Tumour suppressor PTEN enhanced enzyme activity of GPx, SOD and catalase by suppression of PI3K/AKT pathway in non-small cell lung cancer cell lines
Phosphates and tensin homologue deleted on chromosome 10 (PTEN) is a tumour suppressor gene which dephosphorilates phosphoinositol 3,4,5 triphosphates. Therefore PTEN can regulate PI3K/AKT pathway in cells. Because of promoter methylation or gene deletion, PTEN expression is commonly decreased or lost in non-small cell lung cancer (NSCLC) cell lines. Therefore, we hypothesized that PTEN could regulate the activity of superoxide dismutase (CuZnSOD), glutathione peroxidase (GPx) and catalase. We first recreated PTENwt, G129R and G129E expressions in lung cell lines, in which endogenous PTEN expression was not detected. Then, we showed that PTEN could suppress AKT activity by its lipid phosphatase domain. We then examined the effect of recreated PTEN expressions in NSCLC cells. While PTENwt expression caused enhanced activity of SOD, GPx and catalase in transfected cells lines, neither G129R nor G129E expression effected enzyme activities. These results suggest that PTEN can up-regulate SOD, GPx and catalase activity by inhibition of PI3K/AKT pathway in NSCLC cell lines. © 2013 Informa UK, Ltd
Extraction of valuable compounds from ginja cherry by-products: effect of the solvent and antioxidant properties
Purpose To study the composition of extracts of Ginja cherries stems and leaves obtained after extraction using different solvents and to evaluate their antioxidant activities. Stems and leaves are by-products of the Ginjinha cherry liquor production; extracting valuable compounds with the most appropriate solvent would valorise these wastes. Method The extraction was performed using different solvents (acetone, ethanol, methanol, ethyl acetate, water, 2-propanol); liquid chromatography-electro spray ionization-mass spectrometry (LC-ESI/MS) was utilized to identify the predominant phenolic acids and flavonoids present in the resulting extracts. The Total Phenolic Content was determined with the Folin-Ciocolteau method. The antioxidant activity was also tested using the ABTS? essay. Results Stems extracts showed a higher concentration in polyphenols than those from leaves. The solvent affected remarkably the extracts compositions: considering the polyphenols content ethanol and water gave the best results for stems and leaves, respectively. Active front-end rectifier modelling using dynamic phasors for more-electric aircraft applications
The More-Electric Aircraft (MEA) has become a dominant trend for next-generation aircraft. The Electrical Power System (EPS) on-board may take many forms: AC, DC, hybrid, frequency-wild, variable voltage, together with the possibility of novel connectivity topologies. To address the stability, availability and capability issues as well as to assess the performance of the power quality and transient behaviour, extensive simulation work is required to develop the EPS architectures. The paper develops a fast-simulation model of active front-end rectifiers based on the dynamic phasor concept. The model is suitable for accelerated simulation studies of EPS under normal, unbalanced and line fault conditions. The performance and effectiveness of the developed model have been demonstrated by comparison against time-domain models in three-phase and synchronous space-vector representations. The experimental verification of the dynamic phasor model is also reported. The prime purpose of the model is for the simulation studies of MEA power architectures at system level; however it can be directly applied for simulation study of any other EPS interfacing with active front-end rectifiers
A multi-site study on walkability, data sharing and privacy perception using mobile sensing data gathered from the mk-sense platform
Walking is a fundamental part of a physically active lifestyle, it is one of everyday activities that positively impacts health and wellbeing. In this paper we describe the challenges and experiences of conducting a sensing campaign in the wild. We make use of mk-sense; a software platform to facilitate the deployment of collaborative sensing campaigns. We elaborate on two cross-cultural studies conducted in four different countries (Mexico, Turkey, Spain, and Switzerland) with a total of 77 participants. We present a detailed description of the data collected from one of the studies aimed at measuring walkability around three different university campuses. The analysis of the data shows that walkability can be assessed using information from the sensors in the smartphones and results from surveys answered by participants. In addition, we analyze issues about data sharing and privacy awareness
Antioxidant, antimicrobial properties and phenolics of different solvent extracts from bark, leaves and seeds of Pongamia pinnata (L.) Pierre
This study appraises the antioxidant and antimicrobial attributes of various solvent extracts (absolute methanol, aqueous methanol, absolute ethanol, aqueous ethanol, absolute acetone, aqueous acetone, and deionized water) from bark, leaves and seeds of Pongamia pinnata (L.) Pierre. Maximum extraction yield of antioxidant components from bark (16.31%), leaves (11.42%) and seeds (21.51%) of P. pinnata was obtained using aqueous methanol (20:80). Of the extracts tested, the bark extract, obtained with aqueous methanol, exhibited greater levels of total phenolics [6.94 g GAE/100 g dry weight (DW)], total flavonoids (3.44 g CE/100 g DW), inhibition of linoleic acid peroxidation (69.23%) and DPPH radical scavenging activity (IC(50) value, 3.21 μg/mL), followed by leaves and seeds extracts. Bark extract tested against a set of bacterial and fungal strains also revealed the strongest antimicrobial activity with the largest inhibition zone and lowest minimum inhibitory concentration (MIC). HPLC analysis of aqueous methanol extracts from bark, leaves and seeds indicated the presence of protocatechuic, ellagic, ferulic, gallic, gentisic, 4-hydroxybenzoic and 4-hydroxycinnamic acids in bark (1.50-6.70 mg/100 g DW); sorbic, ferulic, gallic, salicylic and p-coumaric acids in leaves (1.18-4.71 mg/100 g DW); vanillic, gallic and tannic acids in seeds (0.52-0.65 mg/100 g DW) as the main phenolic acids. The present investigation concludes that the tested parts of P. pinnata, in particular the bark, have strong potential for the isolation of antioxidant and antimicrobial agents for functional food and pharmaceutical uses
A two-layered mechanical model of the rat esophagus. Experiment and theory
BACKGROUND: The function of esophagus is to move food by peristaltic motion which is the result of the interaction of the tissue forces in the esophageal wall and the hydrodynamic forces in the food bolus. The structure of the esophagus is layered. In this paper, the esophagus is treated as a two-layered structure consisting of an inner collagen-rich submucosa layer and an outer muscle layer. We developed a model and experimental setup for determination of elastic moduli in the two layers in circumferential direction and related the measured elastic modulus of the intact esophagus to the elastic modulus computed from the elastic moduli of the two layers. METHODS: Inflation experiments were done at in vivo length and pressure-diameters relations were recorded for the rat esophagus. Furthermore, the zero-stress state was taken into consideration. RESULTS: The radius and the strain increased as function of pressure in the intact as well as in the individual layers of the esophagus. At pressures higher than 1.5 cmH(2)O the muscle layer had a larger radius and strain than the mucosa-submucosa layer. The strain for the intact esophagus and for the muscle layer was negative at low pressures indicating the presence of residual strains in the tissue. The stress-strain curve for the submucosa-mucosa layer was shifted to the left of the curves for the muscle layer and for the intact esophagus at strains higher than 0.3. The tangent modulus was highest in the submucosa-mucosa layer, indicating that the submucosa-mucosa has the highest stiffness. A good agreement was found between the measured elastic modulus of the intact esophagus and the elastic modulus computed from the elastic moduli of the two separated layers
