    CARMENES input catalogue of M dwarfs IV. New rotation periods from photometric time series

    Aims. The main goal of this work is to measure rotation periods of the M-type dwarf stars being observed by the CARMENES exoplanet survey to help distinguish radial-velocity signals produced by magnetic activity from those produced by exoplanets. Rotation periods are also fundamental for a detailed study of the relation between activity and rotation in late-type stars. Methods. We look for significant periodic signals in 622 photometric time series of 337 bright, nearby M dwarfs obtained by long-time baseline, automated surveys (MEarth, ASAS, SuperWASP, NSVS, Catalina, ASAS-SN, K2, and HATNet) and for 20 stars which we obtained with four 0.2-0.8 m telescopes at high geographical latitudes. Results. We present 142 rotation periods (73 new) from 0.12 d to 133 d and ten long-term activity cycles (six new) from 3.0 a to 11.5 a. We compare our determinations with those in the existing literature; we investigate the distribution of P rot in the CARMENES input catalogue,the amplitude of photometric variability, and their relation to vsin i and pEW(Halfa); and we identify three very active stars with new rotation periods between 0.34 d and 23.6 d.Comment: 34 pages, 43 figures, 2 appendix table

    Metal Abundances of KISS Galaxies. IV. Galaxian Luminosity-Metallicity Relations in the Optical and Near-IR

    We explore the galaxian luminosity-metallicity (L-Z) relationship in both the optical and the near-IR using a combination of optical photometric and spectroscopic observations from the KPNO International Spectroscopic Survey (KISS) and near-infrared photometry from the Two-micron All Sky Survey (2MASS). We supplement the 2MASS data with our own NIR photometry for a small number of lower-luminosity ELGs that are under-represented in the 2MASS database. Our B-band L-Z relationship includes 765 star-forming KISS galaxies with coarse abundance estimates from our follow-up spectra, while the correlation with KISS and 2MASS yields a total of 420 galaxies in our J-band L-Z relationship. We explore the effect that changing the correlation between the strong-line abundance diagnostic R_23 and metallicity has on the derived L-Z relation. We find that the slope of the L-Z relationship decreases as the wavelength of the luminosity bandpass increases. We interpret this as being, at least in part, an effect of internal absorption in the host galaxy. Furthermore, the dispersion in the L-Z relation decreases for the NIR bands, suggesting that variations in internal absorption contribute significantly to the observed scatter. We propose that our NIR L-Z relations are more fundamental than the B-band relation, since they are largely free of absorption effects and the NIR luminosities are more directly related to the stellar mass of the galaxy than are the optical luminosities.Comment: 22 pages, including 6 figures and 6 tables.     Comparative Cytotoxic Activity of Hydroxytyrosol and Its Semisynthetic Lipophilic Derivatives in Prostate Cancer Cells

    A high adherence to a Mediterranean diet has been related to numerous beneficial effects in human health, including a lower incidence and mortality of prostate cancer (PCa). Olive oil is an important source of phenolic bioactive compounds, mainly hydroxytyrosol (HT), of this diet. Because of the growing interest of this compound and its derivatives as a cancer chemopreventive agent, we aimed to compare the in vitro effect of HT isolated from olive mill wastewaters and five semisynthetic alkyl ether, ester, and nitro-derivatives against prostate cancer (PCa) cell lines. The effect in cell proliferation was determined in RWPE-1, LNCaP, 22Rv1, and PC-3 cells by resazurin assay, the effect in cell migration by wound healing assay, and tumorsphere and colony formation were evaluated. The changes in key signaling pathways involved in carcinogenesis were assessed by using a phosphorylation pathway profiling array and by Western blotting. Antiproliferative effects of HT and two lipophilic derivatives [hydroxytyrosyl acetate (HT-Ac)/ethyl hydroxytyrosyl ether (HT-Et)] were significantly higher in cancerous PC-3 and 22Rv1 cells than in non-malignant RWPE-1 cells. HT/HT-Ac/HT-Et significantly reduced migration capacity in RWPE-1 and PC-3 and prostatosphere size and colony formation in 22Rv1, whereas only HT-Ac and HT-Et reduced these functional parameters in PC-3. The cytotoxic effect in 22Rv1 cells was correlated with modifications in the phosphorylation pattern of key proteins, including ERK1/2 and AKT. Consistently, HT-Ac and HT-Et decreased p-AKT levels in PC-3. In sum, our results suggest that HT and its lipophilic derivatives could be considered as potential therapeutic tools in PCa

    Unleashing the Diagnostic, Prognostic and Therapeutic Potential of the Neuronostatin/GPR107 System in Prostate Cancer

    Certain components of the somatostatin-system play relevant roles in Prostate Cancer (PCa), whose most aggressive phenotype (Castration-Resistant-PCa (CRPC)) remains lethal nowadays. However, neuronostatin and the G protein-coupled receptor 107 (GPR107), two novel members of the somatostatin-system, have not been explored yet in PCa. Consequently, we investigated the pathophysiological role of NST/GPR107-system in PCa. GPR107 expression was analyzed in well-characterized PCa patient′s cohorts, and functional/mechanistic assays were performed in response to GPR107-silencing and NST-treatment in PCa cells (androgen-dependent (AD: LNCaP) and androgen-independent (AI: 22Rv1/PC-3), which are cell models of hormone-sensitive and CRPC, respectively), and normal prostate cells (RWPE-1 cell-line). GPR107 was overexpressed in PCa and associated with key clinical parameters (e.g., advance stage of PCa, presence of vascular invasion and metastasis). Furthermore, GPR107-silencing inhibited proliferation/migration rates in AI-PCa-cells and altered key genes and oncogenic signaling-pathways involved in PCa aggressiveness (i.e., KI67/CDKN2D/MMP9/PRPF40A, SST5TMD4/AR-v7/In1-ghrelin/EZH2 splicing-variants and AKT-signaling). Interestingly, NST treatment inhibited proliferation/migration only in AI-PCa cells and evoked an identical molecular response than GPR107-silencing. Finally, NST decreased GPR107 expression exclusively in AI-PCa-cells, suggesting that part of the specific antitumor effects of NST could be mediated through a GPR107-downregulation. Altogether, NST/GPR107-system could represent a valuable diagnostic and prognostic tool and a promising novel therapeutic target for PCa and CRPC

    Chemical evolution of starburst galaxies: How does star formation proceed?

