    Signatures of anti-Thomsen — friedenreich antigen antibody diversity in colon cancer patients

    Aim: To determine whether the structural and functional diversities of naturally occurring antibodies to the Thomsen — Friedenreich (TF) antigen may be of diagnostic and prognostic value in colon cancer. Materials and Methods: Serum samples were taken from patients with colon cancer (n = 94) and healthy controls (n = 64). The level of TF-specific antibody isotypes and their sialylation were determined using ELISA and lectin-ELISA with synthetic TF-polyacrylamide conjugate as an antigen and a sialic acid-specific Sambucus nigra agglutinin (SNA). The avidity was determined using ammonium thiocyanate as a chaotrope. The accuracy of diagnostics was evaluated using the receiver operator characteristic curve analysis and the survival analysis employing the Kaplan — Meier method. Results: Compared to healthy controls, patients with colon cancer exhibited a lower level of anti-TF IgG antibodies, significantly lower ratios of TF-specific IgG/IgM and IgG/IgA, an increased SNA reactivity of anti-TF antibodies, mostly on account of IgG, and a lower avidity of TF-specific antibodies, especially their SNA-reactive subset. An increased SNA reactivity of anti-TF IgG was observed already at the early stages of cancer (p = 0.0004). The decrease of the ratio of IgG/IgM and IgG/IgA showed a good accuracy of diagnostics with about 60% sensitivity at 90% specificity. A similar potential was found for the SNA binding/IgG level index. The high level of TF-specific IgA antibodies was associated with a lower survival rate (hazard ratio = 0.34). Conclusion: This is the first report ever on the colon cancer-related signatures of anti-TF antibody diversity which show diagnostic potential, including in early cancer, and prognostic value. The hypersialylation of TF-specific antibodies appeared to be a common phenomenon in cancer. The signatures may be used as non-invasive antibody-based markers for colon cancer

    A Luminous Red Nova in M31 and its Progenitor System

    We present observations of M31LRN 2015 (MASTER OT J004207.99+405501.1), discovered in M31 in January 2015, and identified as a rare and enigmatic luminous red nova (LRN). Spectroscopic and photometric observations obtained by the Liverpool Telescope showed the LRN becoming extremely red as it faded from its M(V) = -9.4 +/- 0.2 peak. Early spectra showed strong Halpha emission that weakened over time as a number of absorption features appeared, including Na I D and Ba II. At later times strong TiO absorption bands were also seen. A search of archival Hubble Space Telescope data revealed a luminous red source to be the likely progenitor system, with pre-outburst Halpha emission also detected in ground-based data. The outburst of M31LRN 2015 shows many similarities, both spectroscopically and photometrically, with that of V838 Mon, the best studied LRN. We finally discuss the possible progenitor scenarios

    Massive star mergers and the recent transient in NGC 4490: A more massive cousin of V838 Mon and V1309 Sco

    The Galactic transient V1309 Sco was the result of a merger in a low-mass star system, while V838 Mon was thought to be a similar merger event from a more massive B-type progenitor. In this paper, we study a recent optical and infrared (IR) transient discovered in the nearby galaxy NGC4490 named NGC4490-OT2011 (NGC 4490-OT hereafter), which appeared similar to these merger events (unobscured progenitor, irregular multi-peaked light curve, increasingly red colour, similar optical spectrum, IR excess at late times), but which had a higher peak luminosity and longer duration in outburst. NGC4490-OT has less in common with the class of SN 2008S-like transients. A progenitor detected in pre-eruption Hubble Space Telescope (HST) images, combined with upper limits in the IR, requires a luminous and blue progenitor that has faded in late-time HST images. The same source was detected by Spitzer and groundbased data as a luminous IR (2-5 μm) transient, indicating a transition to a self-obscured state qualitatively similar to the evolution seen in other stellar mergers and in luminous blue variables. The post-outburst dust-obscured source is too luminous and too warm at late times to be explained with an IR echo, suggesting that the object survived the event. The luminosity of the enshrouded IR source is similar to that of the progenitor. Compared to proposed merger events, the more massive progenitor of NGC4490-OT seems to extend a correlation between stellar mass and peak luminosity, and may suggest that both of these correlate with duration. We show that spectra of NGC4490-OT and V838 Mon also resemble light-echo spectra of η Car, prompting us to speculate that η Car may be an extreme extension of this phenomenon

