    Heterologous expression of AtNPR1 gene in olive for increasing fungal tolerance

    The NPR1 gene encodes a key component of SAR signaling mediated by salicylic acid (SA). After a pathogen infection, the accumulation of SA releases NPR1 monomers in the cytosol that are translocated to the nucleus, activating the expression of pathogenesis-related (PR) genes. Overexpression of NPR1 has conferred resistance to fungal, viral and bacterial pathogens in several plant species. The aim of this research was to generate transgenic olive plants expressing the gene AtNPR1 from Arabidopsis thaliana to obtain material resistant to fungal pathogens. Three transgenic lines expressing AtNPR1 gene under the control of the constitutive promoter CaMV35S were obtained following the protocol of Torreblanca et al. (2010), using an embryogenic line derived from a seed of cv. Picual. Level of AtNPR1 expression in transgenic calli varied greatly among the different lines, being higher in the line NPR1-780. The elicitation of embryogenic calli in liquid medium with AS did not increase endochitinase activity, a PR protein. However, jasmonic acid induced a transient increase in chitinase activity after 24 h of treatment in all the lines, being the increment higher in transgenic NPR1 than in control. After maturation and germination of transgenic somatic embryos, plants were micropropagated and acclimated to ex vitro conditions. The expression of AtNPR1 did not alter the growth of transgenic plants neither in vitro nor in the greenhouse.     Using Flipped classroom at University to improve the chemistry learning.

    The main objective of this paper was to improve the learning of Chemistry subject using Flipped classroom, because it makes class time more engaging. The lecture portion of General Chemistry courses in engineers degrees have been pushed outside the classroom using pre recording technology and steaming delivery of content, to make classes more interactive and participative. The Flipped classroom model has become one of the main topics in the higher education space in recent years thanks to improvements in technology. This year, the Flipped classroom model with the chemistry students at Malaga State University has been begun experimenting with. Statistical significance of the data has shown, and proved with, that implementing the Flipped classroom model could not only benefit professors, but it could also help us adapt the classes to the various learning styles that exist among the students.     Short-term forecasting of intermodal freight using ANNs and SVR: Case of the Port of Algeciras Bay

    [EN] Forecasting of future intermodal traffic demand is very important for decision making in ports operations management. The use of accurate prediction tools is an issue that awakens a lot of interest among transport researchers. Intermodal freight forecasting plays an important role in ports management and in the planning of the principal port activities. Hence, the study is carried out under the motivation of knowing that modeling the freight transport flows could facilitate the management of the infrastructure and optimize the resources of the ports facilities. The use of advanced models for freight forecasting is essential to improve the port level-service and competitiveness. In this paper, two forecasting-models are presented and compared to predict the freight volume. The models developed and tested are based on Artificial Neural Networks and Support Vector Machines. Both techniques are based in a historical data and these methods forecast the daily weight of the freight with one week in advance. The performance of the models is evaluated on real data from Ro-Ro freight transport in the Port of Algeciras Bay. This work proposes and compares different approaches to determine the best prediction. In order to select the best model a multicomparison procedure is developed using several statistical test.     Flow Cytometry Data Preparation Guidelines for Improved Automated Phenotypic Analysis.

    Advances in flow cytometry (FCM) increasingly demand adoption of computational analysis tools to tackle the ever-growing data dimensionality. In this study, we tested different data input modes to evaluate how cytometry acquisition configuration and data compensation procedures affect the performance of unsupervised phenotyping tools. An analysis workflow was set up and tested for the detection of changes in reference bead subsets and in a rare subpopulation of murine lymph node CD103+ dendritic cells acquired by conventional or spectral cytometry. Raw spectral data or pseudospectral data acquired with the full set of available detectors by conventional cytometry consistently outperformed datasets acquired and compensated according to FCM standards. Our results thus challenge the paradigm of one-fluorochrome/one-parameter acquisition in FCM for unsupervised cluster-based analysis. Instead, we propose to configure instrument acquisition to use all available fluorescence detectors and to avoid integration and compensation procedures, thereby using raw spectral or pseudospectral data for improved automated phenotypic analysis.We thank Irene Palacios, Elena Prieto, Mariano Vito´n, and Raquel Nieto for excellent technical assistance and Dr. Salvador Iborra for helpful discussion of dendritic cell studies.     Patient motivation as a predictor of digital health intervention effects: A meta-epidemiological study of cancer trials