    We compute chemical evolution models to constrain the mode and the history of star formation in starburst galaxies as a whole, i.e. over a large range of mass and metallicity. To this end, we investigate the origin of the dispersion observed in the evolution of both nitrogen-to-oxygen abundance ratio and galaxy luminosity as a function of metallicity for a large sample of starburst galaxies. We find that the variation of the star formation efficiency, in the framework of continuous star formation models, produce a scatter equivalent to what is observed in the N/O versus O/H diagram for low-mass HII galaxies only. However, continous star formation models are unable to reproduce i) the scatter observed for massive starburst and UV-selected galaxies in the N/O versus O/H relation, and ii) the scatter in the luminosity versus O/H scaling relation observed for the whole sample of starburst galaxies. The dispersion associated with the distribution of N/O as a function of metallicity, for both low-mass and massive galaxies, is well explained in the framework of bursting star formation models. It is interpreted as a consequence of the time-delay between the ejection of nitrogen and that of oxygen into the ISM. These models also reproduce the spread observed in the luminosity-metallicity relation. Metal-rich spiral galaxies differ from metal-poor ones by a higher star formation efficiency and starburst frequency. Low-mass galaxies experienced a few bursts of star formation whereas massive spiral galaxies experienced numerous and extended powerful starbursts (abridged version).Comment: 9 pages, 4 figures.     Clinical Utility of Ghrelin-O-Acyltransferase (GOAT) Enzyme as a Diagnostic Tool and Potential Therapeutic Target in Prostate Cancer

    Recent data suggested that plasma Ghrelin O-Acyl Transferase enzyme (GOAT) levels could represent a new diagnostic biomarker for prostate cancer (PCa). In this study, we aimed to explore the diagnostic and prognostic/aggressiveness capacity of GOAT in urine, as well as to interrogate its putative pathophysiological role in PCa. We analysed urine/plasma levels of GOAT in a cohort of 993 patients. In vitro (i.e., cell-proliferation) and in vivo (tumor-growth in a xenograft-model) approaches were performed in response to the modulation of GOAT expression/activity in PCa cells. Our results demonstrate that plasma and urine GOAT levels were significantly elevated in PCa patients compared to controls. Remarkably, GOAT significantly outperformed PSA in the diagnosis of PCa and significant PCa in patients with PSA levels ranging from 3 to 10 ng/mL (the so-called PSA grey-zone). Additionally, urine GOAT levels were associated to clinical (e.g., Gleason-score, PSA levels) and molecular (e.g., CDK2/CDK6/CDKN2A expression) aggressiveness parameters. Indeed, GOAT overexpression increased, while its silencing/blockade decreased cell-proliferation in PCa cells. Moreover, xenograft tumors derived from GOAT-overexpressing PCa (DU145) cells were significantly higher than those derived from the mock-overexpressing cells. Altogether, our results demonstrate that GOAT could be used as a diagnostic and aggressiveness marker in urine and a therapeutic target in PCa

    EN-DALBACEN 2.0 Cohort: real-life study of dalbavancin as sequential/consolidation therapy in patients with infective endocarditis due to Gram-positive cocci

    Objectives: Infective endocarditis (IE) has high mortality and morbidity and requires long hospital stays to deliver the antibiotic treatment recommended in clinical practice guidelines. We aimed to analyse the health outcomes of the use of dalbavancin (DBV) in the consolidation treatment of IEs caused by Gram-positive cocci and to perform a pharmacoeconomic study. Materials and methods: This observational, retrospective, Spanish multicentre study in patients with IE who received DBV as part of antibiotic treatment in consolidation phase were followed for at least 12 months. The study was approved by the Provincial Committee of the coordinating centre. Results: The study included 124 subjects, 70.2% male, with a mean age of 67.4 years and median Charlson index of 4 (interquartile range: 2.5-6). Criteria for definite IE were met by 91.1%. Coagulase-negative staphylococci (38.8%), Staphylococcus aureus (22.6%), Enterococcus faecalis (19.4%), and Streptococcus Spp. (9.7%) were isolated more frequently, all susceptible to vancomycin. Before DVB administration, 91.2% had undergone surgery; 60.5% had received a second regimen for 24.5 d (16.6-56); and 20.2% had received a third regimen for 14.5 d (12-19.5). DBV was administered to facilitate discharge in 95.2% of cases. At 12 months, the effectiveness was of 95.9%, and there was 0.8% loss to follow-up, 0.8% IE-related death, and 3.2% relapse. Adverse events were recorded in 3.2%. The hospital stay was reduced by 14 d, and there was a mean savings of 5548.57 €/patient vs. conventional treatments. Conclusion: DBV is highly effective, safe, and cost-effective as consolidation therapy in patients with IE by Gram-positive cocci, with few adverse events