    The WEBT campaign on the BL Lac object PG 1553+113 in 2013. An analysis of the enigmatic synchrotron emission

    A multifrequency campaign on the BL Lac object PG 1553+113 was organized by the Whole Earth Blazar Telescope (WEBT) in 2013 April-August, involving 19 optical, two near-IR, and three radio telescopes. The aim was to study the source behaviour at low energies during and around the high-energy observations by the Major Atmospheric Gamma-ray Imaging Cherenkov telescopes in April-July. We also analyse the UV and X-ray data acquired by the Swift and XMM-Newton satellites in the same period. The WEBT and satellite observations allow us to detail the synchrotron emission bump in the source spectral energy distribution (SED). In the optical, we found a general bluer-when-brighter trend. The X-ray spectrum remained stable during 2013, but a comparison with previous observations suggests that it becomes harder when the X-ray flux increases. The long XMM-Newton exposure reveals a curved X-ray spectrum. In the SED, the XMM-Newton data show a hard near-UV spectrum, while Swift data display a softer shape that is confirmed by previous Hubble Space Telescope/Cosmic Origins Spectrograph and International Ultraviolet Explorer observations. Polynomial fits to the optical-X-ray SED show that the synchrotron peak likely lies in the 4-30eV energy range, with a general shift towards higher frequencies for increasing X-ray brightness. However, the UV and X-ray spectra do not connect smoothly. Possible interpretations include: (i) orientation effects, (ii) additional absorption, (iii) multiple emission components, and (iv) a peculiar energy distribution of relativistic electrons.     The January 2015 outburst of a red nova in M 31

    © ESO 2015. Context. M31N 2015-01a (or M31LRN 2015) is a red nova that erupted in January 2015 - the first event of this kind observed in M 31 since 1988. Very few similar events have been confirmed as of 2015. Most of them are considered to be products of stellar mergers. Aims. Results of an extensive optical monitoring of the transient in the period January-March 2015 are presented. Methods. Eight optical telescopes were used for imaging. Spectra were obtained on the Large Altazimuth Telescope (BTA), the Gran Telecsopio Canarias (GTC) and the Rozhen 2 m telescope. Results. We present a highly accurate 70 d light curve and astrometry with a 0.05″uncertainty. The colour indices reached a minimum of 2-3 d before peak brightness and rapidly increased afterwards. The spectral type changed from F5I to F0I in 6 d before the maximum and then to K3I in the next 30 d. The luminosity of the transient was estimated to be 8.7-2.2+3.3 × 105 L⊙ during the optical maximum. Conclusions. Both the photometric and the spectroscopic results confirm that the object is a red nova, similar to V838 Monocerotis

    Dynamics and nucleation of dislocations in crystals

    Hydrogen-poor superluminous supernovae (SLSNe-I) have been predominantly found in low-metallicity, star-forming dwarf galaxies. Here we identify Gaia17biu/SN 2017egm as an SLSN-I occurring in a "normal" spiral galaxy (NGC 3191) in terms of stellar mass (several times 10^10 M_sun) and metallicity (roughly Solar). At redshift z=0.031, Gaia17biu is also the lowest redshift SLSN-I to date, and the absence of a larger population of SLSNe-I in dwarf galaxies of similar redshift suggests that metallicity is likely less important to the production of SLSNe-I than previously believed. With the smallest distance and highest apparent brightness for an SLSN-I, we are able to study Gaia17biu in unprecedented detail. Its pre-peak near-ultraviolet to optical color is similar to that of Gaia16apd and among the bluest observed for an SLSN-I while its peak luminosity (M_g = -21 mag) is substantially lower than Gaia16apd. Thanks to the high signal-to-noise ratios of our spectra, we identify several new spectroscopic features that may help to probe the properties of these enigmatic explosions. We detect polarization at the ~0.5% level that is not strongly dependent on wavelength, suggesting a modest, global departure from spherical symmetry. In addition, we put the tightest upper limit yet on the radio luminosity of an SLSN-I with <5.4x10^26 erg/s/Hz (at 10 GHz), which is almost a factor of 40 better than previous upper limits and one of the few measured at an early stage in the evolution of an SLSN-I. This limit largely rules out an association of this SLSNe-I with known populations of gamma-ray burst (GRB) like central engines.Comment: Accepted for publication in ApJ.     Rise and Fall of an Anti-MUC1 Specific Antibody

