    New Eunicellin-Type Diterpenes from the Panamanian Octocoral Briareum Asbestinum

    Gorgonian octocorals are considered a prolific source of secondary metabolites with a wide range of biological activities, including anti-inflammatory activity. In particular, the genus Briareum is known for producing a wealth of diterpenes with complex chemical structures. The chemical study of the methanolic extract of Briareum asbestinum collected in Bocas del Toro, on the Caribbean side of Panama, led to the isolation of three new eunicellin-type diterpenes: briarellin T (1), asbestinin 27 (2), asbestinin 28 (3) and the previously described asbestinin 17 (4). The structures of the new compounds were determined by extensive NMR analyses and HRMS. Anti-inflammatory activity assays showed a significant reduction of the pro-inflammatory cytokines TNF-α, IL-6, IL-1β and IL-8 as well as a downregulation of COX-2 expression in LPS-stimulated THP-1 macrophages. These findings support the potential use of these marine compounds as therapeutic agents in the treatment of inflammatory diseases

    Uprolides N, O and P from the Panamanian Octocoral Eunicea succinea.

    Three new diterpenes, uprolide N (1), uprolide O (2), uprolide P (3) and a known one, dolabellane (4), were isolated from the CH₂Cl₂-MeOH extract of the gorgonian octocoral Eunicea succinea, collected from Bocas del Toro, on the Caribbean coast of Panama. Their structures were determined using spectroscopic analyses, including 1D and 2D NMR and high-resolution mass spectrometry (HRMS) together with molecular modeling studies. Compounds 1-3 displayed anti-inflammatory properties by inhibiting production of Tumor Necrosis Factor (TNF) and Interleukin (IL)-6 induced by lipopolysaccharide (LPS) in murine macrophages

    El presente trabajo tiene por objetivo determinar las principales fortalezas y limitaciones de la producción de café orgánico en Colombia y Brasil mediante el análisis de tres líneas de acción fundamentales en un sistema orgánico: el proceso de certificación, la producción y los canales de comercialización. Está fundamentado en el método cualitativo a partir del desarrollo de una revisión bibliográfica de los temas de interés y el análisis de datos secundarios. El estudio evidenció las fortalezas de ambos países en relación con sus características climatológicas ideales para la producción, su reconocimiento y aceptación mundial como productores y exportadores del commodity y el creciente mercado internacional de café orgánico. También se pudo establecer algunas limitaciones para ambos países como los altos costos y exigencias en el proceso de certificación, un mayor costo de producción y un bajo nivel de asociativismo. No obstante, Colombia y Brasil se constituyen como países con alto potencial para la producción de cafés orgánicos. Abstract: This study aims to determine the strengths and limitations of the production of organic coffee in Colombia and Brazil through the analysis of three fundamental lines of action of an organic system: the certification process, production, and marketing channels. It is based on the qualitative method by developing of a literature review of the topics of interest and analysis of secondary data. The study showed the strengths of both countries in relation to their ideal climatic characteristics for production, its recognition and worldwide acceptance as producers and exporters of the commodity and the growing international market for organic coffee. It was also possible to set some limitations in both countries as the high costs and demands on the certification process, a higher cost of production and low associative. However, Colombia and Brazil are countries with high potential for producing organic coffees

    Development and psychometric properties of the stressors in Breast Cancer Scale.

    Background: A diagnosis of breast cancer generates psychological stress, due not only to treatment and its side effects but also to the impact on different areas of the patient’s daily life. Although there are instruments for measuring psychological stress in the cancer context, there is currently no tool for assessing stressors specific to breast cancer. Aims: The aim of this study was to develop the Stressors in Breast Cancer Scale (SBCS). Method: A panel of experts evaluated the clarity and relevance of scale items, providing validity evidence based on test content. Psychometric properties of the scale were then analyzed. Results: Validity evidence based on the internal structure of the SBCS was obtained through exploratory factor analysis (EFA) and confirmatory factor analysis (CFA), following a cross-validation strategy. The CFA supported a second-order factor model with five dimensions: physical appearance and sex strains, health and daily difficulties, interpersonal relationship strains, healthcare strains, and worries and concerns about the future. This structure was invariant across two groups distinguished by time from cancer diagnosis (less than 3 and 3 years or more from diagnosis). Reliability, based on McDonald’s omega and Cronbach’s alpha coefficients, ranged from 0.83 to 0.89 for factor scores, and reached 0.95 for total scores. Validity evidence was also provided by correlations with depression, anxiety, perceived stress, and perceived health and quality of life. Discussion: The results support the use of the SBCS for measuring stress as a stimulus in the breast cancer context.     The Role of Nrf2 Signaling in PPARβ/δ-Mediated Vascular Protection against Hyperglycemia-Induced Oxidative Stress

