly reduced from pre-to post-intervention in prefrontal regions often linked to impulse control. This decrease in activity was associated with weight loss failure, suggesting an impairment in individuals' ability to exert control and adjust their solid food intake over the intervention period. Our findings highlight the need to implement multidisciplinary approaches when aiming to help individuals lose body weight
Structure and Mechanism of Dimer-Monomer Transition of a Plant Poly(A)-Binding Protein upon RNA Interaction: Insights into Its Poly(A) Tail Assembly
Poly(A)-binding proteins (PABPs) play crucial roles in mRNA biogenesis, stability, transport and translational control in most eukaryotic cells. Although animal PABPs are well-studied proteins, the biological role, three-dimensional structure and RNA-binding mode of plant PABPs remain largely uncharacterized. Here, we report the structural features and RNA-binding mode of a Citrus sinensis PABP (CsPABPN1). CsPABPN1 has a domain architecture of nuclear PABPs (PABPNs) with a single RNA recognition motif (RRM) flanked by an acidic N-terminus and a GRPF-rich C-terminus. The RRM domain of CsPABPN1 displays virtually the same three-dimensional structure and poly(A)-binding mode of animal PABPNs. However, while the CsPABPN1 RRM domain specifically binds poly(A), the full-length protein also binds poly(U). CsPABPN1 localizes to the nucleus of plant cells and undergoes a dimer–monomer transition upon poly(A) interaction. We show that poly(A) binding by CsPABPN1 begins with the recognition of the RNA-binding sites RNP1 and RNP2, followed by interactions with residues of the β2 strands, which stabilize the dimer, thus leading to dimer dissociation. Like human PABPN1, CsPABPN1 also seems to form filaments in the presence of poly(A). Based on these data, we propose a structural model in which contiguous CsPABPN1 RRM monomers wrap around the RNA molecule creating a superhelical structure that could not only shield the poly(A) tail but also serve as a scaffold for the assembly of additional mRNA processing factors
Development of a gas chromatography - mass spectrometry method for the determination of carbon disulfide in the atmosphere
[EN] Carbon disulfide (CS2), a relevant reduced sulfur compound in air, is well-known for its malodor and its significant effect on global atmospheric chemistry. Therefore, a reliable method for determining CS2 in atmospheric samples has been developed based on solid-phase sampling and gas chromatography-mass spectrometry (GC-MS). Two types of solid-phase sampling supports (Orbo-32 and SKC) and the elution with organic solvents - hexane and toluene - were evaluated for low-volume outdoor sampling. Recovery studies and the standard addition method were carried out to demonstrate the proper determination of CS2 in the absence of the influence of interferences such as ozone, hydrogen sulfide or water - important atmospheric pollutants. The proposed methodology was validated by performing experiments in a high-volume smog chamber and by comparison with two reference optical methods, Fourier Transform Infrared (FTIR) and Differential Optical Absorption Spectroscopy (DOAS) installed in these facilities. Satisfactory analytical parameters were reported: fast analysis, a correct repeatability of 6±1% and reproducibility of 14±3%, and low detection limits of 0.3-0.9pgm-3. Finally, the method was successfully applied to industrial samples near a pulp factory area, where a high correlation between industrial emissions and reported carbon disulfide concentrations were observed. © 2011 Elsevier B.V.The research leading to these results received funding from the Centro de Investigacion del Medio Ambiente (CIMA), Consejeria de Medio Ambiente, Gobierno de Cantabria.
