    The individual fibers of any individual muscle vary greatly in enzyme composition, a fact which is obscured when enzyme levels of a whole muscle are measured. The purpose of this study was therefore to assess the changes due to weightless on the enzyme patterns composed by the individual fibers within the flight muscles. In spite of the limitation in numbers of muscles examined, it is apparent that: (1) that the size of individual fibers (i.e., their dry weight) was reduced about a third, (2) that this loss in dry mass was accompanied by changes in the eight enzymes studied, and (3) that these changes were different for the two muscles, and different for the two enzyme groups. In the soleus muscle the absolute amounts of the three enzymes of oxidative metabolism decreased about in proportion to the dry weight loss, so that their concentration in the atrophic fibers was almost unchanged. In contrast, there was little loss among the four enzymes of glycogenolysis - glycolysis so that their concentrations were substantially increased in the atrophic fibers. In the TA muscle, these seven enzymes were affected in just the opposite direction. There appeared to be no absolute loss among the oxidative enzymes, whereas the glycogenolytic enzymes were reduced by nearly half, so that the concentrations of the first metabolic group were increased within the atrophic fibers and the concentrations of the second group were only marginally decreased. The behavior of hexokinase was exceptional in that it did not decrease in absolute terms in either type of muscle and probably increased as much as 50 percent in soleus. Thus, their was a large increase in concentration of this enzyme in the atrophied fibers of both muscles. Another clear-cut finding was the large increase in the range of activities of the glycolytic enzymes among individual fibers of TA muscles. This was due to the emergence of TA fibers with activities for enzymes of this group extending down to levels as low as those found in control soleus muscles. It would be interesting to know if this represents a transition stage, and whether with prolonged weightlessness most of the fibers would be transformed into a low glycogenolytic type

    Results of subject tests indicate the difficulty of obtaining closely balanced rudder surfaces for most tail assemblies with shielded horns and maintaining a near zero rate-of-change of hinge-moment coefficient without an additional balancing device. A comparison is made between shielded and unshielded horn test results. Pressure distribution and tuft tests of flow over different shaped horns showed higher critical speed for medium-taper nosed horn. The trim tab nose shape had little effect on tab test results

    Effects of Monovalent and Divalent Cations on Ca2+ Fluxes Across Chromaffin Secretory Membrane Vesicles

    Abstract: Bovine chromaffin secretory vesicle ghosts loaded with Na+ were found to take up Ca2+ when incubated in K+ media or in sucrose media containing micromolar concentrations of free Ca2+. Li+- or choline+loaded ghosts did not take up Ca2+. The Ca2+ accumulated by Na+-loaded ghosts could be released by the Ca2+ ionophore A23187, but not by EGTA. Ca2+ uptake was inhibited by external Sr2+, Na +, Li +, or choline +. All the 45Ca2+ accumulated by Na+-dependent Ca2+ uptake could be released by external Na +, indicating that both Ca2+ influx and efflux occur in a Na+-dependent manner. Na + -dependent Ca2+ uptake and release were only slightly inhibited by Mg2+. In the presence of the Na+ ionophore Monensin the Ca2+ uptake by Na +-loaded ghosts was reduced. Ca2+ sequestered by the Na+-dependent mechanism could also be released by external Ca2+ or Sr2+ but not by Mg2+, indicating the presence of a Ca2+/Ca2+ exchange activity in secretory membrane vesicles. This Ca2+/Ca2+ exchange system is inhibited by Mg2+, but not by Sr2+. The Na + -dependent Ca2+ uptake system in the presence of Mg2+ is a saturable process with an apparent Km of 0.28 μM and a Vmax= 14.5 nmol min−1 mg protein−1. Ruthenium red inhibited neither the Na+/Ca2+ nor the Ca2+/Ca2+ exchange, even at high concentrations