    Hyperglycemia induces oxidative stress and plays a substantial role in the progression of vascular diseases. Here, we demonstrated the potentiality of peroxisome proliferator-activated receptor (PPAR)β/δ activation in attenuating high glucose-induced oxidative stress in endothelial cells and diabetic rats, pointing to the involvement of nuclear factor erythroid 2-related factor 2 (Nrf2). HUVECs exposed to high glucose showed increased levels of reactive oxygen species (ROS) and upregulated NOX-2, NOX-4, Nrf2, and NQO-1 effects that were significantly reversed by the PPARβ/δ agonists GW0742 and L165041. Both PPARβ/δ agonists, in a concentration-dependent manner, induced transcriptional and protein upregulation of heme oxygenase-1 (HO-1) under low- and high-glucose conditions. All effects of PPARβ/δ agonists were reversed by either pharmacological inhibition or siRNA-based downregulation of PPARβ/δ. These in vitro findings were confirmed in diabetic rats treated with GW0742. In conclusion, PPARβ/δ activation confers vascular protection against hyperglycemia-induced oxidative stress by suppressing NOX-2 and NOX-4 expression plus a direct induction of HO-1; with the subsequent downregulation of the Nrf2 pathway.     Esophageal cancer (EC) is an aggressive tumor that represents the 6th most common cause of cancer death worldwide. The estimated incidence in Spain is 2090 cases/year. Two main pathological subtypes exist, squamous cell carcinoma and adenocarcinoma. The main differences between them are localization and underlying factors which are the principal cause of the recent incidence changes observed in west countries. Staging techniques and treatment options which combine surgery, chemotherapy and radiotherapy, reflected the high complexity of the EC management. An undeniably multidisciplinary approach is, therefore, required. In this guide, we review the status of current diagnosis and treatment, define evidence and propose recommendations

    LRP10, PGK1 and RPLP0: best reference genes in periprostatic adipose tissue under obesity and prostate cancer conditions

    Obesity (OB) is a metabolic disorder characterized by adipose tissue dysfunction that has emerged as a health problem of epidemic proportions in recent decades. OB is associated with multiple comorbidities, including some types of cancers. Specifically, prostate cancer (PCa) has been postulated as one of the tumors that could have a causal relationship with OB. Particularly, a specialized adipose tissue (AT) depot known as periprostatic adipose tissue (PPAT) has gained increasing attention over the last few years as it could be a key player in the pathophysiological interaction between PCa and OB. However, to date, no studies have defined the most appropriate internal reference genes (IRGs) to be used in gene expression studies in this AT depot. In this work, two independent cohorts of PPAT samples (n = 20/n = 48) were used to assess the validity of a battery of 15 literature-selected IRGs using two widely used techniques (reverse transcription quantitative PCR [RT-qPCR] and microfluidic-based qPCR array). For this purpose, ΔCt method, GeNorm (v3.5), BestKeeper (v1.0), NormFinder (v.20.0), and RefFinder software were employed to assess the overall trends of our analyses. LRP10, PGK1, and RPLP0 were identified as the best IRGs to be used for gene expression studies in human PPATs, specifically when considering PCa and OB conditions

    Diverging Effects of Landscape Factors and Inter-Row Management on the Abundance of Beneficial and Herbivorous Arthropods in Andalusian Vineyards (Spain)

    Land use at landscape and field scales can increase the diversity and abundance of natural enemies for pest control. In this study, we investigated interactions between landscape elements (semi-natural vegetation, olive orchards, vineyards, other agricultural areas) and inter-row management (vegetation cover vs. bare soil) in relation to arthropod populations in Andalusian vineyards. Arthropods were collected from grapevine foliage in 15 vineyards using suction sampling. Landscape structure was analyzed within a 750 m radius surrounding the studied vineyards. Arthropods were categorized into functional groups (predators, parasitoids, herbivores), and their responses to the most influencing factors were analyzed by likelihood methods and model selection. Of the total of 650 arthropods collected, 48% were predators, 33% herbivores and 19% parasitoids. Numbers of predatory aeolothrips, parasitoids and herbivorous cicadas in the study vineyards decreased with an increased proportion of vineyards in the surroundings. Spider populations in vineyards increased with increasing proportions of other agricultural fields (non-flowering crops) in the surroundings. Semi-natural elements and olive orchards had no influence on the abundance of collected arthropods. We observed synergistic effects between landscape elements and inter-row management. The total numbers of arthropods, herbivores and parasitoids in vineyards benefitted from inter-row vegetation, while spiders benefitted from bare soil. Our findings underline the importance of both surrounding landscape elements and vineyard ground cover management to promote beneficial arthropods for potential natural pest control

    Effective interactions of colloids on nematic films

    The elastic and capillary interactions between a pair of colloidal particles trapped on top of a nematic film are studied theoretically for large separations dd. The elastic interaction is repulsive and of quadrupolar type, varying as d5d^{-5}. For macroscopically thick films, the capillary interaction is likewise repulsive and proportional to d5d^{-5} as a consequence of mechanical isolation of the system comprised of the colloids and the interface. A finite film thickness introduces a nonvanishing force on the system (exerted by the substrate supporting the film) leading to logarithmically varying capillary attractions. However, their strength turns out to be too small to be of importance for the recently observed pattern formation of colloidal droplets on nematic films.Comment: 13 pages, accepted by EPJ