    The objective of this meta-epidemiological study was to develop a rating that captures participants’ motivation at the study level in digital health intervention (DHI) randomised controlled trials (RCTs). The rating was used to investigate whether participants’ motivation is associated with the effect estimates in DHI RCTs for cancer patients. The development of the rating was based on a bottom-up approach involving the collection of information that captures participants’ baseline motivation in empirical studies from the Smartphone-RCCT Database. We specified three indicators for rating: indicator 1 captures whether the study team actively selects or enhances the motivation of the potential study participants; indicator 2 captures the study participants’ active engagement before the treatment allocation; and indicator 3 captures the potential bond and trust between the study participants and the person/institution referring to the study. The rating of each indicator and the overall rating varies between high motivation, moderate motivation, and low motivation. We applied the rating across 27 DHI RCTs with cancer patients. We performed meta-regression analysis to examine the effect of patient motivation on quality of life (QoL), psychological outcomes, and attrition. The intraclass correlation coefficient (ICC) indicated moderate to poor inter-rater reliability. The meta-regression showed that cancer patients’ overall motivation before engaging in the intervention was associated with the treatment effect of QoL. Patient motivation was not found to be associated with psychological outcomes or attrition. Subgroup analyses revealed that the clinical effects of DHIs were more prevalent in the high-motivation subgroups, whereas the low-motivation subgroups were unlikely to show intervention benefits. The likelihood of dropouts from DHIs seems to be especially high among the low-bond (indicator 3) subgroup. We suggest using single indicators since they reflect specific content. Better reporting about baseline motivation is required to enable meaningful interpretations in not only primary studies but also in evidence syntheses

    Decomposition and nitrogen mineralisation of two wild lupins (Leguminosae) species with potential as green manures

    The amount of biomass produced by various native species of genus Lupinus (L.) growing in Mexico ranges from 2.9- 8.2 Mg/ha of dry matter, which can add up to 200 kg/ha of N to soil as green manure. However, information is scarce on the decomposition and mineralisation this biomass in the soil. The above-ground decomposition and N mineralisation of Lupinus mexicanus Cerv. ex Lag. and Lupinus rotundiflorus M.E. Jones species from Mexico using fine-mesh litter bags was evaluated. Litter bags containing 5 g of above-ground air-dried biomass at the vegetative and flowering stages were buried at a depth of 20 cm. Were dug up every 3 weeks over the course of 4 months, dried and re-weighed to determine the lost mass and total N by the Kjeldahl method. The largest decrease in residue mass occurred during the first 3 weeks of incubation. However, the lost mass was higher in younger green manure (75 days old) than in older plants (85 days old) after 18 weeks of exposure in the field. It was found that 60-75% of the total N in the plant material was released in the first 6 weeks. In L. rotundiflorus green manure, it was found that 79.14% of the initial N in the vegetative stage and 77.6% of N in the flowering stage was released 18 weeks after litter bag installation, whereas L. mexicanus were 74.6% and 74.7%, respectively. It was found that both decomposition and N mineralisation occurred quickly in the green manures evaluated

    Neurodegeneration trajectory in pediatric and adult/late DM1: A follow‐up MRI study across a decade

    Objective: To characterize the progression of brain structural abnormalities in adults with pediatric and adult/late onset DM1, as well as to examine the potential predictive markers of such progression. Methods: 21 DM1 patients (pediatric onset: N = 9; adult/late onset: N = 12) and 18 healthy controls (HC) were assessed longitudinally over 9.17 years through brain MRI. Additionally, patients underwent neuropsychological, genetic, and muscular impairment assessment. Inter-group comparisons of total and voxel-level regional brain volume were conducted through Voxel Based Morphometry (VBM); cross-sectionally and longitudinally, analyzing the associations between brain changes and demographic, clinical, and cognitive outcomes. Results: The percentage of GM loss did not significantly differ in any of the groups compared with HC and when assessed independently, adult/late DM1 patients and their HC group suffered a significant loss in WM volume. Regional VBM analyses revealed subcortical GM damage in both DM1 groups, evolving to frontal regions in the pediatric onset patients. Muscular impairment and the outcomes of certain neuropsychological tests were significantly associated with follow-up GM damage, while visuoconstruction, attention, and executive function tests showed sensitivity to WM degeneration over time. Interpretation: Distinct patterns of brain atrophy and its progression over time in pediatric and adult/late onset DM1 patients are suggested. Results indicate a possible neurodevelopmental origin of the brain abnormalities in DM1, along with the possible existence of an additional neurodegenerative process. Fronto-subcortical networks appear to be involved in the disease progression at young adulthood in pediatric onset DM1 patients.