    Geochemistry and deposition of Be-7 in river‐estuarine and coastal waters

    The atmospheric flux of cosmogenic Be-7 (53.3-day half-life) and the mode of ?Be deposition in river- estuarine and coastal environments have been examined. The atmospheric flux of ?Be commonly sup- ports inventories ranging from 1.0 to 2.0 pCi/cm 2 (1 pCi = 0.037 Bq). Beryllium 7 concentrations in water phase samples, collected across salinity gradients in several estuaries along the eastern coastline of the United States, range from 0.03 to 0.53 pCi/L and primarily reflect variations in Be-7supply and sorption kinetics. The major process controlling the concentration of Be-7 on estuarine suspended particles appears to be the length of time that these particles remain in the water column. Field particle-to- water distribution coefficients for Be-7have a median value of about 4 x 10 \u27• but range over an order of magnitude reflecting short-term variations in 7Be input, particle dynamics, and particulate iron content rather than equilibrium sorption-desorption responses to changes in water salinity or particle type. Residence times of 7Be in the water column range from a few days in estuarine areas of rapid fine-particle deposition, to several weeks in high-energy environments where pronounced sediment resuspension reintroduces deposited 7Be back into the water column. Inventories of ?Be in sediments range-from nondetectable to 3.3 pCi/cm 2, with the highest inventories in areas where fine particles are accumulating rapidly. Such sites are also major repositories for other particle-reactive substances. A ?Be budget for the James estuary indicates that less than 5% of the expected ?Be input is in the water column and that the short-term estuarine trapping efficiency for atmospherically derived ?Be is somewhere between 50 and 100%

    Spitzer observations of the asteroid-comet transition object and potential spacecraft target 107P (4015) Wilson-Harrington

    Context. Near-Earth asteroid-comet transition object 107P/ (4015) Wilson-Harrington is a possible target of the joint European Space Agency (ESA) and Japanese Aerospace Exploration Agency (JAXA) Marco Polo sample return mission. Physical studies of this object are relevant to this mission, and also to understanding its asteroidal or cometary nature. Aims. Our aim is to obtain significant new constraints on the surface thermal properties of this object. Methods. We present mid-infrared photometry in two filters (16 and 22 microns) obtained with NASA's Spitzer Space Telescope on February 12, 2007, and results from the application of the Near Earth Asteroid Thermal Model (NEATM).We obtained high S/N in two mid-IR bands allowing accurate measurements of its thermal emission. Results. We obtain a well constrained beaming parameter (eta = 1.39 +/- 0.26) and obtain a diameter and geometric albedo of D = 3.46 +/- 0.32 km, and pV = 0.059 +/- 0.011. We also obtain similar results when we apply this best-fitting thermal model to single-band mid-IR photometry reported by Campins et al. (1995), Kraemer et al. (2005) and Reach et al. (2007). Conclusions. The albedo of 4015 Wilson-Harrington is low, consistent with those of comet nuclei and primitive C-, P-, D-type asteorids. We establish a rough lower limit for the thermal inertia of W-H of 60 Jm^-2s^(-0.5)K^-1 when it is at r=1AU, which is slightly over the limit of 30 Jm^-2s^(-0.5)K-1 derived by Groussin et al. (2009) for the thermal inertia of the nucleus of comet 22P/Kopff.Comment: 4 pages, 1 figure and 3 tables. Paper accepted for publicatio

    Anthropogenic methane plume detection from point sources in the Paris megacity area and characterization of their δ<sup>13</sup>C signature