    So far, human antibodies with good affinity and specificity for MUC1, a transmembrane protein overexpressed on breast cancers and ovarian carcinomas, and thus a promising target for therapy, were very difficult to generate.A human scFv antibody was isolated from an immune library derived from breast cancer patients immunised with MUC1. The anti-MUC1 scFv reacted with tumour cells in more than 80% of 228 tissue sections of mamma carcinoma samples, while showing very low reactivity with a large panel of non-tumour tissues. By mutagenesis and phage display, affinity of scFvs was increased up to 500fold to 5,7×10(-10) M. Half-life in serum was improved from below 1 day to more than 4 weeks and was correlated with the dimerisation tendency of the individual scFvs. The scFv bound to T47D and MCF-7 mammalian cancer cell lines were recloned into the scFv-Fc and IgG format resulting in decrease of affinity of one binder. The IgG variants with the highest affinity were tested in mouse xenograft models using MCF-7 and OVCAR tumour cells. However, the experiments showed no significant decrease in tumour growth or increase in the survival rates. To study the reasons for the failure of the xenograft experiments, ADCC was analysed in vitro using MCF-7 and OVCAR3 target cells, revealing a low ADCC, possibly due to internalisation, as detected for MCF-7 cells.Antibody phage display starting with immune libraries and followed by affinity maturation is a powerful strategy to generate high affinity human antibodies to difficult targets, in this case shown by the creation of a highly specific antibody with subnanomolar affinity to a very small epitope consisting of four amino acids. Despite these "best in class" binding parameters, the therapeutic success of this antibody was prevented by the target biology

    The hidden symbiotic star SU Lyn-detection of flickering in U band

    We report photometric observations of the hidden symbiotic star SU Lyn in the optical bands. In four nights we detect a weak flickering in U band with amplitude of about 0.05 magnitudes. No intranight variations are found in B, V, g’ and r’ bands. This is one more indication that the secondary component is a white dwarf accreting at a low accretion rate Ẇe also searched for intranight variability of a dozen related objects-RR Boo, RT Boo, AM Cyg, AG Peg, BF Cyg, NQ Gem, StHa190, V627 Cas, XX Oph, FS Cet and Y Gem-however no variability above the observational errors is detected

    Intraperitoneal drain placement and outcomes after elective colorectal surgery: international matched, prospective, cohort study

    Despite current guidelines, intraperitoneal drain placement after elective colorectal surgery remains widespread. Drains were not associated with earlier detection of intraperitoneal collections, but were associated with prolonged hospital stay and increased risk of surgical-site infections.Background Many surgeons routinely place intraperitoneal drains after elective colorectal surgery. However, enhanced recovery after surgery guidelines recommend against their routine use owing to a lack of clear clinical benefit. This study aimed to describe international variation in intraperitoneal drain placement and the safety of this practice. Methods COMPASS (COMPlicAted intra-abdominal collectionS after colorectal Surgery) was a prospective, international, cohort study which enrolled consecutive adults undergoing elective colorectal surgery (February to March 2020). The primary outcome was the rate of intraperitoneal drain placement. Secondary outcomes included: rate and time to diagnosis of postoperative intraperitoneal collections; rate of surgical site infections (SSIs); time to discharge; and 30-day major postoperative complications (Clavien-Dindo grade at least III). After propensity score matching, multivariable logistic regression and Cox proportional hazards regression were used to estimate the independent association of the secondary outcomes with drain placement. Results Overall, 1805 patients from 22 countries were included (798 women, 44.2 per cent; median age 67.0 years). The drain insertion rate was 51.9 per cent (937 patients). After matching, drains were not associated with reduced rates (odds ratio (OR) 1.33, 95 per cent c.i. 0.79 to 2.23; P = 0.287) or earlier detection (hazard ratio (HR) 0.87, 0.33 to 2.31; P = 0.780) of collections. Although not associated with worse major postoperative complications (OR 1.09, 0.68 to 1.75; P = 0.709), drains were associated with delayed hospital discharge (HR 0.58, 0.52 to 0.66; P &lt; 0.001) and an increased risk of SSIs (OR 2.47, 1.50 to 4.05; P &lt; 0.001). Conclusion Intraperitoneal drain placement after elective colorectal surgery is not associated with earlier detection of postoperative collections, but prolongs hospital stay and increases SSI risk