    International audienceMitigating anthropogenic methane emissions is one of the available tools for reaching the near term objectives of the Paris Agreement. Characterizing the isotopic signature of the methane plumes emitted by these sources is needed to improve the quantification of methane sources at the regional scale. Urbanized and industrialized regions such as the Paris megacity are key places to better characterize anthropogenic methane sources. In this study, we present the results of the first mobile surveys in the Paris region, assessing methane point sources from 10 landfills (which in the regional inventory are the main emission sector of methane in the region), 5 gas storage sites (supplying Paris) and 1 waste water treatment (WWT) facility (Europe’s largest, second worldwide). Local atmospheric methane concentration (or mixing ratio) enhancements in the source plumes were quantified and their d13C in CH4 (further noted d13CH4) signature characterized. Among the 10 landfills sampled, at 6 of them we detected atmospheric methane local enhancements ranging from 0.8 to 8.5 parts per million (ppm) with d13CH4 signatures between -63.7 ± 0.3 permils (‰) to - 58.2 ± 0.3 ‰. Among the 5 gas storage sites surveyed, we could observe that 3 of them were leaking methane with local methane concentration enhancements ranging from 0.8 to 8.1 ppm and d13CH4 signatures spanning from -43.4 ± 0.5 ‰ to -33.8 ± 0.4 ‰. Dutch gas with a d13CH4 signature of -33.8 ± 0.4 ‰ (typical of thermogenic gas) was also likely identified. The WWT site emitted local methane enhancements up to 4.0 ppm. For this site, two d13CH4 signatures were determined as -51.9 ± 0.2 ‰ and -55.3 ± 0.1 ‰, typical of a biogenic origin. About forty methane plumes were also detected in the Paris city, leading to local concentration enhancements whose origin was in two cases interpreted as natural gas leaks thanks to their isotopic composition. However, such enhancements were much less common than in cities of North America. More isotopic surveys are needed to discriminate whether such urban methane enhancements are outcoming from gas line leaks and sewer network emanations. Furthermore, our results lead us to the conclusion that the regional emissions inventory could underestimate methane emissions from the WWT sector. Further campaigns are needed to assess the variability and seasonality of the sources and of their isotopic signature, and to estimate their emissions using methods independent of the inventory

    The price of rapid exit in venture capital-backed IPOs

    This paper proposes an explanation for two empirical puzzles surrounding initial public offerings (IPOs). Firstly, it is well documented that IPO underpricing increases during “hot issue” periods. Secondly, venture capital (VC) backed IPOs are less underpriced than non-venture capital backed IPOs during normal periods of activity, but the reverse is true during hot issue periods: VC backed IPOs are more underpriced than non-VC backed ones. This paper shows that when IPOs are driven by the initial investor’s desire to exit from an existing investment in order to finance a new venture, both the value of the new venture and the value of the existing firm to be sold in the IPO drive the investor’s choice of price and fraction of shares sold in the IPO. When this is the case, the availability of attractive new ventures increases equilibrium underpricing, which is what we observe during hot issue periods. Moreover, I show that underpricing is affected by the severity of the moral hazard problem between an investor and the firm’s manager. In the presence of a moral hazard problem the degree of equilibrium underpricing is more sensitive to changes in the value of the new venture. This can explain why venture capitalists, who often finance firms with more severe moral hazard problems, underprice IPOs less in normal periods, but underprice more strongly during hot issue periods. Further empirical implications relating the fraction of shares sold and the degree of underpricing are presented

    Mechanism of age-dependent susceptibility and novel treatment strategy in glutaric acidemia type I

    Glutaric acidemia type I (GA-I) is an inherited disorder of lysine and tryptophan metabolism presenting with striatal lesions anatomically and symptomatically similar to Huntington disease. Affected children commonly suffer acute brain injury in the context of a catabolic state associated with nonspecific illness. The mechanisms underlying injury and age-dependent susceptibility have been unknown, and lack of a diagnostic marker heralding brain injury has impeded intervention efforts. Using a mouse model of GA-I, we show that pathologic events began in the neuronal compartment while enhanced lysine accumulation in the immature brain allowed increased glutaric acid production resulting in age-dependent injury. Glutamate and GABA depletion correlated with brain glutaric acid accumulation and could be monitored in vivo by proton nuclear magnetic resonance (1H NMR) spectroscopy as a diagnostic marker. Blocking brain lysine uptake reduced glutaric acid levels and brain injury. These findings provide what we believe are new monitoring and treatment strategies that may translate for use in human GA-I
